The designed hydrogel is based on chitosan/ PEG600/Gurgam with carbon nanofiller along silane crosslinked (TEOS) with pH sensitive response to controlled release of drug in biomedical materials and agriculture industry. The various concentration of carbon nanofiller is used to analyze its effect on the fabricated hydrogel characteristics by using FTIR, SEM, TGA, swelling studies (water, buffer and ionic solution). Spectra of FTIR reflected both established and newly developed groups (like hydrogel). COOH group presence is clearly observed in this range in the carbon filler reinforced hydrogel. The SEM micrographs show that CPG0.003 had a collection of polysaccharide chains as thin helices, which is attributed to the increase in the size of porosity. TGA shows to increase concentration of nanofiller enhanced the thermal stability of the designed hydrogels at temperature 25˚C to 550˚C mass loss percentage decrease upto 20% and increase thermal stability. This pH response made these resultant hydrogels as fruitful competitor against the many reported controlled release application.展开更多

Analytical expres-sions pertaining to concentration of oxygen, glucose, and gluconic acid for all values of parameters are presented. We have employed Homotopy analysis method to evaluate the approximate analytical solutions of the non-linear boundary value problem. A comparison of the analytical approximation and numerical simulation is also presented. A good agreement between theoretical predictions and numerical results is observed.展开更多
In this study, a p H-sensitive micelle self-assembled from poly(L-histidine) based triblock copolymers of poly(ethylene glycol)–poly(D,L-lactide)–poly(L-histidine)(mPEG-PLA-PHis) was prepared and used as the intrace...In this study, a p H-sensitive micelle self-assembled from poly(L-histidine) based triblock copolymers of poly(ethylene glycol)–poly(D,L-lactide)–poly(L-histidine)(mPEG-PLA-PHis) was prepared and used as the intracellular doxorubicin(Dox) delivery for cancer chemotherapy. Dox was loaded into the micelles by thin-film hydration method and a Box–Behnken design for three factors at three levels was used to optimize the preparations. The optimized mPEG-PLA-Phis/Dox micelles exhibited good encapsulation efficiency of 91.12%,a mean diameter of 45 nm and narrow size distribution with polydispersity index of 0.256.In vitro drug release studies demonstrated that Dox was released from the micelles in a p Hdependent manner. Furthermore, the cellular evaluation of Dox loaded micelles displayed that the micelles possessed high antitumor activity in vitro with an IC50 of 35.30 μg/ml against MCF-7/ADR cells. The confocal microscopy and flow cytometry experiments indicated that m PEG-PLA-Phis micelles mediated efficient cytoplasmic delivery of Dox with the aid of poly(Lhistidine) mediated endosomal escape. In addition, blank m PEG-PLA-Phis micelles were shown to be nontoxic to MCF-7/ADR cells even at a high concentration of 200 μg/ml. The pHsensitive mPEG-PLA-PHis micelles have been demonstrated to be a promising nanosystem for the intracellular delivery of Dox for MDR reversal.展开更多
Smart material can be defined as a material that can dock or convert energy between physical domains or as a material that can generate a response, in their characteristics, properties or geometries, when submitted to...Smart material can be defined as a material that can dock or convert energy between physical domains or as a material that can generate a response, in their characteristics, properties or geometries, when submitted to an external stimulus, for example, to heat, water presence, light, etc. In this paper, the second definition will be approached. Hydrogels are crosslinked materials that can absorb a big amount of water. They generally can be considered as smart materials once they exhibit sensibility to external stimuli like to pH variation, as will be approached in this paper. Thus, chitosan/polyvinylpyrrolidone hydrogels of three different ratios between these two polymers (1:1, 7:3 and 3:7) were synthesized and putted in aqueous solution with different pHs. The pH was adjusted adding drops of NaOH and HCl, slowly. After the collection of results and in order to understand the phenomena in a visual way, models of the molecules were also elaborated using the Avogadro software. Therefore, it was possible to realize that the greater the ratio of chitosan in the hydrogel, the greater its sensitivity to pH. Such characteristic is associated with the amino (-NH2) groups in it structure, which are capable of protonating and deprotonating (depending of the pH), generating charges under the chemical structure of the material, which will expand its volume in order to minimize the repulsion between charges. In addition, it was also noted that the hydrogel expansion is inversely proportional to the pH increase. By practical tests, it was possible to conclude that chitosan/PVP hydrogel with ratio 7:3 is the most interesting once it presented a greater quantity of chitosan in its composition, what implied in more rigidity than the others and greater ease of handling, resulting in more reliable results. The obtained polymers were characterized by ^13C NMR, ^1H NMR and FTIR. The effects of pH on swelling ratio (SR) of the pure crosslinked PDMAEMA membrane and on separation performances of the composite NF membrane were investigated. The results show that the quaternized amino groups produced through interfacial polymerization technique are soluble in both phases, which accelerate the crosslinking reaction as self-catalysts. The swelling/contracting behavior of the pure crosslinked PDMAEMA exhibited a well reversible pH sensitive property. Importantly, the rejection and flux of the composite NF membrane show pH-sensitive behavior in NF process. Furthermore, with the help of a relatively novel method to measure membrane conduction, the true zeta potentials calculated on the basis of the streaming potential measurements proved the pH-sensitive behavior of the NF membrane.展开更多

Methods The...Objective To evaluate the acute toxicity of 2-deoxy-D-glucose (2DG) by oral (p.o.) and intravenous (i.v.) routes, and also the cardio-respiratory effects following high doses of 2DG in animal models. Methods The LD50 of 2DG (in water) was determined in rats and mice by p.o. route and in mice by i.v. route. The effect of 2-DG (250 mg/kg, 500 mg/kg, and 1000 mg/kg, i.v.) was studied on various cardio-respiratory parameters viz., mean arterial blood pressure, heart rate and respiratory rate in anaesthetised rats. The effect of 2DG (500 mg/kg, 1000 mg/kg, and 2000 mg/kg, p.o.) was also studied on various respiratory parameters viz., respiratory rate and tidal volume in conscious rats and mice using a computer program. Results The p.o. LD50 of 2DG was found to be 〉8000 mg/kg in mice and rats, and at this dose no death was observed. The LD50 in mice by i.v. route was found to be 8000 mg/kg. At this dose 2 out of 4 mice died and the death occurred within 6 h. A significant increase in the body weight was observed after p.o. administration of 2DG in rats at 500 mg/kg, 1000 mg/kg, and 2000 mg/kg doses. There was no significant change in the body weight at 4000 mg/kg and 8000 mg/kg by the p.o. route in rats and up to 8000 mg/kg by p.o. as well as i.v. routes in mice. Intravenous administration of 2DG (250 mg/kg, 500 mg/kg, and 1000 mg/kg) in anaesthetised rats showed a time-dependent decrease in the mean arterial blood pressure. There was no change in the heart rate in any of the treatment groups. The tidal volume was not changed significantly by p.o administration in conscious rats, but a significant decrease in the respiratory frequency at 500 mg/kg and 1000 mg/kg doses was observed. In the mice also there was no change in the tidal volume after p.o, administration, but the respiratory frequency decreased significantly at 2000 mg/kg dose. Through changing the feeding modes of reactants, two types of hydrogels(comb-type and reticular-typ...A new kind of biodegradable pH-sensitive drug delivery system was developed via chitosan-albumin conjugate hydrogel. Through changing the feeding modes of reactants, two types of hydrogels(comb-type and reticular-type) were synthesized by amidation reactions between 6-O-succinoylated N-phthaloyl chitosan and albumin. The structures and morphologies of the hydrogels were characterized by SEM. And their water swelling capacity, drug loading and releasing properties at different pH values were also investigated. It was found that the comb-type of hydrogels with looser space construction had better water swelling ratio(more than 400% of its original mass) than the reticular-type of ones did(about 180% of its original mass). In vitro release experiments of Rifampicin show that the hydrogels provided the controlled release of the entrapped drug for more than 50 h. The drug release rates of both types of hydrogels under acidic condition were lower than those under neutral or basic condition. The introduction of albumin not only improved the hydrophilicity of chitosan, but also provided the possibility of the carrier system combining other biologically active materials more easily to fulfill the delivery and therapy functions.展开更多
Series batch experiments were made to investigate the influences of pH and temperature on the activity of acidogenus and acidogenus in glucose-degrading bacteria cultured in an UASB(up-flow anaerobic sludge blanket) r...Series batch experiments were made to investigate the influences of pH and temperature on the activity of acidogenus and acidogenus in glucose-degrading bacteria cultured in an UASB(up-flow anaerobic sludge blanket) reactor for glucose fermentation and hydrogen production. The bacteria exhibited different capability to recover to produce hydrogen at different initial pH and temperature. Hydrogen production, VFA production, COD removal and COD balance were measured at different pH and 20, 37 ℃ respectively with the same glucose and VSS in vials. Results showed that there are different influences on the activity of acidogenic bacteria at varied pH and result in a variety of amount of hydrogen production, specific hydrogen production and VFA production, etc. Through the present study, when nonmalized to the weight of VSS, a maximal biogas and hydrogen production of 1 717 1 ml/g and 870 0 ml/g were obtained when pH equals 9 at 37 ℃ and 679 00 ml/g of biogas, 246 35 ml/g of hydrogen were also got when pH equals 5 at 20 ℃ respectively. The maximal specific hydrogen production (SHA) was 116 56 ml/h,g around 8 of pH value at 37 ℃ and 6 46 ml/h,g around 4 of pH value at 20 ℃, which were obtained by calculating the slope of the accumulated hydrogen gas via time. Butyric acid fermentation was important for hydrogen production. Large quantity of unknown COD was found in the vials when a small quantity of bio-gas was produced, but relative less unknown COD was determined when there was large quantity of hydrogen produced. This revealed a better engineering foreground for application of hydrogen bio-production.展开更多

In vitro release experiments of Rifampicin show that the hydrogels provided the controlled release of the entrapped drug for more than 50 h. The drug release rates of both types of hydrogels under acidic condition were lower than those under neutral or basic condition. The introduction of albumin not only improved the hydrophilicity of chitosan, but also provided the possibility of the carrier system combining other biologically active materials more easily to fulfill the delivery and therapy functions.
文摘Series batch experiments were made to investigate the influences of pH and temperature on the activity of acidogenus and acidogenus in glucose-degrading bacteria cultured in an UASB(up-flow anaerobic sludge blanket) reactor for glucose fermentation and hydrogen production. The bacteria exhibited different capability to recover to produce hydrogen at different initial pH and temperature. Hydrogen production, VFA production, COD removal and COD balance were measured at different pH and 20, 37 ℃ respectively with the same glucose and VSS in vials. Results showed that there are different influences on the activity of acidogenic bacteria at varied pH and result in a variety of amount of hydrogen production, specific hydrogen production and VFA production, etc. Through the present study, when nonmalized to the weight of VSS, a maximal biogas and hydrogen production of 1 717 1 ml/g and 870 0 ml/g were obtained when pH equals 9 at 37 ℃ and 679 00 ml/g of biogas, 246 35 ml/g of hydrogen were also got when pH equals 5 at 20 ℃ respectively. The maximal specific hydrogen production (SHA) was 116 56 ml/h,g around 8 of pH value at 37 ℃ and 6 46 ml/h,g around 4 of pH value at 20 ℃, which were obtained by calculating the slope of the accumulated hydrogen gas via time. Butyric acid fermentation was important for hydrogen production. Large quantity of unknown COD was found in the vials when a small quantity of bio-gas was produced, but relative less unknown COD was determined when there was large quantity of hydrogen produced. This revealed a better engineering foreground for application of hydrogen bio-production.
