Sea cucumber Holothuria leucospilota is one of the most widespread tropical holothurian species.In this study,eukaryotic organism composition in foregut and hindgut contents of H.leucospilota and surrounding sediments...Sea cucumber Holothuria leucospilota is one of the most widespread tropical holothurian species.In this study,eukaryotic organism composition in foregut and hindgut contents of H.leucospilota and surrounding sediments was assessed by 18S rRNA gene high-throughput sequencing.Eukaryon richness and diversity in the habitat sediments were significantly higher than those in foregut and hindgut contents of the sea cucumbers(P<0.05).The foregut content group,hindgut content group,and marine sediment group sequences were respectively assigned to 18.20±1.32,19.40±1.03,and 21.80±0.37 phyla.In the foregut contents,Nematoda(20.18%±9.59%),Mollusca(16.12%±10.49%),Chlorophyta(10.04%±4.85%),Annelida(8.72%±10.93%),Streptophyta(8.46%±4.65%),and Diatomea(5.99%±2.01%)were the predominant phyla,which showed the eukaryotic food sources of H.leucospilota were primarily belong to the above phyla.The predominant phyla in the hindgut contents were Streptophyta(45.55%±17.32%),Mollusca(4.93%±4.82%),Arthropoda(5.37%±3.08%),Diatomea(3.88%±2.34%),and Chlorophyta(3.79%±1.59%);and Annelida(37.80%±17.00%),Arthropoda(24.49%±12.53%),Platyhelminthes(7.14%±3.02%),Nematoda(4.14%±0.91%),and Diatomea(5.11%±1.35%)had large contents in the sediments.The comparatively high content of Paris genus in phylum Streptophyta in foregut contents indicated that land plants were one of the primary food sources of H.leucospilota,however the significantly higher contents of Streptophyta in hindgut contents than that in foregut contents might suggest a large part of the terrigenous detritus ingested might not be digested by H.leucospilota.UPGMA and PCoA analysis revealed that eukaryotic organism composition differed significantly between foregut contents of H.leucospilota and ambient sediments,indicating selective feeding feature of H.leucospilota.This study provided useful references for artificial feed of tropical sea cucumbers and enhanced understanding of the ecological roles of detritus-feeding macrobenthos.展开更多

Objective:To determine lethal median dose(LD_(50)) and histopalhological toxicity of water extract of Hololhuria atra(H,atra) in mice.Methods:The behavioral changes,mortality and histopathology examination on liver we...Objective:To determine lethal median dose(LD_(50)) and histopalhological toxicity of water extract of Hololhuria atra(H,atra) in mice.Methods:The behavioral changes,mortality and histopathology examination on liver were assessed in mice 14 d after the administration(i.p.) of H.atra water extract.Seven doses 110,20,30,50.100.150 and 200 mg/kg) of H.atra were used.The control group was treated with normal saline,Results:In the acute study in mice,the water extracts of H,atra caused dose-dependent general behavior adverse affects and mortality.The main behavioral sign of toxicity was hypoactivity,noticed immediately after administration of the extract which was more obvious at the higher doses and persisted until death.Mortality increased with increasing doses,the calculated LD_(50)was 41 mg/kg in mice.The liver toxicity was confirmed by histopalhological examination,which indicated the presence of abnormal hepatocytes with a distorted shape and undefined cell lining as well as enlarged nuclei in low doses groups.High doses groups indicated a more prominent distortion of the polyhedral hepatocytes with undefined cell lining,massive cytoplasm,pvknotic.karyorhexis and karyolytie nuclei(necrosis of hepatocytes).Control group showed polyhedral hepatocytes with defined cell lining arranged in cords and normal round nuclei,with granular cytoplasm.Conclusions:Because of the relatively low LD_(50) value in the acute study in mice,it may be concluded that the H.atra water extract is toxic.展开更多
[Objectives]Proteomics methods were used to analyze the total proteome of Holothuria leucospilota in the South China Sea,so as to provide a theoretical basis for elucidating the medicinal material basis of sea cucumbe...[Objectives]Proteomics methods were used to analyze the total proteome of Holothuria leucospilota in the South China Sea,so as to provide a theoretical basis for elucidating the medicinal material basis of sea cucumber proteins and peptides.[Methods]H.leucospilota was collected from the South China Sea.Label-free proteomics methods were used to detect the protein profile of enzymatically hydrolyzed H.leucospilota total protein,and identify sea cucumber protein and peptide sequences,and gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)bioinformatics analysis software were used to analyze significantly enriched proteins.The String database was used to construct a protein-protein interaction network of significantly enriched H.leucospilota proteins(PPI network).[Results]Proteomics identified 1200 sea cucumber proteins and 2967 peptides.GO analysis and KEGG analysis showed that the four significantly-enriched sea cucumber proteins hsc70,COX,CYTB and rbbp4 participated in 8 metabolic pathways,and COX1 and CYTB among them were significantly related to cardiac muscle contraction,oxidative phosphorylation and metabolic pathways.The PPI network analysis showed that the 13 significantly-enriched sea cucumber proteins had close protein-protein interaction relationships.[Conclusions]This study used proteomics to conduct preliminary identification and research on the proteins and peptides of H.leucospilota,providing a scientific basis for the application of H.leucospilota proteins and peptides in the fields of functional health food and drug research and development.展开更多

Sea cucumber Holothuria atra is commonly known as the lollyfish. The antifungal activity was detected using agar well diffusion method against the various fungal strains such as Trichoderma viride, Aspergillus niger, Aspergillus flavis, Candida albicans and Penicillium chrysogenum. Relatively high antifungal activity was seen against Candida albicans at 100 μL-1 concentration of extracts. Zone of inhibition was measured at 18 mm of diameter. The anti-tumor activities were detected against the Vero and Hep2 cell lines using MTT assay. The cells were treated with H. atra extract at concentrations 0.078-10 mg m L-1. The extract showed high proliferative activity against the Hep2 cells. The body wall extracts of sea cucumber(H. atra) showed effective antifungal and antitumor activities. All these findings suggest that the extracts could be used for the development of drugs.展开更多

Results Three triterpene glycosides were identified as scabraside D (1), fuscocineroside C (2), and 24-dehydroechinoside A (3). Their inhibition on P-388, A549, MKN-28, HCT116, and MCF-7 cells were significant. Conclusion Scabraside D (1) is a new triterpene glycoside, and compounds 2 and 3 are isolated from H. scabra for the first time. The glycosides 1-3 show the in vitro cytotoxicity against five human tumor cell lines in comparison to 10-hydroxycamptothecin.展开更多
Objectives:The paper evaluates the benefit and risk for human health associated with consumption of sea cucumber Holothuria poli(H.polil from Italian coasts(Central Mediterranean Sea).Materials and Methods:Body wall(B...Objectives:The paper evaluates the benefit and risk for human health associated with consumption of sea cucumber Holothuria poli(H.polil from Italian coasts(Central Mediterranean Sea).Materials and Methods:Body wall(BW),internal tunic(ITu),muscle bands(MBs),alimentary canal(AC),gonad(Gd),and respiratory tree(RT)of H.polii were analyzed for proximate composition.Moreover,amino acids(AAs),fatty acids(FAs)and polycyclic aromatic hydrocar bons(PAHs)were determined with high-performance liquid chromatography coupled with ultraviolet-visible spectroscopy(UPLC UV/Vis),gas chromatography-flame ionization detector(GC-FID)and gas chromatography mass spectrometry(GC-MS),respectively.Results:Differences in the contents of total amino acids(TAAs)occurred based on tissue and sex,with AC and MB of female and Gd of male showing higher contents(range 47.8-60.2 g/kg we weight(ww)).Glycine and glutamic acid were the most abundant.Polyunsaturated fatty acid(PUFA)was the major class of FAs and arachidonic acid and eicosapentenoic acid(EPA)were the predominant PUFA.n-3 PUFA showed higher content in Gd,AC,and RT,indicating higher quality.A favorable n-3/n-6 in the range of 1.04-1.67 was observed.PAHs showed values ranging from 23 to 207μg/kg ww with the highest levels in Gd and AC tissues and the lower in BW.Benzo[alpyrene,the most toxic compound,was detected in all tissues,of both sexes,at levels of 1.5-18μg/kg ww.Conclusion:All tissues of H.poli,although with differences among them,are valuable food and can contribute for a healthy diet.Excess cancer risk(CR)values for Gd and AC tissues were above the considerable CR threshold of one in 10000 established by the United States Environmental Protection Agency(USEPA)for high ingestion rate of this seafood.展开更多

文摘海参幼苗发育需要经历耳状幼体、樽形幼体、五触手幼体及幼苗阶段,而从浮游幼体变态发育至附着幼苗阶段的高死亡率是热带海参繁育中的共性问题,目前有关热带海参变态发育的调控机制仍不清楚。以玉足海参(Holothuria leucospilota)为研究对象,采集小耳幼体(A)、中耳幼体(B)、大耳幼体(C)和樽形幼体(D)4个时期样品进行高通量转录组测序分析,以探究其变态发育的分子机制。结果显示,共产生83.6 GB Raw reads,拼接获得93528个Unigenes。对4个组测序文库的相邻组间(A_vs_B,B_vs_C,C_vs_D)进行两两比较,A_vs_B、B_vs_C和C_vs_D的差异表达基因数目分别为17732、11757和11319个。GO功能富集显示,差异基因主要富集于分子功能、催化活性等与细胞成长相关的GO功能。此外对KEGG通路进行了分析,结果显示差异基因显著富集于PI3K-Akt、细胞周期、癌症途径等与细胞分化增殖、凋亡相关的信号通路中,其中在幼体由浮游的大耳状幼体变态至附着的樽形幼体过程中,癌症途径的富集频率显著上升,表明其在幼体生长发育模式转换上发挥了关键作用。筛选的差异表达基因及预测的功能信息可为热带海参生长发育调控机制研究、人工繁育及分子改良应用提供参考。
