TCM consists of Astragalus membranaceus, Morus alba, Crataegus pinnatifida, Alisma orientale, Salvia miltiorrhiza, and Pueraria lobata. METHODS: The rats were separated randomly into five groups. One (the CD group) was fed a control diet for 10 wk, another (the ED group) fed an ethanol-containing isocaloric liquid diet for 10 wk, and the last three (the TCM group) were fed an ethanol-containing isocaloric liquid diet for 10 wk and dosed orally with TCM (222 mg/kg.d, TCM222; 667 mg/kg·d, TCM667; and 2 000 mg/kg.d, TCM2000, respectively) weekly during the last 4 wk. RESULTS: ED group developed fatty liver according to lipid profile and liver histological findings. Compared with the control group, liver/body weight, serum triglyceride (TG) and total cholesterol (TC), liver TG and TC, serum alanine aminotransferase (ALT) and aspartic aminotransferase (AST) significantly increased in the ED group. Whereas, in the rats administered with TCM, liver/body weight, serum TG and TC, liver TG and TC, serum ALT and AST were significantly decreased, and the degree of hepatic lipid droplets was markedly improved compared with those in the ED group. CONCLUSION: TCM treatment causes significant reduction in alcohol-induced lipid hepatic accumulation, reversing fatty liver and liver damage, and can be used as a remedy for alcoholic fatty liver.展开更多

Methods: TIP was dissolved in 0.1 M NaOH and added to the culture medium at a final concentrations of 50 mg/L, 100 mg/L and 200 mg/L. Some cells (control) were exposed to 0.001 M NaOH. TIP was added to PC12 cells 30 min prior to the administration of MPP^+. TIP and MPP^+ remained in the culture medium for 96 h. D2 dopamine receptor mRNA, preproenkephalin mRNA and prodynorphin mRNA expressions were assayed by real-time quantitative reverse transcription-PCR. Results: The D2 dopamine receptor mRNA and preproenkephalin mRNA expressions were up-regulated in MPP^+ group compared with the control group, and prodynorphin mRNA expression was down-regulated in that. The D2 dopamine receptor mRNA expression being down-regulated and prodynorphin mRNA expression being up-regulated in TIP group compared with the MPP^+ group. And there was no effect of TIP on preproenkephalin gene expression in PC12 cells induced by MPP^+. OBJECTIVE This study aimed to optimize polysaccharides extraction from Urena lobata L.and investigate its antioxidant activity.METHODS The mathematical model was established by re.sponse surface method(RSM) based on t...OBJECTIVE This study aimed to optimize polysaccharides extraction from Urena lobata L.and investigate its antioxidant activity.METHODS The mathematical model was established by re.sponse surface method(RSM) based on the results of single factor experiments,using polysaccha.rides extraction rate as response value,and using the ratio of water to material,cellulase concentra.tion,extraction temperature and time as experimental factors,which was used to screen optimum poly.saccharide extraction conditions from Urena lobata L..Antioxidant activity of polysaccharides was stud.ied by DPPH and ·OH free radical elimination method.RESULTS The optimum conditions obtained by RSM were as follows:the cellulase level was 10.8 g·L^(-1),extraction time duration was 72 min,the ra.tio of water to feedstock was 7 mL·g^(-1),extraction temperature was 43℃,the pH value was 5.0.Under the optimal conditions,there was a difference of less than 5% between predicted extraction rate 13.37% and experimental extraction rate 13.32%.The polysaccharide yield was most significantly af.fected by cellulase concentration,followed by extraction time,water to material ratio and extraction tem.perature.IC50 of DPPH and ·OH were 1.082 g·L^(-1) and 3.202 mg·L^(-1),respectively.Antioxidant activity of sample polysaccharides was weaker than those of vitamin C.CONCLUSION The polysaccharide extraction process from Urena lobata L.by cellulase enzymolysis approach was obtained,which was convenient and feasible,and extracted polysaccharides had good free radical scavenging activity.展开更多
