Adrenaline in pulp capping treatment of reversible pulpitis

BACKGROUND The role of epinephrine in the treatment of pulp capping in patients with reversible pulpitis is not clear.AIM To explore the role of epinephrine in the treatment of pulp capping in patients with reversible pulpitis.METHODS A total of 100 patients with reversible pulpitis who were treated in Anhui Jieshou People's Hospital from January 2020 to December 2021 were included in the study.They were categorized into an observation group(n=50;treatment with adrenaline)and a control group(n=50;treatment with zinc oxide eugenol paste).The 24-h postoperative pain,regression time of gingival congestion and redness,clinical efficacy,and incidence of adverse reactions were compared between the groups.Patients were further categorized into the ineffective and effective treatment groups based on clinical efficacy.Logistic multiple regression analysis explored factors affecting the efficacy of pulp capping treatment.RESULTS A significant difference in 24-h postoperative pain was observed between the groups(P<0.05),with a higher proportion of grade I pain noted in the observation group than in the control group(P<0.01).The regression time of gingival congestion and swelling was lower in the observation group(2.61±1.44 d and 2.73±1.36 d,respectively)than in the control group(3.85±1.47 d and 4.28±1.61 d,respectively)(P<0.05).The 2-wk postoperative total effective rate was lower in the control group(80.00%)than in the observation group(94.00%)(P<0.05).The incidence of adverse reactions was not significantly different between the control(14.00%)and observation(12.00%)groups(P>0.05).The proportion of adrenaline usage was lower(P<0.05)and that of anaerobic digestion by Streptococcus and Fusobacterium nucleatum was higher in the ineffective treatment group than in the effective treatment group(P<0.05).Logistic multiple regression analysis revealed adrenaline as a protective factor(P<0.05)and anaerobic digestion by Streptococcus and F.nucleatum as risk factors for pulp capping in reversible pulpitis(P<0.05).CONCLUSION Adrenaline demonstrated therapeutic efficacy in pulp capping treatment for reversible pulpitis,reducing pain and improving clinical symptoms safely.It is a protective factor for pulp capping,whereas Streptococcus and F.nucleatum are risk factors.Targeted measures can be implemented to improve clinical efficacy. 展开更多

旨在建立检测猪圆环病毒3型(PCV3)抗体的阻断ELISA方法,本研究利用原核表达的PCV3Cap重组蛋白免疫BALB/c小鼠制备获得了一株分泌阻断效果良好抗体的杂交瘤细胞株2E6。以重组Cap蛋白作为包被抗原,以辣根过氧化物酶(HRP)标记的2E6单克隆... 旨在建立检测猪圆环病毒3型(PCV3)抗体的阻断ELISA方法,本研究利用原核表达的PCV3Cap重组蛋白免疫BALB/c小鼠制备获得了一株分泌阻断效果良好抗体的杂交瘤细胞株2E6。以重组Cap蛋白作为包被抗原,以辣根过氧化物酶(HRP)标记的2E6单克隆抗体作为检测抗体,经条件优化后建立了一种检测PCV3抗体的阻断ELISA方法。用建立的阻断ELISA方法检测50份临床阴性血清,计算阻断率(PI)的临界值,以此来确定该方法的判定标准:当PI≤28.30%时,判定结果为阴性;当PI≥35.05%时,判定结果为阳性;当28.30%<PI<35.05%时,判定为可疑,重复一次试验后如果结果仍为可疑,则判定为阳性。特异性试验表明该方法与猪圆环病毒2型(PCV2)、猪伪狂犬病病毒(PRV)、猪繁殖与呼吸综合征病毒(PRRSV)以及猪瘟病毒(CSFV)的阳性血清均无交叉反应;敏感性试验表明其检测效价可达到1:128;重复性试验表明批内与批间的变异系数均小于10%;符合性检验表明该方法与PCV检测金标准免疫过氧化物酶单层试验(IPMA)比对的Kappa值达0.9,具有高度的一致性。综上所述,本研究建立的阻断ELISA方法具有良好的特异性与较高的符合率,可用于后期进行PCV3抗体的检测,为PCV3的流行病学调查与临床诊断提供技术支持。 展开更多

猪圆环病毒2d基因型(Porcine circovirus type 2d,PCV2d)为猪场当前流行的PCV2优势基因型,为了研究针对PCV2d基因型的病毒样颗粒(Virus-like particle,VLP)疫苗,提高猪圆环病毒病的防控效果。本研究利用杆状病毒表达系统表达了能够自组... 猪圆环病毒2d基因型(Porcine circovirus type 2d,PCV2d)为猪场当前流行的PCV2优势基因型,为了研究针对PCV2d基因型的病毒样颗粒(Virus-like particle,VLP)疫苗,提高猪圆环病毒病的防控效果。本研究利用杆状病毒表达系统表达了能够自组装为VLP的PCV2d-Cap蛋白,然后将9头21日龄健康仔猪随机分为VLP组、攻毒对照组和空白对照组(n=3)。VLP组的每头仔猪经颈部肌肉注射400μg Cap蛋白-佐剂复合物(PCV2d-VLP),攻毒组注射等体积的PBS与佐剂混合物,空白组注射等体积的PBS。共接种2次,每次间隔14 d。于第2次免疫后21 d,VLP组和攻毒对照组的仔猪通过鼻腔感染PCV2 HBDX-2018株(106TCID50/头),评价PCV2d-VLP诱导的免疫效果。结果表明PCV2d-VLP能够诱导仔猪产生高水平的特异性IgG抗体和中和抗体,刺激IFN-γ水平升高,引起外周血淋巴细胞增殖能力增强,降低病毒经鼻腔和直肠的排毒率及病毒血症阳性率,减轻腹股沟淋巴结和脾脏的病理损伤,降低组织中的病毒载量。上述研究表明,应用杆状病毒表达的PCV2d-Cap蛋白能自组装为VLP,并能诱导仔猪产生良好的免疫保护效应,具有进一步开发为PCV2d-VLP疫苗的潜力。 展开更多

为建立一种针对鸽圆环病毒(PiCV)的快速诊断方法,试验根据PiCV的Cap保守基因序列,设计合成一对特异性引物,建立了PiCV荧光定量PCR检测方法,对其敏感性、特异性、重复性进行评价并利用该方法对临床样本进行检测。结果表明:在最佳反应条件... 为建立一种针对鸽圆环病毒(PiCV)的快速诊断方法,试验根据PiCV的Cap保守基因序列,设计合成一对特异性引物,建立了PiCV荧光定量PCR检测方法,对其敏感性、特异性、重复性进行评价并利用该方法对临床样本进行检测。结果表明:在最佳反应条件下,所建立的PiCV荧光定量PCR方法在1×10^(2)~1×10^(7) copies/μL标准品范围内具有良好的线性相关性,线性相关系数为0.9846;敏感性试验结果显示,该方法最低可检测到1×10^(2) copies/μL标准品,是常规PCR检测方法的100倍;特异性试验结果显示,该方法与其他常见的鸽病病原不发生交叉反应;重复性评价结果显示,批内与批间的变异系数均小于2%;所建立PiCV荧光定量PCR方法对临床病死鸽肝脏样品检测结果显示,PiCV阳性率达75.7%,高于常规PCR方法的检测阳性率(54.1%),说明该方法具有良好的适用性。可应用于对PiCV的快速检测,为PiCV的及时防控提供有力技术支持。 展开更多

为研究能否用血凝和反向间接血凝方法定量检测重组杆状病毒表达的PCV2 Cap抗原,分别使用不同稀释液和不同的动物红细胞测定PCV2Cap抗原的凝集效价,同时使用PCV2Cap单抗致敏绵羊红细胞,检测PCV2Cap抗原、空杆状病毒、CSFV等的反向间接血... 为研究能否用血凝和反向间接血凝方法定量检测重组杆状病毒表达的PCV2 Cap抗原,分别使用不同稀释液和不同的动物红细胞测定PCV2Cap抗原的凝集效价,同时使用PCV2Cap单抗致敏绵羊红细胞,检测PCV2Cap抗原、空杆状病毒、CSFV等的反向间接血凝效价(RIHA效价)。结果显示,用猪、鸡、绵羊红细胞在不同稀释液条件下测得的同一批次PCV2Cap抗原凝集效价为3log2~9log2,说明PCV2Cap抗原可凝集猪、鸡、绵羊红细胞,但红细胞不同、稀释液不同,测得的PCV2Cap抗原凝集效价也不同。使用PCV2Cap单抗致敏绵羊红细胞,测得的PCV2Cap抗原RIHA效价为14log2,且纯化后和未纯化的PCV2Cap抗原的RIHA效价均与ELISA定量检测结果有平行关系;空杆状病毒、CSFV、PRRSV、PRV、PEDV、TCEV的RIHA效价均小于1log2。说明反向间接血凝方法可定量检测重组杆状病毒表达的PCV2Cap抗原,该方法敏感、特异,适用于疫苗生产中的PCV2Cap抗原的定量检测。 展开更多

【目的】原核截短表达猪圆环病毒3型(Porcine circovirus type 3,PCV3)四川株Cap蛋白(1-197位氨基酸),并建立其抗体间接ELISA检测方法,为PCV3血清学调查提供材料。【方法】以PCV3四川分离株基因组为模板,PCR扩增获得Cap蛋白编码基因片段... 【目的】原核截短表达猪圆环病毒3型(Porcine circovirus type 3,PCV3)四川株Cap蛋白(1-197位氨基酸),并建立其抗体间接ELISA检测方法,为PCV3血清学调查提供材料。【方法】以PCV3四川分离株基因组为模板,PCR扩增获得Cap蛋白编码基因片段,将其克隆至原核表达载体pET-30a(+)中构建重组质粒pET30a-PCV3-Cap。经酶切和测序鉴定后,将重组质粒转化大肠杆菌BL21(DE3)感受态细胞,IPTG诱导表达,Ni-NTA树脂亲和层析纯化,获得具有良好抗原性的重组Cap蛋白。基于纯化的重组Cap蛋白建立间接ELISA方法,确定最佳抗原包被浓度、待检血清最佳稀释比例、酶标抗体稀释比例和阴阳性临界值,对间接ELISA方法的特异性、敏感性、重复性、相关性和符合率进行分析,并对2018-2022年采自川东北地区猪场的351份临床猪血清进行检测,分析该区域PCV3流行情况。【结果】试验成功构建pET30a-PCV3-Cap重组表达载体,实现重组Cap蛋白(1-197位氨基酸)的原核截短表达,蛋白大小约为26 ku,可与PCV3阳性猪血清发生特异性反应;确定最佳抗原包被浓度为1 ng/μL,待检血清最佳稀释比例为1∶100,酶标抗体稀释比例为1∶60000,阴阳性临界值D_(450 nm)为0.684;建立的间接ELISA方法与猪瘟病毒(CSFV)、猪繁殖与呼吸综合征病毒(PRRSV)、猪圆环病毒2型(PCV2)、猪伪狂犬病病毒(PRV)、日本乙型脑炎病毒(JEV)和猪细小病毒1型(PPV1)阳性猪血清均不发生交叉反应,敏感度达1∶1600,批内变异系数和批间变异系数均<10%,与病毒中和试验(VNT)结果呈正相关,与Western blotting方法的阳性符合率为95.8%;2018-2022年川东北地区的351份猪血清样品阳性率为7.12%。【结论】本研究成功截短表达了PCV3四川株Cap蛋白(1-197位氨基酸),建立了特异性强、灵敏度高、重复性好和符合率高的PCV3间接ELISA方法,为PCV3的血清学调查和检测试剂盒的开发提供了材料。 展开更多

为了制备猪圆环病毒2型(Porcine circovirus type 2,PCV-2)Cap蛋白单克隆抗体,试验将Cap基因克隆至原核表达载体pET-28a(+)上并转化至大肠杆菌BL21(DE3)感受态细胞中,将获得的重组表达菌pET28a-Cap/BL21(DE3)用IPTG进行诱导后表达重组蛋... 为了制备猪圆环病毒2型(Porcine circovirus type 2,PCV-2)Cap蛋白单克隆抗体,试验将Cap基因克隆至原核表达载体pET-28a(+)上并转化至大肠杆菌BL21(DE3)感受态细胞中,将获得的重组表达菌pET28a-Cap/BL21(DE3)用IPTG进行诱导后表达重组蛋白,利用His标签对表达的重组蛋白进行纯化;将纯化后的重组蛋白于皮下免疫Balb/c小鼠,免疫结束后测定免疫小鼠血清效价,当小鼠血清抗体效价达到1∶100000时进行细胞融合;采用有限稀释法进行亚克隆,筛选能够稳定分泌抗体且抗体效价高的阳性杂交瘤细胞株,鉴定杂交瘤细胞株抗体亚类,并选择1株抗体效价最高的细胞株再次注射到小鼠腹腔中(5×10^(5)个/只),取腹水采用亲和层析法对单克隆抗体进行纯化;采用间接ELISA方法测定单克隆抗体效价并检测其特异性,分别采用Western-blot和间接免疫荧光试验检测单克隆抗体的反应性。结果表明:PCR扩增得到大小为708 bp的Cap基因,经双酶切与测序验证后,成功构建重组表达质粒pET28a-Cap;重组表达菌pET28a-Cap/BL21(DE3)经诱导后表达的重组蛋白可与His标签抗体发生特异性反应,纯化后的重组蛋白在预期位置(31.7 ku)出现清晰的单一条带;共筛选出8株能够稳定分泌抗体且抗体效价高的阳性单克隆细胞株(1E5、2A9、3C6、4B3、5F7、6D11、7A2、8G1株),抗体亚类鉴定均为IgG1亚类。其中7A2株抗体效价最高,用于单克隆抗体的制备;间接ELISA方法证实7A2株单克隆抗体仅与PCV-2 Cap蛋白发生反应,与猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)、猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)、猪瘟病毒(Classical swine fever virus,CSFV)、猪细小病毒(Porcine parvovirus,PPV)和猪伪狂犬病病毒(Porcine pseudorabies virus,PRV)不发生反应;Western-blot和间接免疫荧光试验均证实PCV-2 Cap蛋白可被7A2株单克隆抗体特异性识别。说明本试验成功制备出具有高纯度、强特异性、良好反应性的PCV-2 Cap蛋白单克隆抗体。 展开更多

目的内皮祖细胞(EPCs)在损伤和缺血组织的修复中起重要作用。前期研究表明,黏附在损伤区域的EPCs会受到生理性周期性张应变的作用,通过上调长链酯酰辅酶A合成酶1(Acsl1)促进线粒体脂肪酸代谢,进而促进EPCs的归巢能力。然而,Acsl1如何感... 目的内皮祖细胞(EPCs)在损伤和缺血组织的修复中起重要作用。前期研究表明,黏附在损伤区域的EPCs会受到生理性周期性张应变的作用,通过上调长链酯酰辅酶A合成酶1(Acsl1)促进线粒体脂肪酸代谢,进而促进EPCs的归巢能力。然而,Acsl1如何感受力学作用促进EPCs归巢的机制尚不明确。方法应用Flexcell张应变加载系统,对EPCs施加10%幅度,1.25 Hz频率的生理性周期性张应变24 h。应用原子力显微镜检测静止组和牵拉组的细胞硬度,利用高内涵细胞成像分析系统和结构照明显微镜检测F-actin的变化,再通过Co IP-MS/MS寻找Acsl1的关键互作蛋白并验证。结果与静态组相比,张应变组EPCs的硬度显著上升。100倍油镜下张应变组EPCs的F-actin微丝更致密且变长,提示张应变促进了EPCs的骨架重塑。过表达Acsl1后,Co IP-MS/MS检测到潜在的互作蛋白,将其中与细胞骨架相关的蛋白分别与Acsl1进行分子对接,预测结果提示,F-actin的加帽蛋白CAPZA1与Acsl1互作的可能性最大,且Co IP验证二者确实存在互作。结论CAPZA1主要抑制F-actin的聚合,周期性张应变可能通过上调Acsl1水平,从而竞争性结合与F-actin互作的CAPZA1,进而促进F-actin的聚合,导致细胞骨架重塑,最终促进EPCs的归巢。 展开更多

It has always been challenging work to reconcile the contradiction between the strength and plasticity of titanium materials.Laser powder bed fusion(LPBF) is a convenient method to fabricate innovative composites including those inspired by gradient layered materials.In this work,we used LPBF to selectively prepare Ti N/Ti gradient layered structure(GLSTi)composites by using different N_(2)–Ar ratios during the LPBF process.We systematically investigated the mechanisms of in-situ synthesis Ti N,high strength and ductility of GLSTi composites using microscopic analysis,TEM characterization,and tensile testing with digital image correlation.Besides,a digital correspondence was established between the N_(2) concentration and the volume fraction of LPBF in-situ synthesized Ti N.Our results show that the GLSTi composites exhibit superior mechanical properties compared to pure titanium fabricated by LPBF under pure Ar.Specifically,the tensile strength of GLSTi was more than 1.5times higher than that of LPBF-formed pure titanium,reaching up to 1100 MPa,while maintaining a high elongation at fracture of 17%.GLSTi breaks the bottleneck of high strength but low ductility exhibited by conventional nanoceramic particle-strengthened titanium matrix composites,and the hetero-deformation induced strengthening effect formed by the Ti N/Ti layered structure explained its strength-plasticity balanced principle.The microhardness exhibits a jagged variation of the relatively low hardness of 245 HV0.2 for the pure titanium layer and a high hardness of 408 HV0.2 for the N_(2) in-situ synthesis layer.Our study provides a new concept for the structure-performance digital customization of 3D-printed Ti-based composites. 展开更多

The function of solid electrolytes and the composition of solid electrolyte interphase(SEI)are highly significant for inhibiting the growth of Li dendrites.Herein,we report an in-situ interfacial passivation combined with self-adaptability strategy to reinforce Li_(0.33)La_(0.557)TiO_(3)(LLTO)-based solid-state batteries.Specifically,a functional SEI enriched with LiF/Li_(3)PO_(4) is formed by in-situ electrochemical conversion,which is greatly beneficial to improving interface compatibility and enhancing ion transport.While the polarized dielectric BaTiO_(3)-polyamic acid(BTO-PAA,BP)film greatly improves the Li-ion transport kinetics and homogenizes the Li deposition.As expected,the resulting electrolyte offers considerable ionic conductivity at room temperature(4.3 x 10~(-4)S cm^(-1))and appreciable electrochemical decomposition voltage(5.23 V)after electrochemical passivation.For Li-LiFePO_(4) batteries,it shows a high specific capacity of 153 mA h g^(-1)at 0.2C after 100 cycles and a long-term durability of 115 mA h g^(-1)at 1.0 C after 800 cycles.Additionally,a stable Li plating/stripping can be achieved for more than 900 h at 0.5 mA cm^(-2).The stabilization mechanisms are elucidated by ex-situ XRD,ex-situ XPS,and ex-situ FTIR techniques,and the corresponding results reveal that the interfacial passivation combined with polarization effect is an effective strategy for improving the electrochemical performance.The present study provides a deeper insight into the dynamic adjustment of electrode-electrolyte interfacial for solid-state lithium batteries. 展开更多

This study offers significant insights into the multi-physics phenomena of the SLM process and the subsequent porosity characteristics of ZK60 Magnesium(Mg)alloys.High-speed in-situ monitoring was employed to visualise process signals in real-time,elucidating the dynamics of melt pools and vapour plumes under varying laser power conditions specifically between 40 W and 60 W.Detailed morphological analysis was performed using Scanning-Electron Microscopy(SEM),demonstrating a critical correlation between laser power and pore formation.Lower laser power led to increased pore coverage,whereas a denser structure was observed at higher laser power.This laser power influence on porosity was further confirmed via Optical Microscopy(OM)conducted on both top and cross-sectional surfaces of the samples.An increase in laser power resulted in a decrease in pore coverage and pore size,potentially leading to a denser printed part of Mg alloy.X-ray Computed Tomography(XCT)augmented these findings by providing a 3D volumetric representation of the sample internal structure,revealing an inverse relationship between laser power and overall pore volume.Lower laser power appeared to favour the formation of interconnected pores,while a reduction in interconnected pores and an increase in isolated pores were observed at higher power.The interplay between melt pool size,vapour plume effects,and laser power was found to significantly influence the resulting porosity,indicating a need for effective management of these factors to optimise the SLM process of Mg alloys. 展开更多

The poor electrochemical performance of all-solid-state batteries(ASSBs),which is assemblied by Ni-rich cathode and poly(ethylene oxide)(PEO)-based electrolytes,can be attributed to unstable cathodic interface and poor crystal structure stability of Ni-rich cathode.Several coating strategies are previously employed to enhance the stability of the cathodic interface and crystal structure for Ni-rich cathode.However,these methods can hardly achieve simplicity and high efficiency simultaneously.In this work,polyacrylic acid(PAA)replaced traditional PVDF as a binder for cathode,which can achieve a uniform PAA-Li(LixPAA(0<x≤1))coating layer on the surface of single-crystal LiNi_(0.83)Co_(0.12)Mn_(0.05)O_(2)(SC-NCM83)due to H^(+)/Li^(+)exchange reaction during the initial charging-discharging process.The formation of PAA-Li coating layer on cathode can promote interfacial Li^(+)transport and enhance the stability of the cathodic interface.Furthermore,the partially-protonated surface of SC-NCM83 casued by H^(+)/Li^(+)exchange reaction can restrict Ni ions transport to enhance the crystal structure stability.The proposed SC-NCM83-PAA exhibits superior cycling performance with a retention of 92%compared with that(57.3%)of SC-NCM83-polyvinylidene difluoride(PVDF)after 200 cycles.This work provides a practical strategy to construct high-performance cathodes for ASSBs. 展开更多

Conservation programs require rigorous evaluation to ensure the preservation of genetic diversity and viability of conservation populations. In this study, we conducted a comparative analysis of two indigenous Chinese chicken breeds, Gushi and Xichuan black-bone, using whole-genome SNPs to understand their genetic diversity, track changes over time and population structure. The breeds were divided into five conservation populations(GS1, 2010, ex-situ;GS2, 2019, ex-situ;GS3, 2019, in-situ;XB1, 2010, in-situ;and XB2, 2019, in-situ) based on conservation methods and generations. The genetic diversity indices of three conservation populations of Gushi chicken showed consistent trends, with the GS3 population under in-situ strategy having the highest diversity and GS2 under ex-situ strategy having the lowest. The degree of inbreeding of GS2 was higher than that of GS1 and GS3. Conserved populations of Xichuan black-bone chicken showed no obvious changes in genetic diversity between XB1 and XB2. In terms of population structure, the GS3 population were stratified relative to GS1 and GS2. According to the conservation priority, GS3 had the highest contribution to the total gene and allelic diversity in GS breed, whereas the contribution of XB1 and XB2 were similar. We also observed that the genetic diversity of GS2 was lower than GS3, which were from the same generation but under different conservation programs(in-situ and ex-situ). While XB1 and XB2 had similar levels of genetic diversity. Overall, our findings suggested that the conservation programs performed in ex-situ could slow down the occurrence of inbreeding events, but could not entirely prevent the loss of genetic diversity when the conserved population size was small, while in-situ conservation populations with large population size could maintain a relative high level of genetic diversity. 展开更多

Revealing the localized corrosion process of Mg alloy is considered as one of the most significant ways for improving its corrosion resistance.The reliable monitor should be high distinguishability and real-time in liquid environment.Herein,Mg-9Al-1Fe and Mg-9Al-1Fe-1Gd alloys were designed to highlight the impact of intermetallic on the corrosion behaviour.In-situ AFM with a special electrolyte circulation system and quasi-in-situ SEM observation were used to monitor the corrosion process of the designed alloys.SEM-EDS and TEM-SAED were applied to identify the intermetallic in the designed alloys,and their volta potentials were measured by SKPFM.According to the real-time and real-space in-situ AFM monitor,the corrosion process consisted of dissolution of anodicα-Mg phase,accumulation of corrosion products around cathodic phase and shedding of some fine cathodic phase.Then,the localized corrosion process of Mg alloy was revealed combined with the results of the monitor of corrosion process and Volta potential difference. 展开更多
