Using subspeciesspecific molecular markers ILP (intron length polymorphism) and Cheng's index, the indica-japonica differentiation was analyzed with special materials including 18 indica-japonica hybrid parents and 39 derived lines, which accumulated different wide compatibility and restoring genes by convergent cross method in 21 years spanning four breeding phases. The indica-japonica differentiation was detected on all tested loci in 57 materials. Among the 18 parental lines, 4 were japonica type, 5 japonicaclinous type, 8 indicaclinous type and one indica type. The japonica proportion indexes in indica restorer lines Minghui 63 and 9308 were 12.50 and 33.33 %, respectively, while that in japonica restorer line C418 was only 31.25%. Among the 39 derived lines from indica-japonica hybridization, one was japonica type, 11 japonicaclinous type, 20 indicaclinous type and 7 indica type. The japonica proportion index in Minghui 502 was only 10.42%. The results of indica and japonica classification by ILP molecular markers and Cheng's index were relatively consistent. The correlation coefficient between the japonica proportion index and morphology index was 0.794**, while that between the indica proportion index and morphology index was -0.7662**. ILP markers could be used to accurately detect the proportion of indica/japonica content in the genome of a rice variety. The results of indica-japonica differentiation analysis could make reasonable explanation for that the hybrids obtained from indica-japonica type restorer lines had obvious heterosis. This conclusion would provide important guidance in efficient use of beneficial genes of inter-subspecific hybrid rice.展开更多

文摘Loquat(Eriobotrya japonica Lindl.)is a subtropical evergreen fruit tree that produces fruits with abundant nutrients and medicinal components.Confirming suitable reference genes for a set of loquat samples before qRT-PCR experiments is essential for the accurate quantification of gene expression. In this study, eight candidate reference genes were selected from our previously published RNA-seq data, and primers for each candidate reference gene were designed and evaluated. The Cq values of the candidate reference genes were calculated by RT-qPCR in 31 different loquat samples, including 12 subgroups of developing or abiotic-stressed tissues.Different combinations of stable reference genes were screened according to a comprehensive rank, which was synthesized from the results of four algorithms, including the geNorm,NormFinder,BestKeeper andΔCt methods. The screened reference genes were verified by normalizing EjLGA1 in each subgroup. The obtained suitable combinations of reference genes for accurate normalization were GAPDH,EF1αand ACT for floral development;GAPDH,UBCE and ACT for fruit setting;EF1α,GAPDH and eIF2B for fruit ripening;ACT,EF1αand UBCE for leaves under heat stress;eIF2B,UBCE and EF1αfor leaves under freezing stress;EF1α,TUA and UBCE for leaves under salt stress;ACT,EF1αand eIF2B for immature pulp under freezing stress;ACT,UBCE and eIF2B for immature seeds under freezing stress;EF1α,eIF2B and UBCE for both immature pulp and seeds under freezing stress;UBCE,TUB and TUA for red-fleshed fruits under cold-storage stress;eIF2B,RPS3 and TUB for white-fleshed fruits under coldstorage stress;and eIF2B,UBCE and RPS3 for both red-and white-fleshed fruits under cold-storage stress. This study obtained different combinations of stable reference genes for accurate normalization in twelve subgroups of developing or abiotic-stressed tissues in loquat. To our knowledge, this is the first report to obtain stable reference genes for normalizing gene expression of abiotic-stressed tissues in E.japonica.The use of the three most stable reference genes could increase the reliability of future quantification experiments.

文摘Saccharina japonica gametophytes can survive a long period under unfavorable environmental conditions, while they also delay in growth and/or reproduction.Although the reproduction in delayed gametophyte of S.japonica was known to be strongly influenced by light intensity, light quality, and photoperiod,no previous studies have evaluated their interactive effects on gametogenesis. To evaluate these effects,we used an orthogonal experiment to expose delayed gametophytes of S.japonica to different light intensities, light qualities, and photoperiods for 12 days. The results showed that changes in light intensity rather than light quality and photoperiod significantly affected the relative growth rates of the delayed gametophytes.Blue light had the greatest promotion on reproduction rate. The optimal light conditions in the early vegetative growth phase in gametogenesis induction for the delayed gametophytes were at 60–80μmol photons m^(−2) s^(−1) with daylength of 12 or 16 hours under white or blue light. When the delayed gametophytes were maintained in a constant light condition from delayed state to gametogenesis, the beneficial photoperiods for vegetative growth and reproductive rate were both 16L(16 hours of light):8D(8 hours of dark). However, when the delayed S.japonica gametophytes achieve the optimal growth state during the first 6 days and then they were cultured at different light conditions for the following 6 days, the reproduction rate increased as the daylength decreased and attained a peak value in group of 8L:16D photoperiod, indicating that photoperiod adjustment at the transition period is crucial in the gametogenesis induction process of delayed gametophyte of S.japonica.

文摘Long non-coding RNAs(lncRNAs)regulate a variety of biological processes, including sexual reproduction and differentiation.Saccharina japonica,a commercially important brown alga in China, shows remarkable sexual dimorphism in haploid gametophytes. The sex of Saccharina japonica gametophytes is determined by UV sexual system. However,no results have been reported on the lncRNAs involved in the sex-related gene regulation of S.japonica.This study identified a number of lncRNAs and assessed their expression levels in male and female gametophytes.Among them,a total of 405 lncRNAs and 211 mRNAs showed differential expressions.Furthermore, the functions of target genes of differentially expressed lncRNAs(DELs)differed from those of differentially expressed genes(DEGs),suggesting that lncRNA may interact with other functional proteins,in addition to DEGs,to involve sex regulation in S.japonica.There were 32 and 90 potential cis-regulatory and trans-regulatory interactions between DELsDEGs,respectively.Five of these lncRNAs(LNC_002974,LNC_021059,LNC_038466,LNC_051584,and LNC_027400)interacted with putative male sex determination region(SDR)genes, suggesting that they act as regulators in gametophytes'sex regulation potentially.Findings from this study contribute to our understanding of the roles of lncRNAs in sex differentiation and lay the foundation for functional studies of candidate lncRNAs in the future.
文摘Three hitherto undescribed Stemona alkaloids, named stemajapines A-C(1-3),along with six known alkaloids(4-9),were isolated and identified from the roots of Stemona japonica(Blume)Miq.(Stemonaceae).Their structures were established by the analysis of the mass data,NMR spectra, and computational chemistry.Stemjapines A and B were degraded maistemonines without spiro-lactone ring and skeletal methyl from maistemonine.Concurrence of alkaloids 1 and 2 revealed an undescribed way to form diverse Stemona alkaloids.Bioassay results disclosed the anti-inflammatory natural constituents stemjapines A and C with IC_(50) values of 19.7 and 13.8μM,respectively,compared to positive control dexamethasone with 11.7μM.The findings may point out a new direction of Stemona alkaloids inaddition to its traditional antitussive and insecticide activities.

文摘[Objectives]This study was conducted to increase the reproduction coefficient of Japanese honeysuckle(Lonicera japonica)to keep the character of optimal benign.[Methods]The young leaves of medicinal Japanese honeysuckle were selected as explants, and MS was used as the basic culture medium.Suitable culture concentrations and conditions were screened through different concentration gradients of growth regulators and cytokinin.[Results]As the concentration of 6-BA in the culture medium increased, the browning rate increased, and the browning phenomenon occurred earlier. On the contrary,a lower concentration of 6-BA was suitable for the differentiation and growth of young leaves, and the browning response was slow. However,if the cultivation time was too long and the materials were not transferred in a timely manner, browning would also occur. The optimal combination of levels was obtained through a 3×3 orthogonal experiment(three parallel groups for each of 6-BA and NAA).The culture conditions included a constant temperature of 26℃and light intensity of 1200 lx.The optimal medium for inducing callus proliferation was MS+6-BA 0.5 mg/L+NAA 0.5 mg/L;and the optimal medium for inducing bud differentiation was MS+6-BA 1.0 mg/L+NAA 0.1 mg/L.[Conclusions]This study provides a theoretical basis for accelerating the development of the honeysuckle industry.

文摘Revealing the indica-japonica differentiation in parents of hybridization between indica and japonica rice and their derived lines can provide theoretical and practical bases for the breeding of practical inter-subspecific hybrid rice. This conclusion would provide important guidance in efficient use of beneficial genes of inter-subspecific hybrid rice.
