Study on the Correlation between Human Papillomavirus and Mycoplasma genitalium Combined with TCT Detection

Objective: This study aims to explore the correlation between human papillomavirus (HPV) and Mycoplasma genitalium (CT) combined with TCT detection in cervical cancer screening. Method: A cross-sectional study design was adopted, and a total of 609 women who came to seek medical treatment were recruited as the study subjects. Combination testing was evaluated on cervical cancer screening by testing the women for HPV, CT with TCT detection and analyzing the relationship of cervical lesions with HPV and CT infection. Results: The study results showed that 21.57% of the subjects were infected with both HPV and CT, and 48.42% of the cases had abnormal TCT results at the same time. Further data analysis showed that HPV infection was significantly associated with abnormal TCT outcomes (p < 0.05), suggesting a possible synergistic effect of the two infections in cervical lesions. The combined sensitivity and specificity of HPV, CT and TCT detection were 21.57% and 48.42%, respectively, which were significantly higher than that of single detection. Conclusion: In summary, the results of this study support the importance of combined HPV, CT, and TCT testing in cervical cancer screening, and propose the hypothesis that combined testing may improve screening effectiveness. However, further large sample studies are needed to confirm this conclusion and explore the prospects of combined testing in clinical practice. 展开更多

Approximately 80% of sexually active women are likely infected with HPV. Cervical cancer is one of the most common malignant tumors and is second in incidence only to breast cancer. Infection with high-risk HPV types is the main risk factor for cervical cancer, which is currently the only malignant tumor with a clearly defined etiology. HPV infection is also closely related to the incidence and development of other malignant tumors. In addition to cervical cancer, HPV can cause other urogenital tumors, as well as tumors in the digestive tract, lungs, eyes, skin, and other organ systems. This paper provides a review of the progress in HPV infection-related research and provides novel ideas for the study of tumor etiology and mechanisms. 展开更多

Background:The role of human papillomavirus(HPV)in the development of vulvar cancer(VC)has been widely studied,but findings have been inconsistent.Despite numerous meta-analyses exploring the potential link between HPV and VC,the association remains controversial due to inherent limitations in meta-analytic methods.Objectives:To address this controversy,the study aims to investigate the potential link between HPV and VC using the Bradford Hill criteria,which offer a more comprehensive framework for establishing causation.Methodology:The study began by extracting all relevant studies on the association between HPV and VC from the PubMed database.The potential links were then assessed by examining the data using the major postulates of the Bradford Hill criteria.To ensure the reliability of the findings,the methodologies of the identified studies were critically evaluated to account for possible false-negative and false-positive results.Results:The assessment of previous studies against the Bradford Hill criteria revealed that the major postulates were not fulfilled.Conclusion:Based on the findings,it can concluded that there is no causal association between HPV and VC. 展开更多

Malakoplakia is a rare granulomatous disease probably caused by infection and characterized histologically by Michaelis-Gutmann bodies.We report a more rarely seen case esophageal malakoplakia in a 54-year-old woman.She presented with coughing while eating and drinking.Gastroscopy showed yellow nodules in the esophagus,and endoscopic ultrasonography showed a space-occupying lesion in the substratum of the esophageal mucosa.All findings highly resembled esophageal cancer.Histopathological examination finally indentified this space-occupying lesion as malakoplakia and not cancer.Immunohistochemistry showed that she had human papillomavirus(HPV) infection in the esophagus,which indicates that infection was responsible for the malakoplakia.This is believed to be the first case of malakoplakia in the esophagus,and more importantly,we established that HPV infection was the initiator of esophageal malakoplakia. 展开更多

The specimens of 111 cervical carcinomas. 68 chronic cervicitis and 43 normal cervical exfoliated epithelial cells were examined for the presence of HSV2 DNA sequences with DNA hybridization using HSV2 BgL Ⅱ N fragment probe labelled by 32PdCTP. The result showed that the infection rates of HSV2 in the samples of cervical cancer.chronic cervicitis and normal epithelial cells were 1 4. 41 %(16/111). 27.94%( 19/68) and 25.58% ( 11/43),respectively. It was implied that early stages carcinogenesis of cervical epithelial cells might be correlated with the HSV2 infection.Sixteen HSV 2 positive samples of cervical carcinomas were also examined for the presence of the sequences homologous to human papillomavirus (HPV) type 6B/11. 16 and 18 DNA using dot blot hybridization (Tm17℃). The result indicated that 13 out of 16 were HPV 16 DNA hybridization positive accounting for 81. 2% of all HSV-2 positive samples and none of them were positive for HPV type 6B/11 and 18. The result indicated that double infection of HSV 2 and HPV16 in the same cervical carcinoma tissues may provide a strong evidence of the viral synergistic interaction in the induction of female cervical 展开更多

To screen patients with early cervical lesions by analyzing the infection of high-risk Human papillomavirus (HR-HPV). Research Methods: The cervical exfoliated cell specimens and their clinical data were collected. The HPV infection types of the collected specimens were detected by fluorescence quantitative PCR, and the correlation between HPV infection and clinicopathological features was analyzed statistically. Results: 725 cases were HR-HPV positive from 2605 cases, including 15 high-risk types of HPV16, 18, 31, 33, 35, 39, 45, 51, 52, 53, 56, 58, 59, 66, 68. Different histological types ranged from NILM to HSIL, and the positive rate of HPV showed an increasing trend with the aggravation of cervical lesions. Conclusion: The positive rate of 15 high-risk HPV types in the collected specimens was 27.8%. Patients with early cervical lesions could be screened for 15 high-risk HPV infection types. 展开更多

目的:探讨多种治疗方案对宫颈高危型人乳头瘤病毒(HPV)感染患者的临床治疗效果,为HPV患者的临床治疗提供新思路。方法:采用回顾性研究方法纳入2022年1月—2023年1月在宜昌市中心人民医院治疗的宫颈高危型HPV感染患者501例,根据患者选取... 目的:探讨多种治疗方案对宫颈高危型人乳头瘤病毒(HPV)感染患者的临床治疗效果,为HPV患者的临床治疗提供新思路。方法:采用回顾性研究方法纳入2022年1月—2023年1月在宜昌市中心人民医院治疗的宫颈高危型HPV感染患者501例,根据患者选取的治疗方法分为三组,A组患者给予保妇康栓治疗(n=167),B组患者给予重组人干扰素α-2b凝胶治疗(n=167),C组给予抗HPV生物蛋白敷料治疗(n=167)。比较三组HPV患者治疗有效率、HPV-DNA水平、治疗后HPV阳性率和治疗过程中不良反应发生率,评价临床治疗效果。结果:C组患者治疗总有效率明显高于A组(90.42%vs 74.85%)和B组(90.42%vs 77.25%)(均P<0.05)。三组患者治疗3个月及6个月HPV-DNA水平较治疗前明显下降,且治疗3个月及6个月时,C组患者的HPV-DNA水平较A组和B组明显降低(均P<0.05)。治疗后C组HPV阳性率明显低于A组(6.60%vs 22.75%)和B组(6.60%vs 20.96%)(均P<0.05)。三组患者不良反应发生情况无明显统计学差异(P>0.05)。结论:抗HPV生物蛋白敷料对于宫颈高危型HPV感染患者的治疗效果优于保妇康栓及重组人干扰素α-2b凝胶,且不良反应发生少,值得临床上进一步推广。 展开更多

7345例女性HPV感染状况及亚型分布特征分析

目的了解于日照市中医医院就诊的女性人乳头瘤病毒(HPV)感染状况及基因型分布特征,为宫颈癌早期筛查防治、疫苗接种提供数据参考。方法收集2020年1月至2022年12月于日照市中医医院行HPV基因分型的7345名女性的检测结果,分析总体及不同... 目的了解于日照市中医医院就诊的女性人乳头瘤病毒(HPV)感染状况及基因型分布特征,为宫颈癌早期筛查防治、疫苗接种提供数据参考。方法收集2020年1月至2022年12月于日照市中医医院行HPV基因分型的7345名女性的检测结果,分析总体及不同年龄组的HPV感染率、基因型分布及疫苗保护的HPV基因型感染情况。结果在7345份检测标本中,检测出阳性标本1563份,阳性率为21.28%。其中来自妇科门诊的标本阳性率25.54%(1097/4295),来自健康中心的标本阳性率15.28%(466/3050),门诊标本阳性率明显高于健康中心的标本阳性率,差异有统计学意义(χ^(2)=112.13,P<0.05)。单一高危型感染率为11.79%(866/7345),多重分型感染率为5.62%(413/7345)。感染率最高的5种高危型依次是HPV16(3.89%)、HPV52(3.05%)、HPV58(2.06%)、HPV 53(1.97%)、HPV 31(1.54%)。HPV感染率在年龄分组分析中出现两个高峰,分别为≤20岁组(39.06%)和≥61岁组(27.24%)。HPV九价疫苗涵盖的基因型感染占比为64.43%。结论于日照市中医医院就诊的女性HPV感染率为21.28%,以单一高危型感染为主,青少年女性感染率最高,适龄女性推荐选择九价疫苗。 展开更多

目的探讨山东省滨州市不同年龄妇女人乳头瘤病毒(HPV)高危型感染率及HPV E6/E7 mRNA表达情况。方法选取2022年1-10月在该院妇产科就诊行HPV-DNA分型检测的女性患者10887例。按年龄分组(<30岁组2180例,30~<40岁组3222例,40~<50... 目的探讨山东省滨州市不同年龄妇女人乳头瘤病毒(HPV)高危型感染率及HPV E6/E7 mRNA表达情况。方法选取2022年1-10月在该院妇产科就诊行HPV-DNA分型检测的女性患者10887例。按年龄分组(<30岁组2180例,30~<40岁组3222例,40~<50岁组3438例,≥50岁组2047例)并统计HPV-DNA分型检测结果。在入选患者中分层随机选取HPV-DNA 16型阳性女性患者250例,各年龄组50例,统计其HPV E6/E7 mRNA检测结果,比较不同年龄组女性患者HPV-DNA分型及HPV E6/E7 mRNA表达的差异。结果10887例女性患者中阳性1666例,感染率为15.3%,其中高危型阳性1393例,占83.6%(1393/1666),低危型感染273例,占16.4%(273/1666)。HPV高危型阳性分布排前5位者依次为HPV 16、52、58、33、18型。不同年龄组女性患者感染率不同,HPV感染年龄分布曲线呈“V”形。不同年龄HPV-DNA 16型阳性女性患者HPV E6/E7 mRNA阳性率不同,40~<50岁组、≥50岁组女性患者HPV E6/E7 mRNA阳性率均高于30~<40岁组,差异均有统计学意义(P<0.05)。HPV-58型E6/E7 mRNA阳性率、拷贝值均高于HPV 16、18、52型,差异均有统计学意义(P<0.05)。HPV高危型混合感染与单一感染E6/E7 mRNA表达存在差异。结论通过开展HPV-DNA筛查工作及研究了解本地区HPV亚型的分布具有非常重要的价值。年轻妇女和围绝经期及以上妇女HPV感染风险较高。随年龄增加HPV 16型感染人群中HPV E6/E7 mRN阳性率逐渐升高,即发生宫颈病变的风险增加。不同HPV高危型感染E6/E7 mRNA阳性率、拷贝值均存在差异,且HPV高危型混合感染者E6/E7 mRNA阳性率、拷贝值均高于单一感染者。 展开更多

Human papillomavirus (HPV) is one of the most common sexually transmitted diseases which comprises a group of small DNA viruses that infect both cutaneous and mucous squamous epithelia. Liquid bead microarray technology (LBMA) were used to evaluate 24 HPV genotypes in confirmed fertile and infertile males of North China so that the effects of HPV infection on semen parameters and relationship with male infertility could be discussed. A total of 1138 subjects were recruited in this study; 142 were HPV-positive (12.48%). Among 523 confirmed fertile males, only 35 were HPV-positive (6.70%), and two of them had multiple infections. Among 615 infertile males, 107 were HPV-positive (17.4%), and 29 of them had multiple infections. Infertile males had a relatively high HPV infection rate compared with confirmed fertile males. Sperm progressive motility (PR) and the normal morphology rate were significantly decreased in HPV-positive subjects. HPV-45, HPV-52, HPV-18, HPV-59 and HPV-16 infections were more frequently in infertile males. Hence, HPV infection is closely related to male infertility which will decrease sperm PR and morphology. HPV-45, HPV-52, HPV-18, HPV-59 and HPV-16 infection seems to be major risk factors. 展开更多

AIM:To detect human papillomavirus(HPV) DNA in esophageal carcinoma(EC) 109 cells and investigate the relationship between HPV and EC.METHODS:Genomic DNA and total RNA from EC109 cells were isolated.HPV DNA was detected by polymerase chain reaction(PCR) with the general primer sets of My09/11 and GP5 +/6 + for the HPV L1 gene and type-specific primer sets for HPV18 E6 and HPV18 E6-E7.Reverse transcription(RT) of mRNA isolated from EC109 cells was performed to produce a cDNA.And then a PCR-based protocol for the amplification of papillomavirus oncogene transcripts was used to analyze HPV18 DNA and integrated transcripts of HPV18 in the chromosomes of EC109 cells.The final nested PCR products were cloned into a pMD-18T vector and sequenced to analyze the chromosomal location of HPV integration.RESULTS:HPV18 DNA was detected in EC109 cells by PCR using the general primer sets of My09/11 and GP5 +/6 + for HPV L1 and the type-specif ic primer sets for HPV18 E6 and E6-E7 to generate products of 450 bp,150 bp,335 bp and 944 bp,respectively.Approximately 600 bp of integrated HPV18-specific transcript was identified.The final nested PCR product of integrated HPV18 DNA was cloned into a pMD-18T vector and sequenced to analyze the chromosomal location of HPV integration.Sequence alignment showed that the HPV18 sequence from EC109 cells was identical to that of the encoded early protein E7-E1 of the standard HPV18 strain X05015,and another partial gene sequence was identical to a partial sequence of human chromosome 8.CONCLUSION:Integration of the HPV genome into the host cell chromosome suggests that persistent HPV infection is vital for malignant cell transformation and carcinogenesis. 展开更多

HPV DNA was identified in formalin-fixed, paraffin-embedded tumor specimens by nested PCR with MY09/MY11 and GP5^+/GP6^+ primer pairs and the HPV genotype was determined by direct DNA sequencing. Association between the HPV status and risk factors for cancer as well as tumor-host characteristics were analyzed.Survival curves were calculated by the Kaplan-Meier method and analyzed using the log-rank test. Results HPV was found in 40.4% of the tumors with HPV16 accounting for 63.5%, HPV18 for 30.8%, HPV6 for 3.9% and HPVll for 1.8%. No infection with more than one HPV genotype was detected. HPV infection was significantly associated with poor histological grade, TNM stage Ⅰ -Ⅱ, alcohol usage and no smoking status. Multivariate analysis showed that HPV had an independent prognostic effect on the overall survival after adjusting other confounding factors such as histological grade, TNM stage and tobacco usage. The presence of HPV was significantly correlated with a better survival in patients with OSCC. Conclusion HPV infection can act as an independent predictor for the survival and prognosis of OSCC. 展开更多

目的分析滨州市某医院妇科自然就诊患者人乳头瘤病毒(human papillomavirus,HPV)基因型分布及HPV感染例数,为当地宫颈癌防治提供参考。方法统计分析2020年1月—2022年12月妇科自然就诊患者HPV基因亚型及单一和多重感染率。结果2020——2... 目的分析滨州市某医院妇科自然就诊患者人乳头瘤病毒(human papillomavirus,HPV)基因型分布及HPV感染例数,为当地宫颈癌防治提供参考。方法统计分析2020年1月—2022年12月妇科自然就诊患者HPV基因亚型及单一和多重感染率。结果2020——2022年滨州市某医院妇科自然就诊患者进行HPV基因型检测,三年共检测HPV 22580例,共检出HPV阳性者4458例,阳性率为19.86%;单一HPV基因型感染者3301例,阳性率14.62%,多重HPV感染者1184例,阳性率5.24%。对2020——2022年HPV感染患者进行HPV基因型统计分析,结果显示:高危型HPV(HR-HPV)前五位是HPV 52型、HPV 16型、HPV 58型、HPV 51型、HPV 68型,低危型HPV感染第一位为HPV 42型;HPV感染最少的基因型是HPV 83型。年龄方面,HPV感染者以21~60岁年龄段人群为主,≤20岁HPV感染者126例,最小年龄者16岁;>60岁HPV感染者153例,最大年龄为87岁,此两年龄组HPV多重感染率均高出21~60岁年龄组。结论本地患者HPV感染者以HPV 52型最多,其次为HPV 16型。HPV感染以单一型为主,多重感染者比例也不容忽视。HPV基因型检测是宫颈癌筛查的重要手段。 展开更多

Globally,about 70% of cervical cancers are associated with human papillomavirus (HPV)-16 or HPV-18 infection.A meta-analysis of epidemiologic studies in China showed that HPV was present in 98% of cervical cancer samples.The HPV-16/18 AS04-adjuvanted vaccine Cervarix has shown a high level of protection against HPV-16/18 infections and associated cervical lesions.This phase Ⅰ trial (NCT00549900) assessed the safety,tolerability,and immunogenicity of the vaccine in Chinese.Thirty healthy Chinese females,aged 15 to 45 years with a median age of 29.5 years,received three doses of Cervarix in Months 0,1,and 6.Safety was assessed via recording solicited local and systemic symptoms within 7 days and unsolicited symptoms within 30 days after each vaccination.Serious adverse events,new onset of chronic diseases,and other medically significant conditions were recorded throughout this trial.As an exploratory objective,HPV-16/18 antibody titers were determined by enzyme-linked immunosorbent assay in serum samples collected in Months 0 and 7.Pain at the injection site was the most frequently reported local symptom.Two subjects reported medically significant adverse events.Both cases were assessed as unrelated to vaccination by the investigator.In Month 7,100% seroconversion was observed for both anti-HPV-16 and anti-HPV-18 with high geometric mean antibody titers.HPV-16/18 AS04-adjuvanted vaccine,evaluated for the first time in Chinese females,was generally well tolerated and immunogenic,as previously shown in global studies. 展开更多

Methods: Recombinant vector of pGADT7/E6 or pGBKT7/hDaxx was con- structed. The interaction of E6 protein and hDaxx was detected by yeast two-hybrid system. Their expression in yeast was detected by Western blotting. The eukaryotic plasmids of E6 and hDaxx were co-transfected into HeLa cells. Apoptosis was induced by 5-FU. The apoptotic rate was measured by flow cytometry (FCM). Results: E6 protein had intracellular interaction with hDaxx. The apoptotic rate was rising with the increase in the transfection quantity of pcDNA3.1 (-) / hDaxx in pcDNA3.1 (-) /E6 and pcDNA3.1 (-) / hDaxx co-transfected cells. The difference was significant ( P < 0. 01). Conclusion: There is intracellular interaction between HPV16 E6 protein and hDaxx. The over-expression of hDaxx can increase the sensitivity of E6 protein positive HeLa cells to 5-FU. The effect was in a dose dependent manner. 基于ARIMA模型的天津地区单中心HPV感染趋势及基因型特征

目的采用自回归移动平均(ARIMA)模型构建时间序列,分析天津地区单中心人乳头瘤病毒(HPV)感染趋势及基因型特征。方法选择2018年1月-2022年12月某院进行HPV检测的7236例女性患者,比较2018-2022年天津地区HPV感染情况及基因型分布。建立AR... 目的采用自回归移动平均(ARIMA)模型构建时间序列,分析天津地区单中心人乳头瘤病毒(HPV)感染趋势及基因型特征。方法选择2018年1月-2022年12月某院进行HPV检测的7236例女性患者,比较2018-2022年天津地区HPV感染情况及基因型分布。建立ARIMA模型时间序列,分析模型拟合。预测2023年HPV感染数,并与实际发生数进行比较,评价模型的预测效果。结果2018-2022年天津地区HPV感染率为14.41%;HPV感染率在31~40岁年龄段最高,感染率为15.47%。阳性标本中HPV单一型别感染比率最高,占比为73.54%(767/1043),以高危型HPV为主。低危型感染占比最高的是HPV-6型,为2.59%,高危型感染占比最高的是HPV-16型,为16.06%。建立ARIMA模型,确定最佳模型为ARIMA(0,1,2)(0,1,1)12,其AIC值和BIC值分别为3.877、4.005,经白噪声检验Ljung-Box Q=8.828差异无统计学意义(P>0.05)。利用模型预测2023年HPV感染数,实际值、预测值的总体趋势基本保持一致,模型RMSE、MAPE、MAE分别为6.289、34.149、4.706,提示模型的预测效果较好。结论天津地区女性人群中,HPV病毒感染类型以单一高危型感染为主,其中HPV-16型感染率最高。天津地区HPV感染存在季节性,ARIMA模型在HPV感染流行趋势的预测中效果较好,适用于短期预测。 展开更多
