In this study, a total of 600 40-week-old Hy-Line variety brown layers were randomized into five groups and fed with a corn-soybean-mixed basal diet supplement with 0, 10, 50, 100, and 500 mg · kg^-1 daidzein, respectively. Then, two PCR-based typing methods(RAPD-PCR and ERIC-PCR) were combined to analyze the ileum content and explore the changes of ileum microflora biodiversity. The results of RAPD-PCR and ERIC-PCR showed that bands under 10 mg · kg^-1 and 50 mg · kg^-1 were the most, and their similarity was the largest. Bands under 500 mg · kg^-1 were the least and similarity with other groups was the minimum. Ileum microflora biodiversity under 10 mg · kg^-1 or 50 mg · kg^-1 was richer than that under 500 mg · kg^-1. A corn-soybean-mixed basal diet supplement with 10 mg · kg^-1 to 50 mg · kg^-1 of daidzein might be beneficial to Hy-Line variety brown layers intestinal bacteria.展开更多
Polyphenolic compound in processing apple (Malus domestica Borkh.) varieties and the relationship between polyphenol content and enzymatic browning were studied to provide reference for raw material selection and pr...Polyphenolic compound in processing apple (Malus domestica Borkh.) varieties and the relationship between polyphenol content and enzymatic browning were studied to provide reference for raw material selection and processing method optimization. The content of polyphenol compound in 10 processing apple varieties (4 cider and 6 juice varieties) were analyzed using the Folin-Ciocalteu method and HPLC. The degree of browning and the activities of polyphenol oxidase were also studied. The content and proportion of the polyphenol varied depending on the variety. Bitter varieties globally showed a higher polyphenol concentration than sweet or acid varieties. Proanthocyanidins, chlorogenic acid, (+)-catechin, (-)-epicatechin were high-concentrated polyphenols in apple fruits. Phloridzin, the unique polyphenol of apple, was abundant in the bitter variety Frequin rouge fruit. Total polyphenols, proanthocyanidins, (+)-catechin, and phloridzin had higher correlations with browning. The correlation was low between chlorogenic acid and browning. The polyphenolic profiles were correlated with the apple types. Cider apples contained more polyphenol than juice apple varieties. The content of flavan-3-ol has a close relationship with fruit browning.展开更多

The structures of bacterial communities in the brown planthopper (BPH) Nilaparvata lugens (St^l) from different geographic and resistant virulent populations were analyzed by using denatured gradient gel electroph...The structures of bacterial communities in the brown planthopper (BPH) Nilaparvata lugens (St^l) from different geographic and resistant virulent populations were analyzed by using denatured gradient gel electrophoresis (DGGE). Results showed that the bacterial communities in BPH nymph from the first to the fifth instars varied with nymphal growth and development. The bacterial communities in the first-instar BPH nymph were similar to those in adults. Nine geographic BPH populations were divided into three groups based on the cluster analysis of DGGE fingerprint. The first group was from the Philippines; the second group was from Thailand and Hainan, Yunnan and Zhejiang provinces of China; and the third group was from Vietnam and Guangxi, Hunan and Jiangxi provinces of China. BPH populations adapted to different resistant rice varieties. Through the marker-assisted selection,it obtained 4 pieces of homozygous resistance gene maintainer line intermediate materials( 100B,101B,102B and 103B),and 2 pieces of resistant sterile line materials( 100A and 103A). All 10 combinations that have testcross with highly resistant sterile 100A showed higher level of brown planthopper resistance: 100A/R2586,100A/KR838,and 100A/KR527 had high resistance level,the others had low to intermediate resistance;100A/KR527,100A/R2586,100A/Minghui 63,100A/Fuhui 838 and 100A/Gui 99 combinations had yield per plant significantly higher than the control group( Teyou 7118),increasing by 14. 45%-49. 26%. The obtained resistant lines are expected to provide a better gene source for the breeding of resistant sterile lines of hybrid rice and the obtained resistant sterile lines can be directly used in the selection of three-line hybrid rice.展开更多
【目的】获取鸡小黄卵泡颗粒细胞(granulosa cells of small yellow follicle,SYFG)全长转录组数据,为深入挖掘鸡卵泡发育内在分子调控信息提供参考。【方法】采用PacBio高通量测序平台,对海兰褐壳蛋鸡SYFG进行测序、过滤、聚类和矫正...【目的】获取鸡小黄卵泡颗粒细胞(granulosa cells of small yellow follicle,SYFG)全长转录组数据,为深入挖掘鸡卵泡发育内在分子调控信息提供参考。【方法】采用PacBio高通量测序平台,对海兰褐壳蛋鸡SYFG进行测序、过滤、聚类和矫正原始数据,获取全长转录本序列。通过生物信息学软件对全长转录本数据进行功能注释、转录因子注释、编码区序列(coding sequence,CDS)预测、长链非编码RNA(long non-coding RNA,lncRNA)预测筛选、简单重复序列(simple sequence repeats,SSR)检测及可变剪接(alternative splicing,AS)和选择性多聚腺苷酸化(alternative polyadenylation,APA)分析。【结果】通过测序共获得52311条海兰褐壳蛋鸡SYFG全长转录本,长度主要分布于1~3500 bp。通过与GO、KEGG、COG、NR、Swiss-Prot和Pfam数据库比对,对转录本序列进行注释,其中GO数据库注释了26525条转录本,KEGG数据库注释了25233条转录本,COG数据库注释了31303条转录本,NR数据库注释了34284条转录本,Swiss-Prot数据库注释了30701条转录本,Pfam数据库注释了24622条转录本。此外,经鉴定或预测,还获得了559个转录因子、40944个CDS、14699个lncRNAs、7种类型的SSR、28423个AS和2747个APA事件。【结论】本研究获得了鸡SYFG全长转录组测序数据,提升了转录本的数量和长度,完善了转录本的功能注释,分析了AS和APA事件的类型,揭示了转录本的复杂性,为进一步从转录组水平解析鸡卵泡发育的分子调节机制提供数据支持。展开更多
