The sulfate groups in agar structure played a good role in the formation of fiber.However,commercially available agar is usually extracted from red algae by alkali treatment to decrease the content of sulfate group for the purpose of high gel strength.In this paper,an alkali-free method of agar extraction from Gracilaria lemaneiformis was proposed for the wet-spinning purpose.This method is environmentally friendly,reduces the extraction steps,saves energy,and reduces the production cost of agar fiber.The improved agar preparation process not only has higher agar yield,but also has higher molecular weight and sulfate group content,which is beneficial to the preparation and forming of fiber and makes the fiber have higher mechanical strength and elongation.Therefore,this extraction technology has broad application prospect in the textile field. 展开更多

Comparative studies of absorption spectra of phycobiliproteins of Gracilaria lemaneiformis Greville and its pigmental mutants were conducted in this study. The results showed that the absorption spectra of phycoerythrins ( PE) from different material changed significantly, while those of phycocyanins (PC) and allophycocyanins (APC) were basically similar. In order to disclose the essence of die difference, partial sequences of die subunit genes of PE of Qingdao strain of G. lemaneiformis (qd) and its pigmental mutants were determined. The amino acid sequences were deduced and used to explain spectral shifts of PE from the pigmental mutants. The amino acid sequences of PE resembled each other, and several residues changed among qd and its pigmental mutants. Residue substitutions were found in a region consisting of amino acids which determined are secondary structure and subunits interactions, thus might influence the confirmation and interaction of subunits, and further caused spectral deviation. 展开更多

Gracilaria lemaneiformis Bory is an economically important alga that is primarily used for agar production.Although tetraspores are ideal seeds for the cultivation of G.lemaneiformis,the most popular culture method is currently based on vegetative fragments,which is labor-intensive and time-consuming.In this study,we optimized the conditions for tetraspore release and evaluated the photosynthetic activities of different colonies formed from the branches of G.lemaneiformis using a PAM(pulse-amplitude-modulated) measuring system.The results showed that variations in temperature and salinityhad significant effects on tetraspore yield.However,variations in the photon flux density(from 15 μmol m-2 s-1 to 480 μmol m-2 s-1) had no apparent effect on tetraspore yield.Moreover,the PAM-parameters Y(I),Y(Ⅱ),ETR(I),ETR(Ⅱ) and Fv/Fm of colonies formed from different branches showed the same trend:parameter values of first generation branches>second generation branches>third generation branches.These results suggest that the photosynthetic activities of different colonies of branches changed with the same trend.Furthermore,photosynthesis in G.lemaneiformis was found to be involved in vegetative reproduction and tetraspore formation.Finally,the first generation branches grew slowly,but accumulated organic compounds to form large numbers of tetraspores.Taken together,these results showed that the first generation branches are ideal materials for the release of tetraspores. 展开更多

In this paper, in vitro anti-influenza virus activities of sulfated polysaccharide fractions from Gracilaria lemaneiforrnis were investigated. Cytotoxicities and antiviral activities of Gracilaria lemaneiformis polysaccharides (PGL), Gracilaria lemaneiformis polysaccharide fraction-1 (GL-1), Gracilaria lemaneiformis polysaccharide fraction-2 (GL-2) and Gracilaria lemaneiformis polysaccharide fraction-3 (GL-3) were studied by the Methyl thiazolyl tetrazolium (MTT) method, and the inhibitory effect against Human influenza virus H1-364 induced cytopathic effect (CPE) on MDCK cells were observed by the CPE method. In addition, the antiviral mechanism of PGL was explored by Plaque forming unit (PFU), MTT and CPE methods. The results showed: i) Cytotoxicities were not significantly revealed, and H1-364 induced CPE was also reduced treated with sulfated polysaccharide fractions from Gracilaria lemaneiformis; ii) Antiviral activities were associated with the mass percentage content of sulfate groups in polysaccharide fractions, which was about 13%, in polysaccharides (PGL and GL-2) both of which exhibited higher antiviral activity; iii) A potential antiviral mechanism to explain these observations is that viral adsorption and replication on host cells were inhibited by sulfated polysaccharides from Gracilaria lemaneiformis. In conclusion, Anti-influenza virus activities of sulfated polysaccharide fractions from Gracilaria lemaneiformis were revealed, and the antiviral activities were associated with content of sulfate groups in polysaccharide fractions 展开更多

Six ISSR primers are employed to display the polymorphism of different phases and sexes of red alga Gracilaria lemaneiformis, and two of them, P1 and P3, amplified distinct band patterns. The ISSR pattern amplified by primer P1 of the female gametophyte is identical to that of tetrasporophyte, but distinct from that of male gametophyte. Of the bands produced by primer P3, one is specific to female gametophyte. Three morphologically similar fronds can be easily identified using ISSR technique. Two specific markers, SM1 andSF3, related to male gametophyte and female gametophyte, are cloned and sequenced. The homologous sequences of SM1 are found to encode a hypothetical protein. There is no homologous sequence of SF3 that can be found in GenBank. 展开更多

This paper aims to evaluate the effects of ultraviolet-Bradiation(UVBR) on Gracilaria lemaneiformis,a commercial red macroalga and an important source of agar. To study the in-vitro effect of UVBR on G. lemaneiformis,this plant was cultivated and exposed to photosynthetically active radiation(PAR) at 40 μmol photons/(m2 ·s) and enhanced UVBR(0,0.36,0.72,1.08,1.44,and 1.80 k J/(m 2 ·d)) for 13 days. The samples were processed for histochemical analysis,and the growth rate,photosynthetic pigment contents,photosynthetic performance,reactive oxygen species levels,membrane permeability,malonyl dialdehyde contents and antioxidant capacity of G. lemaneiformis were investigated. After 13 days of exposure to PAR+UVBR,G. lemaneiformis showed photodamage and photoinhibition of photosynthetic pigments(chlorophylla and phycoerythrin),leading to a decreased photosynthetic efficiency. Further,there was a corresponding decrease in the relative growth rates and depigmentation and partial necrosis of the apical segments were noted after exposure to PAR+UVBR. Additionally,UVBR induced excess production of superoxide radicals and hydrogen peroxide,eliciting a marked cellular membrane damage and antioxidative response. 展开更多

Abstract The removal of cypermethrin with a red macroalga, Gracilaria lemaneiformis, was studied under laboratory conditions. Results showed that the residue contents with G. lemaneiformis were significantly lower than those corresponding groups without the algal thalli after 96 h treatment. The removal rates decreased with increasing concentrations, which were about 50% without G. lemaneiformis after 96h exposure, and increased to 89%, 73%, and 66% in flasks with G. lemaneiformis at the concentrations of 10, 100, and 1000 gg L-1, respectively. The amount of biosorption (absorption and adsorption) by G. lemaneiformis increased with the increasing concentration and exposure time. Adsorption was the main process for the removal by G. lemaneiformis, which accounted for 75%-97% of the total biosorption. However, biosorption only contributed 0.5%-19.3% to the total losses of cypermethrin, which was more efficient under the low concentration. Natural losses contributed the largest portion of losses, which was over 65% in all treatments during the experiment. The unknown pathway of removal, which might be the bio-decomposed by microorganisms attaching the algal thalli, also contributed a lot to the total removal. The results suggested that cultivation of G. lemaneiformis could significantly remove cypermethrin, especially at low concentrations, and large-scale cultivation of G. lemaneiformis has considerable potential of biorestoration of eutrophic and cypermethrin-poUuted coastal sea areas. 展开更多

We investigated the effects of dried macroalga Gracilaria lemaneiform (Rhodophyta) on photosynthesis of the bloom-forming microalga Chaetoceros curvisetus. C. curvisetus was cultured with different amounts of dried G. lemaneiformis under controlled laboratory conditions. We measured the photosynthetic oxygen evolution rate and established the chlorophyll a fluorescence transient (OJIP) curve coupled with its specific parameters. We observed concentration-dependent and time-dependent relationships between dried G. lemaneiformis and inhibition of photosynthesis in C. curvisetus. Co-culture with dried G. lemaneiformis also resulted in a decrease in the light-saturated maximum photosynthetic oxygen evolution rate (P^ax) in C. curvisetus, and a decrease in the OJIP curve along with its specific parameters; the maximum photochemical efficiency of PSII (FJFm), the amount of active PSII reaction centers per excited cross section at t=0 and t=--tFM (RC/CS0 and RC/CSm, respectively), the absorption flux per excited cross section at t=0 (ABS/ CS0), and the efficiency with which a trapped exciton moves an electron into the electron transport chain (~u0). The dark respiration rate (Rd) increased in C. curvisetus co-cultured with dried G. lemaneiformis. The JIP-test and the oxygen evolution results indicated that dried G. lemaneiJbrmis decreased the number of active reaction centers, blocked the electron transport chain, and damaged the oxygen-evolving complex of C. curvisetus. This result indicated that dried fragments of G. lemaneiformis could effectively inhibit photosynthesis of C. curvisetus, and thus, could serve as a functional product to control and mitigate C. curvisetus blooms. 展开更多

The chemical composition, mineral elements, vitamins, free fatty acids and amino acid content of the edible red alga Gracilaria lemaneiformis Bory, grown in the sea near Nan'ao island, Guangdong Province, were analyzed in the present study. Gracilaria lemaneiformis Bory showed a total sugar content of 14.65%. The protein content was 21%, of which approximately 41% was determined to be essential amino acids (EAA). The major amino acid components were glutamic acid, leucine, arginine, and alanine. Of the EAA assayed, methionine and cysteine appeared to be the most limiting amino acids compared with the EAA pattern provided by Food and Agricultural Organization of the United Nations. The total lipids content was 0.87% and comprised a high composition of unsaturated fatty acids (61%), mainly as linoleic acid and oleic acid, and a little amount of polyunsaturated fatty acid; palmitic acid was the main component (39%) of saturated acids. Relatively high levels of vitamin C, iodine, phosphorus, and zinc were also present in G. lemaneiformis. The nutritional composition between G. lemaneiformis and Nostoc flagelliforme, a rare alga that is widely eaten in Chinese society, was compared. The results suggest that N. flagelliforme can be substituted for by G. lemaneiformis, not only because of their similar shape, but also because of their approximate nutritional composition. Gracilaria lemaneiformis may possibly serve as a potential healthy food in human diets in the future. 展开更多
