This study investigated the differences in heat production(HP), respiratory quotient(RQ) and plasma metabolites in pigs in the fed and fasted state, using the techniques of indirect calorimetry and metabolomics.Methods: Nine barrows(45.2 ± 1.7 kg BW) were fed corn-soybean based meal diets and were kept in metabolism crates for a period of 14 d. After 7 d adaptation, pigs were transferred to respiratory chambers to determine HP and RQ based on indirect calorimetry. Pigs were fed the diet at 2,400 k J ME/(kg BW0.6·d) during d 8 to 12. The last 2 d were divided into 24 h fasting and 48 h fasting treatment, respectively. Plasma samples of each pig were collected from the anterior vena cava during the last 3 d(1 d while pigs were fed and 2 d during which they were fasted).The metabolites of plasma were determined by high-resolution mass spectrometry using a metabolomics approach.Results: Indirect calorimetry analysis revealed that HP and RQ were no significant difference between 24 h fasting and 48 h fasting, which were lower than those of fed state(P < 0.01). The nitrogen concentration of urine tended to decrease with fasting(P = 0.054). Metabolomics analysis between the fed and fasted state revealed differences in15 compounds, most of which were not significantly different between 24 h fasting and 48 h fasting. Identified compounds were enriched in metabolic pathways related to linoleic acid metabolism, amino acid metabolism,sphingolipid metabolism, and pantothenate and Co A biosynthesis.Conclusion: These results suggest that the decreases in HP and RQ of growing pigs under fasting conditions were associated with the alterations of linoleic acid metabolism and amino acid metabolism. The integrative analysis also revealed that growing pigs under a 24-h fasting were more appropriate than a 48-h fasting to investigate the metabolic components of maintenance energy expenditure.展开更多
The multifunctionality and the advantages of thermal plasma for the fast inactivation of viable cells and degradation of organic compounds dissolved in waste water are presented.A complete bacterial inactivation proce...The multifunctionality and the advantages of thermal plasma for the fast inactivation of viable cells and degradation of organic compounds dissolved in waste water are presented.A complete bacterial inactivation process was observed and studied using a thermal plasma treatment source with very short application times,in particular for Staphylococcus aureus bundle spore survival.The survival curves and analyses of the experimental data of the initial and final densities of S.aureus bacteria show a dramatic inhibitory effect of the plasma discharge on the residual bacteria survival ratio.As the exposure time increased,the inactivation process rate increased for direct exposure more than it did for indirect exposure.The evaluation of direct and indirect exposure was based on the analysis of the ultraviolet spectrum from the absorbance spectra of the organic compound dye called benzene sulfonate(C(16)H(11)N2Na O4S)and of viable cells called S.aureus.Organic compounds were degraded and viable cells were killed in a short time by thermal plasma.Moreover,analyses of total carbon,total organic carbon,and total inorganic carbon showed a fast decrease in organically bound carbon,however,this was not as fast as the absorbance spectra revealed by the exposure time increasing more for direct exposure than indirect exposure.After 100 s of exposure to the organic compound dye the removal had a maximun of 40%for samples with indirect exposure to the plasma and a maximum of 90%for samples with the direct exposure.For both samples,where some organic contaminants still remained in treated water,four electrolytes(KCl,Na Cl,Na2SO4,and CH3COONa)were added to be effective for complete sterilization,reaching a purity of 100%.A proposal is made for an optimized thermal plasma water purification system(TPWPS)to improve fast inactivation of microbes and the degradation of organic compounds dissolved in water(especially for direct exposure rather than indirect exposure)using a hybrid plasma torch with an electrical power of 125 kW(500 V–250 A)producing a high-temperature(10 000 K–19 000 K)plasma jet with a maximum gas consumption of 28 mg s^-1.展开更多

Plasma-activated phosphate-buffered saline (PBS) causes rapid inactivation of bacteria following generation of oxidative stress. However, dose optimization requires understanding of cellular mechanisms. The objective of this study was to explore genome-wise response to develop gene expression profile of Escherichia coli using DNA microarray following exposure to plasma-activated PBS solution. Upon exposure to plasma-treated PBS solution, E. coli cells had differentially expressed genes involved in oxidative stress, and cell envelope and membrane associated porin and transporters. The genes involved in house-keeping and metabolism, energy generation, motility and virulence were conversely downregulated. This is the first report which demonstrates a severe oxidative stress induced in E. coli cells in response to an exposure to nonequilibrium nonthermal dielectric-barrier discharge plasma-activated PBS solution, and the genes that are responsive to reactive oxygen species appeared to play a role in cellular stress. Such studies are important to identify targets of inactivation, and to understand plasma-treated solution and bacterial cell interactions.展开更多

Such emissions are due to the ...Optical emissions from the major and trace elements embodied in a transparent gel prepared from cooking oil were detected after the gel was spread in a thin film on a metallic substrate. Such emissions are due to the indirect breakdown of the coating layer. The generated plasma, a mixture of substances from the substrate, the layer, and the ambient gas, was characterized using emission spectroscopy. The characteristics of the plasma formed on the metal with and without the coating layer were investigated. The results showed that A1 emission induced from the aluminum substrates coated with oil films extends away from the target surface to ablate the oil film. This finally formed a bifurcating circulation of aluminum vapor against a spherical confinement wall in the front of the plume, which differed from the evolution of the plasma induced from the uncoated aluminum target. The strongest emissions of elements from the oil films can be observed at 2 mm above the target after a detection delay of 1.0μs. A high temperature zone has been observed in the plasma after the delay of 1.0 μs for the plasma induced from the coated metal. This higher temperature determined in the plasma allows the consideration of the sensitive detection of trace elements in liquids, gels, biological samples, or thin films.展开更多
