Methods: 74 colorectal cancer patients who atte...Objective: To investigate the value of peripheral blood helper T cell 17 cell level and monocyte/lymphocyte ratio to predict the prognosis of colorectal cancer patients. Methods: 74 colorectal cancer patients who attended Hospital 960 from January 2021 to January 2022 were retrospectively analyzed. Clinical data of the patients were collected, including gender, age, and histologic type. Immunohistochemical indexes such as Th17 cell level and monocyte/ lymphocyte ratio in the peripheral blood of patients were also collected. The prognosis of patients after treatment, as well as peripheral blood Th17 and MLR levels, were observed and analyzed. Results: After follow-up after treatment, in the final 74 patients, the prognosis was good in 32 patients, accounting for 43.24%, and the prognosis was bad in 42 patients, accounting for 56.76%. There were no significant differences between the average age and tumor diameters of the good prognosis and poor prognosis groups (P > 0.05). However, the TNM staging, intervention taken, differentiation degree, presence of distant metastasis, presence of lymph node metastasis, Th17 level, and MLR level are significantly different between the two groups (P < 0.05). Conclusion: Peripheral blood Th17 and MLR have predictive value for the prognosis of colorectal cancer patients, and high levels of peripheral blood Th17 and MLR imply poor prognosis. The detection of peripheral blood Th17 and MLR levels is simple and convenient and can be used as indicators to provide a reference for the prognostic assessment of colorectal cancer patients.展开更多

We shall f...A correct antibody response requires the participation of both B and T lymphocytes and antigen presenting cells. In this review we address the role of follicular helper T lymphocytes(T FH) in this reaction. We shall focus on the regulation of their development and function in health and disease. T FH can be characterized on the basis of their phenotype and the pattern of secretion of cytokines. This fact is useful to study their participation in the generation of antibody deficiency in primary immunodeficiency diseases such as common variable immunodeficiency, X-linked hyper Ig M syndrome orX-linked lymphoproliferative disease. Increased numbers of T FH have been demonstrated in several autoimmune diseases and are thought to play a role in the development of autoantibodies. In chronic viral infections caused by the human immunodeficiency virus, hepatitis B or C virus, increased circulating T FH have been observed, but their role in the protective immune response to these agents is under discussion. Likewise, an important role of T FH in the control of some experimental protozoan infections has been proposed, and it will be important to assess their relevance in order to design effective vaccination strategies.展开更多

Objective This study was performed to investigate the effect of high mobility group box-1 protein(HMGB1)on immune function of human T lymphocytes in vitro and explore its potential role in cell-mediated immune dysfunc...Objective This study was performed to investigate the effect of high mobility group box-1 protein(HMGB1)on immune function of human T lymphocytes in vitro and explore its potential role in cell-mediated immune dysfunction.Methods Fresh blood was obtained from healthy adult volunteers and peripheral blood mononuclear cells(PBMCs)were isolated,then rhHMGB1 was added to PBMCs.Four-color flow cytometric(FCM)analysis was used for the measurement of intracellular cytokine including interleukin IL-4 and interferon IFN-?ELISA kits were employed for the determination of IL-2 and sIL-2R protein levels in cell culture supernatants.Results(1)Different stimulating time and dosage of rhHMGB1 did not alter the number of IFN-αpositive cells(Th1).rhHMGB1 stimulation provoked a dose-dependent and time-dependent increase in Th2 subset and decrease in ratio of Th1 to Th2.(2)Compared with the untreated cells,when the cells were coincubated with rhHMGB1(10-100ng/ml)for 12 hrs,protein release of IL-2 and sIL-2R were significantly up-regulated.At 48 hrs,in contrast,protein production was relatively lower in cells after exposure to 100-1000 ng/ml rhHMGB1.Conclusions These findings demonstrated that HMGB1 has a dual influence on immune functions of human T lymphocytes.展开更多
The mechanism of action of ribavirin(RBV) as an immunomodulatory and antiviral agent and its clinical significance in the future treatment of patients with hepatitis C virus(HCV) infection are reviewed.RBV up-regulate...The mechanism of action of ribavirin(RBV) as an immunomodulatory and antiviral agent and its clinical significance in the future treatment of patients with hepatitis C virus(HCV) infection are reviewed.RBV up-regulates type 1 and/or 2 cytokines to modulate the T helper(Th) 1/2 cell balance to Th1 dominance.Examination of co-stimulatory signaling indicated that RBV down-modulates inducible co-stimulator on Th cells,which contributes to differentiating na?ve Th cells into Th2 cells while reducing their interleukin-10 production.The effects on T-regulatory(Treg) cells were also investigated,and RBV inhibited the differentiation of na?ve Th cells into adaptive Treg cells by downmodulating forkhead box-P3.These findings indicate that RBV mainly down-regulates the activity of Th2 cells,resulting in the maintenance of Th1 activity that contributes to abrogating HCV-infected hepatocytes.Although an interferon-free treatment regimen exhibits almost the same efficacy without serious complications,regimens with RBV will be still be used because of their ability to facilitate the cellular immune response,which may contribute to reducing the development of hepatocellular carcinogenesis in patients infected with HCV.展开更多
目的探讨果蝇双翅边缘缺刻同源基因(Notch)信号通路在慢性阻塞性肺疾病(COPD)辅助性T细胞1(Helper T cells 1,Th1)和辅助性T细胞2(Helper T cells 2,Th2)失衡中的作用及芪蛭皱肺颗粒的干预机制。方法70只Wistar大鼠随机挑选10只作为空...目的探讨果蝇双翅边缘缺刻同源基因(Notch)信号通路在慢性阻塞性肺疾病(COPD)辅助性T细胞1(Helper T cells 1,Th1)和辅助性T细胞2(Helper T cells 2,Th2)失衡中的作用及芪蛭皱肺颗粒的干预机制。方法70只Wistar大鼠随机挑选10只作为空白对照组,其余大鼠均采用香烟烟雾(CS)联合气管滴注脂多糖(Lipopolysaccharide,LPS)法建立COPD模型,空白对照组及造模组各随机挑选3只大鼠验证造模是否成功。造模结束进行灌胃给药干预,造模组大鼠随机分为模型对照组、阳性对照组(67.5μg·kg^(-1))及芪蛭皱肺颗粒高中低剂量组(3.24、1.62、0.81 g·kg^(-1)),分别给予生理盐水、醋酸地塞米松混悬液、芪蛭皱肺高、中、低剂量混悬液进行灌胃干预,空白对照组同模型对照组,灌胃等体积生理盐水。经28天造模及28天治疗后,采用动物肺功能测试系统检测吸气峰流速(Peak Inspiratory Flow,PIF)和呼气峰流速(Peak Expiratory Flow,PEF),处死大鼠提取肺脏、脾脏、血清及支气管肺泡灌洗液(BALF),苏木素-伊红(HE)染色评价肺组织病理变化,酶联免疫吸附实验法(ELISA)测定血清及BALF中肿瘤坏死因子-α(TNF-α)含量,流式细胞仪检测脾脏Th1/Th2细胞水平,免疫组织化学法(Immunohistochemistry,IHC)及蛋白免疫印迹法(Western blot)检测肺组织Notch1、Hes家族发状分裂相关增强子1(Hes1)、Hey家族发状分裂相关增强子1(Hey1)蛋白水平,实时荧光定量聚合酶链式反应(Real-time PCR,RT-PCR)检测肺组织Notch1、Hes1、Hey1基因表达水平。结果与空白对照组比较,模型对照组大鼠肺功能显著降低(P<0.05),肺组织出现炎性细胞浸润、支气管结构破坏等病变,血清及BALF中TNF-α含量显著升高(P<0.05),脾Th1细胞百分比显著降低(P<0.05),Th2细胞百分比显著升高(P<0.05),肺组织Notch1、Hes1、Hey1蛋白及mRNA表达显著升高(P<0.05),差异均具有统计学意义;与模型对照组比较,各给药组大鼠肺功能显著升高(P<0.05),肺组织病理损伤均有所减轻,血清及BALF中TNF-α含量显著降低(P<0.05),脾Th1细胞百分比显著升高(P<0.05),Th2细胞百分比显著降低(P<0.05),肺组织Notch1、Hes1、Hey1蛋白及mRNA表达显著降低(P<0.05),差异均具有统计学意义。结论芪蛭皱肺颗粒通过抑制Notch信号通路调节Th1/Th2平衡,从而改善COPD大鼠肺功能及病理损伤,影响其免疫功能。展开更多
目的观察血清成纤维生长因子-23(fibroblast growth factor 23,FGF23)/抗衰老基因Klotho、辅助性T淋巴细胞(helper T lymphocytes,Th)1/Th2细胞因子在老年终末期肾病患者中的表达,并分析对医院感染的预测和预后的影响。方法选取2021年1...目的观察血清成纤维生长因子-23(fibroblast growth factor 23,FGF23)/抗衰老基因Klotho、辅助性T淋巴细胞(helper T lymphocytes,Th)1/Th2细胞因子在老年终末期肾病患者中的表达,并分析对医院感染的预测和预后的影响。方法选取2021年1月1日至2022年12月31日自贡市精神卫生中心(自贡市老年病医院)收治的106例老年终末期肾病患者,男65例,年龄范围60~84岁,年龄(67.21±3.07)岁,女41例,年龄范围60~84岁,年龄(67.65±2.98)岁,于入院第2天检测血清FGF23、抗衰老基因Klotho、Th1细胞因子[γ干扰素(interferon-γ,IFN-γ)、白介素2(interleukin-2,IL-2)]、Th2细胞因子(IL-4、IL-10)水平,进行急性生理功能及慢性健康状况评分Ⅱ(acute physiology and chronic health status scoring systemⅡ,APACHEⅡ)评估。根据住院期间是否发生医院感染分为感染组(28例)、未感染组(78例),比较两组各项血清指标及APACHEⅡ评分水平,分析各项指标与APACHEⅡ评分的相关性。随访3个月统计患者生存预后[生存(71例)、死亡(35例)],比较不同预后患者血清FGF23、Klotho、IFN-γ、IL-2、IL-4、IL-10水平,分析各项指标对医院感染及预后的预测价值及临床效用。结果106例老年终末期肾病患者根据住院期间是否发生医院感染分为感染组(28例)、未感染组(78例),随访3个月后生存71例、死亡35例。入院第2天感染组血清FGF23、IL-4、IL-10水平及APACHEⅡ评分分别为(78.64±20.16)ng/mL、(20.14±1.48)μg/L、(22.47±2.56)μg/L、(26.38±6.51)分,未感染组分别为(60.17±16.83)ng/mL、(16.25±1.21)μg/L、(19.52±1.86)μg/L、(22.97±6.45)分,感染组血清FGF23、IL-4、IL-10水平及APACHEⅡ评分均高于未感染组(P<0.05);入院第2天感染组血清Klotho、IFN-γ、IL-2水平分别为(34.95±12.62)ng/mL、(22.19±1.69)μg/L、(28.73±2.95)μg/L,未感染组分别为(51.61±16.08)ng/mL、(25.31±1.74)μg/L、(33.95±1.52)μg/L,感染组血清Klotho、IFN-γ、IL-2水平均低于未感染组(P<0.05);血清FGF23、IL-4、IL-10水平与APACHEⅡ评分呈正相关(相关系数r=0.629、0.597、0.612,P均<0.05),血清Klotho、IFN-γ、IL-2水平与APACHEⅡ评分呈负相关(相关系数r=-0.632、-0.718、-0.701、0.597,P均<0.05);死亡患者入组后1个月血清FGF23、IL-10、IL-4水平分别为(77.49±21.85)ng/mL、(24.76±4.77)μg/L、(24.81±6.28)μg/L,生存患者分别为(58.92±16.94)ng/mL、(18.10±3.82)μg/L、(13.57±4.38)μg/L,死亡患者入组后1个月血清FGF23、IL-10、IL-4水平高于生存患者(P<0.05);死亡患者入组后1个月血清Klotho、IFN-γ、IL-2水平分别为(30.03±11.76)ng/mL、(20.33±2.63)μg/L、(27.19±4.91)μg/L,生存患者分别为(55.68±17.02)ng/mL、(26.54±4.79)μg/L、(35.22±5.64)μg/L,死亡患者入组后1个月血清Klotho、IFN-γ、IL-2水平低于生存患者(P<0.05);血清FGF23、Klotho、IFN-γ、IL-2、IL-4、IL-10预测终末期老年肾病患者发生医院感染、生存预后曲线下面积分别为0.904(OR:0.832~0.953)、0.911(OR:0.840~0.958),且具有良好临床效用(P<0.05)。结论老年终末期肾病患者血清FGF23/Klotho、Th1/Th2细胞因子水平失衡,合并感染患者血清FGF23、IL-10、IL-4水平异常升高,血清Klotho、IFN-γ、IL-2水平表达降低,联合血清FGF23/Klotho、Th1/Th2细胞因子预测医院感染、评估预后的价值较高。展开更多

文摘目的观察血清成纤维生长因子-23(fibroblast growth factor 23,FGF23)/抗衰老基因Klotho、辅助性T淋巴细胞(helper T lymphocytes,Th)1/Th2细胞因子在老年终末期肾病患者中的表达,并分析对医院感染的预测和预后的影响。方法选取2021年1月1日至2022年12月31日自贡市精神卫生中心(自贡市老年病医院)收治的106例老年终末期肾病患者,男65例,年龄范围60~84岁,年龄(67.21±3.07)岁,女41例,年龄范围60~84岁,年龄(67.65±2.98)岁,于入院第2天检测血清FGF23、抗衰老基因Klotho、Th1细胞因子[γ干扰素(interferon-γ,IFN-γ)、白介素2(interleukin-2,IL-2)]、Th2细胞因子(IL-4、IL-10)水平,进行急性生理功能及慢性健康状况评分Ⅱ(acute physiology and chronic health status scoring systemⅡ,APACHEⅡ)评估。根据住院期间是否发生医院感染分为感染组(28例)、未感染组(78例),比较两组各项血清指标及APACHEⅡ评分水平,分析各项指标与APACHEⅡ评分的相关性。随访3个月统计患者生存预后[生存(71例)、死亡(35例)],比较不同预后患者血清FGF23、Klotho、IFN-γ、IL-2、IL-4、IL-10水平,分析各项指标对医院感染及预后的预测价值及临床效用。结果106例老年终末期肾病患者根据住院期间是否发生医院感染分为感染组(28例)、未感染组(78例),随访3个月后生存71例、死亡35例。入院第2天感染组血清FGF23、IL-4、IL-10水平及APACHEⅡ评分分别为(78.64±20.16)ng/mL、(20.14±1.48)μg/L、(22.47±2.56)μg/L、(26.38±6.51)分,未感染组分别为(60.17±16.83)ng/mL、(16.25±1.21)μg/L、(19.52±1.86)μg/L、(22.97±6.45)分,感染组血清FGF23、IL-4、IL-10水平及APACHEⅡ评分均高于未感染组(P<0.05);入院第2天感染组血清Klotho、IFN-γ、IL-2水平分别为(34.95±12.62)ng/mL、(22.19±1.69)μg/L、(28.73±2.95)μg/L,未感染组分别为(51.61±16.08)ng/mL、(25.31±1.74)μg/L、(33.95±1.52)μg/L,感染组血清Klotho、IFN-γ、IL-2水平均低于未感染组(P<0.05);血清FGF23、IL-4、IL-10水平与APACHEⅡ评分呈正相关(相关系数r=0.629、0.597、0.612,P均<0.05),血清Klotho、IFN-γ、IL-2水平与APACHEⅡ评分呈负相关(相关系数r=-0.632、-0.718、-0.701、0.597,P均<0.05);死亡患者入组后1个月血清FGF23、IL-10、IL-4水平分别为(77.49±21.85)ng/mL、(24.76±4.77)μg/L、(24.81±6.28)μg/L,生存患者分别为(58.92±16.94)ng/mL、(18.10±3.82)μg/L、(13.57±4.38)μg/L,死亡患者入组后1个月血清FGF23、IL-10、IL-4水平高于生存患者(P<0.05);死亡患者入组后1个月血清Klotho、IFN-γ、IL-2水平分别为(30.03±11.76)ng/mL、(20.33±2.63)μg/L、(27.19±4.91)μg/L,生存患者分别为(55.68±17.02)ng/mL、(26.54±4.79)μg/L、(35.22±5.64)μg/L,死亡患者入组后1个月血清Klotho、IFN-γ、IL-2水平低于生存患者(P<0.05);血清FGF23、Klotho、IFN-γ、IL-2、IL-4、IL-10预测终末期老年肾病患者发生医院感染、生存预后曲线下面积分别为0.904(OR:0.832~0.953)、0.911(OR:0.840~0.958),且具有良好临床效用(P<0.05)。结论老年终末期肾病患者血清FGF23/Klotho、Th1/Th2细胞因子水平失衡,合并感染患者血清FGF23、IL-10、IL-4水平异常升高,血清Klotho、IFN-γ、IL-2水平表达降低,联合血清FGF23/Klotho、Th1/Th2细胞因子预测医院感染、评估预后的价值较高。
