Increasing root-lower characteristics improves drought tolerance in cotton cultivars at the seedling stage

Drought is an important abiotic stress factor in cotton production.The root system architecture(RSA)of cotton shows high plasticity which can alleviate drought-related stress under drought stress(DS)conditions;however,this alleviation is cultivar dependent.Therefore,this study estimated the genetic variability of RSA in cotton under DS.Using the paper-based growth system,we assessed the RSA variability in 80 cotton cultivars at the seedling stage,with 0 and10%polyethylene glycol 6000(PEG6000)as the control(CK)and DS treatment,respectively.An analysis of 23 aboveground and root traits in the 80 cotton cultivars revealed different responses to DS.On the 10th day after DS treatment,the degree of variation in the RSA traits under DS(5–55%)was greater than that of CK(5–49%).The 80 cultivars were divided into drought-tolerant cultivars(group 1),intermediate drought-tolerant cultivars(group 2),and drought-sensitive cultivars(group 3)based on their comprehensive evaluation values of drought resistance.Under DS,the root lengthlower,root area-lower,root volume-lower,and root length density-lower were significantly reduced by 63,71,76,and 4%in the drought-sensitive cultivars compared to CK.Notably,the drought-tolerant cultivars maintained their root lengthlower,root area-lower,root volume-lower,and root length density–lower attributes.Compared to CK,the root diameter(0–2 mm)-lower increased by 21%in group 1 but decreased by 3 and 64%in groups 2 and 3,respectively,under DS.Additionally,the drought-tolerant cultivars displayed a plastic response under DS that was characterized by an increase in the root-lower characteristics.Drought resistance was positively correlated with the root area-lower and root length density-lower.Overall,the RSA of the different cotton cultivars varied greatly under DS.Therefore,important root traits,such as the root-lower traits,provide great insights for exploring whether drought-tolerant cotton cultivars can effectively withstand adverse environments. 展开更多

Sorghum(Sorghum bicolor(L.)Moench)is a world cereal crop used in China for producing Baijiu,a distilled spirit.We report a telomere-to-telomere genome assembly of the Baijiu cultivar Hongyingzi,HYZ-T2T,using ultralong reads.The 10 chromosome pairs contained 33,462 genes,of which 93%were functionally annotated.The 20 telomeres and 10 centromeric regions on the HYZ-T2T chromosomes were predicted and two consecutive large inversions on chromosome 2 were characterized.A 65-gene reconstruction of the metabolic pathway of tannins,the flavor substances in Baijiu,was performed and may advance the breeding of sorghum cultivars for Baijiu production. 展开更多

The continued expansion of the world population,increasingly inconsistent climate and shrinking agricultural resources present major challenges to crop breeding.Fortunately,the increasing ability to discover and manipulate genes creates new opportunities to develop more productive and resilient cultivars.Many genes have been described in papers as being beneficial for yield increase.However,few of them have been translated into increased yield on farms.In contrast,commercial breeders are facing gene decidophobia,i.e.,puzzled about which gene to choose for breeding among the many identified,a huge chasm between gene discovery and cultivar innovation.The purpose of this paper is to draw attention to the shortfalls in current gene discovery research and to emphasise the need to align with cultivar innovation.The methodology dictates that genetic studies not only focus on gene discovery but also pay good attention to the genetic backgrounds,experimental validation in relevant environments,appropriate crop management,and data reusability.The close of the gaps should accelerate the application of molecular study in breeding and contribute to future global food security. 展开更多

Studying on the genetic diversity and genetic relationship of flowering cherry cultivars is extremely important for germplasm conservation, cultivar identification and breeding. Flowering cherry is widely cultivated as an important woody ornamental plant in worldwide, especially Japan, China. However, owning to the morphological similarity, many cultivars are distinguished hardly in non-flowering season. Here, we evaluated the genetic diversity and genetic relationship of 40 flowering cherry cultivars, which are mainly cultivated in China. We selected 13 polymorphicprimers to amplify to allele fragments with fluorescent-labeled capillary electrophoresis technology. The population structure analysis results show that these cultivars could be divided into 4 subpopulations. At the population level, N<sub>a</sub> and N<sub>e</sub> were 6.062, 4.326, respectively. H<sub>o</sub> and H<sub>e</sub> were 0.458 and 0.670, respectively. The Shannon’s information index (I) was 1.417. The Pop3, which originated from P. serrulata, had the highest H<sub>o</sub>, H<sub>e</sub>, and I among the 4 subpopulations. AMOVA showed that only 4% of genetic variation came from populations, the 39% variation came from individuals and 57% (p < 0.05) came from intra-individuals. 5 polymorphic SSR primers were selected to construct molecular ID code system of these cultivars. This analysis on the genetic diversity and relationship of the 40 flowering cherry cultivars will help to insight into the genetic background, relationship of these flowering cherry cultivars and promote to identify similar cultivars. 展开更多

Capsicum is a nutritious vegetable and its cultivation in farms is getting popular in Bangladesh. Although many efforts have lain to explore better yielding and nutritionally rich cultivars with suitable modern cultivation techniques but still have to find the desired outcome. Thus, it’s necessary to conduct further research to identify the high-yielding and nutritious capsicum cultivars in Bangladesh. An experiment was conducted from July 2021 to June 2022 at the Bangladesh Institute of Research and Training on Applied Nutrition (BIRTAN) research field with three cultivars of capsicum: B<sub>0</sub> = California Wonder, B<sub>1</sub> = BARI Misti Morich-1 and B<sub>2</sub> = BARI Misti Morich-2 and three mulching: T<sub>0</sub> = No mulching, T<sub>1</sub> = Water hyacinth, T<sub>2</sub> = Poly Mulching in randomized complete block design with three replications to identify better quality capsicum cultivar and suitable mulching material. Among cultivars the BARI Misti Morich-2 (B<sub>2</sub>) showed increased agronomic parameters like number of branches and effective branches per plant, leaves length and width, consequently yield and yield contributing traits were also enhanced like fruits per plant, fruit length, fruit diameter and yield per plant (25.97%, 4.54%, 3.64% and 21.43%, respectively). Poly Mulching (T<sub>2</sub>) increased agronomic traits, yield traits and yield (0.61 kg) than BARI Misti Morich-1 (T<sub>1</sub>). The combined effect of B<sub>2</sub>T<sub>2</sub> increased the number of branches per plant, effective branches per plant, leaves length and breadth by 40%, 90%, 15.57% and 26.22%, respectively, hence resulting in an increased yield of 20%. BARI Misti Morich-2 cultivar showed an increase in Fe, Zn and Vitamin-C content of 26.24% and 23.10%, 8.82% and 5.14%, and 6.03% and 5.74% than B0 and B1 cultivars, respectively. Therefore, BARI Misti Morich-2 exhibited the improved agronomic, yield and nutritional traits of capsicum under poly mulching among other cultivars in Bangladesh. 展开更多

Effect of N Fertilization on Yield, N Absorption and Utilization of Two Species of Super High-Yielding Summer Maize 被引量:6

[Objective] The aim was to study on effects of N fertilizer on yield, N absorption and utilization of different cultivars of super high-yielding summer maize, in order to provide reference for reasonable N fertilization in accordance with different cultivars. [Method] Field experiment was conducted to study on effects of different N fertilizers on yield, N absorption and use efficiency of Zhengdan 958 and Xundan 20, in order to learn the effect differences at different N fertilizer levels. [Result] After N was applied, yields of the two summer maize increased significantly. Zhengdan 958 achieved the highest in yield and proceeds at 12 051.18 kg/hm2 and 1 722.40 yuan/hm2, respectively in low N level. In contrast, Xundan 20 achieved the highest at 13 166.00 kg/hm2 and 1 343.92 yuan/hm2 in the above two aspects in high N level. Compared with Zhengdan 958, Xundan 20 increased by 9.90%, 5.20% and 12.00% in N levels of 0, 240, and 450 kg/hm2, respectively. When N fertilizers were applied, protein yield of Xundan 20 was significantly higher than that of Zhengdan 958, so that higher N fertilizers contributed higher protein yield for Xundan 20. In high N level, N efficiency, N-fertilizer utilization and partial productivity of Xundan 20 were significantly higher than that of Zhengdan 958. [Conclusion] Lower N-fertilizer was suitable for Zhengdan 958 and Xundan 20 would get a good harvest if more N-fertilizers were applied. [Result] Rice increased by 596.4-809.55 kg/hm2, labor saving reached 120-135 per hm2, and profit improved in the range from 5 521.39 to 8 727.48 yuan/hm2 in the treatment groups by combinations of shallow rotary tillage, mechanical transplanting, and controlled-release fertilizer, of shallow rotary tillage, mechanical transplanting, and quantitative fertilization, and of deep ploughing, mechanical transplanting, and controlled release fertilizer. [Method] Key technologies affecting the seed production of japonica hybrid rice were investigated based on manual transplanting experiment, mechanical transplanting experiment, paclobutrazol chemical control experiment, leaf-clipping experiment, gibberellic acid treatment experiment and experiment of different row ratios of male and female parents. [Result] The duration from seeding to heading was significantly shortened with sowing date postponing. The sowing date of mechanical-transplanting parents should be 3 -4 d earlier than that of manual-transplanting parents. Spraying paclobutrazol had significantly control effect on plant height of rice seedlings, and the spraying dose should be determined based on the sensitivities of different parents to paclobutrazol, with an appropriate range of 600-900 g/hm2 . Leaf clipping had certain improving effect on the seed-setting rate. The appropriate spraying dose of gibberellin for japonica hybrid rice was 75-90 g/hm2 . The yield would be relatively high when row ratio of male and female parent was 6∶2. [Conclusion] This study laid the foundation for improving the yield of produced japonica hybrid rice seeds. 展开更多

Rice culm carbohydrate transport can simultaneously affect grain filling and stem lodging resistance by regulating non-structural carbohydrate(NSC) and structural carbohydrate(SC) contents. However, the relationship between carbohydrate transposition and culm strength is not well documented. Accordingly, a high-yielding hybrid rice cultivar(Y Liangyou 2) was tested under different N fertilization regimes at two locations, Taoyuan(a special high-yield eco-site), Yunnan province and Danyang(a representative eco-site of the middle and lower Yangtze), Jiangsu province, China. Significantly higher grain yield and basal stem strength were found at Taoyuan than Danyang under all N rates throughout the two-year experiment. At heading stage, soluble sugars, starch, cellulose and lignin contents of the basal culm at Taoyuan were significantly 132.0%, 73.7%, 1.2%, and 62.7% higher than those at Danyang, respectively. At 20 days after heading, soluble sugars and starch content at Taoyuan decreased significantly compared to Danyang, but lignin content remained higher. Culm carbohydrate transport to kernels at Taoyuan was significantly greater than that at Danyang, and the proportion of soluble sugars and starch was correspondingly 62.9%lower. However, the proportion of lignin and cellulose was 22.7% higher at Taoyuan than that at Danyang. Soluble sugars and starch partitioning were significantly reduced under an increased nitrogen application rate, but SC partitioning was little affected. There were significant positive correlations between basal culm bending stress and dry weight and cellulose and lignin proportions at both locations under all N rates, suggesting that the higher SC proportion at 20 days after heading was primarily responsible for culm strength.These results suggest that high-yielding rice populations with greater culm strength require both moderate NSC transport and greater SC accumulation. 展开更多

The morphological characters of each population were investigated at the heading stage, and the data were analyzed by using ANOVY and other statistic methods. The plant ideal morphological characters at the heading stage were established as follows: 1 ) for the early-season cropping, 90-105 cm plant height; 11-12 tillers per plant; 35-40 em length and 2.1-2.2 cm width of flag leaf; 46-50 cm length and 1.8-2.1 cm width of the second leaf from the top (L2); 59-64 cm length and 1.4-1.9 cm width of the third leaf from the top (L3); 7°-14°, 18° and 200-33° for the ideal leaf angles of the flag leaf, L2 and L3, respectively; 2) for the late-season cropping, 90-100 cm plant height; 9-15 tillers per plant; 30-41 cm length and 1.8-2.0 cm width of flag leaf; 53-61 cm length and 1.3-1.8 cm width of L2; 52-58 cm length and 1.2-1.5 cm width of L3; 9°-19°, 15°-37° and 16°-49° for the ideal leaf angles of the flag leaf, L2 and L3, respectively. Super high-yielding soybean cultivar Liaodou 14, soybean cultivars from Ohio in the United States, and the common soybean cultivars from Liaoning Province, China, with similar geographic latitudes and identical pod-bearing habits were used as the study materials for a comparison of morphological traits and production characteristics to provide a theoretical basis for the breeding of improved super high-yielding soybean cultivars. Using a randomized block design, different soybean cultivars from the same latitude were compared under conventional and unconventional treatments for their production characteristics, including morphological traits, leaf area index (LAI), net photosynthesis rate, and dry matter accumulation. The specific characteristics of the super high-yielding soybean cultivar Liaodou 14 were analyzed. The results showed that the plant height of Liaodou 14 was significantly lower than that of the common cultivars from Liaoning, whereas the number of its main-stem nodes was higher than that of the cultivars from Ohio or Liaoning. A high pod density was observed in Liaodou 14 under conventional treatments. Under both conventional and unconventional treatments, the branch number of Liaodou 14 was markedly higher than that of the common cultivars from Liaoning, and its branch length and leaf inclination angle were significantly higher than those of common cultivars from Liaoning or Ohio. Only small changes in the leaf inclination angle were observed in Liaodou 14 treated with conventional or unconventional methods. Under each treatment, Liaodou 14 exhibited the lowest amplitude of reduction in SPAD values and net photosynthesis rates from the grain-filling to ripening stages; the cultivars from Ohio and the common cultivars from Liaoning exhibited more significant reductions. Liaodou 14 reached its peak LAI later than the other cultivars but maintained its LAI at a higher level for a longer duration. Under both conventional and unconventional treatments, Liaodou 14 produced a higher yield than the other two cultivars, with significant differences from the Ohio cultivars. In summary, super high-yielding soybean cultivars have several main features: suitable plant height, high pod density, good leaf structure with strong functionality, and slow leaf senescence at the late reproductive stage, which is conducive to the accumulation of dry matter and improved yield. 展开更多

[Methed] This survey was carried out in two demonstration bases, one for investigating the relationship between different cultivated varieties with growth and production, the other for investigating the relationship between different planting densities with growth and production. [Result] For different cultivated varieties, Cenruan 2 showed tree height, ground diameter and crown product significantly higher than other two varieties, besides, the per unit area fruit yield of Cenruan 2 was also significantly higher than other two varieties. From flower bud differentiation,the best variety was Changlin 4, which showed number of differentiated flower buds significantly higher than Changlin 27, but there were no significant differences between Cenruan 2 with Changlin 4 and Changlin 27. For different planting densities, plant height and crown product at planting density of 1.5 m×2 m were significantly higher than those of other two planting densities, but from ground diam- eter, the best planting density was 1.5 m×1.0 m, with a ground diameter significantly higher than other two planting densities. From per unit area fruit yield, the best planting density was 1.5 m×2.0 m, under which early plant growth and fruit production were better. Therefore, the best planting density is 2.0 m ×3.0 m when constructing early-bearing high-yielding model plantations. For variety Cenruan 3, it's fruit production is high in the early pedod, and the best planting density is 1.5 m× 2.0 m or 1.0 m×1.5 m when constructing early-bearing high-yielding model plantations, and after 4-6 years when it put into production, the plants could be transplanted. 展开更多

In a field experiment, rice cultivar Shanyou 63 was used as a check cultivar to investigate the changes in activity of peroxidase and contents of malondialdehyde and soluble protein after full heading stage in flag leaves of two super high-yielding inbred rice, including Shengtai 1 and Yuxiangyouzhan. The results showed that the peroxidase activities and soluble protein contents in the leaves of the three cultivars tended to increase gradually to peak values and thereafter descended during stage after full heading. Moreover, both the peroxidase activities and soluble protein contents of Shengtai 1 and Yuxiangyouzhan peaked at 8 days after full heading, which was compared with that of Shanyou 63 at 4 days later. Meanwhile, the malondialdehyde contents in the leaves of the three cultivars increased gradually after full heading stage. In addition, activities of peroxidase and contents of malondialdehyde and soluble protein in different cultivars were various at the same time. In general, for Shengtai 1 and Yuxiangyouzhan, the peroxidase activities and soluble protein contents were higher than those of Shanyou 63, whereas the malondialdehyde contents were lower compared with that of Shanyou 63. And these features could make the senescence of leaf become later and slower and photosynthetic product was more in Shengtai 1 and Yuxiangyouzhan than that in Shanyou 63. 展开更多

[Result] The performance of ear, grain and weight in Zhongzheyou No.8 showed significant difference among yield levels. The enhancement of effective ears would restrain the number of total grains per ear. It was found that the direct contributions of main economic traits to yield were: number of fil ed grains per ear 〉 number of effective ears 〉1 000-grain weight 〉number of total grains per ear. [Conclusion] The promotion of high-yielding techniques was suggested as fol ows: on the basis of certain number of effective ears, number of fil ed grains per ear should be mainly focused with a consideration to grain weight. 展开更多

Commercial production of spinach(Spinacia oleracea L.)is centered in California and Arizona in the US,where downy mildew caused by Peronospora effusa is the most destructive disease.Nineteen typical races of P.effusa have been reported to infect spinach,with 16 identified after 1990.The regular appearance of new pathogen races breaks the resistance gene introgressed in spinach.We attempted to map and delineate the RPF2 locus at a finer resolution,identify linked single nucleotide polymorphism(SNP)markers,and report candidate downy mildew resistance(R)genes.Progeny populations segregating for RPF2 locus derived from resistant differential cultivar Lazio were infected using race 5 of P.effusa and were used to study for genetic transmission and mapping analysis in this study.Association analysis performed with low coverage whole genome resequencing-generated SNP markers mapped the RPF2 locus between 0.47 to 1.46 Mb of chromosome 3 with peak SNP(Chr3_1,221,009)showing a LOD value of 61.6 in the GLM model in TASSEL,which was within 1.08 Kb from Spo12821,a gene that encodes CC-NBS-LRR plant disease resistance protein.In addition,a combined analysis of progeny panels of Lazio and Whale segregating for RPF2 and RPF3 loci delineated the resistance section in chromosome 3 between 1.18–1.23 and 1.75–1.76 Mb.This study provides valuable information on the RPF2 resistance region in the spinach cultivar Lazio compared to RPF3 loci in the cultivar Whale.The RPF2 and RPF3 specific SNP markers,plus the resistant genes reported here,could add value to breeding efforts to develop downy mildew resistant cultivars in the future. 展开更多

Background Under K deficiency the uptake and distribution pattern in plant cells is mediated through different transport proteins and channels which were controlled by specific gene family.Therefore,a hydroponic experiment was conducted under control condition for testing the gene expression pattern of the K transporter under adequate and low K supply levels.After that,a 2-year field experiment was conducted to evaluate five selected cotton cultivars(four K-efficient cultivars,viz.,CIM-554,CYTO-124,FH-142,IUB-2013,and one K non-efficient,BH-212) screened from the initial hydroponics culture experiment and two levels of potassium(0 K_(2)O kg·ha^(-1) and 50 K_(2)O kg·ha^(-1)) were tested under reduced irrigation(50% available water content;50 AWC) and normal irrigation conditions(100% available water content;100 AWC).Result Results revealed that the transcript levels of GhHAK5aD in roots were significantly higher in K^(+) efficient cultivars than that in K^(+) non-efficient cultivars.The GhHAK5aD expression upon K^(+) deficiency was higher in roots but lower in shoots,indicating that GhHAK5aD could have a role in K^(+) uptake in roots,instead of transport of K^(+) from root to shoot.Similarly,under field conditions the cultivar FH-142 showed an increase of 22.3%,4.9%,2.4%,and 1.4% as compared with BH-212,IUB-2013,CYTO-124,and CIM-554,respectively,in seed cotton yield(SCY) with K application under reduced irrigation conditions.With applied K,the FH-142 showed an increase in net photosynthetic rate by 57.3% as compared with the rest of the cultivars under reduced irrigation over K control.However,the overall performance indicators of K-efficient cultivars like FH-142,CYTO-124,CIM-554,and IUB-2013 were better than BH-212(K in-efficient) under reduced irrigation conditions with applied K at 50 kg·ha^(-1).Fiber quality trait improved significantly with K application under water deficit.The increase in micronaire was 3.6%,4.7%,7.8%,3.4%,and 6.7% in BH-212,IUB-2013,CIM-554,CYTO-124,and FH-142,respectively,with K application at 50 kg·ha^(-1) over without K application under reduced irrigation conditions during the cotton growing season.Similarly,the cultivars FH-142 increased by 12% with K application under reduced irrigation as compared with other cultivars.The performance of K-efficient cultivars under reduced irrigation conditions was 30% better in SCY and quality traits with the application of K at 50 kg·ha^(-1) as compared with K-non-efficient cultivars.Similarly,water use efficiency(WUE)(40.1%) and potassium use efficiency(KUE)(20.2%) were also noted higher in case of FH-142 as compared with other cultivar with K application under reduced conditions.Conclusion Higher expression of GhHAK5aD gene was observed in K-efficient cultivars as compared with K-nonefficient cultivars in roots indicates that GhHAK5aD may be contributing to genotypic differences for K^(+) efficiency in cotton.K-efficient cotton cultivars can be used for the low-K environments and can also be recommended for general cultivars. 展开更多
