METHODS: We analyzed the association between immunohistochemically detected LNγ2, LNβ3, and MMP7 expression in biliary tract cancer and clinicopathological characteristics. Activity of MMP7 was analyzed by casein zymography. An in vitro invasion assay after treatment with MMP7-specific siRNA was performed. RESULTS: LNγ2 expression was predominantly observed in carcinoma cells at the invasive front. LNγ2 expression was seen in 57% of patients with biliary tract cancer, and was associated with depth of invasion, histologic type, and advanced stage. The expression pattern of LNβ3 was classified Into two types: invasive front dominant type (38%) and diffuse type (28%).The invasive front dominant type was associated with histologic type and advanced stage. MMP7 positivity was correlated with LNγ2 or LNβ3 expression but not with clinicopathological characteristics. Active MMP7 detected by casein zymography was correlated with depth of invasion and advanced stage. Downregulation of MMP7 expression by siRNA resulted in a significant decrease in biliary tract cancer cell invasion in vitro.CONCLUSION: Our results suggest that LNγ2 and LNβ3, in conjunction with MMP7, play a key role in the progression of biliary tract cancer.展开更多
Platinum (Pt) implants coated with poly (3, 4-ethylenedioxythiophene)/carbon nanotube (PEDOT/CNT) composite films were implanted into the brain of rats, and the brain response was evaluated 6 weeks after the imp...Platinum (Pt) implants coated with poly (3, 4-ethylenedioxythiophene)/carbon nanotube (PEDOT/CNT) composite films were implanted into the brain of rats, and the brain response was evaluated 6 weeks after the implantation. The surface morphology of Pt implants with and without the PEDOT/CNT coating was studied using scanning electron microscopy (SEM). After 6 weeks post-implantation, the expression of laminin (vascular endothelial marker) and neuronal nuclei (NeuN, neuronal marker) were evaluated by immnohistochemistry. It is revealed that the obvious improvements of the surface density of blood vessels and neurons aound the Pt implants with the coating, which were evidenced by laminin and NeuN staining in the zone within the distance of 150 μm to the implant interface. These results suggest the PEDOT/CNT composite films can improve the biocompatibility of the Pt electrodes while it is implanted in brain.展开更多

The expressions of epithelial-cadherin (E-cad), laminin receptor(LNR) and type Ⅳcollagenase(Ⅳ col) in two kinds of HCC cell lines, HT29 and LoVo cells, were observed quantitatively. The intracellular CD44V6 expressed in mRNA and in protein of the HCC cell lines were detected with in situ hybridization and immunohistochemistry. Results: 100% of nude mice had liver metastasis, which demonstrated thatLoVo is a high metastatic HCC cell line. However, HT29 is a relatively low metastatic HCC cell line becauseonly 42. 9 % of nude mice transplanted HT29 cells had liver metastasis. The expressions of E--cad, LNR andIVcol of HT29 and LoVo cells in vitro were detected quantitatively. HT29 cells expressed E-cad with weakpositive on cell membrane. and LoVo cells almost expressed none. LoVo cells expressed higher level of LNRand tV col than HT29 cells did. The expression of CD44V6 in LoVo cells whether in mRNA or in protein werehigher than that of HT29 cells. Conclusion: LoVo is a high metastatic HCC cell line while HT29 is a relatively low metastatic HCC cell line. E-cadherin, LNR, IV col and CD44V6 can determine the metastatic potentialof HCC cells.展开更多
