[Method] Four microsatellite loci including BMS1591, TEXAN-2, FCB128 and BM81124 mapped on GDF8 region of chromosome No. 2 of sheep that may be correlated with growth performance were chosen to detect the molecular genetics foundation of growth performance of Hu sheep. [ Result] Four microsatellite loci detected were high in heterozygosity, more in effective alleles number and rich in polymorphic information, all the three indices passed through the high polymorphic level (PIC 〉0.5). [ Conclusion ] The four microsatellite loci detected could be used to estimate the genetic polymorphism of growth performance of Hu sheep. 展开更多

Relationship Between the mRNA Expression Level of TGF-β Receptor Genes in Tissues and Ovulation Rate in Hu Sheep 被引量:5

Eight ewes of Hu sheep which bred multi-lamb were used as the high-fecundity group and the other eight ewes of Hu sheep which bred single lamb were used as the control group to investigate the relationship between the mRNA expression level of TGF-β receptor genes in tissues and ovulation rate in Hu sheep. Cloprostenol sodium was injected to make the synchronization of estrus treatment, then all ewes were slaughtered within 24-36 h after empathema and the ovaries were collected. Furthermore, the number of ovulation points was counted to determine ovulation rate for each sheep. Tissue expression analysis was conducted by RT-PCR for one ewe form the high-fecundity group and the relationship between the mRNA expression of genes encoding TGF-β receptors and ovulation rate was detected by real-time fluorescent quantitative PCR. The results showed that the relative expression level of TGF-flR I gene in the reproductive organ was significantly higher than in the lung and muscle (P 〈 0.01), while relative expression level of TGF-βR H in reproductive organ was significantly higher than that of other tissues (P 〈 0.01), indicating that these are highly expressed genes in the ovary. In addition, mRNA expression level of TGF-βR I and TGF-flRH in the ovaries of the high-fecundity group were significantly higher (P〈 0.01) and obviously higher (P= 0.011) than the control group, respectively. The mRNA expression level of TGF-βR I and TGF-βR H had a positive correlation with ovulation rate and the correlation coefficients were 0.562 (P〉 0.05) and 0.711 (P〈 0.05), respectively. It is suggested that TGF-β receptors have close relationship with highfecundity in Hu sheep. 展开更多

Hu Sheep is a local sheep breed in the plain of Taihu, China. Due to the natural conditions in Taihu and human domestication, Hu Sheep is gradually featured with high adaptability, fast growth rate, early maturity and high productivity, which make them to be a unique and rare breed of sheep. In the present study, an experiment was conducted by means of instantaneous scan sampling, focal animal sampling and all-occurrence sampling to learn about Hu Sheep's behavior and activity pattern under certain feeding management and ecological condition. The result showed that the activity time budgets for several behaviors were different between rams and ewes, with respect to feeding (rams 30.60% vs. ewes 33.23%), ruminating (33.43% vs.35.30%); sleeping (19.92% vs. 13.69%), standing (4.13% vs. 5.26%), moving (8.10% vs. 7.46%), and other behaviors (3.86% vs.5.06%). A circadian activity rhythm was shown in some behaviors; e.g., total feeding time was greater in the daytime than at night, whereas resting time showed the opposite tendency. Moving mostly occurred in the daytime, and ruminating mainly occurred at night. Additionally, feeding activity occurred 32.22 times per day in rams and 31.73 times per day in ewes, whereas ruminating activity after feeding occurred 19.83 times per day in rams and 24.34 times per day in ewes. For each food bolus,the average ruminating time was 51.22 s in rams and 49.52 s in ewes, with 68.22 bouts of rumination per day in rams and57.35 bouts of rumination in ewes. By conducting the experiment, we studied and grasped the local Hu Sheep's main behavior and regularity which are of great significance on improving the feeding management and reducing diseases. 展开更多

The phenotypic diversity resulting from artificial or natural selection of sheep has made a significant contribution to human civilization.Hu sheep are a local sheep breed unique to China with high reproductive rates and rapid growth.Genomic selection signatures have been widely used to investigate the genetic mechanisms underlying phenotypic variation in livestock.Here,we conduct whole-genome sequencing of 207 Hu sheep and compare them with the wild ancestors of domestic sheep(Asiatic mouflon)to investigate the genetic characteristics and selection signatures of Hu sheep.Based on six signatures of selection approaches,we detect genomic regions containing genes related to reproduction(BMPR1B,BMP2,PGFS,CYP19,CAMK4,GGT5,and GNAQ),vision(ALDH1A2,SAG,and PDE6B),nervous system(NAV1),and immune response(GPR35,SH2B2,PIK3R3,and HRAS).Association analysis with a population of 1299 Hu sheep reveals that those missense mutations in the GPR35(GPR35 g.952651 A>G;GPR35 g.952496 C>T)and NAV1(NAV1 g.84216190 C>T;NAV1 g.84227412 G>A)genes are significantly associated(P<0.05)with immune and growth traits in Hu sheep,respectively.This research offers unique insights into the selection characteristics of Hu sheep and facilitates further genetic improvement and molecular investigations. 展开更多

Background In the modern sheep production systems,the reproductive performance of ewes determines the economic profitability of farming.Revealing the genetic mechanisms underlying differences in the litter size is important for the selection and breeding of highly prolific ewes.Hu sheep,a high-quality Chinese sheep breed,is known for its high fecundity and is often used as a model to study prolificacy traits.In the current study,animals were divided into two groups according to their delivery rates in three consecutive lambing seasons(namely,the high and low reproductive groups with≥3 lambs and one lamb per season,n=3,respectively).The ewes were slaughtered within 12 h of estrus,and unilateral ovarian tissues were collected and analyzed by 10×Genomics single-cell RNA sequencing.Results A total of 5 types of somatic cells were identified and corresponding expression profiles were mapped in the ovaries of each group.Noticeably,the differences in the ovary somatic cell expression profiles between the high and low reproductive groups were mainly clustered in the granulosa cells.Furthermore,four granulosa cell subtypes were identified.GeneSwitches analysis revealed that the abundance of JPH1 expression and the reduction of LOC101112291 expression could lead to different evolutionary directions of the granulosa cells.Additionally,the expression levels of FTH1 and FTL in mural granulosa cells of the highly reproductive group were significantly higher.These genes inhibit necroptosis and ferroptosis of mural granulosa cells,which helps prevent follicular atresia.Conclusions This study provides insights into the molecular mechanisms underlying the high fecundity of Hu sheep.The differences in gene expression profiles,particularly in the granulosa cells,suggest that these cells play a critical role in female prolificacy.The findings also highlight the importance of genes such as JPH1,LOC101112291,FTH1,and FTL in regulating granulosa cell function and follicular development. 展开更多

背景:有研究表明基于腰椎CT的HU(hounsfield units)值可以筛查骨质疏松症,而目前因肺部感染就诊的患者增加,肺部感染合并2型糖尿病的患者也随之增加,增加了胸部CT的使用率。目的:探讨胸部CT检查中L_1椎体HU值在2型糖尿病骨质疏松症筛查... 背景:有研究表明基于腰椎CT的HU(hounsfield units)值可以筛查骨质疏松症,而目前因肺部感染就诊的患者增加,肺部感染合并2型糖尿病的患者也随之增加,增加了胸部CT的使用率。目的:探讨胸部CT检查中L_1椎体HU值在2型糖尿病骨质疏松症筛查中的作用。方法:回顾性分析2020年6月至2022年6月成都医学院第一附属医院收治的244例2型糖尿病患者的临床资料。利用双能X射线骨密度仪获得骨密度T值,按WHO的骨质疏松症诊断标准,将研究对象分为非骨质疏松组(n=120)及骨质疏松组(n=124),比较两组患者一般情况、骨密度T值及胸部CT检查中L_(1)椎体的HU值,分析HU值与各部位T值的关系并评估2型糖尿病骨质疏松症的准确性。结果与结论:(1)两组患者性别、年龄、体质量指数、糖化血红蛋白、平均血糖、钙、磷、2型糖尿病患病时间、高血压病史、高脂血症病史之间对比差异无显著性意义(P>0.05);(2)HU值与最低T值呈正相关(r=0.619,P<0.01),与髋部T值呈正相关(r=0.584,P<0.01),与股骨颈T值呈正相关(r=0.641,P<0.01);当HU值取98时,其预测2型糖尿病骨质疏松症具有良好的准确性,敏感性为70.8%;(3)提示基于胸部CT检查的L_1椎体HU值对2型糖尿病患者的骨质疏松症筛查具有良好的价值,可作为2型糖尿病骨质疏松症一种机会性、无成本的补充筛查方法。 展开更多

Objective:To investigate the therapeutic effect of Bai Hu Ren Shen Soup(BHRSS)on type 2 diabetes mellitus(T2DM)rats and explore whether its therapeutic effect is achieved by promoting autophagy in the pancreas tissue of T2DM rats.Methods:Thirty male standard deviation rats were equally divided into normal control group,T2DM group and BHRSS group.Firstly,the therapeutic effect of BHRSS on T2DM was evaluated by biochemical indices in serum and pathological changes in the pancreas tissue of rats in each group after modeling and medication.Then,the levels of microtubule-associated protein 1 light chain 3(LC3)protein and p62 protein in the pancreas tissue of rats in each group after BHRSS intervention were detected by Western blot.Results:BHRSS intervention significantly reduced the levels of blood sugar,liver and kidney function,and other related biochemical indices in the serum of T2DM rats,and effectively improved the pathological changes in the pancreas tissue of T2DM rats.In addition,Western blot results showed that the expression levels of autophagy-related protein LC3 protein in the pancreas tissue of rats were increased,and the expression level of p62 protein was decreased after BHRSS intervention.Conclusion:BHRSS has a clear therapeutic effect on T2DM model rats,which may be related to that increase of autophagy protein LC3 and the decrease of p62 protein level in the pancreas tissue. 展开更多

Objective:To explore the characteristics of Xiao Chai Hu(XCH)decoction in randomized controlled trials(RCTs)using a bibliometric method.Methods:Seven databases including PubMed,Web of Science,Embase,Cochrane library,China National Knowledge Infrastructure(CNKI),Chinese Scientific Journal Database(VIP)and Wanfang database were retrieved from inception to May 27,2022.In the study,XCH decoction(or modified)used alone or combined with conventional Western medicine as an intervention measure was included.The basic characteristics,funding support,relevant diseases,intervention methods,and adverse events(AEs)were analyzed.Data analysis was performed using SPSS 26.0 software.Results:A total of 813 RCTs were included,published from 1989 to 2022.There was only one Englishlanguage literature with the recent impact factor of 5.374.There were 147 studies were from Chineselanguage core journals,with the highest impact factor of 2.414.Only 6.15%of the literatures mentioned funding support.96.31%of the included literature reported the statistical significance of using XCH decoction.The diseases treated mainly included chronic hepatitis B(9.35%),cough variant asthma(5.66%),dizziness(5.54%),bile reflux gastritis(4.43%),and fever(4.18%).However,the overall research design of the included literature was poor,and large sample size,multicenter RCTs are needed.The incidence of AEs of XCH decoction alone was 8.86%,which was significantly lower than that of conventional Western medicine treatment.The combination of XCH decoction and conventional Western medicine treatment could reduce the incidence of AEs,and no serious adverse event was reported.Conclusion:Although the included studies show that XCH decoction is widely used,and has good efficacy and few AEs.Due to the low quality of the included RCTs,there may be some bias,and its rational use based on the specific conditions is recommended in clinical practice. 展开更多

在人机交互领域中手势识别的应用前景十分广阔,在诸多领域中均为人类带来了无限便利。基于数理统计特征设计一种人机交互图像手势识别方法,实现了基于数理统计特征的手势信息获取与基于识别结果的人机交互。对人机交互图像实施图像灰度... 在人机交互领域中手势识别的应用前景十分广阔,在诸多领域中均为人类带来了无限便利。基于数理统计特征设计一种人机交互图像手势识别方法,实现了基于数理统计特征的手势信息获取与基于识别结果的人机交互。对人机交互图像实施图像灰度化处理、二值化处理、平滑处理、边缘检测和轮廓提取处理等一系列预处理。通过OpenCV提取二值化人机交互图七个具有尺度不变性、旋转不变性、平移不变性的Hu矩,前四个矩描述手势的图像椭圆、主轴方向角、面积、旋转半径这四个物理量,后三个矩描述的是图像对称性、重心、中心距。基于Darknet-19改进YOLO-V2网络的骨干网络,使模型能够预测的anchor box数量达到16×16×N个。基于改进YOLO-V2网络设计手势识别模型,模型的输入为人机交互图、提取的手势轮廓与Hu矩,实现交互图像手势识别。测试结果表明,设计方法在室内和室外的手势识别准确率均较高,通过该方法的手势识别结果能够实现人机交互。 展开更多

不同于传统的接触和射频两种方式识别存储介质,利用X射线透照目标对象作为检测手段,这种检测方法符合无损检测的特征。首先采用灰度分组(GLG)的图像增强方法和基于单像素提取边缘实现图像预处理,然后利用改进的Hu不变矩得到适合非线性... 不同于传统的接触和射频两种方式识别存储介质,利用X射线透照目标对象作为检测手段,这种检测方法符合无损检测的特征。首先采用灰度分组(GLG)的图像增强方法和基于单像素提取边缘实现图像预处理,然后利用改进的Hu不变矩得到适合非线性不连续空间的矩不变特征向量,新的不变矩被证明作用于图像的平移、旋转和缩放具有不变性。K-means分类器根据计算不同的形态变换后图像的特征值判断存储介质种类,准确率达到95.83%,证明改进的Hu不变矩算法对被检存储介质种类的分类和识别有较好的鲁棒性和可靠性。实验结果表明,该识别方法可以使涉密的移动存储介质处于安全监控状态下,该技术应用于邮政连续包分机小像素X射线检测设备,可大大减少物联网时代通过邮路途径实施非法媒介传输。 展开更多

目的探讨抗Hu抗体检测在神经系统副肿瘤综合征诊断中的临床意义。方法采用间接免疫荧光方法和蛋白免疫印迹法对送检至北京协和医院神经病理实验室的1500余例患者的血清和脑脊液进行抗Hu抗体检测,回顾性分析抗Hu抗体阳性患者的临床资料... 目的探讨抗Hu抗体检测在神经系统副肿瘤综合征诊断中的临床意义。方法采用间接免疫荧光方法和蛋白免疫印迹法对送检至北京协和医院神经病理实验室的1500余例患者的血清和脑脊液进行抗Hu抗体检测,回顾性分析抗Hu抗体阳性患者的临床资料及诊断。神经系统副肿瘤综合征的诊断标准参照Graus等的诊断标准。结果共有27例患者抗体阳性,其中血清抗Hu抗体阳性25例,脑脊液抗Hu抗体阳性8例。临床主要表现为感觉神经元神经病、亚急性小脑变性、Lambert-Eaton综合征和抗利尿激素分泌不当综合征引起的脑病等,其中20例(74.1%)患有肿瘤,包括肺癌17例,胃癌2例,不明性质腹部肿瘤1例。7例患者未发现恶性肿瘤,包括多发性肌炎和系统性红斑狼疮(SLE)合并神经肌肉病各1例。根据Graus等神经系统副肿瘤综合征的诊断标准,27例中22例可确诊神经系统副肿瘤综合征或相关的肿瘤,抗Hu抗体的阳性预测值为81.5%。结论抗Hu抗体阳性对神经系统副肿瘤综合征的诊断具有一定意义;其相关肿瘤以肺癌,特别是小细胞肺癌最常见。其他自身免疫性疾病偶可见抗Hu抗体阳性,需要全面检查和密切随访以排除恶性肿瘤的可能。 展开更多
