Methods: The expression ofLNR and Ⅳ col in human colorectal carcinoma(HCC) cell lines were detected quantitatively with different potential metastasis. By using human amniotic membrane(HAM) invasion model, we observed the expressionof Ⅳ col and invasion in both HCC cell lines affected by LNR monoclonal antibodies. Results: High metastaticpotential cell line Lovo expressed higher levels of LNR and Ⅳ col than HT29, a low metastatic potential HCCcell line. The number of cells adhered to HAM of Lovo were more than that of HT29. When the cells of thetwo HCC cell lines blocked by LNR antibodies, the expression of Ⅳcol were decreased, and the cells adheredto HAM expressed Ⅳ col were also decreased. Conclusion: LNR might affect the regulation of expression ofⅣ col in tumor cells by cellular signal communication, and then affect the invasion and metastasis of tumor.展开更多
In order to determine the usefulness value of the antibodies to cytokeratins(CK) of'bile duct type'in the differential diagnosis between hepatocellular carcinoma(HCC) and cholangiocellular carcinoma(CC),we hav...In order to determine the usefulness value of the antibodies to cytokeratins(CK) of'bile duct type'in the differential diagnosis between hepatocellular carcinoma(HCC) and cholangiocellular carcinoma(CC),we have made an immunocytochemical investigation,using the antibodies specifically recognizing CK19 and CK18,seperately,in liver,and laminin(LN) antibody.All the CC examined(10 cases) were found CK19-positive;interestingly,CK19-positive cancer cells were also observed in 38% of HCCs(14/37).Therefore,CK19 was not a reliable marker in differentiating HCC from CC,in our consideration.The CK19 expression in HCC was showed to be irrelevant to their differentiation degres,but related to the histologic subtypes which indicated the directions of their differentiation.CK19 expression was observed in all the HCC cell nests with glandular differentiation,and an untontinuous LN-Positive basement membrane-like structure was immunolocalized around these cells.Which indicated that the glandular differentiation and CK19 expression in HCC were also related to the LN deposition,as in fetal liver and some chronic liver disorders.展开更多

Methods: Experimental metastatic potentiality of two kinds ofcell lines, HT29 and LoVo cells were compared by using nude mice model for human colorectal carcinoma liver metastasis. The expressions of epithelial-cadherin (E-cad), laminin receptor(LNR) and type Ⅳcollagenase(Ⅳ col) in two kinds of HCC cell lines, HT29 and LoVo cells, were observed quantitatively. The intracellular CD44V6 expressed in mRNA and in protein of the HCC cell lines were detected with in situ hybridization and immunohistochemistry. Results: 100% of nude mice had liver metastasis, which demonstrated thatLoVo is a high metastatic HCC cell line. However, HT29 is a relatively low metastatic HCC cell line becauseonly 42. 9 % of nude mice transplanted HT29 cells had liver metastasis. The expressions of E--cad, LNR andIVcol of HT29 and LoVo cells in vitro were detected quantitatively. HT29 cells expressed E-cad with weakpositive on cell membrane. and LoVo cells almost expressed none. LoVo cells expressed higher level of LNRand tV col than HT29 cells did. The expression of CD44V6 in LoVo cells whether in mRNA or in protein werehigher than that of HT29 cells.
