In this article, a cross was made between CN19-1 and Thai4-6, and the F2segregation population was also constructed through F1selfing. The genetic characteristics of the first female flower node and the first male flower node were analyzed by adopting the major gene plus polygene mixed genetic model. The data analysis results showed that the first female flower node and the first male flower node were continuous distribution in the F2segregation population. E-2 model was the most suitable model for the genetic analysis of the first female flower node and the first male flower node. The additive effect values of the 2 pairs of major genes controlling the first female flower node were 2.722 and 1.862 8 respectively, the dominant effect values were-2.721 6 and-0.171 8, respectively. The additive effect value of polygene was-0.839 2, and the dominant effect value of polygene was 2.225 4. The heritability of major genes and polygene were 83.73% and 1.54%, respectively. The additive effect values of the 2 pairs of major genes controlling the first male flower node were 17.746 9 and 3.972, respectively, the dominant effect values were 5.191 6 and-3.972, respectively. The additive effect value of polygene was-20.530 5, and the dominant effect value was-4.141 4. The heritability of major genes and polygene was 92.34% and 4.7%, respectively. This study could provide a theoretical basis for bitter gourd breeding.展开更多
The red swamp crayfish,Procambarus clarkii,is an economically important species especially in China.Their exoskeleton places serious constraints on growth and culture management.Their growth is achieved through interm...The red swamp crayfish,Procambarus clarkii,is an economically important species especially in China.Their exoskeleton places serious constraints on growth and culture management.Their growth is achieved through intermittent molting/ecdysis.The longitudinal genetic dynamics for growth-related traits at different ecdysial points in P.clarkii has been unclear to date.In this study,conditional genetic analysis was carried out for growth-related traits(body weight,body length,chela length,and cephalothorax length)based upon a mixed genetic model with conditional additive,dominance,and genotype by environment effects in P.clarkii.A complete diallel cross was made among three geographic populations of P.clarkii for the genetic mating design.Results of the conditional genetic analysis showed that from 4 th molt to 9 th molt the conditional additive variations were increased significantly whereas the conditional non-additive genetic variations(dominance and genotype by environment interaction)were decreased significantly for these growth-related traits.This indicated that lots of new expression of additive effect genes for body weight,body length,chela length,and cephalothorax length occurred during ontogeny,and environment played a signifi cant role in the expression of genes affecting these growth-related traits.Growth of the four traits was mainly affected by non-additive genetic effects in early developmental stage(prior to 4 th molt).The cumulative conditional additive variation for the growth-related traits from 4 th molt to 9 th molt accounted for a large majority of the total conditional additive variations from 2 nd molt to 9 th molt,indicating that this period was very important for the growth of this species.Using the conditional analysis method,dynamics of growth-related traits during an important ontogenetic phase of red swamp crayfish was uncovered.Our results provide valuable insights into refining production of this species.展开更多
The objective of this study was to estimate genetic parameters of body width(BW)to body length(BL)ratio(BW/BL)and of body weight traits(BWT)in turbot,and to elucidate the genetic mechanism of the two traits during ont...The objective of this study was to estimate genetic parameters of body width(BW)to body length(BL)ratio(BW/BL)and of body weight traits(BWT)in turbot,and to elucidate the genetic mechanism of the two traits during ontogeny by dynamic genetic analysis.From 3 to 27 months,BW,BL and BWT of each communally stocked fish were measured every 3 months.The BW/BL ratio was measured at different sampling ages.A twotrait animal model was used for genetic evaluation of traits.The results showed that the heritability values of BW/BL ratio ranged from 0.2168 to 0.3148,corresponding to moderate heritability.The BWT heritability values ranged from 0.2702 to 0.3479 corresponding to moderate heritability.The heritability of BW/BL ratio was lower than that of BWT,except at 3 months of age.Genetic correlation between BW/BL ratio and BWT decreased throughout the measurement period.Genetic correlations were higher than the phenotypic correlations.The current results for estimating genetic parameters demonstrate that the BW/BL ratio could be used as a phenotypic marker of fast-growing turbot,and the BW/BL ratio and BWT could be improved simultaneously through selective breeding.展开更多
Heading date of 26 native japonica rice cultivars in southwest China was investigated,and their basic vegetative growth(BVG),photoperiod-sensitivity(PS) and temperature-sensitivity(TS) were analyzed under artificial s...Heading date of 26 native japonica rice cultivars in southwest China was investigated,and their basic vegetative growth(BVG),photoperiod-sensitivity(PS) and temperature-sensitivity(TS) were analyzed under artificial short-day and natural long-day conditions in Nanjing,as well as artificial high-temperature and natural low-temperature conditions in winter in Hainan.The results showed that the PS and TS varied among different cultivars.The BVG of all the japonica cultivars was well situated,but differed within cultivars.Regression analysis showed a significant correlation between heading date and PS,indicating that PS was the main factor affecting heading date of japonica cultivars in southwest China.Genetic analysis was conducted on these 26 cultivars using a set of heading date near isogenic lines as test lines.All the japonica cultivars carried the dominant early-heading gene Ef-1 or Ef-1t,and most of these cultivars carried the dominant photoperiod sensitivity allele E1 or E1t,the PS of which was slightly weaker than E1.For the Se-1 locus,these cultivars mainly carried recessive photoperiod insensitivity gene Se-1e.In addition,the PS of 22 japonica cultivars could be repressed or weakened by the recessive allele hd2,inhibiting the expression of E1 and Se-1.These results indicated that the genotypes of heading date determined different PS and well situated BVG in japonica rice cultivars in southwest China.展开更多

The field test of their blast resistance was conducted in 858 Farm, which showed that Aichi Asahi, BL1and Digu were highly resistant to either leaf blast or neck blast and could be used to develop molecular breeding by design, and genetic analysis indicated that the field resistance of Aichi Asahi, BL1and Digu to leaf blast and neck blast was controlled by a single dominant gene, and the leaf blast and neck blast resistance in the donor cv. Pai-kan-tao was inherited as a single recessive gene, the neck blast resistance of the donor cv. Oryzica Llanos 5 was controlled by a single recessive gene while its leaf blast resistance was not controlled by this gene. These results suggested that five parental varieties positively contributed to resistance to either leaf blast or neck blast and could be used to expand the genetic germplasms resistant to blast in cold region using molecular assisted selection.展开更多

The results showed that the internode length was continuous distribution in the F2 population. The optimum model for internode length genetic analysis was B-1. The additive effect values of two pairs of major genes controlling the internode length were 3.206 3 and 2.638 3, respectively, which showed a positive effect. The dominant effect values were-3.434 6 and-1.459 6, respectively, which showed a negative effect. The dominant degree of major genes is-1.07 and-0.55, respectively, indicating that the first pair of major genes was over dominant and the second pair of major genes was incompletely dominant. The heritability of the major gene was 82.25%, indicating the genetic stability was high. This study could provide a theoretical reference for the breeding of bitter gourd.展开更多

A 24-year-old woman, coming from Bolivia two years before, was diagnosed as cystic echinococcosis (CE) by unstained wet mount and cytological findings through aspiration cytology from the liver, suggested as liver abscess by diagnostic images. The patient was classified as CE1, smaller than 5 cm, and was diagnosed as P1N0M0, stage I. The phylogenetic trees of Echinococcus spp., based on cox1 genes showed that the isolates on the patient belonged to E. ortleppi. The patient was conservatively treated with two courses of albendazole therapy. E. ortleppi has been reported to be in regions including Latin America, except with Japan. Although the patient came from Bolivia before two years, this is the first case report of E. ortleppi from liver echinococcosis in Japan.展开更多

Alpha linolenic acid(ALA)is an essential polyunsaturated fatty acids that can improve human health.Rapeseed is the second largest oil crop in the world but the ALA content in its seed fatty acids is only about 10%.Two...Alpha linolenic acid(ALA)is an essential polyunsaturated fatty acids that can improve human health.Rapeseed is the second largest oil crop in the world but the ALA content in its seed fatty acids is only about 10%.Two rapeseed germplasms YH25005 and R8Q10 with high content(up to 21%)ALA were developed by intervarietal crossing.They were used as the maternal parent(P_(1))when crossed with a low ALA parent SW(P2)to produce the seeds of the F1 hybrid,F2,and backcrosses to P1(BCP1)and P2(BCP2).A multigeneration joint segregation analysis was conducted to determine major gene t polygene effects of the content of three major unsaturated fatty acids including oleic(OA),linoleic(LA)and ALA.The results showed that,although some genes favorable to ALA accumulation were not allelic in R8Q10 and YH25005,all the inheritances of OA,LA and ALA in YH25005×SW and R8Q10×SW followed a genetic model of‘two pairs of additive major genes t additive-dominant polygenes’.It is suggested that the contents of OA,LA and ALA are closely related and the major genes in the parent SW containing loss-of-function mutations in FAD2 and FAD3 loci had strong effect to reduce ALA and elevate OA.However,total genetic effect of the polygenes was greater than that of the two major genes,especial on ALA content.It indicated that it is necessary to employ a larger F2 population to find the plant that accumulate enough minor-effect polygenes for high ALA content.The results are useful for high ALA rapeseed breeding and future work of gene mapping.展开更多
A biparental soybean population of 364 recombinant inbred lines(RILs)derived from Zhongdou 41×ZYD02.878 was used to identify quantitative trait loci(QTL)associated with hundred-seed weight(100-SW),pod length(PL),...A biparental soybean population of 364 recombinant inbred lines(RILs)derived from Zhongdou 41×ZYD02.878 was used to identify quantitative trait loci(QTL)associated with hundred-seed weight(100-SW),pod length(PL),and pod width(PW).100-SW,PL,and PW showed moderate correlations among one another,and 100-SW was correlated most strongly with PW(0.64–0.74).Respectively 74,70,75 and19 QTL accounting for 38.7%–78.8%of total phenotypic variance were identified by inclusive composite interval mapping,restricted two-stage multi-locus genome-wide association analysis,3 variancecomponent multi-locus random-SNP-effect mixed linear model analysis,and conditional genome-wide association analysis.Of these QTL,189 were novel,and 24 were detected by multiple methods.Six loci were associated with 100-SW,PL,and PW and may be pleiotropic loci.A total of 284 candidate genes were identified in colocalizing QTL regions,including the verified gene Seed thickness 1(ST1).Eleven genes with functions involved in pectin biosynthesis,phytohormone,ubiquitin-protein,and photosynthesis pathways were prioritized by examining single nucleotide polymorphism(SNP)variation,calculating genetic differentiation index,and inquiring gene expression.The prediction accuracies of genomic selection(GS)for 100-SW,PL,and PW based on single trait-associated markers reached 0.82,0.76,and 0.86 respectively,but selection index(SI)-assisted GS strategy did not increase GS efficiency and inclusion of trait-associated markers as fixed effects reduced prediction accuracy.These results shed light on the genetic basis of 100-SW,PL,and PW and provide GS models for these traits with potential application in breeding programs.展开更多
Single nucleotide polymorphism(SNP)genotyping arrays provide an optimal high-throughput platform for genetic research and molecular breeding programs in both animals and plants.In this study,a highquality and custom-d...Single nucleotide polymorphism(SNP)genotyping arrays provide an optimal high-throughput platform for genetic research and molecular breeding programs in both animals and plants.In this study,a highquality and custom-designed Rice3K56 SNP array was developed with the resequencing data of 3024 rice accessions worldwide,which was then tested extensively in 192 representative rice samples.Printed on the Gene Titan chips of Affymetrix Axiom each containing 56,606 SNP markers,the Rice3K56 array has a high genotyping reliability(99.6%),high and uniform genome coverage(an average of 6.7-kb between adjacent SNPs),abundant polymorphic information and easy automation,compared with previously developed rice SNP arrays.When applied in rice varietal differentiation,population diversity analysis,gene mapping of 13 complex traits by a genome-wide association study analysis(GWAS),and genome selection experiments in a recombinant inbred line and a multi-parent advanced generation inter-cross populations,these properties of the Rice3K56 array were well demonstrated for its power and great potential to be a highly efficient tool for rice genetic research and genomic breeding.展开更多
BACKGROUND Osteopetrosis is a rare genetic disorder characterized by increased bone density due to defective bone resorption of osteoclasts.Approximately,80%of autosomal dominant osteopetrosis type II(ADO-II)patients ...BACKGROUND Osteopetrosis is a rare genetic disorder characterized by increased bone density due to defective bone resorption of osteoclasts.Approximately,80%of autosomal dominant osteopetrosis type II(ADO-II)patients were usually affected by heterozygous dominant mutations in the chloride voltage-gated channel 7(ClCN7)gene and present early-onset osteoarthritis or recurrent fractures.In this study,we report a case of persistent joint pain without bone injury or underlying history.CASE SUMMARY We report a 53-year-old female with joint pain who was accidentally diagnosed with ADO-II.The clinical diagnosis was based on increased bone density and typical radiographic features.Two heterozygous mutations in the ClCN7 and Tcell immune regulator 1(TCIRG1)genes by whole exome sequencing were identified in the patient and her daughter.The missense mutation(c.857G>A)occurred in the CLCN7 gene p.R286Q,which is highly conserved across species.The TCIRG1 gene point mutation(c.714-20G>A)in intron 7(near the splicing site of exon 7)had no effect on subsequent transcription.CONCLUSION This ADO-II case had a pathogenic CLCN7 mutation and late onset without the usual clinical symptoms.For the diagnosis and assessment of the prognosis for osteopetrosis,genetic analysis is advised.展开更多

文摘The objective of this study was to estimate genetic parameters of body width(BW)to body length(BL)ratio(BW/BL)and of body weight traits(BWT)in turbot,and to elucidate the genetic mechanism of the two traits during ontogeny by dynamic genetic analysis.From 3 to 27 months,BW,BL and BWT of each communally stocked fish were measured every 3 months.The BW/BL ratio was measured at different sampling ages.A twotrait animal model was used for genetic evaluation of traits.The results showed that the heritability values of BW/BL ratio ranged from 0.2168 to 0.3148,corresponding to moderate heritability.The BWT heritability values ranged from 0.2702 to 0.3479 corresponding to moderate heritability.The heritability of BW/BL ratio was lower than that of BWT,except at 3 months of age.Genetic correlation between BW/BL ratio and BWT decreased throughout the measurement period.Genetic correlations were higher than the phenotypic correlations.The current results for estimating genetic parameters demonstrate that the BW/BL ratio could be used as a phenotypic marker of fast-growing turbot,and the BW/BL ratio and BWT could be improved simultaneously through selective breeding.

文摘Heading date of 26 native japonica rice cultivars in southwest China was investigated,and their basic vegetative growth(BVG),photoperiod-sensitivity(PS) and temperature-sensitivity(TS) were analyzed under artificial short-day and natural long-day conditions in Nanjing,as well as artificial high-temperature and natural low-temperature conditions in winter in Hainan.The results showed that the PS and TS varied among different cultivars.The BVG of all the japonica cultivars was well situated,but differed within cultivars.Regression analysis showed a significant correlation between heading date and PS,indicating that PS was the main factor affecting heading date of japonica cultivars in southwest China.Genetic analysis was conducted on these 26 cultivars using a set of heading date near isogenic lines as test lines.All the japonica cultivars carried the dominant early-heading gene Ef-1 or Ef-1t,and most of these cultivars carried the dominant photoperiod sensitivity allele E1 or E1t,the PS of which was slightly weaker than E1.For the Se-1 locus,these cultivars mainly carried recessive photoperiod insensitivity gene Se-1e.In addition,the PS of 22 japonica cultivars could be repressed or weakened by the recessive allele hd2,inhibiting the expression of E1 and Se-1.These results indicated that the genotypes of heading date determined different PS and well situated BVG in japonica rice cultivars in southwest China.

文摘A rumpled and twisted leaf 1(rtl1) mutant was generated from a japonica cultivar Nipponbare by ethyl methanesulfonate treatment,which was characterized as rumpled and twisted leaf at the seedling stage.The F2 populations were constructed by crossing with indica cultivars TN1 and Zhefu 802,respectively.Genetic analysis demonstrated that the phenotype was controlled by a single recessive nuclear gene.The closely linked simple sequence repeat(SSR) marker RM1155 was obtained from bulked segregant analysis.Subsequently,sequence tagged site(STS) markers were developed using the published rice genome sequence.Finally,RTL1 was located between an STS marker T1591 and an SSR marker RM1359,at the distances of 0.48 cM and 0.96 cM,respectively.These results will facilitate the cloning of the target gene in further studies.

文摘Five F2 segregation populations, derived from crosses between the susceptible japonica cultivars (cvs.) Kongyu131 and donor cvs. Aichi Asahi (AA), BL1, Digu, Pai-kan-tao (PKT) and Oryzica Llanos 5 (ORL5), were used to evaluate their natural resistance to blast in cold region. The field test of their blast resistance was conducted in 858 Farm, which showed that Aichi Asahi, BL1and Digu were highly resistant to either leaf blast or neck blast and could be used to develop molecular breeding by design, and genetic analysis indicated that the field resistance of Aichi Asahi, BL1and Digu to leaf blast and neck blast was controlled by a single dominant gene, and the leaf blast and neck blast resistance in the donor cv. Pai-kan-tao was inherited as a single recessive gene, the neck blast resistance of the donor cv. Oryzica Llanos 5 was controlled by a single recessive gene while its leaf blast resistance was not controlled by this gene. These results suggested that five parental varieties positively contributed to resistance to either leaf blast or neck blast and could be used to expand the genetic germplasms resistant to blast in cold region using molecular assisted selection.

文摘[Objectives]To explore the genetic mechanism that leaf length and leaf width in bitter gourd has realistic guiding significance for formulating breeding strategy.[Methods]In this paper,the cross was made between CN19-1 and Thai4-6 and an F2segregation population was constructed.The genetic characteristics of leaf length and leaf width were analyzed by employing the major gene plus polygene mixed genetic model.[Results]The data analysis results demonstrated that the leaf length and leaf width of the F_(2) segregation population showed continuous distribution.The optimum model for leaf length genetic analysis was A-1,the additive effect value of the major gene was 3.1766,the dominant effect value was-3.1766,and the heritability was 74.51%.The optimum model for leaf width genetic analysis was B-1.The additive effect values of 2 pairs of major genes controlling the leaf width were 3.1917 and 0.9623,respectively.The dominant effect values were-1.3135 and-0.3463,respectively.The additive×additive interaction effect was-0.3591,the additive×dominant interaction effect was-0.2570,the dominant×additive interaction effect was 1.9916,the dominant×dominant interaction effect was 1.9686.The heritability of major genes was 96.67%.[Conclusions]This study is expected to provide a theoretical basis for the breeding of bitter gourd.

文摘Performing the genetic mechanism research of internode length in bitter gourd has important guiding significance for making breeding strategy. A cross was made between CN19-1 and Thai4-6, and an F2 segregation population was constructed. The genetic characteristic of the internode length was analyzed through employing the major gene plus polygene mixed genetic model. The results showed that the internode length was continuous distribution in the F2 population. The optimum model for internode length genetic analysis was B-1. The additive effect values of two pairs of major genes controlling the internode length were 3.206 3 and 2.638 3, respectively, which showed a positive effect. The dominant effect values were-3.434 6 and-1.459 6, respectively, which showed a negative effect. The dominant degree of major genes is-1.07 and-0.55, respectively, indicating that the first pair of major genes was over dominant and the second pair of major genes was incompletely dominant. The heritability of the major gene was 82.25%, indicating the genetic stability was high. This study could provide a theoretical reference for the breeding of bitter gourd.

文摘Echinococcosis is a global and zoonotic helminthic disease caused by Echinococcus sp. A 24-year-old woman, coming from Bolivia two years before, was diagnosed as cystic echinococcosis (CE) by unstained wet mount and cytological findings through aspiration cytology from the liver, suggested as liver abscess by diagnostic images. The patient was classified as CE1, smaller than 5 cm, and was diagnosed as P1N0M0, stage I. The phylogenetic trees of Echinococcus spp., based on cox1 genes showed that the isolates on the patient belonged to E. ortleppi. The patient was conservatively treated with two courses of albendazole therapy. E. ortleppi has been reported to be in regions including Latin America, except with Japan. Although the patient came from Bolivia before two years, this is the first case report of E. ortleppi from liver echinococcosis in Japan.

文摘Thirty-one polymorphic decamer primers were selected to genotype 92 progenies from the cross be- tween Yiben No.4, a monoembryonic diploid F1 hybrid of Citrus reticulata Blanco cv Huanongbendizao tangerine and C. ichangensis Swingle, and [Hamlin sweet orange + Rough lemon], an allotetraploid somatic hybrid of Citrus sinensis Osbeck cv. Hamlin and C. jambhiri Lush cv. Rough Lemon. χ2 (Chi-square) analysis of RAPD markers in the progenies indicated they were randomly transmitted from the four donor parents, without significant difference between the diploids and triploids. However, these progenies were clustered into three major groups using dendrogram constructed by UPGMA, skewed to three parents in certain degrees, 15 (13 triploids and 2 diploids) to Hamlin, 16 (9 and 7) to Yiben No. 4, and 61 (57 and 4) to [Hamlin sweet orange + Rough Lemon] from which genomic contribu- tion was predominant in progenies, respectively.

文摘Some novel members of extremely halophilic archaea,strains AJ11,AJ12 and AJ13,were isolated from the Aularz Lake located in the Altun Mountain National Nature Reserve of Xinjiang,Uygur Autonomous Region in China.Partial DNA fragments encoding a bacteriorhodopsin(BR),as well as for 16S rRNA of isolated strains,were amplified by PCR and their DNA sequences were determined subsequently.On the basis of homology and phylogenetic analysis of the 16S rDNA,we thought that the isolated strains forming a microbiological population are the members of the genus Natrinema.The results of genetic analysis,such as GC content,transition/transversion(Ti/Tv)rate ratios and synonymous substitution rates(Ks)indicate that the br fragments,with a high level of genetic divergence,are faced with both purifying selection and bias mutation pressure.The study provides the basis for use of species and BR proteins resources.

文摘Alpha linolenic acid(ALA)is an essential polyunsaturated fatty acids that can improve human health.Rapeseed is the second largest oil crop in the world but the ALA content in its seed fatty acids is only about 10%.Two rapeseed germplasms YH25005 and R8Q10 with high content(up to 21%)ALA were developed by intervarietal crossing.They were used as the maternal parent(P_(1))when crossed with a low ALA parent SW(P2)to produce the seeds of the F1 hybrid,F2,and backcrosses to P1(BCP1)and P2(BCP2).A multigeneration joint segregation analysis was conducted to determine major gene t polygene effects of the content of three major unsaturated fatty acids including oleic(OA),linoleic(LA)and ALA.The results showed that,although some genes favorable to ALA accumulation were not allelic in R8Q10 and YH25005,all the inheritances of OA,LA and ALA in YH25005×SW and R8Q10×SW followed a genetic model of‘two pairs of additive major genes t additive-dominant polygenes’.It is suggested that the contents of OA,LA and ALA are closely related and the major genes in the parent SW containing loss-of-function mutations in FAD2 and FAD3 loci had strong effect to reduce ALA and elevate OA.However,total genetic effect of the polygenes was greater than that of the two major genes,especial on ALA content.It indicated that it is necessary to employ a larger F2 population to find the plant that accumulate enough minor-effect polygenes for high ALA content.The results are useful for high ALA rapeseed breeding and future work of gene mapping.

文摘A biparental soybean population of 364 recombinant inbred lines(RILs)derived from Zhongdou 41×ZYD02.878 was used to identify quantitative trait loci(QTL)associated with hundred-seed weight(100-SW),pod length(PL),and pod width(PW).100-SW,PL,and PW showed moderate correlations among one another,and 100-SW was correlated most strongly with PW(0.64–0.74).Respectively 74,70,75 and19 QTL accounting for 38.7%–78.8%of total phenotypic variance were identified by inclusive composite interval mapping,restricted two-stage multi-locus genome-wide association analysis,3 variancecomponent multi-locus random-SNP-effect mixed linear model analysis,and conditional genome-wide association analysis.Of these QTL,189 were novel,and 24 were detected by multiple methods.Six loci were associated with 100-SW,PL,and PW and may be pleiotropic loci.A total of 284 candidate genes were identified in colocalizing QTL regions,including the verified gene Seed thickness 1(ST1).Eleven genes with functions involved in pectin biosynthesis,phytohormone,ubiquitin-protein,and photosynthesis pathways were prioritized by examining single nucleotide polymorphism(SNP)variation,calculating genetic differentiation index,and inquiring gene expression.The prediction accuracies of genomic selection(GS)for 100-SW,PL,and PW based on single trait-associated markers reached 0.82,0.76,and 0.86 respectively,but selection index(SI)-assisted GS strategy did not increase GS efficiency and inclusion of trait-associated markers as fixed effects reduced prediction accuracy.These results shed light on the genetic basis of 100-SW,PL,and PW and provide GS models for these traits with potential application in breeding programs.

文摘Single nucleotide polymorphism(SNP)genotyping arrays provide an optimal high-throughput platform for genetic research and molecular breeding programs in both animals and plants.In this study,a highquality and custom-designed Rice3K56 SNP array was developed with the resequencing data of 3024 rice accessions worldwide,which was then tested extensively in 192 representative rice samples.Printed on the Gene Titan chips of Affymetrix Axiom each containing 56,606 SNP markers,the Rice3K56 array has a high genotyping reliability(99.6%),high and uniform genome coverage(an average of 6.7-kb between adjacent SNPs),abundant polymorphic information and easy automation,compared with previously developed rice SNP arrays.When applied in rice varietal differentiation,population diversity analysis,gene mapping of 13 complex traits by a genome-wide association study analysis(GWAS),and genome selection experiments in a recombinant inbred line and a multi-parent advanced generation inter-cross populations,these properties of the Rice3K56 array were well demonstrated for its power and great potential to be a highly efficient tool for rice genetic research and genomic breeding.

文摘BACKGROUND Osteopetrosis is a rare genetic disorder characterized by increased bone density due to defective bone resorption of osteoclasts.Approximately,80%of autosomal dominant osteopetrosis type II(ADO-II)patients were usually affected by heterozygous dominant mutations in the chloride voltage-gated channel 7(ClCN7)gene and present early-onset osteoarthritis or recurrent fractures.In this study,we report a case of persistent joint pain without bone injury or underlying history.CASE SUMMARY We report a 53-year-old female with joint pain who was accidentally diagnosed with ADO-II.The clinical diagnosis was based on increased bone density and typical radiographic features.Two heterozygous mutations in the ClCN7 and Tcell immune regulator 1(TCIRG1)genes by whole exome sequencing were identified in the patient and her daughter.The missense mutation(c.857G>A)occurred in the CLCN7 gene p.R286Q,which is highly conserved across species.The TCIRG1 gene point mutation(c.714-20G>A)in intron 7(near the splicing site of exon 7)had no effect on subsequent transcription.CONCLUSION This ADO-II case had a pathogenic CLCN7 mutation and late onset without the usual clinical symptoms.For the diagnosis and assessment of the prognosis for osteopetrosis,genetic analysis is advised.
