The Chinese Herbal Formula Weichang’an Reduces the Proliferation of Human Gastric Cancer Cells via Mitochondrial Apoptosis

Objective The aim of the study was to investigate the effects of Chinese herbal formula Weichang’an(WCA)on the proliferation and mitochondria-mediated apoptosis of human gastric cancer cells.Methods Cell Counting Kit-8(CCK8)was used to evaluate the antiproliferative activity ofWCA on MKN45 cells;Giemsa staining was used to investigate cell colony formation;flow cytometry was used to analyze cell cycle,apoptosis rate,and caspases activation;and Hoechst staining was used to analyze the morphology of cell nuclei.The mitochondrial membrane potential was analyzed by both flow cytometric measurements and fluorescence microscopy.Western blot was used to analyze the protein levels of pro-caspase-3,Bcl-2,Bax,and Bcl-X.MKN45 cells were subcutaneously injected into the right forelimb of 16 nude mice to establish the tumor xenograft model,and a total of 12 nude mouse tumor xenograft models were successfully created.The nude mice were divided into the control group and the WCA-treated group.The two groups received normal saline and WCA treatment at 35.49 g/kg by a single daily oral gavage for 21 days.The body weight and tumor size of the nude mice were measured twice a week.The ultrastructural changes of subcutaneous tumors were observed by a transmission electron microscope.Results WCA can suppress cell proliferation and colony formation.It can also induce changes in the mitochondrial membrane potential and increase the activities of caspases-3,caspases-8,and caspases-9 in MKN45 cells.WCA induced S-phase arrest and apoptosis in MKN45 cells,and decreased the expression levels of antiapoptotic proteins Bcl-2 and Bcl-X in MKN45 cells,while increasing the expression levels of the proapoptotic proteins Bax and pro-caspase-3 compared with the control group.WCA inhibited the growth of a xenografted MKN45 tumor in nude mice,and the protein levels of Bax and pro-caspase-3 were significantly increased,while Bcl-X and Bcl-2 were reduced compared with the control group.The differences are statistically significant(p<0.05).Conclusion WCA could suppress the proliferation of gastric cancer cells via mitochondrial apoptosis. 展开更多

Background:Insomnia is a prevalent clinical condition and Shangxia Liangji formula(SXLJF)is a well-established method of treatment.Nevertheless,the specific mechanism of action of SXLJF remains unclear.Methods:The mouse model of insomnia was established by intraperitoneal injection of para-chlorophenylalanine.Forty-two mice were randomly divided into a negative control group,model group,SXLJF group(18.72 g/kg/day),and positive control group(diazepam,2 mg/kg)and treated with the corresponding drugs for 7 consecutive days.The open field test and pentobarbital-induced sleeping test were conducted.LC-MS-based untargeted metabolomics and network pharmacology were applied to explore the potential targets of SXLJF for treating insomnia.Finally,key targets were validated using RT-qPCR.Results:Behavioral tests demonstrated that SXLJF reduced the total distance,average velocity,central distance,and sleep latency,and prolonged sleep duration.Metabolomics and network pharmacology revealed potential targets,signaling pathways,metabolic pathways,and metabolites associated with the anti-insomnia effects of SXLJF.Specifically,tyrosine hydroxylase(TH)and tyrosine metabolism emerged as crucial metabolic pathways and targets,respectively.RT-qPCR results supported the role of TH in the mechanism of SXLJF in treating insomnia.Conclusion:In conclusion,TH and tyrosine metabolism may represent significant targets and pathways for SXLJF in treating insomnia. 展开更多

为研究氧化铟锡(indium tin oxide,ITO)导电玻璃材料的去除机理,采用单磨粒对材料进行切削仿真,建立了ITO导电玻璃的材料模型,根据加工表面形貌、应力和切削力情况分析了材料去除机理,之后研究了切削参数对切削力和残余应力的影响,并与... 为研究氧化铟锡(indium tin oxide,ITO)导电玻璃材料的去除机理,采用单磨粒对材料进行切削仿真,建立了ITO导电玻璃的材料模型,根据加工表面形貌、应力和切削力情况分析了材料去除机理,之后研究了切削参数对切削力和残余应力的影响,并与钠钙玻璃进行对比分析。结果表明:在磨粒的切削过程中,材料的去除受ITO薄膜层、玻璃基底和内聚力接触行为的共同影响,会产生分层、通道开裂和层间断裂等失效形式;随着磨粒的进给,切削力在一定范围内波动,且呈现上升、稳定、降低的变化,同时磨粒的切削力与切削速度和切削深度呈正相关;薄膜上残余应力相比玻璃基底,数值更大且波动更剧烈;当切削深度接近ITO薄膜厚度时,薄膜的存在对磨粒切削行为的影响显著。 展开更多

基于ITO玻璃较低的背景电流、免打磨预处理等优势,结合还原氧化石墨烯(RGO)与纳米金/聚吡咯纳米复合物(AuNPs/Ppy)的电控吸附能力,构建了一种可代替传统固体(金、玻碳)电极的新型电化学传感器(RGO/AuNPs/Ppy-ITO),用于测定水环境中的痕... 基于ITO玻璃较低的背景电流、免打磨预处理等优势,结合还原氧化石墨烯(RGO)与纳米金/聚吡咯纳米复合物(AuNPs/Ppy)的电控吸附能力,构建了一种可代替传统固体(金、玻碳)电极的新型电化学传感器(RGO/AuNPs/Ppy-ITO),用于测定水环境中的痕量Cr(Ⅵ)。利用修饰电极表面大量含氮结构与正电位下的电控吸附作用,采用差分脉冲溶出伏安法记录Cr(Ⅵ)的还原峰曲线,在经优化的pH、富集电压与时间等条件下,测得Cr(Ⅵ)还原峰电流与检测浓度在1~1500μg/L范围之内线性关系良好,相关系数R 2=0.9973,检测限为0.83μg/L。相较于金电极测试结果,不仅解决了金电极表面状态不稳定导致重现性较差的缺点,且具备更高的灵敏度与较低的成本耗材,在对水环境中Cr(Ⅵ)的现场快速、痕量一次性检测方面具有广阔的应用前景。 展开更多

目的通过双膜层结构设计制备ITO/SiO_(2)薄膜,研究了靶面磁场强度、镀膜工件移动速度、镀膜功率对ITO/SiO_(2)薄膜膜电阻均匀性的影响,并研究了优化条件制备的ITO/SiO_(2)薄膜物相、形貌和结构,并通过元素分布分析探讨了SiO_(2)薄膜的... 目的通过双膜层结构设计制备ITO/SiO_(2)薄膜,研究了靶面磁场强度、镀膜工件移动速度、镀膜功率对ITO/SiO_(2)薄膜膜电阻均匀性的影响,并研究了优化条件制备的ITO/SiO_(2)薄膜物相、形貌和结构,并通过元素分布分析探讨了SiO_(2)薄膜的作用。方法利用磁控溅射法在TN玻璃基板上沉积生成SiO_(2)薄膜,然后再沉积氧化铟锡(ITO)薄膜,制备ITO/SiO_(2)薄膜样品。利用X射线衍射仪(XRD)、ST-21L型薄膜膜电阻测试仪、原子力显微镜(AFM)、透射电镜(TEM)等仪器,研究了靶面磁场强度、镀膜工件移动速度、镀膜功率对ITO/SiO_(2)薄膜膜电阻均匀性的影响,并研究了优化条件制备的ITO/SiO_(2)薄膜样品的物相结构、表面形貌、截面膜层结构、元素分布与有膜电阻均匀性的关系,探讨了SiO_(2)薄膜结构与晶粒尺寸效应可能发挥的作用。结果(1)磁控溅射优化条件下,磁场强度为780~820 Gs,镀膜工件移动速度为1.2 m/min,镀膜功率为2.5 kW(A21)和3 kW(A23)时,ITO/SiO_(2)薄膜膜电阻极差最小为10~11Ω/sq,平均值为75~76Ω/sq,ITO/SiO_(2)薄膜膜电阻的均匀性最好。(2)ITO/SiO_(2)薄膜表现出晶体谱线和非晶谱线的叠加,In_(2)O_(3)和SnO_(2)特征峰发生轻微左偏移现象,SiO_(2)特征峰较宽,说明薄膜中的SiO_(2)处于非晶态结构,且可能部分发生晶粒尺寸效应,以微晶或纳米晶的形式存在。(3)ITO/SiO_(2)薄膜表面形貌比起SiO_(2)薄膜更均匀、连续、平滑且较致密,且具有明显的双膜层结构,其中ITO薄膜表面均匀平整且膜厚均匀,这与膜电阻均匀性一致;SiO_(2)薄膜与ITO薄膜和玻璃基底都形成了界面层,应该也是ITO薄膜结合力较好的原因;In元素的流失受到一定阻隔,应该与薄膜中的SiO_(2)的非晶态结构或发生晶粒尺寸效应,以及多晶ITO结构有关。结论通过优化控制靶面磁场强度、镀膜工件移动速度和镀膜功率等工艺因素,可以提高ITO/SiO_(2)薄膜膜电阻均匀性,同时通过控制SiO_(2)薄膜成膜质量可以改善ITO薄膜质量,并起到阻隔In元素流失的作用。 展开更多

Background:To observe the inhibitory effects of Yigan Jiangzhi formula(liver-supplementing lipid-lowering formula,YGJZF)on hepatic metastasis of colorectal cancer.Methods:The hepatic metastasis BALB-c mice model of colorectal cancer was established by injecting HCT-116 cell suspension following splenectomy.Next,we randomly divided BALB-c mice into 5 groups,which were blank group,model group(tumor only),traditional Chinese medicine group(YGJZF),western medicine group(capecitabine),combined group(YGJZF and capecitabine),respectively.After 14 days treatment,the effects of different treatments were evaluated the serological indicators were evaluated,including the levels of alanine transaminase,aspartate transaminase,triglyceride,total cholesterol and vascular endothelial growth factor(VEGF)by ELISA.Results:Except the blank group,the other groups showed diffuse hepatic metastasis.Compared with the model group,the total number and weight of the metastatic tumors as well as serological indicators in medicine-given groups were sharply reduced(P<0.05 for all)the combined group had better effects.The serum levels of these indicators showed no statistical difference among each medicine-given group(P>0.05),but the combined group has a remarkable decrease of all the indicators than in western medicine group.Conclusion:YGJZF not only can inhibit the hepatic metastasis of colorectal cancer in BALB-c mice,but also affect the levels of hepatic enzymes and lipids.Meanwhile,it can reduce the expression of VEGF.All these results indicate that YGJZF,which lowers down lipids and protects hepatic functions,has the potential to block VEGF pathway,contributing to the inhibition of hepatic metastasis. 展开更多

采用射频与直流磁控交替溅射法在石英玻璃载玻片上制备了氧化铟锡(ITO)/Cu/Al掺杂ZnO(AZO)(45 nm/10 nm/45 nm)组合结构的透明导电薄膜,并在不同退火温度下对薄膜进行真空热处理。利用X射线衍射仪(XRD)、紫外-可见分光光度计、四探针电... 采用射频与直流磁控交替溅射法在石英玻璃载玻片上制备了氧化铟锡(ITO)/Cu/Al掺杂ZnO(AZO)(45 nm/10 nm/45 nm)组合结构的透明导电薄膜,并在不同退火温度下对薄膜进行真空热处理。利用X射线衍射仪(XRD)、紫外-可见分光光度计、四探针电阻测试仪等表征手段,系统地研究了退火温度对ITO/Cu/AZO复合薄膜晶体结构和光电性能的影响。结果显示,经过不同温度的真空退火处理,薄膜的晶体结构和导电性能得到显著改善和提高,薄膜可见光平均透过率随着退火温度的升高先增加后降低。对比发现,在气压5×10^(-3)Pa、温度150℃下退火制备的ITO/Cu/AZO结构薄膜表现出最佳的综合性能,薄膜具有较强的(222)和(440)晶面衍射峰,在400~800 nm光波范围平均透过率约为80.5%,电导率约为1.76×10^(3) S/cm,综合品质因数达到约2.12×10^(-3)/Ω。 展开更多

BACKGROUND Fanlian Huazhuo Formula(FLHZF)has the functions of invigorating spleen and resolving phlegm,clearing heat and purging turbidity.It has been identified to have therapeutic effects on type 2 diabetes mellitus(T2DM)in clinical application.Non-alcoholic fatty liver disease(NAFLD)is frequently diagnosed in patients with T2DM.However,the therapeutic potential of FLHZF on NAFLD and the underlying mechanisms need further investigation.AIM To elucidate the effects of FLHZF on NAFLD and explore the underlying hepatoprotective mechanisms in vivo and in vitro.METHODS HepG2 cells were treated with free fatty acid for 24 hours to induce lipid accumulation cell model.Subsequently,experiments were conducted with the different concentrations of freeze-dried powder of FLHZF for 24 hours.C57BL/6 mice were fed a high-fat diet for 8-week to establish a mouse model of NAFLD,and then treated with the different concentrations of FLHZF for 10 weeks.RESULTS FLHZF had therapeutic potential against lipid accumulation and abnormal changes in biochemical indicators in vivo and in vitro.Further experiments verified that FLHZF alleviated abnormal lipid metabolism might by reducing oxidative stress,regulating the AMPKα/SREBP-1C signaling pathway,activating autophagy,and inhibiting hepatocyte apoptosis.CONCLUSION FLHZF alleviates abnormal lipid metabolism in NAFLD models by regulating reactive oxygen species,autophagy,apoptosis,and lipid synthesis signaling pathways,indicating its potential for clinical application in NAFLD. 展开更多

随着HJT太阳电池的发展,对其氧化铟锡(ITO)薄膜的研究日益增多。通过直流磁控溅射法在玻璃衬底上制备ITO薄膜,研究了氧含量、溅射功率、沉积温度及溅射气压对ITO薄膜光电性能的影响。研究结果显示:1)ITO薄膜的光电性能对氧含量较为敏感... 随着HJT太阳电池的发展,对其氧化铟锡(ITO)薄膜的研究日益增多。通过直流磁控溅射法在玻璃衬底上制备ITO薄膜,研究了氧含量、溅射功率、沉积温度及溅射气压对ITO薄膜光电性能的影响。研究结果显示:1)ITO薄膜的光电性能对氧含量较为敏感,随着氧含量增加,ITO薄膜的电阻率也随之增加,而透过率则呈先上升后下降,然后基本保持不变的趋势;2)随着沉积温度升高,ITO薄膜的透过率也随之升高,而电阻率则呈先下降后上升的趋势;3)随着溅射气压的升高,ITO薄膜的电阻率呈上升趋势,而透过率则是先降低再略微升高;4)当氧含量在1.8%~2.0%,溅射功率在3000~4000 W,沉积温度在150~190℃,溅射气压在0.5~0.7 Pa时,ITO薄膜具有较优的光电性能。因此,在合理范围内提高沉积温度,其则会具有退火作用,有助于进一步改善ITO薄膜的光电性能。 展开更多

We discuss two applications of this result, which are useful in the study of SPDEs driven by a space-time Lévy noise with finite variance: a maximal inequality for the p-th moment of the stochastic integral, and the It&ocirc;representation theorem leading to a chaos expansion similar to the Gaussian case. 展开更多
