目的探讨HSP70高表达对肿瘤细胞过氧基异丙苯敏感性的影响及其机理。方法将KB细胞在40℃预热不同时间,诱导细胞HSP70高表达,用RT-PCR检测细胞HSP70mRNA水平;将细胞暴露于0.001mM、0.002mM、0.004mM、0.008mM的过氧基异丙苯16h,... 目的探讨HSP70高表达对肿瘤细胞过氧基异丙苯敏感性的影响及其机理。方法将KB细胞在40℃预热不同时间,诱导细胞HSP70高表达,用RT-PCR检测细胞HSP70mRNA水平;将细胞暴露于0.001mM、0.002mM、0.004mM、0.008mM的过氧基异丙苯16h,用MTT比色法检测细胞活力,观察预热对不同水平过氧基异丙苯敏感性的影响;保持过氧基异丙苯浓度为0.002mM不变,观察不同预热时间对细胞过氧基异丙苯敏感性的影响。结果40℃预热后细胞的HSP70含量升高,在预热120min后达到最高,以后缓慢下降,可维持10h以上;预热细胞对不同浓度过氧基异丙苯的敏感性均显著低于未预热细胞;,对于相同浓度的过氧基异丙苯而言,预热120min的细胞敏感性最低。结论预热可以引起细胞HSP70表达增高,降低细胞对过氧基异丙苯的敏感性。 展开更多

目的:探讨白屈菜碱(chelidonine)对人口腔上皮样癌细胞KB细胞株的抗肿瘤作用机制。方法:以不同浓度的白屈菜碱作用于体外培养的KB细胞株24、48、72 h,应用MTT检测细胞生长抑制率;Transwell小室检测肿瘤细胞的侵袭作用;流式细胞仪检测细... 目的:探讨白屈菜碱(chelidonine)对人口腔上皮样癌细胞KB细胞株的抗肿瘤作用机制。方法:以不同浓度的白屈菜碱作用于体外培养的KB细胞株24、48、72 h,应用MTT检测细胞生长抑制率;Transwell小室检测肿瘤细胞的侵袭作用;流式细胞仪检测细胞凋亡率;免疫印迹实验分析Caspase-3、Bax、Bcl-2、MMP-2、MMP-9信号通路相关蛋白的表达水平。结果:MTT结果显示白屈菜碱对KB细胞株的增殖具有明显的抑制作用,且呈时间剂量依赖性;侵袭试验结果表明,白屈菜碱能够抑制KB细胞侵袭作用,且随着浓度的增大,抑制侵袭作用增强;流式双染检测白屈菜碱作用KB细胞24 h后细胞的凋亡率,与对照组相比差异有统计学意义;蛋白免疫印迹试验显示白屈菜碱组的Caspase-3、Bax表达明显上调,Bcl-2、MMP-2、MMP-9表达下调,与对照组相比差异有统计学意义。结论:白屈菜碱对KB细胞具有明显的抑制增殖、侵袭和促进凋亡的作用;其抗肿瘤机制可能与下调Bcl-2、MMP-2、MMP-9,上调Caspase-3、Bax的表达有关。 展开更多

目的:探讨乌司他丁对人口腔表皮样癌细胞系KB的转移和侵袭能力的影响。方法:分别通过划痕实验、Transwell小室侵袭实验探讨乌司他丁对人口腔表皮样癌细胞系KB的转移和侵袭的影响;明胶酶谱实验研究乌司他丁对人口腔表皮样癌细胞系KB分泌... 目的:探讨乌司他丁对人口腔表皮样癌细胞系KB的转移和侵袭能力的影响。方法:分别通过划痕实验、Transwell小室侵袭实验探讨乌司他丁对人口腔表皮样癌细胞系KB的转移和侵袭的影响;明胶酶谱实验研究乌司他丁对人口腔表皮样癌细胞系KB分泌活性MMP-9的影响。结果:划痕实验结果显示:与对照组转移细胞数(130.22±14.99)相比,乌司他丁处理后的KB细胞(69.78±11.79)转移能力明显受到抑制(P<0.05);Transwell小室侵袭结果显示,乌司他丁治疗组KB细胞侵袭的细胞数目(223.44±28.54)明显低于阴性对照组(501.11±30.75),(P<0.05);明胶酶谱结果显示,乌司他丁处理后的KB细胞活性MMP-9的表达明显降低(P<0.05)。结论:乌司他丁可能通过抑制MMP-9的活性从而抑制口腔癌细胞的转移和侵袭。 展开更多

AIM: To identify whether JTE-522 can induce apoptosis in AGS cells and ROS also involved in the process, and to investigate the changes in NF-kB, p53, bcl-2 and caspase in the apoptosis process. METHODS: Cell culture, MTT, Electromicroscopy, agarose gel electrophoresis, lucigenin, Western blot and electrophoretic mobility shift assay (EMSA) analysis were employed to investigate the effect of JTE-522 on cell proliferation and apoptosis in AGS cells and related molecular mechanisms. RESULTS: JTE-522 inhibited the growth of AGS cells and induced the apoptosis. Lucigenin assay showed the generation of ROS in cells under incubation with JTE-522. The increased ROS generation might contribute to the induction of AGS cells to apoptosis. EMSA and Western blot revealed that NF-kB activity was almost completely inhibited by preventing the degradation of IkBalpha. Additionally, by using Western blot we confirmed that the level of bcl-2 was decreased, whereas p53 showed a great increase following JTE-522 treatment. Their changes were in a dose-dependent manner. CONCLUSION: These findings suggest that reactive oxygen species, NF-kB, p53, bcl-2 and caspase-3 may play an important role in the induction of apoptosis in AGS cells after treatment with JTE-522. 展开更多

利用二苄基二氯化锡和二硫代氨基甲酸盐反应,合成了13种新的二苄基锡二硫代氨基甲酸衍生物.通过元素分析、红外光谱、核磁共振氢谱和质谱对其结构进行了表征.初步生物活性测试结果表明,该类化合物对KB癌细胞具有一定的抑制活性.

Although previous studies showed that the principal oncoprotein encoded by Epstein-Barr virus, latent membrane protein 1 (LMP1), could induce the nasopharyngeal carcinoma cells in G2/M phase increased, littleis known about the target molecules and mechanisms. The present study demonstrated that LMP1 couldinduce the accumulation of p53 protein and upregulate its transactivity in a dose dependent manner, whichresulted in the decrease of the kinase activity of cdc2/cyclin B complex and inducing arrest at G2/M phasethrough the activation of NF-kB and AP-1 signaling pathways, and the effect of NF-kB was more obviousthan that of AP-1. This study provided some significant evidence for further elucidating the molecular mechanisms that LMP1 had effects on the surveillance mechanism of cell cycle and promoting the survivalof transformed cells and tumorigenesis. 展开更多
