Recombinant Galectins of Hemonchus contortus Inhibit Goat Cytokine mRNA Transcription of Peripheral Blood Mononuclear Cells in vitro

The effects of recombinant galectins of male and female Hemonchus contortus (Hco-gal-m/f) on the mRNA levels of IL-1β, IL-2, IL-4, IL-6, interferon-γ(IFN-γ), and tumor necrosis factor-γ (TNF-γ) of goat peripheral blood mononuclear cells (PBMCs) were examined in this study. Blood samples were collected from five randomly selected two-year-old healthy goats. PBMCs were separated and cultured in vitro with varying concentration galectins (Hco-gal-m/f) of 0, 10, 20, and 40 lag mL-L Semi-quantitative reverse transcription RT-PCR was employed to test the synthesis of mRNA. The results showed that the mRNA transcriptions of IL-1β, IL-2, IL-4, IL-6, IFN-γ, and TNF-γ in the PBMCs were inhibited by Hco-galm in a dose-dependent manner. Hco-gal-f inhibited mRNA synthesis of IL- 1β, IL-4, IFN-γ, and TNF-γ of the same cells in similar fashion. The findings suggested that the recombinant galectin proteins of H. contortus could decrease the transcription of cytokines in vitro. 展开更多

【目的】以体外试验观察雌雄捻转血矛线虫半乳糖凝集素重组蛋白(rHco-gal-m/f)对山羊外周血单核细胞多种细胞因子mRNA转录水平的影响及其量效关系。【方法】选用2岁健康山羊5只,分别从颈静脉无菌采血,分离外周血单核细胞(PBMCs),以ConA... 【目的】以体外试验观察雌雄捻转血矛线虫半乳糖凝集素重组蛋白(rHco-gal-m/f)对山羊外周血单核细胞多种细胞因子mRNA转录水平的影响及其量效关系。【方法】选用2岁健康山羊5只,分别从颈静脉无菌采血,分离外周血单核细胞(PBMCs),以ConA或LPS刺激的同时分别加入0μg·ml-1,10μg·ml-1、20μg·ml-1和40μg·ml-1的rHco-gal-m/f,进行体外培养。用RT-PCR方法,以肌动蛋白基因(β-actin)作内标,定量分析单核淋巴细胞白介素-1β(IL-1β)、白介素-2(IL-2)、白介素-4(IL-4)、白介素-6(IL-6)、干扰素-γ(IFN-γ)和肿瘤坏死因子-α(TNF-α)的mRNA丰度。【结果】重组雄性捻转血矛线虫半乳糖凝集素(rHco-gal-m)可以抑制上述细胞因子mRNA的转录,并呈现出明显的剂量依赖性;重组雌性捻转血矛线虫半乳糖凝集素(rHco-gal-f)可抑制IL-1β、IL-4、IFN-γand TNF-α的转录,也呈现出明显的剂量依赖性。【结论】重组捻转血矛线虫半乳糖凝集素能降低多种细胞因子在体外的转录。 展开更多
