We noted the following important observations regarding the adhesive ability between different Lactobacillus strains and three cell types: the adhesive interaction between the SDnB7, SDnE1 and SDnA3 lactobacillus strains and CaCo-2cells was greater compared to controls, the adhesive effect between SDnB1 and CEF cells was also greater than controls and Lactobacillus showed only minimal adherence to MDCK cells. Incubation time also affected Lactobacillus adherence to CaCo-2cells: adhesive ability was optimal at 37°C when incubated for 2 days and this was confirmed by a local increase in the concentration of Lactobacillus around CaCo-2 cells when incubated for 24 h as opposed to 3 h. Adherence ability in lactobacillus was also tested at various concentrations (108, 107, 106, 105 and 104 ). The number of Lactobacillus that adhered around the cells was significantly increased in the treatment with 108 bacterial cells. Transmission electron microscopy revealed that the cellularity of the junction between CaCo-2cells and Lactobacillus was not compromised. Transmission electron microscopy also revealed that the thalli fabric structure remained intact.展开更多

Their toxic activities were assayed through leaf puncturing on corn differentials (OH43, OH43Ht1, OH43Ht2, and OH43HtN), and the results revealed that eight fractions were toxic and fraction 6 was specifically toxic to OH43Ht1, which could be taken as a gene-selective toxin fraction. Fraction 6 was finely purified via HPLC and condensed by freeze desiccation. Its chemical structure was analyzed with EI-MS, IR, HMBC, ^1H-NMR, and two-dimensional NMR. The prese...Pennellid copepod Peniculus fistula fistula(Nordmann, 1832)(Synonym: Peniculus fistula Nordmann, 1832,Aphia ID: 745880), a worldwide distributed species, has been recovered from at least 19 teleost families. The present paper reports for the first time from the Malabar coast(South India), not only the existence of a new host family, Clupeidae, hosting this parasitic copepod species(P. fistula fistula) but also their season dependent hosting. A total of 123 marine fish species, belonging to 77 genera and 38 families surveyed along the Malabar coast, only the clupeid, Anadontostoma chacunda(Hamilton, 1822) was shown to be infected by this copepod species;all the recovered(copepod) parasites were invariably found attached at the mid portion of the caudal fin lobes and lying parallel to the host body, indicating the strict site-specific parasitisation. There is a discrete seasonality in the prevalence(P<0.05) as the sign of infection was noticed during the period from September to May with relatively high prevalence during winter months(November–January). During the monsoon months(June–August), the host fish was found completely free from Peniculus infection. Interestingly, all the 229 recovered specimens(P. fistula fistula) were gravid females having paired uniserrate egg sacs with the length more than its own body length.展开更多
Background:In Traditional Chinese Medicine(TCM)theory,"phlegm,blood stasis and toxin"are the pathogenesis of carotid atherosclerosis(CAS).The rabbit carotid atherosclerosis(CAS),which is induced by high-chol...Background:In Traditional Chinese Medicine(TCM)theory,"phlegm,blood stasis and toxin"are the pathogenesis of carotid atherosclerosis(CAS).The rabbit carotid atherosclerosis(CAS),which is induced by high-cholesterol diet combined with carotid artery balloon injury,is a classic model for studying CAS.Many studies indicate that gut microbiota and host metabolic disorders are involved in the pathogenesis of rabbit CAS.However,the TCM pathological features and syndromes of this classic rabbit CAS model have not been reported.Objective:To explore the pathogenesis of the rabbit CAS model and its TCM syndrome types from the perspective of"phlegm,blood stasis,and toxin".Methods:Twelve male New Zealand white rabbits were randomly divided into NC group and CAS group according to their body weight,followed by feeding of basic feed and a 1%high cholesterol diet,respectively.After two weeks,the rabbits in the CAS group underwent common carotid artery(CCA)balloon injury,while the rabbits in the NC group underwent only CCA separation without balloon injury.The two groups received differential feed postoperatively for six more weeks,after which,changes in lipids,hemorheology,inflammation,oxidative stress,and CAS phenotypes were analyzed.In addition,the colon contents and serum were collected for 16S rRNA sequencing and ^(1)H-NMR metabonomic analysis.Results:The CAS rabbits were observed to form noticeable abnormalities in lipid metabolism and blood rhe-ology,a sharp increase in oxidative stress levels,excessive release of inflammatory factors and apparent CAS plaque formation.Furthermore,10 specific gut microbiota(such as Akkermansia muciniphila,Barnesiellaceae and Faecalibacterium)and 14 characteristic metabolites(such as trimethylamine oxide,acetic acid and L-carnitine)were identified in the CAS rabbits,which were significantly related to the CAS phenotypes.The pathway func-tion analysis showed that the gut microbiota and its metabolites mainly affected cholesterol metabolism,energy metabolism,inflammation and oxidative stress.Conclusion:The rabbit CAS model conforms to the“phlegm,blood stasis and toxin damage”theory.The gut microbiota and host metabolic disorders of the CAS rabbits interact and promote internal and external toxins,aggravating the progression of CAS.Our study provided experimental evidence for the application of this model in the TCM-based research of CAS.展开更多

The ITS1 and ITS2 sequences of B. acheilognathi varied among 561-639 bp and 559-648 bp, and ten simple sequence repeat loci (microsatellites) were detected in the ITS regions, which contributed to significant sequence length variation. Phylogenetic analyses revealed 4 genetic clades (A, B, C, D) in samples with significantly different fish host spectra and geographic distribution. Clade A possessed a wide host range and geographic distribution, including all the samples ofB. acheilognathi determined in previous report. Clades B, C, and D only infected the small cyprinid fishes Opsariichthys bidens and Zacco platypus, and were limited to different locality. Interesting, Clades A and D were detected coexisting in same water body, and even in same fish host O. bidens in Danjiangkou Reservoir. The relatively higher genetic divergence and wide geographic distribution orB. acheilognathi parasitic in O. bidens indicated that O. bidens is likely to be the primary host. Sympatric occurrence of the two genetically distinct clades suggests the possibility of allopatric speciation and second encounter events.展开更多
