Immunomodulation and liver protection of Yinchenhao decoction against concanavalin A-induced chronic liver injury in mice 被引量:11

OBJECTIVE:This study investigated the immunoregulatory and protective roles of Yinchenhao decoction,a compound of Chinese herbal medicine,in a mouse model of concanavalin A(Con A)-induced chronic liver injury.METHODS:Female Bal B/c mice were randomly divided into 4 groups:normal control,Con A model,Con A model treated with Yinchenhao decoction(400 mg/kg,orally),and Con A model treated with dexamethasone(0.5 mg/kg,orally).All treatments were given once a day for 28 d.Except of the normal control,mice received tail vein injection of Con A(10 mg/kg)on days 7,14,21,and 28,at 1 h after treatment with Yinchenhao decoction or dexamethasone or saline to induce chronic liver injury.RESULTS:Repeated Con A injection induced chronic liver injury,which was evidenced by infl ammatory cell infi ltration and necrosis,increased serum alanine aminotranferease activities,decreased albumin levels,and an imbalanced expression of immunoregulatory genes in the liver tissues including signifi cantly enhanced interferon-γ,interleukin-4,monocyte chemotactic protein-1,and cluster of differentiation 163 m RNA levels,and reduced tumor necrosis factor-αand interleukin-6 m RNA levels.Treatment with Yinchenhao decoction signifi cantly reversed the Con A-induced changes in immunoregulatory gene expression in the liver tissues,reduced serum alanine aminotranferease activity,enhanced serum albumin level,and attenuated the extent of liver infl ammation and necrosis.Furthermore,Yinchenhao decoction did not result in hepatocyte degeneration and spleen weight loss that were observed in mice received long-term treatment with dexamethasone.CONCLUSION:Yinchenhao decoction treatment protected liver against the Con A-induced chronic liver damage and improved liver function,which were associated with the modulation of gene expression related to immune/infl ammatory response. 展开更多

Decoction vs extracts-mixed solution: effect of Yiqihuoxue formula on non-alcoholic fatty liver disease in rats

OBJECTIVE: To investigate the effects on non-alcoholic fatty liver disease (NAFLD) in rats of the decoction of Yiqihuoxue formula and the solution prepared with the extracts from the individual herbal medicines of the formula. METHODS: The rat models of NAFLD were established with high-fat diet (HFD) for 10 weeks. Thirty-two rats were randomly divided into 4 groups: the control group, the model group, the decoction group and the solution group, 8 for each group. From the 6th week, drinking water, the decoction and the solution were intragastrically administrated accordingly to the rats for 5 weeks. The pathological changes of the liver tissues were observed with Hematoxylin and eosin staining, triglyceride levels in liver tissues measured, serum alanine aminotransferase (ALT) activity measured, and serum gastrin and motilin tested. RESULTS: Fatty degeneration and vacuole-like changes to various degrees occurred in hepaticcells of the model group. Indicators for fat metabolism, serum ALT activity and hepatic triglyceride level significantly increased, while serum gastrin and motilin levels significantly decreased. Serum ALT activity and the fatty deposition in hepatocytes were significantly reduced. In the meantime, the expressions of gastrin and motilin in the serum rose significantly in the treatment groups. CONCLUSION: Both the decoction and the extracts-mixed solution had effect on NAFLD of protecting the liver function and reducing the fatty deposition in liver, which might be achieved by regulating the expression of gastrin and motilin. 展开更多

Short-Term Efficacy of Treating Hepatitis B Virus-Related Acute-on-Chronic Liver Failure Based on Cold Pattern Differentiation with Hot Herbs: A Randomized Controlled Trial 被引量:5

Objective: To evaluate the clinical efficacy and safety of Yinchen Zhufu Decoction(茵陈术附汤, YCZFD) in the treatment of acute-on-chronic liver failure caused by hepatitis B virus(HBV-ACLF) with cold pattern in Chinese medicine(CM). Methods: This is a multi-center randomized controlled trial of integrative treatment of CM and Western medicine(WM) for the management of HBV-ACLF patients. A total of 200 HBV-ACLF patients with cold pattern were equally randomly assigned to receive YCZFD and WM(integrative treatment) or WM conventional therapy alone respectively for 4 weeks. The primary end point was the mortality for HBV-ACLF patients. Secondary outcome measures included Model for End-Stage Liver disease(MELD) score, liver biochemical function, coagulation function and complications. Adverse events during treatment were reported. Results: The mortality was decreased 14.28% in the integrative treatment group compared with WM group(χ^2=6.156, P=0.013). The integrative treatment was found to significantly improve the MELD score(t=2.353, P=0.020). There were statistically significant differences in aspartate transaminase, total bilirubin, indirect bilirubin, direct bilirubin and prothrombin time between the two groups(P〈0.05 or P〈0.01). The complications of ascites(χ^2=9.033, P=0.003) and spontaneous bacteria peritonitis(χ^2=4.194, P=0.041) were improved significantly in the integrative treatment group. No serious adverse event was reported. Conclusions: The integrative treatment of CM and WM was effective and safe for HBV-ACLF patients with cold pattern in CM. The Chinese therapeutic principle "treating cold pattern with hot herbs" remains valuable to the clinical therapy.(Trial registration No. Chi CTR-TRC-10000766) 展开更多
