绿豆(Vigna radiata L.)是一种严格闭花授粉作物,其人工授粉效率低、成本高,通过杂交获取新品种较为困难。目前,国内外绿豆遗传转化体系鲜见报道,这极大地限制了绿豆基因功能的研究和新品种的开发,因此尝试建立绿豆遗传转化途径具有重... 绿豆(Vigna radiata L.)是一种严格闭花授粉作物,其人工授粉效率低、成本高,通过杂交获取新品种较为困难。目前,国内外绿豆遗传转化体系鲜见报道,这极大地限制了绿豆基因功能的研究和新品种的开发,因此尝试建立绿豆遗传转化途径具有重要意义。首先对绿豆谷胱甘肽还原酶基因(GR)进行生物信息学分析,并以此为基础,构建基因编辑重组体,同时进行发根农杆菌的遗传转化。结果表明,GR蛋白相对分子质量为58.38 kDa,稳定性较好,为亲水蛋白,有多个磷酸化位点,不含跨膜结构,定位于细胞质中,二级结构主要包括ɑ-螺旋和延伸链,且含有Pyr_redox_2和Pyr_redox_dim两个结构域;系统进化分析发现该蛋白与赤豆(Vigna angularis)亲缘关系最近。在此基础上构建四靶点CRISPR/Cas9-GR-gRNA基因编辑载体,通过发根农杆菌遗传转化绿豆子叶节获取毛状根20株,后经PCR检测获取阳性克隆毛状根12株,其毛状根转化率约为60%,其研究结果可为绿豆后续基因编辑研究奠定重要基础。 展开更多

研究了不同冷轧变形量和退火工艺对Gr.39钛合金带卷显微组织和力学性能的影响。结果表明,对于热轧退火态Gr.39钛合金带卷,当冷轧变形量从0升高到59.4%时,显微组织由完全退火的等轴组织逐渐变形为被拉长的纤维状组织,并且随着变形量的增... 研究了不同冷轧变形量和退火工艺对Gr.39钛合金带卷显微组织和力学性能的影响。结果表明,对于热轧退火态Gr.39钛合金带卷,当冷轧变形量从0升高到59.4%时,显微组织由完全退火的等轴组织逐渐变形为被拉长的纤维状组织,并且随着变形量的增加,材料加工硬化程度逐渐增加。考虑到轧制过程的稳定性,建议单轧程冷轧变形量控制在60%以内。Gr.39钛合金带卷在700℃退火发生完全再结晶,退火温度升高至相变点以上时,显微组织转变为粗大魏氏组织。随着退火温度的升高,抗拉强度和屈服强度逐渐降低,延伸率逐渐升高。推荐冷轧Gr.39钛合金带卷退火制度为700℃/8 h/AC。 展开更多

The as-cast and as-extruded Mg–9Li–1Al–xCa alloys(x=0,0.2;wt%)were prepared by a simple alloying process followed by hot extrusion with an extrusion ratio of 28.2.The microstructures of the as-cast and as-extruded Mg–9Li–1Al–xCa alloys were observed to investigate the effect of calcium(Ca)element on the Mg–9Li–1Al(LA91)alloy,and the crystallographic calculations between Al_(2)Ca and the matrix(α-Mg andβ-Li phases)were examined on the basis of the edge-to-edge matching model.The experimental results indicate that the addition of 0.2 wt%Ca into LA91 alloy reduce the size of theα-Mg phases in the as-cast alloy and that ofβ-Li phases in the as-extruded alloy due to the Al_(2)Ca particles distributed inside the matrix.Crystallographic calculation results suggested that there is a good crystallographic matching between the matrix and Al_(2)Ca,which confirmed that Al_(2)Ca particles can act as a heterogeneous nucleation site for bothα-Mg andβ-Li phases and were effective grain refiners for LA91 alloy. 展开更多

Creep aging forming(CAF) is a potential process used to manufacture large integral components of magnesium(Mg) alloys. The selected stress plays a crucial role in creep aging processes but the mechanism by which stress loading method affects creep aging of Mg alloys is still unclear. In this paper, the microstructural evolution of precipitated phases and precipitation-free zones(PFZ) at grain boundaries with different stress loading modes(unstressed, unidirectional tensile stress, and cyclic stress) at 250 ℃ were investigated along with changes in mechanical properties. The results showed that the addition of stress during aging effectively promoted the precipitation of precipitated phases, while unaffecting grain size. Unidirectional tensile stress caused directional growth of β phase([1010]), as well as rotation of weave towards the basal plane texture, resulting in namely stress orientation effect. Solute atoms diffused in the direction of tensile stress while vacancies moved perpendicular to the direction of tensile stress, resulting in PFZ at grain boundaries(157.06 nm). By contrast, cyclic stresses led to the growth of β phase in three directions([1010], [1100] and [0110]). The solute atoms and vacancies were uniformly distributed in the Mg matrix instead of directional diffusion, effectively reducing the width of PFZ(112.39 nm) at the grain boundary. These features significantly improved the mechanical properties of alloy specimens after cyclic stress creep aging when compared to unidirectional stress creep aging, with yield strength(YS), ultimate tensile strength(UTS), and elongation(EL) enhanced from 171.6 MPa, 305.5 MPa, and 4.4%to 174.8 MPa, 326.3 MPa, and 6.9%, respectively. 展开更多

The structure and magnetic properties of Fe76.5Si13.5B9Cu1 alloys with a nanocrystalline (NC) bcc Fe(Si) phase trom about 23 to 46 nm in diameter, which were first formed into amorphous ribbons and then annealed at various temperatures between 703 and 773 K, have been investigated. At annealing temperatures from 703 to 748 K, the single NC bcc(Si) phase is obtained in the crystallized alloys. The grain size and the Si-content in the NC bcc Fe(Si) phase for the alloys annealed at different temperatures are presented. The soft magnetic properties and the saturation magnetostriction for the alloys with the NC bcc Fe(Si) phase are also measured. The results show that, the saturation magnetizotion and the permeability are improved for the alloys with only the NC bcc Fe(Si) phase and become better with decreasing of the NC bcc phase size, and the saturation magnetostriction declines for the alloys with increasing Si-content in the NC bcc Fe(Si) phase. 展开更多

Based on the deforming technique of severe plastic deformation(SPD), the grain refinement of a Mg-9Gd-3Y-2Zn-0.5Zr alloy treated with decreasing temperature reciprocating upsetting-extrusion(RUE) and its influence on the mechanical properties and wear behavior of the alloy were studied. The RUE process was carried out for 4 passes in total, starting at 0 ℃ and decreasing by 10 ℃ for each pass. The results showed that as the number of RUE passes increased, the grain refinement effect was obvious, and the second phase in the alloy was evenly distributed. Room temperature tensile properties of the alloy and the deepening of the RUE degree showed a positive correlation trend, which was due to the grain refinement, uniform distribution of the second phase and texture weakening. And the microhardness of the alloy showed that the microhardness of RUE is the largest in 2 passes. The change in microhardness was the result of dynamic competition between the softening effect of DRX and the work hardening effect. In addition, the wear resistance of the alloy showed a positive correlation with the degree of RUE under low load conditions. When the applied load was higher, the wear resistance of the alloy treated with RUE decreased compared to the initial state alloy. This phenomenon was mainly due to the presence of oxidative wear on the surface of the alloy, which could balance the positive contribution of severe plastic deformation to wear resistance to a certain extent. 展开更多

目的:研究Delicaflavone对人吉非替尼耐药肺癌PC-9/GR细胞迁移与侵袭作用及机制。方法:采用MTT法检测Delicaflavone对PC-9/GR细胞存活率的影响;采用细胞划痕实验和Transwell实验测定Delicaflavone对PC-9/GR细胞迁移和侵袭能力的影响;采... 目的:研究Delicaflavone对人吉非替尼耐药肺癌PC-9/GR细胞迁移与侵袭作用及机制。方法:采用MTT法检测Delicaflavone对PC-9/GR细胞存活率的影响;采用细胞划痕实验和Transwell实验测定Delicaflavone对PC-9/GR细胞迁移和侵袭能力的影响;采用Western blotting法检测Delicaflavone对PC-9/GR细胞基质金属蛋白酶9(matrix metalloproteinases 9,MMP-9)、基质金属蛋白酶2(matrix metalloproteinases 2,MMP-2)、钙黏连蛋白(E-cadherin,N-cadherin)、波形蛋白(vimentin)及PI3K/Akt/mTOR信号通路相关蛋白表达情况。结果:与对照组比较,20 mg/L Delicaflavone作用24 h能显著抑制PC-9/GR细胞的细胞活性(P<0.05),而浓度在10 mg/L以下时则对细胞活性无明显影响。Delicaflavone能够浓度依赖性地抑制PC-9/GR细胞的迁移率、侵袭细胞数量(P<0.05和P<0.01),并能浓度依赖性地减少PC-9/GR细胞迁移标记蛋白(MMP-9和MMP-2)的表达(P<0.01),上调E-cadherin、下调N-cadherin和Vimentin的表达(P<0.01)。此外,Delicaflavone还浓度依赖性地减少p-PI3K、p-Akt和p-mTOR表达(P<0.01)。结论:Delicaflavone具有抑制PC-9/GR细胞迁移及侵袭能力的作用,该作用与Delicaflavone通过PI3K/Akt/mTOR通路调控上皮-间质转化有关。 展开更多

目的:观察miR-21抑制物对人肺腺癌PC9/GR细胞株增殖、凋亡及迁移能力的影响,探讨miR-21抑制物在PC9/GR细胞吉非替尼耐药的生物学行为中的作用。方法:实验分为miR-21 inhibitor组、阴性对照组和正常细胞对照组3组。MiR-21 inhibitor组转... 目的:观察miR-21抑制物对人肺腺癌PC9/GR细胞株增殖、凋亡及迁移能力的影响,探讨miR-21抑制物在PC9/GR细胞吉非替尼耐药的生物学行为中的作用。方法:实验分为miR-21 inhibitor组、阴性对照组和正常细胞对照组3组。MiR-21 inhibitor组转染miR-21抑制物抑制其细胞内miR-21表达,阴性对照组转染microRNA inhibitor NC。加入吉非替尼处理后,采用MTT比色实验测定3组细胞第1、2、3、4、5天的吸光度(OD值),绘制生长曲线。采用流式细胞仪检测细胞凋亡,Transwell法检测细胞迁移。结果:两对照组间细胞的增殖速度无明显差异(P>0.05),与对照组相比,实验组细胞增殖速度明显减慢(P<0.05)。实验组细胞凋亡率为(40.06±2.32)%,明显增高(P<0.05);阴性对照组[(7.39±0.79)%]和空白对照组[(6.96±0.68)%]之间细胞凋亡无明显差异(P>0.05)。实验组细胞迁移力受到明显抑制(P<0.05)。空白对照组与阴性对照组细胞迁移力相比无明显差异(P>0.05)。结论:在PC9/GR细胞中,下调miRNA-21的表达后,PC9/GR细胞对GR的敏感性提高,凋亡程度增强,部分逆转了肺癌PC9/GR细胞对GR的耐药性。为miRNA-21与化疗联合治疗肺癌提供了实验依据。 展开更多

The effect of melt over-heating on the morphology of Al_9FeNi phase in 2618aluminum alloy with high contents of Fe and Ni and 0.22 wt. percent zirconium has been investigatedby optical microscopy. SEM and TEM. The mechanical properties of 2618 aluminum alloy after hotextrusion and quenching/aging have been tested. The results show: melt over-heating treatment of2618 alloy with high contents of Fe and Ni at 960 deg C led to finer and better-distributedneedle-like Al_9FeNi phase in cast microstructure and fine Al_9FeNi particles after hot extrusion;the grain size of the alloy after hot extrusion could also be refined evidently by alloying ofzirconium; the ambient and high temperature tensile strength and elongation of 2618 alloy have beenapparently enhanced due to fine Al_9FeNi particles and dispersed Al_3Zr as well as fine grain size. 展开更多

A continuous unidirectional solidification equipment with the advantages of electric slag remelting, induction heating, continuous casting and unidirectional solidification was built to study the QAl9 4 Cu Al alloy. The results show that the electro slag induction continuous unidirectional solidification process can be used for the steady continuous unidirectional solidification of QAl9 4, and revitalizes the down pulling continuous unidirectional solidification process; that the temperature distribution in the mold wall reflects that of the molten metal in the mold, thus the temperature distribution in the mold wall can be used to control the electric slag induction continuous unidirectional solidification process; and that the mutual matching of the technological parameters is the key to stabilize the solidification process. 展开更多

研究了火次变形量和退火温度对Gr.9合金管材显微组织和力学性能的影响,探讨了高强Gr.9合金管材的制备方法。结果表明:采用取消成品轧制道次前退火工序以增加火次变形量的方法及优化去应力退火温度可制备出满足标准要求的高强Gr.9合金管... 研究了火次变形量和退火温度对Gr.9合金管材显微组织和力学性能的影响,探讨了高强Gr.9合金管材的制备方法。结果表明:采用取消成品轧制道次前退火工序以增加火次变形量的方法及优化去应力退火温度可制备出满足标准要求的高强Gr.9合金管材。增加火次变形量使管材的强度提高,但塑性有所下降;同时使管材流线组织更为明显,晶粒破碎程度更为充分。经过470℃×90 min去应力退火后,火次变形量较大的管材强塑性可达到较优的匹配,从而满足Rm≥862 MPa,Rp0.2≥724 MPa,A50≥12%。 展开更多
