Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root growth.Endogenous hormones in leaf tips and bases had an impact on rooting. The activities of endogenous hormones mainly existed in leaf tips because they play a major role in the production and consumption of IAA and its ABA content increased during rooting. The ZR in leaf tips influenced the rooting of cuttings, especially in the callus formation and rooting stage. Leaf tips were the main source of GA.展开更多

However, the molecular mechanisms underlying phase change in fruit trees remain largely unknown. Shikimic acid (ShA) pathway is a main metabolic pathway closely related to the synthesis of hormones and many important secondary metabolites participating in plant phase change. So,whether ShA regulates phase change in plants is worth clarifying. Here, the distinct morphological characteristics and the underlying mechanisms of phase change in jujube (Ziziphus jujuba Mill.), an important fruit tree native to China with nutritious fruit and outstanding tolerance abiotic stresses, were clarified. A combined transcriptome and metabolome analysis found that ShA is positively involved in jujube(Yuhong’×Xing 16’) phase change. The genes in the upstream of ShA synthesis pathway (ZjDAHPS, ZjDHQS and ZjSDH), the contents of ShA and the downstream secondary metabolites like phenols were significantly upregulated in the phase change period. Further, the treatment of spraying exogenous ShA verified that ShA at a very low concentration (60 mg·L^(-1)) can substantially speed up the phase change and flowering of jujube and other tested plants including Arabidopsis, tomato and wheat. The exogenous ShA (60 mg·L^(-1)) treatment in jujube seedlings could increase the accumulation of endogenous ShA, enhance leaf photosynthesis and the synthesis of phenols especially flavonoids and phenolic acids, and promote the expression of genes (ZjCOs, ZjNFYs and ZjPHYs) involved in flowering pathway. Basing on above results, we put forward a propose for the underlying mechanism of ShA regulating phase change, and a hypothesis that ShA could be considered a phytohormone-like substance because it is endogenous, ubiquitous, movable and highly efficient at very low concentrations. Fertilization strategies should maximize the efficiency of fertilizers, including the choice of the optimal timing of N supply. ^15N-urea was applied to winter jujubes on Jinsixiaozao jujubes rootstock to evaluate the effect of application timing on Nstorage and remobilization in mature trees in pot culture. The treatments consisted of ground application before budding (BB), during fruit core-hardening stage (FCH), and fruit rapid-swelling stage (FRS). Nitrogen-use efficiency of treatments were significantly different, which were 2.42% (BB), 9.77% (FCH), and 9.01% (FRS) in the dormant and 5.20% (BB), 16.16% (FCH), and 10.30% (FRS) in the following full-bloom. N supply in the pre-harvest helped to increase N-reserves of trees and then translocate to the new growth organs the following year. The largest amount of ^15N was detected in the roots and trunks. In all the treatments, the partition rates were highest in coarse roots, which were 30.43% (BB), 38.61% (FCH), and 40.62% (FRS), respectively. ^15N stored in roots and trunks was used by jujube trees to sustain new growth in the following full-bloom. ^15N applied before budding resulted in lower Ndff% in perennial organs (trunks and coarse roots) sampled in the following full-bloom, but fine roots had highest Ndff% (1.28%). Other organs recovered similar amount of Ndff%. In contrast, FCH and FRS treatments led to higher Ndff% (4.01-5.15%) in the new growth organs (new growth branches, deciduous spurs, leaves and flowers), but lower Ndff% in perennial branches (1.49-2.89%). With the delay of ^15N-urea application time, ^15N increased the partitioning to roots. FCH treatment increased N-storage in perennial organ during winter, which should be remobilized to sustain new growth the following spring.展开更多
Jujube(Ziziphus jujube Mill.)is a traditional economic forest crop and is widely cultivated in hilly areas of the Loess Plateau,China.However,soil desiccation was discovered in jujube plantations.Pruning is recognized...Jujube(Ziziphus jujube Mill.)is a traditional economic forest crop and is widely cultivated in hilly areas of the Loess Plateau,China.However,soil desiccation was discovered in jujube plantations.Pruning is recognized as a water-saving method that can reduces soil water consumption.In this study,we monitored the jujube plots with control(CK),light(C1),medium(C2)and high(C3)pruning intensities during the jujube growing period of 2012-2015 to explore the effect of pruning intensity on soil moisture and water use efficiency(WUE)of jujube plantations in the hilly Loess Plateau Region.The results showed that pruning is an effective method for soil water conservation in jujube plantations.Soil moisture increased with increasing pruning intensity during the jujube growing period of 2012-2015.C1,C2 and C3 pruning intensities increased soil water storage by 6.1-18.3,14.4-40.0 and 24.3-63.3 mm,respectively,compared to CK pruning intensity.Pruning promoted soil moisture infiltration to deeper soil layer.Soil moisture infiltrated to soil depths of 240,280 and>300 cm under C3 pruning intensity,220,260 and 260 cm under C2 pruning intensity,200,240 and 220 cm under C1 pruning intensity,and 180,200 and 160 cm under CK pruning intensity in 2013,2014 and 2015,respectively.Soil water deficit was alleviated by higher pruning intensity.In 2013-2015,soil water change was positive under C2(6.4 mm)and C3(26.8 mm)pruning intensities but negative under C1(-20.5 mm)and CK(-40.6 mm)pruning intensities.Moreover,pruning significantly improved fresh fruit yield and WUE of jujube plants.Fresh fruit yields were highest under C1 pruning intensity with the values of 6897.1-13,059.3 kg/hm^2,which were 2758.4-4712.8,385.7-1432.1 and 802.8-2331.5 kg/hm2 higher than those under CK,C2,and C3 pruning intensities during the jujube growing period of 2012-2015,respectively.However,C3 pruning intensity had the highest WUE values of 2.92-3.13 kg/m3,which were 1.6-2.0,1.1-1.2 and 1.0-1.1 times greater than those under CK,C1 and C2 pruning intensities,respectively.Therefore,C3 pruning intensity is recommended to jujube plantations for its economic and ecological benefits.These results provide an alternative strategy to mitigate soil desiccation in jujube plantations in the hilly Loess Plateau Region,which is critical for sustainable cultivation of economic forest trees in this region.展开更多
Objective: To evaluate the lethal and emergence inhibitory effect of alkaloid, phenolic and terpenoid extracts of Ziziphus jujuba(Z. jujuba; Rhamnaceae) against Aedes aegypti(Diptera: Culicidae), and to explore the ef...Objective: To evaluate the lethal and emergence inhibitory effect of alkaloid, phenolic and terpenoid extracts of Ziziphus jujuba(Z. jujuba; Rhamnaceae) against Aedes aegypti(Diptera: Culicidae), and to explore the effect of the most effective fraction on developmental and biochemical parameters of the dengue vector. Methods: The fourth instar larvae of Aedes aegypti were exposed to alkaloid, phenolic and terpenoid extracts from Z. jujuba leaves to test their toxicity and emergence inhibitory effects. Phenolic extract, being the most effective was further tested against the mosquitoes for their growth inhibitory effect supported by biochemical changes in the parent and F1 generation. Results: While the different secondary metabolite fractions i.e., alkaloid, phenolics and terpenoid caused mortality at larval and pupal stages, the LC_(50) value was the lowest for phenolic fraction. Further study carried out with the phenolic fraction revealed that it affected growth by decreasing adult life span, fertility and fecundity of the mosquitoes. The reduction in growth was also accompanied by decrease in carbohydrate and lipid levels. Conclusions: It is concluded that the phenolic extract of the leaves of Z. jujuba is a potential candidate for control of Aedes mosquitoes.展开更多

The respiration rate, the weight change of single fruit during growth development and the losing water of postharvest of fruit were studied. The results showed that the curve of fruit growth development was a double sigmoid characteristic. When the surface colour of fruit changed form jade-green to alizarin crimson, the respiration rate tended to fall during a entire growth development of fruit, which showed a fluctuation phenomenon. The respiration rate descended when the surface color changed from coloring to baby red, but after a crimson stage the respiration rate recurred to its original downtrend. With increasing of single fruit weight, phenomenon of respiration climacteric has not happened and the respiration rate curve showed a concavity characteristic at some stages. As a result, there was not a phenomenon of respiration climacteric with a double sigmoid growth development curve characteristic of fruit.展开更多

In previous study, we have isolated and iden...The fruit of Ziziphus jujuba Mill., known as Hongzao(or Hong-Zao) in Chinese and cultivated in China for more than 4 000 years, has shown to have hepatoprotective property. In previous study, we have isolated and identified 27 known compounds from Z. jujuba fruits, which demonstrated anti-tumor activity. In this study, a high-performance liquid chromatography-diode-array detection-mass spectrometry(HPLC-DAD-MS) method was successfully applied to the simultaneous characterization and quantitation of 18 constituents in 28 Z. jujuba samples, comprised of 12 cultivars from different regions in China, by comparing their HPLC retention times, MS spectra, UV spectra, and NMR data with those of reference compounds. The quantitative method was validated with excellent linearity(R^(2 )> 0.999 1), preferable intra-and inter-day precisions(RSD < 2.78%), and good recoveries(94.96%–102.65%). The content variation of 18 compounds was analyzed by a chemometric method(hierarchical cluster analysis). In addition, these constituents showed protection against carbon tetrachloride(CCl_(4)) intoxicated Hep G2 cell lines by decreasing lactic dehydrogenase(LDH) levels. Results in this study illustrated that the content of all 18 compounds examined has significant difference and variation among cultivars and extracts. The proposed method can serve as a prerequisite for quality control of bioactive compounds in Z. jujuba products.展开更多
Objective:To evaluale physico-chemical properties and antimicrobial potential of indigenous honey samples against different reference strains including Escherichia coli ATCC 8739.Enterobacter aerogenes ATCC 13048.Pseu...Objective:To evaluale physico-chemical properties and antimicrobial potential of indigenous honey samples against different reference strains including Escherichia coli ATCC 8739.Enterobacter aerogenes ATCC 13048.Pseudomonas aeruginosa ATCC 9027.Bacillus subtilis ATCC6633.Staphylococcus aureus ATCC 25923.Salmonella typhi ATCC 14028,Klebsiella pneumonia ATCC 13883.Aspergillus niger ATCC 16404.Rhizopus oligasparus PCSIR1.Candida albicans ATCC14053 and Candida utilis ATCC 9950.Methods:By using standard methods samples were evaluated for their antimicrobial properties including additive effect of starch and non—peroxidase activity,antioxidative properties(phenol contents,flavonoid contents,1,1-diphenyl-2-pierylhydrazyl radical scavenging activity).Prior to this evaluation,complete physico-chemical properties including pH,color,ash contents,protein contents,moisture contents,hydroxymethyl furfural contents,total sugar contents,reducing sugar and non-reducing sugar contents were analyzed.Results:Relatively higher ash contents were found in the Siddar honey i.e.(0.5900±0.0336)%and small honey showed relatively higher protein contents i.e.(777.598±9.880) mg/kg.The moisture contents of tested honey samples ranged between 13.8%—16,6%,total sugar contents foam 61.672%-72.420%and non-reducing sugar contents from 1.95%—3.93%.Presences of phenolic contents indicate higher antioxidant potential of these honey samples.All bacteria showed clear inhibition zones in response to tested honey samples whereas fungi and yeast showed inhibition at higher concentrations of these honey samples.For Escherichia coli.Bacillus subtilis.Salmonella typhi.Pseudomonas aeruginosa and Aspergillus niger,overall the small honey showed the higher activity than other honey samples.Conclusion:Physico-chemical analysis of honey samples confirmed good quality of honey according to the standards set by European Union Commission and Codex Alimcntarius Commission.Evaluation of these honey samples confirms antimicrobial potential of particular types of honeys indigenous to Pakistan.展开更多

N supply in the pre-harvest helped to increase N-reserves of trees and then translocate to the new growth organs the following year. The largest amount of ^15N was detected in the roots and trunks. In all the treatments, the partition rates were highest in coarse roots, which were 30.43% (BB), 38.61% (FCH), and 40.62% (FRS), respectively. ^15N stored in roots and trunks was used by jujube trees to sustain new growth in the following full-bloom. ^15N applied before budding resulted in lower Ndff% in perennial organs (trunks and coarse roots) sampled in the following full-bloom, but fine roots had highest Ndff% (1.28%). Other organs recovered similar amount of Ndff%. In contrast, FCH and FRS treatments led to higher Ndff% (4.01-5.15%) in the new growth organs (new growth branches, deciduous spurs, leaves and flowers), but lower Ndff% in perennial branches (1.49-2.89%). With the delay of ^15N-urea application time, ^15N increased the partitioning to roots. FCH treatment increased N-storage in perennial organ during winter, which should be remobilized to sustain new growth the following spring.

文摘Jujube(Ziziphus jujube Mill.)is a traditional economic forest crop and is widely cultivated in hilly areas of the Loess Plateau,China.However,soil desiccation was discovered in jujube plantations.Pruning is recognized as a water-saving method that can reduces soil water consumption.In this study,we monitored the jujube plots with control(CK),light(C1),medium(C2)and high(C3)pruning intensities during the jujube growing period of 2012-2015 to explore the effect of pruning intensity on soil moisture and water use efficiency(WUE)of jujube plantations in the hilly Loess Plateau Region.The results showed that pruning is an effective method for soil water conservation in jujube plantations.Soil moisture increased with increasing pruning intensity during the jujube growing period of 2012-2015.C1,C2 and C3 pruning intensities increased soil water storage by 6.1-18.3,14.4-40.0 and 24.3-63.3 mm,respectively,compared to CK pruning intensity.Pruning promoted soil moisture infiltration to deeper soil layer.Soil moisture infiltrated to soil depths of 240,280 and>300 cm under C3 pruning intensity,220,260 and 260 cm under C2 pruning intensity,200,240 and 220 cm under C1 pruning intensity,and 180,200 and 160 cm under CK pruning intensity in 2013,2014 and 2015,respectively.Soil water deficit was alleviated by higher pruning intensity.In 2013-2015,soil water change was positive under C2(6.4 mm)and C3(26.8 mm)pruning intensities but negative under C1(-20.5 mm)and CK(-40.6 mm)pruning intensities.Moreover,pruning significantly improved fresh fruit yield and WUE of jujube plants.Fresh fruit yields were highest under C1 pruning intensity with the values of 6897.1-13,059.3 kg/hm^2,which were 2758.4-4712.8,385.7-1432.1 and 802.8-2331.5 kg/hm2 higher than those under CK,C2,and C3 pruning intensities during the jujube growing period of 2012-2015,respectively.However,C3 pruning intensity had the highest WUE values of 2.92-3.13 kg/m3,which were 1.6-2.0,1.1-1.2 and 1.0-1.1 times greater than those under CK,C1 and C2 pruning intensities,respectively.Therefore,C3 pruning intensity is recommended to jujube plantations for its economic and ecological benefits.These results provide an alternative strategy to mitigate soil desiccation in jujube plantations in the hilly Loess Plateau Region,which is critical for sustainable cultivation of economic forest trees in this region.
文摘Objective: To evaluate the lethal and emergence inhibitory effect of alkaloid, phenolic and terpenoid extracts of Ziziphus jujuba(Z. jujuba; Rhamnaceae) against Aedes aegypti(Diptera: Culicidae), and to explore the effect of the most effective fraction on developmental and biochemical parameters of the dengue vector. Methods: The fourth instar larvae of Aedes aegypti were exposed to alkaloid, phenolic and terpenoid extracts from Z. jujuba leaves to test their toxicity and emergence inhibitory effects. Phenolic extract, being the most effective was further tested against the mosquitoes for their growth inhibitory effect supported by biochemical changes in the parent and F1 generation. Results: While the different secondary metabolite fractions i.e., alkaloid, phenolics and terpenoid caused mortality at larval and pupal stages, the LC_(50) value was the lowest for phenolic fraction. Further study carried out with the phenolic fraction revealed that it affected growth by decreasing adult life span, fertility and fecundity of the mosquitoes. The reduction in growth was also accompanied by decrease in carbohydrate and lipid levels. Conclusions: It is concluded that the phenolic extract of the leaves of Z. jujuba is a potential candidate for control of Aedes mosquitoes.

文摘[Objectives]To study and compare the processing methods of Ziziphus jujuba Mill.[Methods]Z.jujuba was processed by seven methods:cleaning,yellowish frying,burnt frying,carbonized frying,moistening by hot sand,steaming,and boiling.Ultraviolet-visible spectrophotometry was used to determine the content of total flavonoids in each processed Z.jujuba.[Results]The content of total flavonoids measured by various processing methods was different.The content of total flavonoids in the processed Z.jujuba by moistening by hot sand was relatively high and the absorption peak spectrum was almost the same as that of the cleaned processed Z.jujuba and the rutin reference substance.[Conclusions]The moistening by hot sand is the optimal method for processing of Z.jujuba.
文摘Lingwu Changzao (Ziziphus. jujube Mill) is a Ningxia local variety, it has a good table quality despite its short postharvest life. The respiration rate, the weight change of single fruit during growth development and the losing water of postharvest of fruit were studied. The results showed that the curve of fruit growth development was a double sigmoid characteristic. When the surface colour of fruit changed form jade-green to alizarin crimson, the respiration rate tended to fall during a entire growth development of fruit, which showed a fluctuation phenomenon. The respiration rate descended when the surface color changed from coloring to baby red, but after a crimson stage the respiration rate recurred to its original downtrend. With increasing of single fruit weight, phenomenon of respiration climacteric has not happened and the respiration rate curve showed a concavity characteristic at some stages. As a result, there was not a phenomenon of respiration climacteric with a double sigmoid growth development curve characteristic of fruit.

文摘One new ceanothane-type triterpene and one new sesquiterpene, together with two known triterpenes were isolated from the fruits of Ziziphusjujuba Mill. (Rhamnaceae). Based on the spectral analyses, the structures of two new compounds were elucidated as 2α-aldehydo-A(1)-norlup-20(29)-en-27,28-dioic acid (zizyberanal acid) (1), and zizyberanone (2). While the two known triterpenes were identified as zizyberanalic acid (3) and ursolic acid (4).

文摘The fruit of Ziziphus jujuba Mill., known as Hongzao(or Hong-Zao) in Chinese and cultivated in China for more than 4 000 years, has shown to have hepatoprotective property. In previous study, we have isolated and identified 27 known compounds from Z. jujuba fruits, which demonstrated anti-tumor activity. In this study, a high-performance liquid chromatography-diode-array detection-mass spectrometry(HPLC-DAD-MS) method was successfully applied to the simultaneous characterization and quantitation of 18 constituents in 28 Z. jujuba samples, comprised of 12 cultivars from different regions in China, by comparing their HPLC retention times, MS spectra, UV spectra, and NMR data with those of reference compounds. The quantitative method was validated with excellent linearity(R^(2 )> 0.999 1), preferable intra-and inter-day precisions(RSD < 2.78%), and good recoveries(94.96%–102.65%). The content variation of 18 compounds was analyzed by a chemometric method(hierarchical cluster analysis). In addition, these constituents showed protection against carbon tetrachloride(CCl_(4)) intoxicated Hep G2 cell lines by decreasing lactic dehydrogenase(LDH) levels. Results in this study illustrated that the content of all 18 compounds examined has significant difference and variation among cultivars and extracts. The proposed method can serve as a prerequisite for quality control of bioactive compounds in Z. jujuba products.

文摘Objective:To evaluale physico-chemical properties and antimicrobial potential of indigenous honey samples against different reference strains including Escherichia coli ATCC 8739.Enterobacter aerogenes ATCC 13048.Pseudomonas aeruginosa ATCC 9027.Bacillus subtilis ATCC6633.Staphylococcus aureus ATCC 25923.Salmonella typhi ATCC 14028,Klebsiella pneumonia ATCC 13883.Aspergillus niger ATCC 16404.Rhizopus oligasparus PCSIR1.Candida albicans ATCC14053 and Candida utilis ATCC 9950.Methods:By using standard methods samples were evaluated for their antimicrobial properties including additive effect of starch and non—peroxidase activity,antioxidative properties(phenol contents,flavonoid contents,1,1-diphenyl-2-pierylhydrazyl radical scavenging activity).Prior to this evaluation,complete physico-chemical properties including pH,color,ash contents,protein contents,moisture contents,hydroxymethyl furfural contents,total sugar contents,reducing sugar and non-reducing sugar contents were analyzed.Results:Relatively higher ash contents were found in the Siddar honey i.e.(0.5900±0.0336)%and small honey showed relatively higher protein contents i.e.(777.598±9.880) mg/kg.The moisture contents of tested honey samples ranged between 13.8%—16,6%,total sugar contents foam 61.672%-72.420%and non-reducing sugar contents from 1.95%—3.93%.Presences of phenolic contents indicate higher antioxidant potential of these honey samples.All bacteria showed clear inhibition zones in response to tested honey samples whereas fungi and yeast showed inhibition at higher concentrations of these honey samples.For Escherichia coli.Bacillus subtilis.Salmonella typhi.Pseudomonas aeruginosa and Aspergillus niger,overall the small honey showed the higher activity than other honey samples.Conclusion:Physico-chemical analysis of honey samples confirmed good quality of honey according to the standards set by European Union Commission and Codex Alimcntarius Commission.Evaluation of these honey samples confirms antimicrobial potential of particular types of honeys indigenous to Pakistan.

文摘[Objective] The aim was to get optimized conditions for date powder with good color and taste.[Method] The pectinase enzymolysis and vacuum freeze-drying technology were used in the extract process of date powder.[Result] The production boasts of superior quality were with rich fragrance and uniform particle under certain process conditions.The optimized conditions for pectinase enzymolysis were:the amount of pectinase reached to 0.1% of that of date syrup,enzymolysis temperature and time were 50 ℃ and 50 min respectively,the pH value was 4.0.The optimized conditions for vaccum freeze-drying were:10% altodextrin,three times volumn of water with the thickness of 7 mm.[Conclusion] The optimized conditions were obtained in this study to produce instant date powder from Ziziphus jujuba.
