目的探讨IL-27和TGF-β1在肺纤维化患者血清中的表达及临床意义。方法采用ELISA法分别检测20例肺纤维化患者及15例健康对照组血清中的IL-27和TGF-β1的表达水平。结果 (1)肺纤维化重度缺氧组均具有更高的IL-27与TGF-β1水平,并且不同缺... 目的探讨IL-27和TGF-β1在肺纤维化患者血清中的表达及临床意义。方法采用ELISA法分别检测20例肺纤维化患者及15例健康对照组血清中的IL-27和TGF-β1的表达水平。结果 (1)肺纤维化重度缺氧组均具有更高的IL-27与TGF-β1水平,并且不同缺氧程度的肺纤维化IL-27与TGF-β1水平均逐渐升高,从对照组、肺纤维化轻度缺氧组、中度缺氧组、重度缺氧组均不同,差异均有统计学意义(F=21.58,P<0.001;F=18.75,P<0.001)。(2)肺纤维化血清IL-27与TGF-β1水平均随Pa O2升高而明显降低,经直线相关分析发现二者与Pa O2均分别呈负相关(r=-0.754,P<0.05;r=-0.793,P<0.05);血清IL-27和TGF-β1之间呈正相关(r=0.854,P<0.05)。结论IL-27和TGF-β1参与了肺纤维化发病,二者检测对其诊治有着重要的临床意义。 展开更多

胸腔积液红细胞沉降率、转化生长因子-β1及白细胞介素-27对结核性胸膜炎预后的评估

目的探讨分析胸腔积液红细胞沉降率(erythrocyte sedimentation rate,ERS)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)、白细胞介素-27(interleukin-27,IL-27)对结核性胸膜炎预后的评估价值。方法选择2021年3月至2023... 目的探讨分析胸腔积液红细胞沉降率(erythrocyte sedimentation rate,ERS)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)、白细胞介素-27(interleukin-27,IL-27)对结核性胸膜炎预后的评估价值。方法选择2021年3月至2023年3月无锡五院收治的184例结核性胸膜炎患者,根据患者治疗预后情况将患者分为预后良好组(n=146)和预后不良组(n=38)。收集并记录2组患者临床资料,比较2组患者胸腔积液ESR、TGF-β1、IL-27水平,对可能影响患者临床预后的各因素进行单因素及多因素Logistic回归分析,采用受试者工作特征曲线(receiver operating characteristic,ROC)曲线分析患者胸腔积液ESR、TGF-β1、IL-27水平对不良预后预测价值。结果2组患者在性别、糖尿病、高血压以及冠心病方面比较,差异均无统计学意义(P均>0.05),而2组患者在年龄、就诊时间、胸腔积液细胞数、胸腔积液蛋白总量方面比较,差异均具有统计学意义(P均<0.05)。预后不良组患者胸腔积液ESR、TGF-β1、IL-27水平均显著高于预后良好组(P均<0.05)。多因素Logistic回归分析结果显示,胸腔积液ESR、TGF-β1、IL-27以及胸腔积液细胞数、胸腔积液蛋白总量进入回归模型(P<0.05)。采用ROC曲线分析患者胸腔积液ESR、TGF-β1、IL-27水平对不良预后的预测价值,曲线下面积分别为0.787、0.852、0.892(P<0.05)。结论结核性胸膜炎患者胸腔积液ESR、TGF-β1、IL-27可在一定程度上预测患者预后,从而为患者病情评估以及治疗效果的评估提供一定的参考价值,值得临床进一步深入研究分析。 展开更多

Objective:To explore the clinical efficacy of Danshen injection in combined with local injection of triamcinolone acetonide in the treatment of oral submucous fibrosis (OSF) and the effect on serum TGF-β1 and IL-6.Methods:A total of 70 patients with OSF who were admitted in our hospital from October, 2015 to October, 2016 were included in the study and randomized into the observation group and the control group (n=35). The patients in the control group were given multipoint injection of triamcinolone acetonide (40 mg/time) in the basal region of oral mucosal regions. On this basis, the patients in the observation group were given additional Danshen injection (4 mL/time), 1 time/week, for 12 times. VAS was used for evaluation. The oral mucosa lesion area and mouth opening were measured. The morning fasting peripheral venous blood before and after treatment in the two groups was collected. The blood rheometer was used to detect the whole blood viscosity [high shear (HS), moderate shear (MS), and low shear (LS)], plasma viscosity (PV), and fibrinogen (FIB). ELISA was used to detect the serum TGF-β1 and IL-6 levels.Results: VAS score in the observation group was significantly reduced (P<0.05), mucosal lesion area was significantly less than that in the control group (P<0.05), and mouth opening was significantly greater than that in the control group (P<0.05). LS, MS, PV, and FIB after treatment in the observation group were significantly reduced when compared with before treatment (P<0.05), while LS, MS, HS, PV, and FIB after treatment in the control group were not significantly changed (P>0.05). TGF-β1 and IL-6 levels after treatment in the observation group were significantly lower than those in the control group (P<0.05).Conclusions: Danshen injection in combined with local injection of triamcinolone acetonide in the treatment of OSF can effectively reduce the serum TGF-β1 and IL-6 levels, inhibit the connective tissue fibrosis, and improve the pain, oral mucosal lesion area, and mouth opening, with a satisfactory clinical effect. 展开更多

Objective:To assess the effect of D-pinitol on pulmonary fibrosis induced by bleomycin.Methods:Sprague-Dawley rats received intratracheal bleomycin(6 IU/kg)to induce pulmonary fibrosis,followed by administration of either D-pinitol(5,10,or 20 mg/kg)or vehicle or methylprednisolone(10 mg/kg)over 28 days after bleomycin administration.Lung function,biochemical parameters,serum biochemistry,mRNA expressions,and histological features were observed.Results:D-pinitol at 10 and 20 mg/kg significantly(P<0.05)attenuated bleomycin-induced bronchoalveolar lavage fluid,decreased myeloperoxidase,nitric oxide,malondialdehyde levels,and increased glutathione and superoxide dismutase level.D-pinitol also improved lung function(enhanced pause,frequency of breathing,expired volume,and tidal volume).Besides,D-pinitol significantly(P<0.05)upregulated Nrf2 and downregulated mRNA expressions of TGF-β,collagen-1,and Smad-3.Furthermore,considerably less inflammation(peribronchial,perivascular,and total),Ashcroft,and interstitial fibrosis scores were observed in the D-pinitol group.Conclusions:D-pinitol exerts its effect against bleomycin-induced pulmonary fibrosis via antioxidative and anti-fibrotic pathways. 展开更多

Epidemiological evidence presents that dust storms are related to respiratory diseases,such as pulmonary fibrosis(PF).However,the precise underlying mechanisms of SPM-elicited adverse effects still need to be investigated.Epithelial-mesenchymal transition(EMT)process is a characteristic of PF.We discussed whether suspended particulate matter(SPM)is involved in EMT induction via transforming growth factor-β1(TGF-β1).In this study,a detailed elemental analysis(55 elements),particle size,and morphology were determined.To investigate the toxicity of SPM,an MTT test was performed to detect cell viability.Next,A549cells were exposed to selected concentrations of SPM(20 and 40μg/mL)for single and repeated exposures.The DCFH-DA assay showed that exposure to SPM could produce reactive oxygen species(ROS).The ELISA assay demonstrated increased levels of interleukin-8(IL-8)and TGF-β1 in the supernatant.Western blot was used to detect the expression of proteins associated with EMT and the SMAD3-dependent pathway.Results of western blot demonstrated that E-cadherin was reduced,whereas p-SMAD3,vimentin,andα-smooth muscle actin were elevated.Our findings indicated that SPM triggered EMT by induction of oxidative stress,inflammation,and the TGF-β1/SMAD3 pathway activation. 展开更多
