In this paper, using the quasi-3D coupled current method, the influences of structure of cold crucible, the power frequency, the electricity property of melt, the coil position and current on the electromagnetic field (EMF) and the levitation characteristics in the melting processes are analyzed. It is shown that in the processes of levitation melting with cold crucible, the power frequency and cold crucible structure are the decisive factors for the ability of magnetic flux penetrating into cold crucible. The magnetic flux density in cold crucible is reduced as the increasing of power frequency, and this tendency becomes stronger when the power frequency is higher than 100kHz. The segmented structure of cold crucible can reduce the induction eddy in itself effectively, and the higher the power frequency is, the better the result is. So, a cold crucible can be segmented into 16-20 sectors for high frequency electromagnetic field and/or 8-12 sectors for lower frequency one. It is also shown that the levitation force of melting charge is related to coil current as a parabolic function. 展开更多

Herein, a multi-index analysis of the nickel content of an alloy, output rate of the alloy, nickel recovery rate, and iron recovery rate during the melting of laterite metallized pellets was performed. The thermodynamic reduction behavior of oxides such as NiO, FeO, Fe_3 O_4, and Cr_2 O_3 was studied using the FactSage software, which revealed that SiO_2 is not conducive to the reduction of iron oxides, whereas the addition of basic oxides such as CaO and MgO is beneficial for the reduction of iron oxides. On the basis of a comprehensive analysis to achieve greater nickel recovery and lower iron recovery rates, the optimum experimental parameters in the orthogonal experiment were A3 B1 C3(t = 30 min, C/O = 0.4, R = 1.2); the indicators wNi, φalloy, ηNi, and ηFe had values of 15.0 wt%, 12.1%, 44.9%, and 96.4%, respectively. In single-factor experiments, increasing basicity(R) substantially improved the separation effect in the low-basicity range 0.5 ≤ R ≤ 0.8 but not in the high-basicity range 0.8 ≤ R ≤ 1.2. Similar results were obtained for the effect of the C/O ratio. Moreover, the recovery rate of nickel increased with increasing recovery rate of iron. 展开更多

High Resolution Melting Curve Analysis for Rapid Detection of Pyrazinamide Resistance in Mycobacterium tuberculosis Clinical Isolates

Background: Pyrazinamide (PZA) is one of the most important drugs for tuberculosis (TB) treatment, however, its susceptibility is not routinely tested. High-resolution melting (HRM) curve analysis has been widely used for many applications. In this study, HRM assay was developed and evaluated for the detection of PZA resistance in Mycobacterium tuberculosis clinical isolates. Methods: Ninety five M. tuberculosis clinical isolates with different susceptibility patterns to anti-TB drugs were used to evaluate this assay. Isolates were phenotypically (Bactec MGIT 960) and genotypically (HRM and pncA gene sequencing) analysed for PZA resistance. Results: Bactec MGIT 960 analysis revealed that 29 of the 95 M. tuberculosis isolates were PZA resistant. In comparison to the Bactec MGIT 960, HRM showed a sensitivity of 47.7% and specificity of 74.6%, and the overall agreement between the two methods was 68.4%. Based on DNA sequencing, a correlation of 0.67 (significant at p-value pncA mutations was observed. PZA resistance was strongly associated with multi-drug resistant (MDR)-TB as it was shown in 79.3% of the MDR isolates included in the study. Conclusion: HRM is simple and useful for screening clinical M. tuberculosis isolates for PZA resistance, however, further modifications to improve its performance are required. 展开更多

The Ross-Amundsen sector is experiencing an accelerating warming trend and a more intensive advective influx of marine air streams.As a result,massive surface melting events of the ice shelf are occurring more frequently,which puts the West Antarctica Ice Sheet at greater risk of degradation.This study shows the connection between surface melting and the prominent intrusion of warm and humid air flows from lower latitudes.By applying the Climate Feedback-Response Analysis Method(CFRAM),the temporal surge of the downward longwave(LW)fluxes over the surface of the Ross Ice Shelf(RIS)and adjacent regions are identified for four historically massive RIS surface melting events.The melting events are decomposed to identify which physical mechanisms are the main contributors.We found that intrusions of warm and humid airflow from lower latitudes are conducive to warm air temperature and water vapor anomalies,as well as cloud development.These changes exert a combined impact on the abnormal enhancement of the downward LW surface radiative fluxes,significantly contributing to surface warming and the resultant massive melting of ice. 展开更多

The melt onset dates(MOD)over Arctic sea ice plays an important role in the seasonal cycle of sea ice surface properties,which impacts Arctic surface solar radiation absorbed by the ice-ocean system.Monitoring interannual variations in MOD is valuable for understanding climate change.In this study,we investigated the spatio-temporal variability of MOD over Arctic sea ice and 14 Arctic sub-regions in the period of 1979 to 2017 from passive microwave satellite data.A set of mathematical and statistical methods,including the Sen’s slope and Mann-Kendall mutation tests,were used to comprehensively assess the variation trend and abrupt points of MOD during the past 39 years for different Arctic sub-regions.Additionally,the correlation between Arctic Oscillation(AO)and MOD was analyzed.The results indicate that:(1)all Arctic sub-regions show a trend toward earlier MOD except the Bering Sea and St.Lawrence Gulf.The East Siberian Sea exhibits a significantly earlier trend,with the highest rate of-9.45 d/decade;(2)the temporal variability and statistical significance of MOD trend exhibit large interannual differences with different time windows for most regions in the Arctic;(3)during the past 39 years,the MOD changed abruptly in different years for different sub-regions;(4)the seasonal AO has more influence on MOD than monthly AO.The findings in this study can improve our knowledge of MOD changes and are beneficial for further Arctic climate change study. 展开更多

Food allergies represent a clear threat to the general health and wellbeing of those affected which place increasing pressure on food producers and regulatory authorities. Current analytical techniques typically find difficulties distinguishing between closely related Prunus species which include almond (Prunus dulcis), an EU listed allergenic species. This study describes a proof of principle real-time PCR approach utilising DNA melt analyses that targets the internal transcribed spacer (ITS) sequence to differentiate between a panel of Prunus test species. The method was successfully applied to the characterisation of a commercial paprika sample suspected of having being adulterated with almond, referred to the UK Government Chemist in 2015 in its advisory capacity. Subject to further validation work, the method appears to specifically amplify Prunus species and is capable of discrimination based on the resultant melt profiles. The developed method provides analysts with a simple and broad molecular tool to identify common Prunus species for food authenticity and allergen testing purposes. Initial development work demonstrates a promising approach with the potential to improve discrimination between Prunus species not easily resolved by routine analytical methods. 展开更多

In this work,the evolution of melt pool under single-point and single-line printing in the laser engineered net shaping(LENS)process is analyzed.Firstly,the basic structure of the melt pool model of the LENS process is established and the necessary assumptions are made.Then,the establishment process of the multi-physical field model of the melt pool is introduced in detail.It is concluded that the simulation model results are highly consistent with the online measurement experiment results in terms of melt pool profile,space temperature gradient,and time temperature gradient.Meanwhile,some parameters,such as the 3D morphology and surface fluid field of the melt pool,which are not obtained in the online measurement experiment,are analyzed.Finally,the influence of changing the scanning speed on the profile,peak temperature,and temperature gradient of the single-line melt pool is also analyzed,and the following conclusions are obtained:With the increase in scanning speed,the profile of the melt pool gradually becomes slender;The relationship between peak temperature and scanning speed is approximately linear in a certain speed range;The space temperature gradient at the tail of the melt pool under different scanning speeds hardly changes with the scanning speed,and the time temperature gradient at the tail of the melt pool is in direct proportion to the scanning speed. 展开更多

As a type of metamaterial, pentamode materials (PMs) approximate the elastic properties of liquids. In this study, a finite-element analysis was conducted to predict the mechanical properties of PM structures by altering the thin wall thicknesses and layer numbers to obtain an outstanding load-bearing capacity. It was found that as the thin wall thickness increased from 0.15 to 0.45 mm, the compressive modulus of the PM structures increased and the Poisson’s ratio decreased. As the layer number increased, the Poisson’s ratio of the PM structures increased rapidly and reaches a stable value ranging from 0.50 to 0.55. Simulation results of the stress distribution in the PM structures confirmed that stress concentrations exist at the junctions of the thin walls and weights. For validation, Ti–6Al–4V specimens were fabricated by selective laser melting (SLM), and the mechanical properties of these specimens (i.e., Poisson’s ratio and elastic modulus) were experimentally studied. Good consistency was achieved between the numerical and experimental results. This work is beneficial for the design and development of PM structures with simultaneous load-bearing capacity and pentamodal properties. 展开更多

AIM: To investigate the metabolic profiles of xenograft pancreatic cancer before and after radiotherapy by high-resolution magic angle spinning proton magnetic resonance spectroscopy (HRMAS 1H NMR) combined with principal components analysis (PCA) and evaluate the radiotherapeutic effect. METHODS: The nude mouse xenograft model of human pancreatic cancer was established by injecting human pancreatic cancer cell SW1990 subcutaneously into the nude mice. When the tumors volume reached 800 mm3 , the mice received various radiation doses. Two weeks later, tumor tissue sections were prepared for running the NMR measurements. 1H NMR and PCA were used to determine the changes in the metabolic profiles of tumor tissues after radiotherapy. Metabolic profiles of normal pancreas, pancreatic tumor tissues, and radiationtreated pancreatic tumor tissues were compared. RESULTS: Compared with 1H NMR spectra of the normal nude mouse pancreas, the levels of choline, taurine, alanine, isoleucine, leucine, valine, lactate, and glutamic acid of the pancreatic cancer group were increased, whereas an opposite trend for phosphocholine, glycerophosphocholine, and betaine was observed. The ratio of phosphocholine to creatine, and glycerophosphocholine to creatine showed noticeable decrease in the pancreatic cancer group. After further evaluation of the tissue metabolic profile after treatment with three different radiation doses, no significant change in metabolites was observed in the 1H NMR spectra, while the inhibition of tumor growth was in proportion to the radiation doses. However, PCA results showed that the levels of choline and betaine were decreased with the increased radiation dose, and conversely, the level of acetic acid was dramatically increased. CONCLUSION: The combined methods were demonstrated to have the potential for allowing early diagnosis and assessment of pancreatic cancer response to radiotherapy. 展开更多

Cancer growth in the bone due to its random shape disables bone strength and thus changes its capacity to support body weight or muscles,which can crucially affect the quality of human life in terms of normal walking or daily activities.For successful patient recovery,it is necessary to remove the cancer-affected minimal bone area and quickly replace it with a biocompatible metal implant within less than 2 weeks.An electron beam-melted Ti-6Al-4V implant was designed and applied in a patient to preserve the natural knee joint close to the bone tumor.The developed implant fits the bone defect well,and the independent ambulatory function of the natural knee joint was restored in the patient within six weeks after surgery.A delayed fracture occurred six months after the successful replacement of cancer-affected bone with Ti-6Al-4V implant at the proximal meshed junction of the implant because of a minor downward slip.Microstructural,mechanical,and computational analyses were conducted for the fractured area to find the main reason for the delayed fracture.Our findings pertaining to the mechanical and material investigation can help realize the safe implantation of the three-dimensionally printed titanium implant to preserve the natural joints of patients with massive bone defects of the extremities. 展开更多

Re-melting process has been utilized to mitigate the residual stress level in the selective laser melting(SLM)process in recent years.However,the complex consolidation mechanism of powder and the different material behavior after the first laser melting hinder the direct implementation of the re-melting process.In this work,the effects of re-melting on the temperature and residual stress evolution in the SLM process are investigated using a thermo-mechanically coupled finite element model.The degree of consolidation is incorporated in the energy balance equation based on the thermodynamically-consistent phase-field approach.The drastic change of material properties due to the variation of temperature and material state is also considered.Using the proposed simulation framework,the single-track scanning is simulated first to predict the melt pool dimension and validate the proposed model with the existing experimental data.The obtained thermal histories reveal that the highest cooling rate is observed at the end of the local solidification time which acts as an important indicator for the alleviation of temperature gradient.Then,the scanning of a whole single layer that consists of multiple tracks is simulated to observe the stress evolution with several re-melting processes.After the full melting of powder material in the first scanning process,the increase of residual stress level is observed with one remelting cycle.Moreover,the predicted stress level with the re-melting process shows the variation trend attributable to the accumulated heat in the tracks.The numerical issues and the detailed implementation process are also introduced in this paper. 展开更多

Objective: To detect and identify filarial parasites in dried blood spots(DBS) collected from domestic cats using high resolution melting real-time PCR(HRM RT-PCR). Methods: A total of 208 DBS were collected from domestic cats in a brugian filariasis endemic areas in Surat Thani Province, southern Thailand. Microfilariae were found in 9 blood slides using Giemsa-stained thick blood film. The extracted DNA from blood spot volumes of 10 and 20 μL DBS with positive filarial parasites in cats were performed using HRM RT-PCR method. The primers were designed based on the partial mitochondrial 12S rRNA gene for identifying Brugia malayi, Brugia pahangi, Dirofilaria immitis. All purified samples were then detected. Results: Using different volumes of 10 μL and 20 μL DBS could easily distinguish filarial parasites and showed similar results. PCR amplicons of Brugia malayi, Brugia pahangi and Dirofilaria immitis were determined at melting peak(temperature) of 75.70℃, 77.46 ℃, and 73.56 ℃, respectively. All 9 positive DBS samples showed positive Brugia pahangi and similar nucleotide sequences. Conclusions: This HRM RT-PCR method is able to diagnose, identify and discriminate filarial parasites collected from DBS, which is simple and inexpensive compared with other probe-based genotyping methods. Furthermore, this method is useful to survey, prevent and control filariasis. 展开更多

Discrimination among grapevine varieties based on quantitative traits,such as flowering,veraison and ripening dates is crucial for variety selection in the context of climate change and in breeding programs.These traits are under complex genetic control for which 6 linked SSR loci(VVS2,VVIn16,VMC7G3,VrZAG29,VMC5G7,and VVIB23)have been identified.Using these markers in HRM-PCR analysis,we assessed genetic diversity among a large collection of 192 grapevine varieties.The grapevine germplasm used encompasses the majority of Greek vineyard with 181 varieties,3 prominent foreign varieties and 11 varieties of Palestinian origin.The SSR markers used were highly polymorphic,displaying unique melting curves for unusually higher number of samples than generally observed in SSR analysis.This prompted us to examine sequence composition for selected samples and found that variation present as SNPs in the flanking sequences of SSR motifs was responsible for the observed polymorphism.Hence,HRM-PCR proved to be a tool of higher analytical power to distinguish genotypes surpassing the discrimination power of conventional gel-based SSR analysis.The study provides a better understanding of genetic variation of SSR marker loci associated to phenological traits in grapevine varieties,signifying an analytical methodology that may be of higher discrimination power in detection of polymorphism for utilization in breeding programs. 展开更多

Aiming at dealing with the problem in extraction of dominant tree species,we tested the highly hot method-object-based analysis. Based on the ALOS image data,we combined multi-resolution in e Cognition software and fuzzy classification algorithm. Through analyzing the segmentation results,we basically extract the spruce,the pine,the birch and the oak of the study area. Both the spectral and spatial characteristics were derived from those objects,and with the help of GLCM,we got the differences of each species. Automatic road detection, in dense urban areas, is a challenging application in the remote sensing community. This is mainly because of physical and geometrical variations of road pixels, their spectral similarity to other features such as buildings, parking lots and sidewalks, and the obstruction by vehicles and trees. These problems are real obstacles in precise detection and identification of urban roads from high-resolution satellite imagery. One of the promising strategies to deal with this problem is using multi-sensors data to reduce the uncertainties of detection. In this paper, an integrated object-based analysis framework was developed for detecting and extracting various types of urban roads from high-resolution optical images and Lidar data. The proposed method is designed and implemented using a rule-oriented approach based on a masking strategy. The overall accuracy (OA) of the final road map was 89.2%, and the kappa coefficient of agreement was 0.83, which show the efficiency and performance of the method in different conditions and interclass noises. The results also demonstrate the high capability of this object-based method in simultaneous identification of a wide variety of road elements in complex urban areas using both high-resolution satellite images and Lidar data. 展开更多

The melting phenomenon in two-dimensional(2 D)flow of fourth-grade material over a stretching surface is explored.The flow is created via a stretching surface.A Darcy-Forchheimer(D-F)porous medium is considered in the flow field.The heat transport is examined with the existence of the Cattaneo-Christov(C-C)heat flux.The fourth-grade material is electrically conducting subject to an applied magnetic field.The governing partial differential equations(PDEs)are reduced into ordinary differential equations(ODEs)by appropriate transformations.The solutions are constructed analytically through the optimal homotopy analysis method(OHAM).The fluid velocity,temperature,and skin friction are examined under the effects of various involved parameters.The fluid velocity increases with higher material parameters and velocity ratio parameter while decreases with higher magnetic parameter,porosity parameter,and Forchheimer number.The fluid temperature is reduced with higher melting parameter while boosts against higher Prandtl number,magnetic parameter,and thermal relaxation parameter.Furthermore,the skin friction coefficient decreases against higher melting and velocity ratio parameters while increases against higher material parameters,thermal relaxation parameter,and Forchheimer number. 展开更多

Machine learning methods, one type of methods used in artificial intelligence, are now widely used to analyze two-dimensional (2D) images in various fields. In these analyses, estimating the boundary between two regions is basic but important. If the model contains stochastic factors such as random observation errors, determining the boundary is not easy. When the probability distributions are mis-specified, ordinal methods such as probit and logit maximum likelihood estimators (MLE) have large biases. The grouping estimator is a semiparametric estimator based on the grouping of data that does not require specific probability distributions. For 2D images, the grouping is simple. Monte Carlo experiments show that the grouping estimator clearly improves the probit MLE in many cases. The grouping estimator essentially makes the resolution density lower, and the present findings imply that methods using low-resolution image analyses might not be the proper ones in high-density image analyses. It is necessary to combine and compare the results of high- and low-resolution image analyses. The grouping estimator may provide theoretical justifications for such analysis. 展开更多
