[Objective] This study aimed to obtain IL-10 (interleukin 10) full-length cDNA of common carp (Cyprinus carpio L.) and conduct the sequence analysis. [Method] The differentially expressed cDNA fragment was obtained by...[Objective] This study aimed to obtain IL-10 (interleukin 10) full-length cDNA of common carp (Cyprinus carpio L.) and conduct the sequence analysis. [Method] The differentially expressed cDNA fragment was obtained by DD-RTPCR (differential display RT-PCR). The cDNA library of peripheral blood leukocytes which were separated from common carp and stimulated by mitogen was screened with a probe labeled with DIG (digoxigenin). The IL-10 full-length cDNA was cloned from 0.8×104 pfu of recombinant phages, and the sequence analysis and homology comparison were carried out. [Result] Sequence analysis indicated that the IL-10 full-length cDNA of common carp was 1 117 bp long, containing a 55 bp 5’-UTR, a 522 bp 3’-UTR, and a 540 bp open reading frame(ORF) encoding 179 amino acids. In addition, there were three mRNA instability motifs (ATTTA) in the 3’-untranslated region. The deduced protein sequence shared typical sequence features of the IL-10 family. Homology comparison indicated that the obtained sequence shared 89.1% homology with the carp IL-10 gene from GenBank. [Conclusion] This study laid foundation for further study of the expression manner, functional characteristic and regulation mechanism of IL-10 in vivo and the interaction mechanism in the inflammatory reaction and immune response.展开更多
目的研究术前NLR家族Pyrin域蛋白3(NLR family pyrin domain-containing protein 3,NLRP3)、白细胞介素(Interleukin,ILs)表达谱特征对神经胶质瘤患者手术预后的影响。方法以2019年1月-2022年12月新疆医科大学第二附属医院神经外科收治...目的研究术前NLR家族Pyrin域蛋白3(NLR family pyrin domain-containing protein 3,NLRP3)、白细胞介素(Interleukin,ILs)表达谱特征对神经胶质瘤患者手术预后的影响。方法以2019年1月-2022年12月新疆医科大学第二附属医院神经外科收治的86例神经胶质瘤患者为研究对象,根据患者术后1年内随访情况将患者划分为手术预后良好组(n=52)和手术预后不良组(n=34)。对比两组患者一般临床资料及术前NLRP3、ILs(IL-1β、IL-4、IL-12、IL-17、IL-33)表达谱的表达水平差异。采用Spearman相关性分析,经单因素及多因素Logistic回归分析筛选神经胶质瘤患者手术预后不良的危险因素,通过受试者工作特征(Receiver operating characteristic,ROC)曲线评价各危险因素对患者手术预后的影响。结果与手术预后良好组比较,手术预后不良组患者肿瘤大小、肿瘤周围水肿患者比例、合并癫痫发作史患者比例、血清NLRP3、IL-1β、IL-4、IL-33水平均升高,术前卡氏功能状态评分、血清IL-12水平均降低,差异有统计学意义(P<0.05);Spearman相关性分析结果表明,肿瘤大小、肿瘤周围水肿、有癫痫发作史、血清中NLRP3、IL-1β、IL-4、IL-33水平均与手术预后不良呈正相关性,卡氏功能状态评分、IL-12与手术预后不良呈负相关性(P<0.05);单因素及多因素Logistic回归分析表明血清中NLRP3、IL-1β、IL-4水平均是神经胶质瘤患者手术预后不良的重要危险因素,IL-12是神经胶质瘤患者手术预后不良的重要保护因素(P<0.05);ROC曲线分析表明NLRP3、IL-1β、IL-4、IL-12独立及联合对于神经胶质瘤患者手术预后不良均具有较高预测效能(P<0.05)。结论神经胶质瘤患者NLRP3、ILs水平较高均是手术预后不良的重要危险因素,通过早期干预减轻神经胶质瘤患者炎症水平可能有利于改善手术治疗效果。展开更多

文摘目的探究血清瓜氨酸组蛋白H3(citrullinated histone H3,CitH3)、降钙素原(procalcitonin,PCT)及白细胞介素-10(interleukin-10,IL-10)对体外心肺复苏(extracorporeal cardiopulmonary resuscitation,ECPR)后急性肺损伤(acute lung injury,ALI)的预测价值。方法择取2021年1月至2024年4月行ECPR的患者126例,入院次日空腹采血测定患者血清CitH3、PCT、IL⁃10水平。根据ECPR后ALI发生与否分为ALI组与非ALI组,对比两组临床资料及血清CitH3、PCT、IL-10水平,Logistic多因素回归分析ECPR后ALI发生的危险因素。另外受试者工作特征(receive operating characteristic,ROC)曲线分析血清CitH3、PCT、IL-10对ALI的预测价值。结果因6例患者未配合完成相关检查等被剔除,最终纳入120例患者,其中住院期间ALI发生28例(ALI组),ALI未发生92例(非ALI组)。ALI组入院时急性生理学和慢性健康状况评分(Acute Physiology and Chronic Health Scoring System,APACHEⅡ)、序贯器官衰竭评估(Sequential Organ Failure Assessment,SOFA)、心肺复苏到自主循环恢复时间均大于非ALI组(P<0.05);ALI组血清CitH3、PCT、IL-10水平均高于非ALI组(P<0.05)。Logistic多因素回归分析显示心肺复苏到自主循环恢复时间、血清CitH3、PCT及IL-10是ALI发生的独立危险因素(P<0.05)。ROC曲线显示,血清CitH3、PCT、IL-10单一及3项联合预测ALI发生的曲线下面积(area under curve,AUC)分别为0.886、0.837、0.852、0.951,3项联合的AUC均大于PCT、IL-10单一检测(P<0.05)。结论ECPR后ALI发生患者血清CitH3、PCT、IL-10水平上升,对ALI发生有一定的预测价值,特别是联合检测效能更显著。
