目的:建立以药物活性和专属性菧类成分longistylin A与longistylin C为评价指标的树豆(木豆)药材质量控制方法。方法:采用硅胶柱色谱分离纯化有关成分,光谱技术鉴定化学结构;反相高效液相色谱法测定指标成分的含量。结果:从树豆叶分离... 目的:建立以药物活性和专属性菧类成分longistylin A与longistylin C为评价指标的树豆(木豆)药材质量控制方法。方法:采用硅胶柱色谱分离纯化有关成分,光谱技术鉴定化学结构;反相高效液相色谱法测定指标成分的含量。结果:从树豆叶分离鉴定了2种指标成分longistylin A和longistylin C;采用Thermo BDS Hypersil C18色谱柱(4.6 mm×250 mm,5μm),以甲醇-水(80∶20)为流动相可使指标成分获得良好分离;longistylin A和longistylin C分别在0.002 88~0.057 6μg和0.011 2~0.224μg呈良好的线性关系,平均回收率分别为98.9%,97.2%,RSD分别为2.4%,2.2%。结论:建立了以树豆叶中药物活性菧类成分为指标的药物分析方法,可用于树豆(木豆)药材的质量控制。 展开更多

The present study was designed to identify potent anti-tumor compounds from a series of new longistylin C derivatives.Ten longistylin C derivatives were synthesized and their structures were confirmed by ~1H NMR,MS,and elemental analyses.Their cytotoxicity in vitro against three human cancer cell lines(A549,HepG2,and MCF-7) were evaluated by the MTT assay.Among these compounds,DT-6 and DT-9 displayed much better cytotoxicity against A549,HepG2,and MCF-7 cells,DT-1 exhibited selective cytotoxicity against HepG2,and the structure-activity relationships were investigated.In conclusion,Compounds DT-6 and DT-9 may serve as potential lead compounds for the discovery of new anti-cancer drugs. 展开更多
