The aim of this study was to investigate the extraction process and antioxidant activity of polyphenols from Yunnan Mengzi pomegranate peel.Single factor and orthogonal test were used as optimize the extract rate(ER)o...The aim of this study was to investigate the extraction process and antioxidant activity of polyphenols from Yunnan Mengzi pomegranate peel.Single factor and orthogonal test were used as optimize the extract rate(ER)of polyphenols from pomegranate peel(PGP).The polyphenols antioxidant activity was evaluated by DPPH·,·OH and total reducing ability,and compared with that of Vc.The optimum extraction conditions of polyphenols were as follows:ethanol concentration of 50%,liquid-solid ratio of 50:1,extraction temperature of 90°C,extraction for 4 h.Under these conditions,the ER of polyphenols from GPG was 8.15%.The results showed that the scavenging effects of polyphenols and Vc on DPPH·,·OH increased with increasing concentration,and the scavenging effect of polyphenols on DPPH·was higher than Vc in rang of 0.1-1.6µg/mL.When the concentration is greater than 0.6µg/mL,the scavenging effects of plyphenols on·OH is higher than Vc.The total reducing ability of polyphenols from PGP improved with the increase of concentration,and when the concentration was 8µg/mL,the total reducing ability was 114 U/mL.展开更多

[Method] Or...[ Objective] This study aimed to provide theoretical reference for the extraction of theabrownine from Liubao tea, determination of the biological activity, and development and utilization of theabrownine. [Method] Orthogonal experimental design was adopted to investigate the effects of solid-liquid ratio, extraction temperature, extraction duration, and extraction frequency on the yield of theabrownine extracted from Liubao tea. The extraction technology was optimized, and the antioxidant activity of theabrownine was analyzed. [ Result] The results showed that extraction frequency and extraction temperature had significant effects on the yield of theabrownine extracted from Liubao tea, while solid-liquid ratio and extraction duration had no significant effect. The optimal conditions for extracting theabrownine from Liubao tea were determined as follows : solid-liquid ratio was 1 : 30, extraction temperature was 100℃, extraction duration was 80 min, and extraction frequency was 4 times. Under these conditions, the yield of theabrownine reached 22.92%. Results of antioxidant activity determination showed that hydroxyl radical-scavenging activity of theabrownine was as high as 74%, and the scavenging effects was significantly higher than that of Vitamin C under low concentrations; nitroso-scavenging activity of theabrownine reached 73%, with no significant difference from that of Vitamin C; superoxide anion free radical-scavenging activity of theabrownine could be above 60%, which was lower than that of Vitamin C. [Condusion] Liubao tea contains abundant theabrownine that has relatively high antioxidant activity with good prospects in antioxidant application, which lays solid foundation for in-depth development of Liubao tea.展开更多

Optimum extraction process was under extraction temperature of 80 ℃,extraction time of 2. 5 h,ultrasonic power of 180 W,solid-liquid ratio of 1∶ 25 g / m L; the total antioxidant activity was increased with the increasing concentration of polysaccharides. The extraction process is simple and efficient. The extraction rate of lychee pericarp polysaccharides with antioxidant activities is high.展开更多
[Objectives]This study was conducted to study the total polyphenol content and antioxidant capacity of different extraction fractions of Pholidota chinensis.[Methods]Different polar fractions of P.chinensis were obtai...[Objectives]This study was conducted to study the total polyphenol content and antioxidant capacity of different extraction fractions of Pholidota chinensis.[Methods]Different polar fractions of P.chinensis were obtained by ultrasonic extraction method,and determined for polyphenol contents by UV spectrophotometry,and their antioxidant activity was evaluated by DPPH free radical scavenging and ferric ion reducing antioxidant power(FRAP).SPSS analysis software was used to analyze the correlation between total polyphenol contents and antioxidant activity of different extracts.[Results]The contents of total polyphenols in different polar fractions ranged from 1.52 to 6.41 mg/g,and the order from low to high was:water<petroleum ether<ethyl acetate<95%ethanol.The reduction capacity of the total antioxidant capacity method ranged from 12.54 to 22.45μmol/L,and the order was 95%ethanol>ethyl acetate>petroleum ether>water.The DPPH median inhibition concentration ranged from 0.35 to 2.65 mg/ml,and the order was water>petroleum ether>ethyl acetate>95%ethanol.The total polyphenol content of P.chinensis was best in the 95%ethanol extraction fraction,which showed the strongest total antioxidant capacity,so the total polyphenol content of the 95%ethanol fraction was positively correlated with its antioxidant capacity,while the DPPH free radical-scavenging capacity was better in the water fraction,indicating that the free radical-scavenging capacity of the water fraction was better.[Conclusions]This study provides a theoretical basis for the further study of the chemical components of P.chinensis and the development and utilization of natural antioxidants.展开更多

Therefore,the microwave assisted extraction was selected as the main method,the ethanol aqueous solution was used as the solvent,and the single factor experiment was carried out with microwave power,ethanol volume fraction,solid-to-liquid ratio and microwave time as the variables. The optimal extraction process of flavonoids was microwave power 400 W,solid-to-liquid ratio 1 ∶ 30,extraction time60 s,ethanol volume fraction 70%,under this condition,the extraction yield was 10. 65 mg/g. The antioxidant activity of flavonoids in D. indicum was determined. The results showed that the highest scavenging rate of DPPH·free radicals was 47. 89%,and the highest scavenging rate of hydroxyl radicals was 98. 00%. [Conclusions]The flavonoids in D. indicum have excellent antioxidant activity. This study is expected to provide a basis for further development of D. indicum products.展开更多

This study aimed to investigate the flavonoid profiles and antioxidant activity of Paeonia lactiflora petal flavonoid extract(PPF). The UHPLC-ESI-Q-Exactive HF MS/MS method was established for characterization, and 21 predominant flavonoid compounds were tentatively identified in PPF. Among them, isoscutellarein-7-(6’-acetylallosyl-(1->2)-glucoside) and scutellarin methylester were discovered in PPF for the first time. Pretreatment with PPF significantly reduced H2O2-induced cell damage, ROS accumulation, and malondialdehyde content and increased the activity of SOD, CAT, and GSH-Px in buffalo rat liver 3A(BRL3A) cells. Moreover, the expression of nuclear Factor E2-related factor(Nrf2) was upregulated by PPF, whose expression trend was consistent with that of heme oxygenase-1(HO-1), glutamate-cysteine ligase catalytic subunit(GCLC), and NAD(P)H quinone oxidoreductase-1(NQO1). These findings suggested that herbaceous peony flavonoids can be used as a natural bioactive agent to prevent oxidative stress.展开更多
[Objectives]To explore the optimal extraction and purification process of the flavonoids in Amaranthus caudatus L.and to study the antioxidant activity in vitro of the flavonoids in A.caudatus.[Methods]Taking A.caudat...[Objectives]To explore the optimal extraction and purification process of the flavonoids in Amaranthus caudatus L.and to study the antioxidant activity in vitro of the flavonoids in A.caudatus.[Methods]Taking A.caudatus as the raw material,flavonoids were extracted by alcohol extraction method,and AB-8 macroporous adsorption resin was selected for purification.The hydroxyl radical scavenging ability,DPPH radical scavenging ability,and O^2-radical scavenging ability were used as evaluation indicators,to explore the antioxidant activity in vitro of the flavonoids in A.caudatus.[Results]The optimal extraction process conditions of flavonoids in A.caudatus are:liquid-to-material ratio 40:1,extraction temperature 60℃,ethanol concentration 60%,ultrasonic power 320 W,extraction time 50 min.Under these conditions,the extraction yield of flavonoids in A.caudatus is(1.35±0.01)%.The optimal purification process conditions of flavonoids in A.caudatus are 2.5 g AB-8 macroporous adsorption resin,sample volume 5 mL,mass concentration of adsorption solution 1.60 mg/mL,pH value of adsorption solution 3.0,sample flow rate 3 BV/h,ethanol concentration in desorption process is 70%and the desorption flow rate is 3 BV/h.Under these conditions,the recovery rate reaches 88.35%±0.68%.[Conclusions]A.caudatus has a high content of flavonoids and has excellent free radical scavenging ability in vitro.This study is intended to provide important technical support for the research of flavonoid activity of A.caudatus and the development of functional products.展开更多
Macroalgae serve as a potential feedstock for fucoxanthin extraction.Fucoxanthin,a bioactive pigment found in the chloroplasts of marine algae,exhibits significant pharmacological properties.As a member of the caroten...Macroalgae serve as a potential feedstock for fucoxanthin extraction.Fucoxanthin,a bioactive pigment found in the chloroplasts of marine algae,exhibits significant pharmacological properties.As a member of the carotenoid family,fucoxanthin plays a crucial role in both the food and pharmaceutical industries.This research explores the effects of ultrasonics on the extraction of fucoxanthin from the marine macroalga Padina australis.In addition,various extraction techniques and the influence of solvents on the efficient separation of fucoxanthin from algae have been studied and compared.Using methanol,chloroform,and a combination of methanol and chloroform(1:1,v/v),conventional fucoxanthin extraction from Padina australis yielded 8.12 mg of fucoxanthin per gram of biomass.However,the ultrasonic-assisted extraction resulted in a significantly higher yield of 16.9 mg of fucoxanthin per gram of biomass,demonstrating that the use of ultrasonics enhances the extraction rate compared to conventional methods.Therefore,the efficient separation of fucoxanthin from Padina australis is highly dependent on ultrasonic-assisted extraction.The process conditions for the extraction were optimized to maximize the yield of fucoxanthin from seaweeds.The following parameters were selected for optimization studies:moisture content,particle size,mixing speed,extraction temperature,extraction duration,and solid-to-solvent ratio.The extracted fucoxanthin exhibited various biological activities,including antimicrobial and antioxidant properties,and its structure was elucidated through FTIR and NMR spectroscopy.Additionally,thin-layer chromatography of the crude algae extracts confirmed the presence of fucoxanthin in the marine algae.Given these findings,the optimized extraction process holds the potential for scaling up to large-scale fucoxanthin production.Fucoxanthin,as a potent pharmacological agent,offers promising applications in the treatment of various ailments.展开更多

[ Method ] With water extraction and alcohol precipitation method, the optimal conditions for extracting crude polysaccharides from wild L. volemus Ft. were screened by single-factor and orthogonal experiments. The antioxidant activity of the extracted crude polysac- charides was determined with DPPH assay. [ Result ] The optimal conditions for pigment removal with activated carbon were: activated carbon amount of 20 g/L, water bath time of 40 min, water bath temperature of40 ℃ ; the optimal conditions for extracting crude polysaccharides from wild L. vo/emus Fr. with hot water ex- traction method were: hot water extraction time of 3 h, solid-liquid ratio of 1: 45, extraction frequency of twice. Under the optimized extraction conditions, the yield of crude polysaccharides was 21.33 mg/g. In addition, the antioxidant activity of 0. 665 mg/ml crude polysaccharides was 52.46% ; the amount of crude polysac- eharides was proportional to the antioxidant activity. [ Conclusion] Hot water extraction method can be used as a high-efficiency extraction technology of crude pol- ysaecharides from wild L. volemus Fr. with simple operation and low costs. Capitata crude polysaccharide was as follows: solid-to-liquid ratio was 40∶ 1,the extraction temperature was 50℃,the ultrasonic time was 80 min. Under such conditions,the extraction rate of C. Capitata crude polysaccharide was 10. 51%,and the purity was 56. 9%; C. Capitata crude polysaccharide has good scavenging ability to DPPH and ABTS^+ free radicals,and its IC_(50) values are 0. 468 and 0. 231 mg/mL,respectively. Conventional solvent extraction (CSE), microwave assisted extraction (MAE) and autoclave assisted pretreated solvent extraction (APSE). In the present experiment, the total phenolic content and antioxidant activity of the various extracts w</span><span style="font-family:"">ere</span><span style="font-family:""> measured. Thiobarbituric acid reactive substances (TBARS) assay was used to evaluate the ability of the BMR to protect lipid peroxidation in corn oil at 185</span><span style="font-family:"">°</span><span style="font-family:"">C frying temperature. The formation of TBARS at 5 hours of heat treated corn oil ha</span><span style="font-family:"">s</span><span style="font-family:""> shown similar antioxidant levels in 150 ppm butylated hydroxytoluene (BHT) or MAE free phenolic extract added to corn oil. [Objectives]To optimize the extraction process of total flavonoids from Eucommiae Cortex,establish the extraction and content determination methods of its medicinal materials,and study its in vitro antioxidant activit...[Objectives]To optimize the extraction process of total flavonoids from Eucommiae Cortex,establish the extraction and content determination methods of its medicinal materials,and study its in vitro antioxidant activity.[Methods]The total flavonoids from Eucommiae Cortex were extracted by reflux extraction method,and the effects of extraction method,extraction solvent concentration,extraction volume,and extraction time on the total flavonoids content of the medicinal materials were explored through the single factor experiment.Orthogonal experiment was carried out to optimize the extraction process conditions,and the optimal extraction process for total flavonoids from Eucommiae Cortex was screened out.The total antioxidant capacity index was used to determine the free radical scavenging ability of the total flavonoids from Eucommiae Cortex.[Results]The optimal extraction process of total flavonoids from Eucommiae Cortex was 50%ethanol,1∶40 solid-to-liquid-ratio,and 1.5 h reflux extraction time.Through the antioxidant experiment in vitro,it is found that the total flavonoids from Eucommiae Cortex have a higher scavenging ability for the total antioxidant capacity,and there is a certain positive correlation with the mass concentration of total flavonoids.[Conclusions]This method can effectively determine the total flavonoids content of Eucommiae Cortex medicinal material,and provide a certain scientific basis for the quality standard research of the medicinal material.The total flavonoids from Eucommiae Cortex have good in vitro antioxidant activity.And the method for extracting total flavonoids from Eucommiae Cortex medicinal material in this experiment has good repeatability,high stability and feasibility.展开更多
The aim of this study was to investigate the extraction method for R.tomentosa and C.zeylanicum leaves and the evaluation of antibacterial and antioxidant activities of crude extracts.The results of the study sh...The aim of this study was to investigate the extraction method for R.tomentosa and C.zeylanicum leaves and the evaluation of antibacterial and antioxidant activities of crude extracts.The results of the study showed that the active ingredients of crude extracts were clearly separated by Thin-layer chromatography and the presence of rhodomyrtone in R.tomentosa crude extract and cinnamaldehyde in C.zeylanicum crude extract.R.tomentosa crude extract was antibacterial activity against Staphylococcus aureus with 13.1 mm of inhibition zone,but is not effective against Salmonella Typhimurium.C.zeylanicum leaf extract did not show antibacterial activity on both S.aureus and S.Typhimurium.At a dilution of 1/2 of the R.tomentosa crude extract can completely inhibit S.aureus growth.This study also indicated the presence of antioxidant compounds such as flavonoids,tannins,phenols and terpenoids in C.zeylanicum and R.tomentosa crude extracts.The results showed that R.tomentosa and C.zeylanicum crude extracts should be used as a biotherapy alternative to antibiotic therapy.However,further study would be needed to investigate the antibacterial activity of crude extracts in vivo.展开更多

This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. The promising results showed that extracted SNP could be a potent natural antioxidant.展开更多
Objective:To isolate and analyse the chemical composition in the essential oils and free radical scavenging activity of different crude extracts from the fresh and dry leaves of vegetable plants of Lactuca sativa L.(L...Objective:To isolate and analyse the chemical composition in the essential oils and free radical scavenging activity of different crude extracts from the fresh and dry leaves of vegetable plants of Lactuca sativa L.(L.sativa).Methods:The essential oils and volatile chemical constituents were isolated from the fresh and dry leaves of L.sativa(lettuce)grown in Sultanate of Oman by hydro distillation method.The antioxidant activity of the crude extracts was carried out by well established free radical scavenging activity(DPPH)method.Results:About 20 chemical compounds of different concentration representing 83.07%and 79.88%respectively were isolated and identified by gas chromatography-mass spectroscopy in the essential oils isolated from the fresh and dry leaves asα-pinene(5.11%and 4.05%).γ-cymene(2.07%and 1.92%),thymol(11.55%and 10.73%),durenol(52.00%and 49.79%),α-terpinene(1.66%and 1.34%),thymol acetate(0.99%and 0.67%),caryophyllene(2.11%and 1.98%),spathulenol(3.09%and 2.98%).camphene(4.11%and3.65%),limonene(1.28%and 1.11%)representing these major chemical compounds.However,some other minor chemical constituents were also isolated and identified from the essential oil of lettuce including p-pinene,α-terpinolene,linalool,4-terpineol,α-terpineol,o-methylthymol,L-alloaromadendrene and viridiflorene.Conclusions:The chemical consliluents in the essential oils from the locally grown lettuce were identified in the following classes or groups of chemical compounds such as monoterpenes,sesquiterpenes volatile organic compounds and their oxygenated hydrocarbons.Therefore,the essential oils and the crude extracts from Omani vegetable species of lettuce are active candidates which would be used as antioxidant,antifungal or antimicrobial agents in new drugs preparation for therapy of infectious diseases.展开更多

A new ultrahigh pressure extr...The present study was designed to establish and optimize a new method for extracting chlorogenic acid and cynaroside from Lonicera japonica Thunb. through orthogonal experimental designl. A new ultrahigh pressure extraction (UPE) technology was applied to extract chlorogenic acid and cynaroside from L. japonica. The influential factors, including solvent type, ethanol concentration, extraction pressure, time, and temperature, and the solid/liquid ratio, have been studied to optimize the extraction process, The optimal conditions for the UPE were developed by quantitative analysis of the extraction products by HPLC-DAD in comparison with standard samples. In addition, the microstructures of the medicinal materials before and after extraction were studied by scanning electron microscopy (SEM). Furthermore, the extraction efficiency of different extraction methods and the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities of the extracts were investigated. The optimal conditions for extracting chlorogenic acid and cynaroside were as follows: ethanol concentration, 60%; extraction pressure, 400 MPa; extraction time, 2 rain; extraction temperature, 30 ℃; and the solid/liquid ratio, 1 : 50. Under these conditions, the yields of chlorogenic acid and cynaroside were raised to 4.863% and 0.080%, respectively. Compared with other extraction methods, such as heat reflux extraction (HRE), ultrasonic extraction (UE), and Sohxlet extraction (SE), the UPE method showed several advantages, including higher extraction yield, shorter extraction time, lower energy consumption, and higher purity of the extracts. This study could help better utilize L. japonica flower buds as a readily accessible source of natural antioxidants in food and pharmaceutical industries.展开更多
To study the changes of volatile compounds and antioxidant activity of fast fermented shrimp head paste during the fermentation processing,Penaeus vannamei boone head was used as the raw material,and compound starter ...To study the changes of volatile compounds and antioxidant activity of fast fermented shrimp head paste during the fermentation processing,Penaeus vannamei boone head was used as the raw material,and compound starter was added for fermen-tation.During the 14 days of fermentation,the changes of free amino acids,volatile flavor compounds and main components were determined by SPME-GC-MS and electronic nose.The oxidation-resistance changes of water extract at different fermentation stages were evaluated.The results showed that the total free amino acids increased from original 2320 to 5640 mg(100 mL)^(−1).SPME-GC-MS analysis found that aldehydes,pyrazine compounds increased from 10.80%,1.94%to 24.35%,13.63%respectively during the fermentation process.The HO·scavenging ability of shrimp head paste increased from 52%to 86%.Our results showed that the shrimp paste produced from shrimp head fermentation could obtain good flavor and possess certain antioxidant activity of aquatic fermented condiment.展开更多

(Asteraceae) extracts obtained by classical and ultrasound solvent extraction were compared. The dry aerial part of plant was extracted by 70% etha-nol at a solid-to-liquid ratio of 1︰10 (m/v) and 25°C. The extract obtained by classical solvent extraction contained 29% larger amounts of phenolic compounds and 20% higher content of flavonoids. 2,2-diphenyl-1-picril hydrazyl radical (DPPH) scavenging reached 93.6% and the values of EC50 were (34.16±0.65) μg·ml-1 and (65.48±1.12) μg·ml-1 for the extracts obtained by the classical and ultrasound extractions, respectively. The extracts, independent of the extraction technique applied, showed a considerable growth inhibition on Candida albicans and Saccharo-myces cerevisiae, while no growth inhibition zones were observed for Aspergillus niger. The diameters of inhibition zone observed for all the microorganisms were larger for extracts obtained by classical extraction than those by ul-trasound extraction.展开更多
The polyphenol content,antioxidant and antimicrobial activities of the extracts obtained by classical, ultrasonic and Soxhlet extractions from dry aerial parts of two Artemisia species(Artemisia vulgaris and Artemisia...The polyphenol content,antioxidant and antimicrobial activities of the extracts obtained by classical, ultrasonic and Soxhlet extractions from dry aerial parts of two Artemisia species(Artemisia vulgaris and Artemisia campestris)were compared.Ultrasound positively affected the yield of extractive substance and the kinetics of extraction,but the extract obtained by the classical extraction showed the highest antioxidant activities and contained higher total contents of phenolic compounds and flavonoids than the extracts obtained by two other extraction techniques.Both flavonoid aglycones(apigenin,quercetin,quercetin 3,3′-dimethyl ether)and flavonoid glycosides(rutin,hyperoside and kaempferol 3-rhamnoside)were identified by thin layer chromatograph(TLC)analysis in the extracts from both species.A.campestris extracts were richer in quercetin than A.vulgaris and its antimicrobial activity was also better than A.vulgaris.Extracts obtained from both species were found to be more effective on the tested yeasts than bacteria.The kinetics of the total extractive substances,such as phenolic,flavonoids and quercetin extraction,was successfully described by the model of unsteady-state diffusion.展开更多

[Methods]FCPwas obtained from C. platycladi by a new set extraction process of microwave-assisted extraction,freeze thawing,superfine crushingand freeze drying technology and purification. On the basis of single-factor tests,the quadratic regression parasitism model was established by the extraction amount of total flavonoids with microwave power,alcohol concentration,and liquid-to-solid ratio through the response surface methodology. [Results] The optimal extraction conditions were microwave power of 757 W,alcohol concentration of 69%,liquid-to-solid ratio of21∶ 1,with the optimal extraction yield of 15. 302 9 mg/g. Under the real conditions( microwave power of 800 W,alcohol concentration of69%,liquid-to-solid ratio of 21∶ 1) in our laboratory,the maximum yield of total flavonoids was 15. 9 mg/g,the results fitwell with the prediction of quadratic regression parasitism model. The good antioxidant activity has been determined byscavenging effect of FCP on 1,1-diphenyl-2-picrylhydrazyl( DPPH) radical and superoxide anion free radical with the scavenging yield of 70. 2% and 68. 8% respectively. 文摘The aim of this study was to investigate the extraction process and antioxidant activity of polyphenols from Yunnan Mengzi pomegranate peel.Single factor and orthogonal test were used as optimize the extract rate(ER)of polyphenols from pomegranate peel(PGP).The polyphenols antioxidant activity was evaluated by DPPH·,·OH and total reducing ability,and compared with that of Vc.The optimum extraction conditions of polyphenols were as follows:ethanol concentration of 50%,liquid-solid ratio of 50:1,extraction temperature of 90°C,extraction for 4 h.Under these conditions,the ER of polyphenols from GPG was 8.15%.The results showed that the scavenging effects of polyphenols and Vc on DPPH·,·OH increased with increasing concentration,and the scavenging effect of polyphenols on DPPH·was higher than Vc in rang of 0.1-1.6µg/mL.When the concentration is greater than 0.6µg/mL,the scavenging effects of plyphenols on·OH is higher than Vc.The total reducing ability of polyphenols from PGP improved with the increase of concentration,and when the concentration was 8µg/mL,the total reducing ability was 114 U/mL.

文摘[ Objective] This study aimed to provide theoretical reference for the extraction of theabrownine from Liubao tea, determination of the biological activity, and development and utilization of theabrownine. [Method] Orthogonal experimental design was adopted to investigate the effects of solid-liquid ratio, extraction temperature, extraction duration, and extraction frequency on the yield of theabrownine extracted from Liubao tea. The extraction technology was optimized, and the antioxidant activity of theabrownine was analyzed. [ Result] The results showed that extraction frequency and extraction temperature had significant effects on the yield of theabrownine extracted from Liubao tea, while solid-liquid ratio and extraction duration had no significant effect. The optimal conditions for extracting theabrownine from Liubao tea were determined as follows : solid-liquid ratio was 1 : 30, extraction temperature was 100℃, extraction duration was 80 min, and extraction frequency was 4 times. Under these conditions, the yield of theabrownine reached 22.92%. Results of antioxidant activity determination showed that hydroxyl radical-scavenging activity of theabrownine was as high as 74%, and the scavenging effects was significantly higher than that of Vitamin C under low concentrations; nitroso-scavenging activity of theabrownine reached 73%, with no significant difference from that of Vitamin C; superoxide anion free radical-scavenging activity of theabrownine could be above 60%, which was lower than that of Vitamin C. [Condusion] Liubao tea contains abundant theabrownine that has relatively high antioxidant activity with good prospects in antioxidant application, which lays solid foundation for in-depth development of Liubao tea.

文摘The aim was to optimize ultrasonic extraction conditions and study antioxidant activities of lychee pericarp polysaccharides. The effects of extraction temperature,time,ultrasonic power,solid-liquid ratio on polysaccharides yield were investigated by single-factor experiment and orthogonal experiment to obtain optimum extraction conditions; the antioxidant activities of lychee pericarp polysaccharides were evaluated by the total antioxidant activity. Optimum extraction process was under extraction temperature of 80 ℃,extraction time of 2. 5 h,ultrasonic power of 180 W,solid-liquid ratio of 1∶ 25 g / m L; the total antioxidant activity was increased with the increasing concentration of polysaccharides. The extraction process is simple and efficient. The extraction rate of lychee pericarp polysaccharides with antioxidant activities is high.

文摘[Objectives]This study was conducted to study the total polyphenol content and antioxidant capacity of different extraction fractions of Pholidota chinensis.[Methods]Different polar fractions of P.chinensis were obtained by ultrasonic extraction method,and determined for polyphenol contents by UV spectrophotometry,and their antioxidant activity was evaluated by DPPH free radical scavenging and ferric ion reducing antioxidant power(FRAP).SPSS analysis software was used to analyze the correlation between total polyphenol contents and antioxidant activity of different extracts.[Results]The contents of total polyphenols in different polar fractions ranged from 1.52 to 6.41 mg/g,and the order from low to high was:water<petroleum ether<ethyl acetate<95%ethanol.The reduction capacity of the total antioxidant capacity method ranged from 12.54 to 22.45μmol/L,and the order was 95%ethanol>ethyl acetate>petroleum ether>water.The DPPH median inhibition concentration ranged from 0.35 to 2.65 mg/ml,and the order was water>petroleum ether>ethyl acetate>95%ethanol.The total polyphenol content of P.chinensis was best in the 95%ethanol extraction fraction,which showed the strongest total antioxidant capacity,so the total polyphenol content of the 95%ethanol fraction was positively correlated with its antioxidant capacity,while the DPPH free radical-scavenging capacity was better in the water fraction,indicating that the free radical-scavenging capacity of the water fraction was better.[Conclusions]This study provides a theoretical basis for the further study of the chemical components of P.chinensis and the development and utilization of natural antioxidants.

文摘[Objectives] To determine the optimal extraction technology of flavonoids in Dendranthema indicum,and analyze the antioxidant activity of total flavonoids in D. indicum. [Methods]D. indicum was used as raw material to extract the total flavonoids by water extraction,alcohol extraction,freeze-thaw,microwave assisted extraction and ultrasonic extraction. [Results]Microwave assisted extraction had the highest extraction rate. Therefore,the microwave assisted extraction was selected as the main method,the ethanol aqueous solution was used as the solvent,and the single factor experiment was carried out with microwave power,ethanol volume fraction,solid-to-liquid ratio and microwave time as the variables. The optimal extraction process of flavonoids was microwave power 400 W,solid-to-liquid ratio 1 ∶ 30,extraction time60 s,ethanol volume fraction 70%,under this condition,the extraction yield was 10. 65 mg/g. The antioxidant activity of flavonoids in D. indicum was determined. The results showed that the highest scavenging rate of DPPH·free radicals was 47. 89%,and the highest scavenging rate of hydroxyl radicals was 98. 00%. [Conclusions]The flavonoids in D. indicum have excellent antioxidant activity. This study is expected to provide a basis for further development of D. indicum products.

文摘Flavonoids have attracted considerable attention due to their health benefits. This study aimed to investigate the flavonoid profiles and antioxidant activity of Paeonia lactiflora petal flavonoid extract(PPF). The UHPLC-ESI-Q-Exactive HF MS/MS method was established for characterization, and 21 predominant flavonoid compounds were tentatively identified in PPF. Among them, isoscutellarein-7-(6’-acetylallosyl-(1->2)-glucoside) and scutellarin methylester were discovered in PPF for the first time. Pretreatment with PPF significantly reduced H2O2-induced cell damage, ROS accumulation, and malondialdehyde content and increased the activity of SOD, CAT, and GSH-Px in buffalo rat liver 3A(BRL3A) cells. Moreover, the expression of nuclear Factor E2-related factor(Nrf2) was upregulated by PPF, whose expression trend was consistent with that of heme oxygenase-1(HO-1), glutamate-cysteine ligase catalytic subunit(GCLC), and NAD(P)H quinone oxidoreductase-1(NQO1). These findings suggested that herbaceous peony flavonoids can be used as a natural bioactive agent to prevent oxidative stress.

文摘[Objectives]To explore the optimal extraction and purification process of the flavonoids in Amaranthus caudatus L.and to study the antioxidant activity in vitro of the flavonoids in A.caudatus.[Methods]Taking A.caudatus as the raw material,flavonoids were extracted by alcohol extraction method,and AB-8 macroporous adsorption resin was selected for purification.The hydroxyl radical scavenging ability,DPPH radical scavenging ability,and O^2-radical scavenging ability were used as evaluation indicators,to explore the antioxidant activity in vitro of the flavonoids in A.caudatus.[Results]The optimal extraction process conditions of flavonoids in A.caudatus are:liquid-to-material ratio 40:1,extraction temperature 60℃,ethanol concentration 60%,ultrasonic power 320 W,extraction time 50 min.Under these conditions,the extraction yield of flavonoids in A.caudatus is(1.35±0.01)%.The optimal purification process conditions of flavonoids in A.caudatus are 2.5 g AB-8 macroporous adsorption resin,sample volume 5 mL,mass concentration of adsorption solution 1.60 mg/mL,pH value of adsorption solution 3.0,sample flow rate 3 BV/h,ethanol concentration in desorption process is 70%and the desorption flow rate is 3 BV/h.Under these conditions,the recovery rate reaches 88.35%±0.68%.[Conclusions]A.caudatus has a high content of flavonoids and has excellent free radical scavenging ability in vitro.This study is intended to provide important technical support for the research of flavonoid activity of A.caudatus and the development of functional products.
文摘Macroalgae serve as a potential feedstock for fucoxanthin extraction.Fucoxanthin,a bioactive pigment found in the chloroplasts of marine algae,exhibits significant pharmacological properties.As a member of the carotenoid family,fucoxanthin plays a crucial role in both the food and pharmaceutical industries.This research explores the effects of ultrasonics on the extraction of fucoxanthin from the marine macroalga Padina australis.In addition,various extraction techniques and the influence of solvents on the efficient separation of fucoxanthin from algae have been studied and compared.Using methanol,chloroform,and a combination of methanol and chloroform(1:1,v/v),conventional fucoxanthin extraction from Padina australis yielded 8.12 mg of fucoxanthin per gram of biomass.However,the ultrasonic-assisted extraction resulted in a significantly higher yield of 16.9 mg of fucoxanthin per gram of biomass,demonstrating that the use of ultrasonics enhances the extraction rate compared to conventional methods.Therefore,the efficient separation of fucoxanthin from Padina australis is highly dependent on ultrasonic-assisted extraction.The process conditions for the extraction were optimized to maximize the yield of fucoxanthin from seaweeds.The following parameters were selected for optimization studies:moisture content,particle size,mixing speed,extraction temperature,extraction duration,and solid-to-solvent ratio.The extracted fucoxanthin exhibited various biological activities,including antimicrobial and antioxidant properties,and its structure was elucidated through FTIR and NMR spectroscopy.Additionally,thin-layer chromatography of the crude algae extracts confirmed the presence of fucoxanthin in the marine algae.Given these findings,the optimized extraction process holds the potential for scaling up to large-scale fucoxanthin production.Fucoxanthin,as a potent pharmacological agent,offers promising applications in the treatment of various ailments.

文摘[ Objective ] This study aimed to investigate the optimal extraction process of crude polysaccharides from wild Lactarius volemus Fr. in Yunnan Province and preliminarily analyzed its antioxidant activity in vitro. [ Method ] With water extraction and alcohol precipitation method, the optimal conditions for extracting crude polysaccharides from wild L. volemus Ft. were screened by single-factor and orthogonal experiments. The antioxidant activity of the extracted crude polysac- charides was determined with DPPH assay. [ Result ] The optimal conditions for pigment removal with activated carbon were: activated carbon amount of 20 g/L, water bath time of 40 min, water bath temperature of40 ℃ ; the optimal conditions for extracting crude polysaccharides from wild L. vo/emus Fr. with hot water ex- traction method were: hot water extraction time of 3 h, solid-liquid ratio of 1: 45, extraction frequency of twice. Under the optimized extraction conditions, the yield of crude polysaccharides was 21.33 mg/g. In addition, the antioxidant activity of 0. 665 mg/ml crude polysaccharides was 52.46% ; the amount of crude polysac- eharides was proportional to the antioxidant activity. [ Conclusion] Hot water extraction method can be used as a high-efficiency extraction technology of crude pol- ysaecharides from wild L. volemus Fr. with simple operation and low costs. Crude polysaccharides extracted from L. volemus Ft. exhibited certain antioxidant activi- ty in vitro.

[Methods] The single factor orthogonal design was applied to explore the effects of ultrasonic time,solid-to-liquid ratio,extraction time and ethanol precipitation concentration on the extraction rate of crude polysaccharides from C. Capitata,to obtain the optimal extraction process; DPPH and ABTS^+ free radical scavenging ability was detected to evaluate its in vitro antioxidant activity.[Results]The optimal extraction process of C. Capitata crude polysaccharide was as follows: solid-to-liquid ratio was 40∶ 1,the extraction temperature was 50℃,the ultrasonic time was 80 min. Under such conditions,the extraction rate of C. Capitata crude polysaccharide was 10. 51%,and the purity was 56. 9%; C. Capitata crude polysaccharide has good scavenging ability to DPPH and ABTS^+ free radicals,and its IC_(50) values are 0. 468 and 0. 231 mg/mL,respectively. [Conclusions]The optimized process is simple and feasible,and the C. Capitata polysaccharide has good antioxidant activity in vitro. This experiment provides a basis for the further development and utilization of the C. Capitata polysaccharide.
文摘In this study the antioxidant activity of barley malt rootlet (BMR) extracts w</span><span style="font-family:"">as</span><span style="font-family:""> evaluated in heat treated corn oil up to 5 hours at 185</span><span style="font-family:"">°</span><span style="font-family:"">C frying temperature. The antioxidant activity </span><span style="font-family:"">of </span><span style="font-family:"">BMR extracts was measured at 25, 50, 100 and 150 ppm concentrations. The free and bound antioxidant phenolics were extracted from BMR using three different extraction methods. Conventional solvent extraction (CSE), microwave assisted extraction (MAE) and autoclave assisted pretreated solvent extraction (APSE). In the present experiment, the total phenolic content and antioxidant activity of the various extracts w</span><span style="font-family:"">ere</span><span style="font-family:""> measured. Thiobarbituric acid reactive substances (TBARS) assay was used to evaluate the ability of the BMR to protect lipid peroxidation in corn oil at 185</span><span style="font-family:"">°</span><span style="font-family:"">C frying temperature. The formation of TBARS at 5 hours of heat treated corn oil ha</span><span style="font-family:"">s</span><span style="font-family:""> shown similar antioxidant levels in 150 ppm butylated hydroxytoluene (BHT) or MAE free phenolic extract added to corn oil. TBARS value for BHT was 1.896 ± 0.013 μg/mL of corn oil and for MAE was 1.896 ± 0.034 μg/mL of corn oil. The highest level of antioxidant activity was found for the free phenolic extracts. The order of inhibition of oxidation was found to be for free phenolics as follows: BHT (100 ppm) > APSE (50 ppm) > MAE (100 ppm) > CSE (100 ppm).

文摘[Objectives]To optimize the extraction process of total flavonoids from Eucommiae Cortex,establish the extraction and content determination methods of its medicinal materials,and study its in vitro antioxidant activity.[Methods]The total flavonoids from Eucommiae Cortex were extracted by reflux extraction method,and the effects of extraction method,extraction solvent concentration,extraction volume,and extraction time on the total flavonoids content of the medicinal materials were explored through the single factor experiment.Orthogonal experiment was carried out to optimize the extraction process conditions,and the optimal extraction process for total flavonoids from Eucommiae Cortex was screened out.The total antioxidant capacity index was used to determine the free radical scavenging ability of the total flavonoids from Eucommiae Cortex.[Results]The optimal extraction process of total flavonoids from Eucommiae Cortex was 50%ethanol,1∶40 solid-to-liquid-ratio,and 1.5 h reflux extraction time.Through the antioxidant experiment in vitro,it is found that the total flavonoids from Eucommiae Cortex have a higher scavenging ability for the total antioxidant capacity,and there is a certain positive correlation with the mass concentration of total flavonoids.[Conclusions]This method can effectively determine the total flavonoids content of Eucommiae Cortex medicinal material,and provide a certain scientific basis for the quality standard research of the medicinal material.The total flavonoids from Eucommiae Cortex have good in vitro antioxidant activity.And the method for extracting total flavonoids from Eucommiae Cortex medicinal material in this experiment has good repeatability,high stability and feasibility.
文摘The aim of this study was to investigate the extraction method for R.tomentosa and C.zeylanicum leaves and the evaluation of antibacterial and antioxidant activities of crude extracts.The results of the study showed that the active ingredients of crude extracts were clearly separated by Thin-layer chromatography and the presence of rhodomyrtone in R.tomentosa crude extract and cinnamaldehyde in C.zeylanicum crude extract.R.tomentosa crude extract was antibacterial activity against Staphylococcus aureus with 13.1 mm of inhibition zone,but is not effective against Salmonella Typhimurium.C.zeylanicum leaf extract did not show antibacterial activity on both S.aureus and S.Typhimurium.At a dilution of 1/2 of the R.tomentosa crude extract can completely inhibit S.aureus growth.This study also indicated the presence of antioxidant compounds such as flavonoids,tannins,phenols and terpenoids in C.zeylanicum and R.tomentosa crude extracts.The results showed that R.tomentosa and C.zeylanicum crude extracts should be used as a biotherapy alternative to antibiotic therapy.However,further study would be needed to investigate the antibacterial activity of crude extracts in vivo.

文摘Marine organisms constitute unlimited resource of bioactive substances due to their high biodiversity and represent a valuable source of new compounds. This study optimized the alkali-extraction conditions and antioxidant activities of soluble polysaccharides from the body wall of Sipunculus nudus. The effects of solid-liquid ratio, extraction duration, extraction temperature, and alkali concentration on the yield of S. nudus polysaccharides(SNP) were examined, according to which the optimal combination of extraction parameters was obtained by an orthogonal test. The relative influencing importance of different extraction parameters on the yield of SNP followed the order as solid-liquid ratio > extraction temperature > alkali concentration > extraction duration. The highest extraction yield, 1.98%, was achieved under an optimal extraction condition: temperature 60℃, solid-liquid ratio 1:6 g mL^(-1), duration 5 h, and alkali(Na OH) mass fraction 6%. The in vitro antioxidant activities examination showed that extracted SNP under this optimized condition had strong power in reducing certain hydroxyl and superoxide radical scavenging abilities. The promising results showed that extracted SNP could be a potent natural antioxidant.

文摘Objective:To isolate and analyse the chemical composition in the essential oils and free radical scavenging activity of different crude extracts from the fresh and dry leaves of vegetable plants of Lactuca sativa L.(L.sativa).Methods:The essential oils and volatile chemical constituents were isolated from the fresh and dry leaves of L.sativa(lettuce)grown in Sultanate of Oman by hydro distillation method.The antioxidant activity of the crude extracts was carried out by well established free radical scavenging activity(DPPH)method.Results:About 20 chemical compounds of different concentration representing 83.07%and 79.88%respectively were isolated and identified by gas chromatography-mass spectroscopy in the essential oils isolated from the fresh and dry leaves asα-pinene(5.11%and 4.05%).γ-cymene(2.07%and 1.92%),thymol(11.55%and 10.73%),durenol(52.00%and 49.79%),α-terpinene(1.66%and 1.34%),thymol acetate(0.99%and 0.67%),caryophyllene(2.11%and 1.98%),spathulenol(3.09%and 2.98%).camphene(4.11%and3.65%),limonene(1.28%and 1.11%)representing these major chemical compounds.However,some other minor chemical constituents were also isolated and identified from the essential oil of lettuce including p-pinene,α-terpinolene,linalool,4-terpineol,α-terpineol,o-methylthymol,L-alloaromadendrene and viridiflorene.Conclusions:The chemical consliluents in the essential oils from the locally grown lettuce were identified in the following classes or groups of chemical compounds such as monoterpenes,sesquiterpenes volatile organic compounds and their oxygenated hydrocarbons.Therefore,the essential oils and the crude extracts from Omani vegetable species of lettuce are active candidates which would be used as antioxidant,antifungal or antimicrobial agents in new drugs preparation for therapy of infectious diseases.

文摘The present study was designed to establish and optimize a new method for extracting chlorogenic acid and cynaroside from Lonicera japonica Thunb. through orthogonal experimental designl. A new ultrahigh pressure extraction (UPE) technology was applied to extract chlorogenic acid and cynaroside from L. japonica. The influential factors, including solvent type, ethanol concentration, extraction pressure, time, and temperature, and the solid/liquid ratio, have been studied to optimize the extraction process, The optimal conditions for the UPE were developed by quantitative analysis of the extraction products by HPLC-DAD in comparison with standard samples. In addition, the microstructures of the medicinal materials before and after extraction were studied by scanning electron microscopy (SEM). Furthermore, the extraction efficiency of different extraction methods and the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities of the extracts were investigated. The optimal conditions for extracting chlorogenic acid and cynaroside were as follows: ethanol concentration, 60%; extraction pressure, 400 MPa; extraction time, 2 rain; extraction temperature, 30 ℃; and the solid/liquid ratio, 1 : 50. Under these conditions, the yields of chlorogenic acid and cynaroside were raised to 4.863% and 0.080%, respectively. Compared with other extraction methods, such as heat reflux extraction (HRE), ultrasonic extraction (UE), and Sohxlet extraction (SE), the UPE method showed several advantages, including higher extraction yield, shorter extraction time, lower energy consumption, and higher purity of the extracts. This study could help better utilize L. japonica flower buds as a readily accessible source of natural antioxidants in food and pharmaceutical industries.

文摘To study the changes of volatile compounds and antioxidant activity of fast fermented shrimp head paste during the fermentation processing,Penaeus vannamei boone head was used as the raw material,and compound starter was added for fermen-tation.During the 14 days of fermentation,the changes of free amino acids,volatile flavor compounds and main components were determined by SPME-GC-MS and electronic nose.The oxidation-resistance changes of water extract at different fermentation stages were evaluated.The results showed that the total free amino acids increased from original 2320 to 5640 mg(100 mL)^(−1).SPME-GC-MS analysis found that aldehydes,pyrazine compounds increased from 10.80%,1.94%to 24.35%,13.63%respectively during the fermentation process.The HO·scavenging ability of shrimp head paste increased from 52%to 86%.Our results showed that the shrimp paste produced from shrimp head fermentation could obtain good flavor and possess certain antioxidant activity of aquatic fermented condiment.

文摘Antioxidant and antimicrobial activities of Echinacea purpurea L. (Asteraceae) extracts obtained by classical and ultrasound solvent extraction were compared. The dry aerial part of plant was extracted by 70% etha-nol at a solid-to-liquid ratio of 1︰10 (m/v) and 25°C. The extract obtained by classical solvent extraction contained 29% larger amounts of phenolic compounds and 20% higher content of flavonoids. 2,2-diphenyl-1-picril hydrazyl radical (DPPH) scavenging reached 93.6% and the values of EC50 were (34.16±0.65) μg·ml-1 and (65.48±1.12) μg·ml-1 for the extracts obtained by the classical and ultrasound extractions, respectively. The extracts, independent of the extraction technique applied, showed a considerable growth inhibition on Candida albicans and Saccharo-myces cerevisiae, while no growth inhibition zones were observed for Aspergillus niger. The diameters of inhibition zone observed for all the microorganisms were larger for extracts obtained by classical extraction than those by ul-trasound extraction.

文摘The polyphenol content,antioxidant and antimicrobial activities of the extracts obtained by classical, ultrasonic and Soxhlet extractions from dry aerial parts of two Artemisia species(Artemisia vulgaris and Artemisia campestris)were compared.Ultrasound positively affected the yield of extractive substance and the kinetics of extraction,but the extract obtained by the classical extraction showed the highest antioxidant activities and contained higher total contents of phenolic compounds and flavonoids than the extracts obtained by two other extraction techniques.Both flavonoid aglycones(apigenin,quercetin,quercetin 3,3′-dimethyl ether)and flavonoid glycosides(rutin,hyperoside and kaempferol 3-rhamnoside)were identified by thin layer chromatograph(TLC)analysis in the extracts from both species.A.campestris extracts were richer in quercetin than A.vulgaris and its antimicrobial activity was also better than A.vulgaris.Extracts obtained from both species were found to be more effective on the tested yeasts than bacteria.The kinetics of the total extractive substances,such as phenolic,flavonoids and quercetin extraction,was successfully described by the model of unsteady-state diffusion.

文摘[Objectives]To explore the optimal extraction process and the antioxidant activity of flavonoidsof Cacumen platycladi. [Methods]FCPwas obtained from C. platycladi by a new set extraction process of microwave-assisted extraction,freeze thawing,superfine crushingand freeze drying technology and purification. On the basis of single-factor tests,the quadratic regression parasitism model was established by the extraction amount of total flavonoids with microwave power,alcohol concentration,and liquid-to-solid ratio through the response surface methodology. [Results] The optimal extraction conditions were microwave power of 757 W,alcohol concentration of 69%,liquid-to-solid ratio of21∶ 1,with the optimal extraction yield of 15. 302 9 mg/g. Under the real conditions( microwave power of 800 W,alcohol concentration of69%,liquid-to-solid ratio of 21∶ 1) in our laboratory,the maximum yield of total flavonoids was 15. 9 mg/g,the results fitwell with the prediction of quadratic regression parasitism model. The good antioxidant activity has been determined byscavenging effect of FCP on 1,1-diphenyl-2-picrylhydrazyl( DPPH) radical and superoxide anion free radical with the scavenging yield of 70. 2% and 68. 8% respectively. [Conclusions]All results established a good foundation for furtherdevelopment of C. platycladi products.
