Human ovarian neoplasm cell CD147 stimulates production and activation of matrix metalloproteinases in co-cultures with mouse fibroblasts

Objective: To investigate the expression of CD147 on human ovarian neoplasm cell lines and its influence on production and activation of matrix metallproteinases(MMPs). Methods: The expression of CD147 on different human ovarian neoplasm cell lines was studied by western blotting. Co-culture was carried out to investigate the stimulative effect of the positive expression CD147 cell HO-8910 on the production of MMPs of fibroblast cell in vitro. Zymography and immune blotting were used to study the production and activity of positive MMPs, at the time, to explore the relation between CD147 and MMPs. Results: CD147 was positively presented in 2 ovarian neoplasm cell lines(HO-8910,3-AO), but in SKOV3, TC-1,NIN3T3 cell was negative. MMP-2 and MMP-9 were detected by HO-8910 cell line, mouse fibroblast cell and co-culture cells; but the expression in co-culture cell is obviously higher than individual cultures of each type alone.CD147 stimulated MMPs in dose-dependent manner. Conclusion: CD147 causes increased production and activation of MMP-2, MMP-9.CD147 is probably a indirect marker of some ovarian cancer cells with invasion and metastasis. 展开更多

目的研究长期应用眼表抗青光眼药物后人的Tenon's囊的成纤维细胞中基质金属蛋白酶(MMP-3)及其抑制剂(TIMP-2)的表达,以了解长期局部应用抗青光眼药物后结膜下组织的病理改变。方法收集长期应用抗青光眼药物及无长期应用抗青光眼药... 目的研究长期应用眼表抗青光眼药物后人的Tenon's囊的成纤维细胞中基质金属蛋白酶(MMP-3)及其抑制剂(TIMP-2)的表达,以了解长期局部应用抗青光眼药物后结膜下组织的病理改变。方法收集长期应用抗青光眼药物及无长期应用抗青光眼药物在小梁切除术中切除的结膜及结膜下组织各20例(20只眼),进行HE染色和马松染色后,在光镜下观察组织学形态,筛选组织标本,进行免疫组织化学检查。结果两组在年龄、性别、诊断之间无差异。用药组A组:用药时间13年;未用药组B组:用药时间790 d。小梁切除术前用两种抗青光眼药的患者为15例,3种以上的患者为5例,未用抗青光眼药的患者20例,用药1年以上的患者20例。在光镜下观察HE和马松染色显示:长期应用抗青光眼药结膜上皮排列不规则,浅基质层为增生的胶原纤维和大量的新生血管伴有炎细胞浸润。在免疫组织化学染色切片,两种抗体染色阳性细胞浆呈程度不同、基本均色棕黄色粗大颗粒反应,反之则为染色阴性。MMP-3和TIMP-2在A、B两组间的表达有差异,P<0.01,有统计学意义。结论长期应用眼表抗青光眼药物后人的Tenon's囊的成纤维细胞中MMP-3及其TIMP-2的表达异常,其临床意义在于长期应用局部抗青光眼药物可以增加组织的改变和增加滤过手术术后的瘢痕形成。 展开更多

MMP-28, the new me... There are striking similiarities present between the behavior of invasive placentaL cells and that of invasive cancer cells. Matrix metalloproteinases (MMPs) are one of the most important mediators. MMP-28, the new member of MMPs, was sequenced and identified recently. Expression of MMP-28 mRNA and protein in the cytotrophoblast cells and a choriocarcinoma cell line, JEG-3 cell, was conducted by zymography, RT-PCR and Northern blot. There is MMP-28 mRNA expression in both the cytotrophoblast cells and JEG-3 cells by RT-PCR. The activity of MMP-28 in cytotrophoblast cells was significantly weaker than that in JEG-3 (P 【 0.01) by zymography. Furthermore, mRNA expression of MMP-28 was significantly stronger (P 【 0.001) in JEG-3 than in human cytotrophoblast cells in a time-dependent way by Northern Blot. Our results suggest that MMP-28 may play a role in some of the tissue-remodeling events associated with normal pregnancy and tumor progression. 展开更多
