为了研究链球菌的分子发病机制,试验筛选了马链球菌随机基因组文库,得到1个阳性克隆,对拯救质粒进行测序,确定插入片段编码成熟蛋白阅读框架的分子质量为39.6 ku,经序列分析确定为IgG内切酶蛋白(IdeE),与白细胞C3b受体Mac(CD11b)具有同源性,设计特异性引物PCR扩增该基因,并对该蛋白进行克隆表达和功能分析。结果表明:该蛋白不能分解IgG,但可以结合嗜中性粒细胞,呈现剂量依赖性抗吞噬活性。

functional analysis Airstract: To study the molecular pathogenesis of Streptococcus equi, one positive clone was obtained using the method for screening random ge- nomic library of Streptococcus equi. The molecular mass of the open reading frame encoding the mature protein for the inserted segement was de- termined as 39.6 ku by sequencing the rescued plasmids. It was confirmed by sequence analysis to be a igg endopoptidase (IdeE) protein, which was homologous to leukocyte C3b receptor Mac (CD11 b). Based on the designed specific primers, the IdeE gene was amplified by PCR, and then cloned and used for the protein functional analysis. The results showed that recombinant IdeE protein could not cleave igg, hut could bind neutrophil to show a dose -dependent anti -phagocytic activity.
