目的探讨补体凝集素H-ficolin表达与丙型肝炎病毒(hepatitis C virus,HCV)病毒感染及肝病病程发展的相关性。方法采用酶联免疫吸附实验(ELISA)检测丙肝、肝硬化、肝癌患者血清中H-ficolin的表达水平,并用免疫组化检测正常肝组织、肝硬... 目的探讨补体凝集素H-ficolin表达与丙型肝炎病毒(hepatitis C virus,HCV)病毒感染及肝病病程发展的相关性。方法采用酶联免疫吸附实验(ELISA)检测丙肝、肝硬化、肝癌患者血清中H-ficolin的表达水平,并用免疫组化检测正常肝组织、肝硬化组织、肝癌组织中H-ficolin的表达。结果丙肝患者血清中H-ficolin表达高于健康对照者,而肝硬化、肝癌患者血清H-ficolin水平低于健康对照者;正常肝细胞高表达H-ficolin,当肝细胞硬化甚至癌变后,则H-ficolin表达下降或不表达。结论 H-ficolin表达与丙型肝炎病程迁延,肝细胞发生硬化或癌变,具有一定相关性。 展开更多

目的了解肺炎支原体肺炎患儿急性期和恢复期血浆中H-ficolin、L-ficolin水平的变化。方法采用ELISA法检测85例肺炎支原体肺炎患儿急性期和恢复期血浆H-ficolin、L-ficolin水平,并进行相关性分析。结果急性期H-ficolin水平18~252uq/m... 目的了解肺炎支原体肺炎患儿急性期和恢复期血浆中H-ficolin、L-ficolin水平的变化。方法采用ELISA法检测85例肺炎支原体肺炎患儿急性期和恢复期血浆H-ficolin、L-ficolin水平,并进行相关性分析。结果急性期H-ficolin水平18~252uq/ml,平均(119±59)ug/ml,恢复期H-ficolin水平0.3~26.5ug/ml,平均(10.1±57)ug/ml,急性期明显高于恢复期(Z=2.161.P=0.031)。急性期L-ticolin水平0.2~106μg/ml,平均(4.5±2.3)ug/ml,恢复期L-ficolin水平0.03~102μg/ml,平均(4.6±22)uq/ml,急性期和恢复期无统计学差异(t=0.245,P=0.807)。患儿急性期血浆H-ficoNn水平与L-ficolin水平明显相关(r=0.22,P=0.039)。结论H-ficolin可能参与儿童肺炎支原体肺炎疾病过程。 展开更多

Ficolins are serum complement lectins,with a structure similar to mannose-binding lectin (MBL) and lung surfactant protein (SP)-A and SP-D.Ficolins activate the lectin complement system and play important roles in host innate immunity.Ficolins are members of the collectin family of proteins,which act as pattern recognition receptors (PRRs).They are soluble oligomeric defense proteins with lectin-like activity,and are able to recognize pathogen-associated molecular patterns (PAMPs),which are carbohydrate molecules on the surface of pathogens,and of apoptotic,necrotic,and malignant cells.Upon binding to their specific PAMPs,ficolins may trigger activation of the immune system either (1) by initiating activation of complement via the lectin pathway,(2) by a primitive type of opsonophagocytosis,or (3) by stimulating secretion of the inflammatory cytokines interferon (IFN)-γ,interleukin (IL)-17,IL-6,and tumor necrosis factor (TNF)-α,and production of nitric oxide (NO)by macrophages,thus limiting the infection and concurrently orchestrating the subsequent adaptive immune response.Recently,a number of reports have shown that dysfunction or abnormal expression of ficolins may play crucial roles in viral and bacterial diseases and in inflammation.This review summarizes the reports on the roles of ficolins in the infectious diseases,and provides insight into ficolins as novel innate immune therapeutic options to treat these diseases. 展开更多
