Inhibiting the death receptor 3(DR3)signaling pathway in group 3 innate lymphoid cells(ILC3s)presents a promising approach for promoting mucosal repair in individuals with ulcerative colitis(UC).Paeoniflorin,a prominent component of Paeonia lactiflora Pall.,has demonstrated the ability to restore barrier function in UC mice,but the precise mechanism remains unclear.In this study,we aimed to delve into whether paeoniflorin may promote intestinal mucosal repair in chronic colitis by inhibiting DR3 signaling in ILC3s.C57BL/6 mice were subjected to random allocation into 7 distinct groups,namely the control group,the 2%dextran sodium sulfate(DSS)group,the paeoniflorin groups(25,50,and 100 mg/kg),the anti-tumor necrosis factor-like ligand 1A(anti-TL1A)antibody group,and the IgG group.We detected the expression of DR3 signaling pathway proteins and the proportion of ILC3s in the mouse colon using Western blot and flow cytometry,respectively.Meanwhile,DR3-overexpressing MNK-3 cells and 2%DSS-induced Rag1^(-/-)mice were used for verification.The results showed that paeoniflorin alleviated DSS-induced chronic colitis and repaired the intestinal mucosal barrier.Simultaneously,paeoniflorin inhibited the DR3 signaling pathway in ILC3s and regulated the content of cytokines(interleukin-17A,granulocyte-macrophage colony stimulating factor,and interleukin-22).Alternatively,paeoniflorin directly inhibited the DR3 signaling pathway in ILC3s to repair mucosal damage independently of the adaptive immune system.We additionally confirmed that paeoniflorin-conditioned medium(CM)restored the expression of tight junctions in Caco-2 cells via coculture.In conclusion,paeoniflorin ameliorates chronic colitis by enhancing the intestinal barrier in an ILC3-dependent manner,and its mechanism is associated with the inhibition of the DR3 signaling pathway. 展开更多

简介了题示示踪剂井间监测技术的原理、示踪剂用量设计及所得资料解释方法。在胜坨油田11南断块沙二8砂层组,相隔22或23天,将3组不同的非分配型+分配型示踪剂分别注入3-9-271注水井的沙二81、沙二82、沙二83层,在周围生产不同层位的9口... 简介了题示示踪剂井间监测技术的原理、示踪剂用量设计及所得资料解释方法。在胜坨油田11南断块沙二8砂层组,相隔22或23天,将3组不同的非分配型+分配型示踪剂分别注入3-9-271注水井的沙二81、沙二82、沙二83层,在周围生产不同层位的9口采油井进行监测。以沙二81层为例说明示踪剂产出浓度曲线拟合效果较好。监测资料表明沙二8层的平面和纵向非均质性均较强,有几口井内有窜槽现象。由监测资料确定了3个小层的主要水驱方向。通过监测数据的综合解释,得到该井组的井间高渗层渗透率、厚度、喉道半径、主流通道、波及体积及系数等储层参数,表明井间主流道均为长期注水冲刷形成的高渗层。井区非均质性强,83层尤其突出。根据综合解释结果修正地质模型,应用示踪数值模拟方法,得到了井组3个小层的剩余油饱和度分布图。图6表5参3。 展开更多

The study of the chlorite coatings always attracts scholars in China and other countries because the chlorite coatings play an important role in the preservation of residual primary pores in sandstone reservoirs.At present,the study of the origin and the controlling factors is relatively few.The occurrence,time of formation,genesis,controlling factors,and the mechanism of chlorite coatings inhibiting quartz overgrowths were studied in detail with thin section and SEM analysis.Samples were from the sandstone reservoirs of the T3x Group in the Baojie area,the transitional zone from the middle to the south of Sichuan Basin.The results indicate that the chlorite coatings on the walls of the pore spaces are oriented perpendicular to grain surfaces in the form of isopachous(even-thickness) grain-coating,while the chlorite coatings at the contacts between adjacent detrital grains are arranged with a preferred orientation tangential to the surface of detrital grains.The chlorite coatings were formed in the eogenetic stage.They were formed by recrystallization of Fe-rich clay films during the syndepositional period,and chlorite cements would be recrystallized after the coatings’ formation.The formation of chlorite coatings was mainly controlled by the depositional environment,provenance conditions,and diagenetic environment.The presence of chlorite coatings could result in the preservation of primary pores in deeply buried sandstone reservoirs by effectively inhibiting quartz overgrowths and the development of compaction and pressure solution. 展开更多

In order to effectively solve the problems of low accuracy,large amount of computation and complex logic of deep learning algorithms in behavior recognition,a kind of behavior recognition based on the fusion of 3 dimensional batch normalization visual geometry group(3D-BN-VGG)and long short-term memory(LSTM)network is designed.In this network,3D convolutional layer is used to extract the spatial domain features and time domain features of video sequence at the same time,multiple small convolution kernels are stacked to replace large convolution kernels,thus the depth of neural network is deepened and the number of network parameters is reduced.In addition,the latest batch normalization algorithm is added to the 3-dimensional convolutional network to improve the training speed.Then the output of the full connection layer is sent to LSTM network as the feature vectors to extract the sequence information.This method,which directly uses the output of the whole base level without passing through the full connection layer,reduces the parameters of the whole fusion network to 15324485,nearly twice as much as those of 3D-BN-VGG.Finally,it reveals that the proposed network achieves 96.5%and 74.9%accuracy in the UCF-101 and HMDB-51 respectively,and the algorithm has a calculation speed of 1066 fps and an acceleration ratio of 1,which has a significant predominance in velocity. 展开更多

To gain an overall picture of the evolution of MYB genes in relict plants, we cloned nine novel MYB genes in Taxodiaceae plants ( Taxodium distichum, Taxodium ascendens, Cryptomeria japonica var. Sinensis, Cryptomeria japonica cv. Araucarioides, Cryptomer Ja- ponica, Metasequoia glyptostroboides, Cunninghamia lanceolata, Tai- wania cryptomerioides and Glyptostrobus pensilis). The deduced amino acid sequences for MYBs showed that the nine MYB proteins contained two DNA binding domains. The first domain is from amino acid position 29 to 78, wherein three tryptophanes at 33, 53 and 73 were separated by 19 amino acids, respectively. The second domain is from amino acid position 82 to 127, wherein three tryptophanes at 86, 105 and 124 were separated by 18 amino acids, respectively, whereas the first tryptophane at amino acid position 86 is replaced by a phenylalanine. The characteri- zation of these conserved domains at nine MYBs indicated that they all belong to the R2R3-MYB group. The secondary structure analysis showed that a-helix and 13-turn are the major motifs of the predicted secondary structure of MYBs. The three dimensional model of each MYB protein showed that the structure is like clip, making it more flexi- ble and mobile. The similarities between the nine MYB proteins in Taxodiaceae were calculated. The highest identical value of 99% is be- tween CjsMYB, CjMYB and CjaMYB, whereas the lowest value of 82% is between TaMYB and C1MYB. According to the phylogenetic tree, the distances between different genera were relatively large whereas those within genera were relatively small. As expected, accessions of the same genus formed a subgroup before being grouped with other genera. 展开更多

The ZnAl<sub>x</sub>Fe<sub>2-x</sub>O<sub>4</sub> (x = 0.0, 0.2 and 0.4) spinel ferrites were prepared by the conventional solid state reaction in air at 1350°C. The X-ray diffraction of all the three samples showed sharp Bragg peaks indicating the formation of a single phase spinel structure. The lattice parameters of the samples were determined from the X-ray diffraction data using the Nelson-Riley extrapolation method. The lattice parameters, cation distribution and oxygen position parameters have also been determined by refining the data by the Rietveld method. Rietveld refinement of the XRD data reveals all the samples to possess cubic symmetry corresponding to the space group Fd3m. Lattice parameters were found to decrease with increasing Al concentration, i.e. 8.4322, 8.4002, and 8.3984 &Aring;for x = 0.0, 0.2 and 0.4, respectively. 展开更多

SU(3) Simple Group Model and New Z' Properties in Future Linear Colliders

In the SU(3) simple group model, the new neutral gauge boson Z' couples to pairs of SM fermions with couplings fixed in terms of the SM gauge couplings and depending only on the choice of the fermion embedding. In this paper, we calculate the contributions of this new particle to the processes e^+e^-→l^+l^-, bb^-, and cc^- and study the possibility of detecting this new particle via these processes in the future high-energy linear e^+e^- collider(LC) experiments with √s= 500 GeV and £int= 340 fb^-1. We find that the new gauge boson Z' is most sensitive to the process e^+e^-→b^+b^-. As long as Mz,≤2 TeV , the absolute values of the relative correction parameter are larger than 5%. We calculate the forward-backward asymmetries and left-right asymmetries for the process e^+e^-→c^+c^-, with both the universal and anomaly-free fermion embeddings. Bounds on Z' masses are also estimated within 95% confidence level. 展开更多

The Higgs-like boson discovered at CERN in 2012 is tentatively assigned to a newly found bound state of two charged gauge bosons W<sup>+</sup>W<sup>-</sup> with a mass of E<sub>B</sub> ≈ 117 GeV, much closer to the measured 125 GeV than 110 GeV predicted in a paper with the same title earlier this year. The improvement is due to a shift from the earlier SU(2) representation assignment for the gauge bosons to the more realistic SU(3) one and that the computations are carried out with much greater accuracy. 展开更多

Luteolin ameliorates ulcerative colitis in mice via reducing the depletion of NCR+ILC3 through Notch signaling pathway

The disorder of group 3 innate lymphoid cells(ILC3)subgroup,such as the predominance of NCR-ILC3 but the deple-tion of NCR+ILC3,is unfavorable to damaged intestinal barrier repair,which leads to the prolongations and obstinacy of ulcerative colitis(UC).Our previous studies had shown that luteolin promoted NCRILC3 differentitating into NCR+ILC3 to improving the de-pletion of NCR+ILC3 in UC mice,while the mechanism is unclear.This article aimed to explore the underlying mechanism of luteolin enhancing the proportion NCR+ILC3.UC mice model was established with 2%DSS and Notch signaling was blocked,then luteolin was used to intervene.The results showed that the effect of luteolin on ameliorating disease symptoms in UC mice,including inhibit-ing the weight loss,reducing the pathological damage of colon mucosa,etc.,was diminished with blocking Notch signaling pathway.In addition,luteolin increased the proportion of NCR+ILC3,NCR+MNK3 and IL-22+ILC3,decreased intestinal permeability,pro-moted mucin secretion,and promoted ZO-1 and Occludin expression,the above effect of luteolin was neutralized by Notch inhibitor LY-411575.Luteolin activated the abnormally blocked Notch signaling pathway in UC mice.And molecular docking predicted the af-finity of luteolin for RBPJ to be-7.5 kcal·mol^(-1 )in mouse,respectively;the affinity of luteolin for Notch1 and RBPJ was respectively scored to be-6.4 kcal·mol^(-1) and-7.7 kcal·mol^(-1) homo sapiens.These results proved that luteolin is positive for enhancing the propor-tion of NCR+ILC3 via Notch signaling,and it provides a basis for targeting NCR+ILC3 for restoring intestinal barrier function to alle-viating ulcerative colitis. 展开更多

OX40L is one of the co-stimulatory molecules that can be expressed by splenic lymphoid tissue inducer(Lti)cells,a subset of group 3 innate lymphoid cells(ILC3s).OX40L expression in subsets of intestinal ILC3s and the molecular regulation of OX40L expression in ILC3s are unknown.Here,we showed intestinal ILC3s marked as an OX40L high population among all the intestinal leukocytes and were the dominant source of OX40L in Rag1–/–mice.All ILC3 subsets expressed OX40L,and NCR–ILC3s were the most abundant source of OX40L.The expression of OX40L in ILC3s could be upregulated during inflammation.In addition to tumor necrosis factor(TNF)-like cytokine 1A(TL1A),which has been known as a trigger for OX40L,we found that Poly(I:C)representing viral stimulus promoted OX40L expression in ILC3s via a cell-autonomous manner.Furthermore,we demonstrated that IL-7-STAT5 signaling sustained OX40L expression by ILC3s.Intestinal regulatory T cells(Tregs),most of which expressed OX40,had defective expansion in chimeric mice,in which ILC3s were specifically deficient for OX40L expression.Consistently,co-localization of Tregs and ILC3s was found in the cryptopatches of the intestine,which suggests the close interaction between ILC3s and Tregs.Our study has unveiled the crosstalk between Tregs and ILC3s in mucosal tissues through OX40–OX40L signaling,which is crucial for the homeostasis of intestinal Tregs. 展开更多

Objective: To investigate the effects of composite Sophora colon-soluble Capsule(CSCC) on gut microbiota-mediated short-chain fatty acids(SCFAs) production and downstream group 3 innate lymphoid cells(ILC3s) of dextran sulfate sodium(DSS)-induced ulcerative colitis(UC) mice model. Methods: The main components of CSCC were analyzed by hybrid ultra-high-performance liquid chromatography ion mobility spectromety quadrupole time-of-flight mass spectrometry(UHPLC-IM-QTOF/MS). Twenty-four male BALB/c mice were randomly divided into 4 groups(n=6) by using a computer algorithm-generated random digital, including control, DSS model, mesalazine, and CSCC groups. A DSS-induced colitis mice model was established to determine the effects of CSCC by recording colonic weight, colonic length, index of colonic weight, and histological colonic score. The variations in ILC3s were assessed by immunofluorescence and flow cytometry. The results of gut microbiota and SCFAs were acquired by 16s r DNA and gas chromatography-mass spectrometry(GC-MS) analysis. The expression levels of NCR^(+)ILC3^(-), CCR6^(+)Nkp46-(Lti) ILC3^(-), and ILCreg-specific markers were detected by enzyme-linked immunosorbent assay, and real-time quantitative polymerase chain reaction and Western blot, respectively. Results: The main components of CSCC were matrine, ammothamnine, Sophora flavescens neoalcohol J, and Sophora oxytol U. After 7 days of treatment, CSCC significantly alleviated colitis by promoting the reproduction of intestinal probiotics manifested as upregulation of the abundance of Bacteroidetes species and specifically the Bacteroidales_S24-7 genus(P<0.05). Among the SCFAs, the content of butyric acid increased the most after CSCC treatment. Meanwhile, compared with the model group, Lti ILC3s and its biomarkers were significantly downregulated and NCR^(+) ILC3s were significantly elevated in the CSCC group(P<0.01). Further experiments revealed that ILC3s were differentiated from Lti ILC3s to NCR^(+)ILC3s, resulting in interleukin-22 production which regulates gut epithelial barrier function. Conclusion: CSCC may exert a therapeutic effect on UC by improving the gut microbiota, promoting metabolite butyric acid production, and managing the ratio between NCR^(+)ILC3s and Lti ILC3s. 展开更多

Spectroscopic study of water splitting by neutral metal clusters is crucial to understanding the microscopic mechanism of catalytic processes but has been proven to be a challenging experimental target due to the difficulty in size selection.Here,we report a size-specific infrared spectroscopic study of the reactions between neutral group 3 metals and water molecules based on threshold photoionization using a vacuum ultraviolet laser.Quantum chemical calculations were carried out to identify the structures and to assign the experimental spectra.All the M_(2)O_(4)H_(4)(M=Sc,Y,La)products are found to have the intriguing M_(2)(μ_(2)-O)(μ_(2)-H)(μ_(2)-OH)(η^(1)-OH)_(2) structures,indicating that the H-OH bond breaking,the M-O/M-H/M-OH bond formation,and hydrogen production proceed efficiently in the reactions between laser-vaporized metals and water molecules.The joint experimental and theoretical results on the atomic scale demonstrate that the water splitting by neutral group 3 metals is both thermodynamically exothermic and kinetically facile in the gas phase.These findings have important implications for unravelling the structure-reactivity relationship of catalysts with isolated metal atoms/clusters dispersed on supports. 展开更多

This paper proposes an Equivariant Filtering(EqF)framework for the inertial-integrated state estimation.As the kin-ematic system of the inertial-integrated navigation can be naturally modeled on the matrix Lie group SE_(2)(3),the sym-metry of the Lie group can be exploited to design an equivariant filter which extends the invariant extended Kalman filtering on the group-affine system and overcomes the inconsitency issue of the traditional extend Kalman filter.We firstly formulate the inertial-integrated dynamics as the group-affine systems.Then,we prove the left equivariant properties of the inertial-integrated dynamics.Finally,we design an equivariant filtering framework on the earth-centered earth-fixed frame and the local geodetic navigation frame.The experiments show the superiority of the proposed filters when confronting large misalignment angles in Global Navigation Satellite Navigation(GNSS)/Inertial Navigation System(INS)loosely integrated navigation experiments. 展开更多
