Capillary electrochromatography(CEC)plays a significant role in chiral separation via the double separation principle,partition coefficient difference between the two phases,and electroosmotic flow-driven separation.Given the distinct properties of the inner wall stationary phase(SP),the separation ability of each SP differs from one another.Particularly,it provides large room for promising applications of open tubular capillary electrochromatography(OT-CEC).We divided the OT-CEC SPs developed over the past four years into six types:ionic liquids,nanoparticle materials,microporous materials,biomaterials,non-nanopolymers,and others,to mainly introduce their characteristics in chiral drug separation.There also added a few classic SPs that occurred within ten years as supplements to enrich the features of each SP.Additionally,we discuss their applications in metabolomics,food,cosmetics,environment,and biology as analytes in addition to chiral drugs.OT-CEC plays an increasingly significant role in chiral separation and may promote the development of capillary electrophoresis(CE)combined with other instruments in recent years,such as CE with mass spectrometry(CE/MS)and CE with ultraviolet light detector(CE/UV). 展开更多

Development of effective chromatographic or electrophoretic separation involves judicious deciding of selection of optimal experimental conditions that can provide an adequate resolution at a reasonable run time for the separation of interested components. Box-Behnken factorial design was effectively applied for the separation optimization of eight structurally related sulfonamides using capillary zone electrophorosis and reverse high performance liquid chromatography. Optimum values for volume ratio of THF to H2O in eluent, column temperature and flow rate of eluent are found as 12 to 88, 35℃ and 1.0 mL/min, respectively. Box-Behnken modified optimization model is extended to separation by capillary electrophoresis (CE). While using CE, a satisfactory separation is achieved with a minimum resolution larger than 1.0 for a separation time less than 10 min. 展开更多

The optimal mobile phase compositions of fluoxetine separation on each column were hexane/isopropanol/diethyl amine (98/2/0.2, v/v/v), hexane/isopropanol/diethyl amine (99/1/0. l, v/v/v), hexane/isopropanol/diethyl amine (98/2/0.2, v/v/v), methanol/0.2% triethylamine acetic acid (TEAA) (25/75, v/v; pH 3.8) and hexane/isopropanol/diethyl amine (98/2/0.2, v/v/v), respectively. Experimental results demonstrated that baseline separation (Rs〉1.5) of fluoxetine enantiomers was obtained on chiralcel OD-H, chiralpak AD-H, and cyclobond I 2000 DM while the best separation was obtained on the last one. The eluate orders of fluoxetine enantiomers on the columns were determined. The first eluate by chiralcel OJ-H and kromasil CHI-TBB is the S-enantiomer, while by chiralpak AD-H and cyclobond 12000 DM is the R-enantiomer. 展开更多

Separation and determination of metal ions based on the formation of chelate anions with 1-Nitroso-2, 7-dihydrexynaphthalene-3, 6-disulfonic acid (1N2, 7O3, 6S) was studied by rising HPCE of the nine metal ions examined, the ions that can be detected sensitively with 1-Nitroso-2, 7-dihydrexynaphthalene-3, 6-disulfonic acid were Fe2+, Co2+, Cu2+, Ni2+, Zn2+ and Pd2+. The cobalt chelate could exist in two oxidation states of cobalt. When TBA(+) were added in electrophoretic solutions, the drastic changes in elertrophoretic mobilities of chelate were observed, which was due to the ion association between chelates anions and TBA(+). The ion association constants of chelate anions with TBA(+) were determined by using the change in electrophoretic mobilities of chelates, metal ions tested were separated within 10 min using 30cm silica capillary (50 m i. d). 展开更多

Its pharmacological activity resides only in S-(+)-enantiomer, and R-()-enantiomer is not only inactive but also has many side effects. Thus it is necessary to separate R-enantiomer from racemic ibuprofen. We studied optical separation of racemic Ibuprofen with chiral high performance liquid chromatography (HPLC). Out of three different chiral stationary phases, which were selected on the basis of structure and availability, two were found to be ef-fective. There was optimum eluent composition for each stationary phase for good resolution in optical separation. Resolution decreased with increase of eluent flow rate, but effect of injection volume on resolution was insignificant at high eluent flow rate. 展开更多

手性药物的对映体理化性质极其相似,而药理、毒理作用常常存在显著差异,因此探索提高对映体分离性能的创新策略对手性分离具有重要意义。以往关于毛细管电泳(CE)中β-受体阻断药美托洛尔(MET)对映体分离的研究主要集中在仅添加手性选择... 手性药物的对映体理化性质极其相似,而药理、毒理作用常常存在显著差异,因此探索提高对映体分离性能的创新策略对手性分离具有重要意义。以往关于毛细管电泳(CE)中β-受体阻断药美托洛尔(MET)对映体分离的研究主要集中在仅添加手性选择剂环糊精(CD)及其衍生物,而对于在CD的基础上添加辅助添加剂的研究较少。本研究将3种深共晶溶剂(DESs)氯化胆碱-葡萄糖、氯化胆碱-果糖和乳酸-葡萄糖作为辅助添加剂,用于MET的CE手性分离,推测了DES对基于CD的毛细管电泳分离MET对映体的协同作用。首先考察了CD种类、CD浓度、缓冲液pH值和缓冲液浓度对MET分离的影响,优化得到了最优条件(15 mmol/L羧甲基-β-环糊精、pH=3.0、40 mmol/L磷酸盐缓冲液)。其次制备了3种DESs作为添加剂,研究不同DESs及其质量分数对手性分离的作用,最终确定了最佳条件:采用质量分数1.5%的氯化胆碱-果糖型DES,此时MET的分离度从不添加DES时的1.30提升至2.61,实现了基线分离。最后对分离效果以及机理进行了推测。本文所建立的MET对映体手性分离分析方法不仅对提高手性药物质量、保证临床用药安全有效具有重要意义,还有助于将DES协同用于CD衍生物毛细管电泳手性分离的进一步研究与发展。 展开更多

设计了一种可以进行高效毛细管电泳分析的微流控芯片,微流控芯片具有一个十字架结构的沟道,沟道横截面的尺寸为100μm×50μm。利用5-氨基甲基荧光素作为荧光探针,使用乙腈溶液与磷酸盐缓冲液作为流动相进行高效液相色谱分析,等量... 设计了一种可以进行高效毛细管电泳分析的微流控芯片,微流控芯片具有一个十字架结构的沟道,沟道横截面的尺寸为100μm×50μm。利用5-氨基甲基荧光素作为荧光探针,使用乙腈溶液与磷酸盐缓冲液作为流动相进行高效液相色谱分析,等量洗脱确认了丙酸到十八烷酸(C3~C18)的衍生物色谱图,并通过高效毛细管电泳分析将丙酸(C3)、己酸(C6)、癸酸(C10)进行分离,成功分离C3和C10,C3和C10的分离度为2.2,分离效果良好。此外,针对高效毛细管电泳分析出现的不稳定电渗现象而导致实验数据不稳定的情况,加入阳离子聚合物六甲溴铵和阴离子聚合物硫酸葡聚糖钠盐形成连续的多个离子聚合物涂层,使得微流控芯片对羧酸检测的稳定性大大提高。 展开更多

Chiral separation of dansyl amino acids by capillary electrophoresis using mixed selectors of Mn(II)- L -alanine complex and β -cyclodextrin ( β -CD) was studied. Resolution was considerably superior to that obtained by using either Mn(II)- L -alanine complex or β -CD alone. The effects of separation parameters,such as pH value of buffer solution,capillary temperature,the concentration of Mn(II)- L -alanine complex,the types of CD and ligand on the migration times and resolutions were investigated. Six different transition metal complexes, Cu(II), Zn(II),Co(II),Ni(II),Hg(II) and Cd(II)- L -alanine complexes have been employed and compared with Mn(II) complex. Differences in retention and selectivity were found. The substitution of Cu(II),Zn(II),Co(II) and Ni(II) for Mn(II) resulted in a better chiral resolution while Hg(II) and Cd(II) showed poorer resolution abilities.
