Natural nano-hydroxyapatite(HA)was extracted from Pacific cod(Gadus macrocephalus)bone with a thermostable col-lagenolytic protease in the present study.Conditions for the enzymatic reaction were optimized to be 60℃and pH 7.0,and a desir-able extraction efficiency was achieved by using the crude collagenolytic protease.Dynamic light scattering,transmission electron microscopy and energy-dispersive X-ray analysis revealed that nano-HA are anionic spherical(about 110nm)particles mainly com-prised of calcium and phosphorus at an approximate ratio of 5:3.As evaluated with the mouse ex vivo intestinal segments,the extracted nano-HA displayed comparable level of intestinal bioavailability to the positive control CaCl_(2).By treating with inhibitors(NaN3,ami-loride)and low temperature(4℃),clathrin-mediated endocytosis was assumed to involve the intestinal absorption of nano-HA.Over-all,the application of thermostable collagenolytic protease is proved to be a promising alternative method for nano-HA extraction from natural resource with improved ecological and biological value. 展开更多

以0号柴油水溶性成分(water soluble fraction of No.0 fuel oil,WSF)为污染物,采用暴露试验法研究了黑鲷(Sparus macrocephalus)肝脏、鳃和肌肉组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽硫转移酶(GST)、过氧化物酶(POD)... 以0号柴油水溶性成分(water soluble fraction of No.0 fuel oil,WSF)为污染物,采用暴露试验法研究了黑鲷(Sparus macrocephalus)肝脏、鳃和肌肉组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽硫转移酶(GST)、过氧化物酶(POD)活性的变化和脂质氧化水平(MDA)。利用整合生物标志物响应(integrated biomarker responses,IBR,RIB)指数定量化评价0号柴油污染胁迫对黑鲷不同组织的毒性效应。结果表明,黑鲷受0.60、0.30和0.015 mg·L-1WSF胁迫后,肝脏、鳃和肌肉中SOD、CAT和GST酶活性呈现诱导-抑制的规律性变化,POD活性呈现抑制-诱导的规律性变化,MDA含量则呈现先降低后升高的变化趋势。各组织表现出一定的差异性,且肝脏较早诱导酶活性。依据RIB值大小分析得到各组织氧化应激能力水平由高到低依次为肝胰腺、鳃和肌肉。RIB值总体表现为前期升高后期下降的变化趋势,肝脏组织RIB值与WSF浓度具有较好的正相关关系。 展开更多

采用分步降温法冷冻保存太平洋鳕(Gadus macrocephalus)精液,并用扫描电镜和透射电镜技术研究了精子的超微结构损伤。分析了添加剂(蛋黄)及五种抗冻剂(PG(丙二醇)、DMSO(二甲基亚砜)、EG(乙二醇)、GLY(甘油)、MeOH(甲醇))不同浓度(8%、... 采用分步降温法冷冻保存太平洋鳕(Gadus macrocephalus)精液,并用扫描电镜和透射电镜技术研究了精子的超微结构损伤。分析了添加剂(蛋黄)及五种抗冻剂(PG(丙二醇)、DMSO(二甲基亚砜)、EG(乙二醇)、GLY(甘油)、MeOH(甲醇))不同浓度(8%、10%、12%、14%、16%、18%)对太平洋鳕精子冷冻保存效果的影响。实验结果表明,蛋黄能够显著提高太平洋鳕冷冻保存效果(P<0.05);12%PG中添加10%蛋黄保存的冻融精子运动率最高(85.87%±1.6%),显著高于其他冻存组(P<0.05)。添加蛋黄的12%浓度的DMSO组解冻后精子运动速率较高,平均直线速度、平均曲线速度、平均路径速度分别达到了(120.39±20.78)μm/s、(120.39±20.78)μm/s、(155.64±12.02)μm/s,与其他各实验组差异显著(P<0.05)。通过扫描电镜和透射电镜观察新鲜和冻融后的鳕鱼精子发现,鲜精中75.5%精子形态结构正常、24.5%精子形态结构异常;运动率最高的冻精中67%精子形态结构正常、33%精子形态结构异常。超低温保存对太平洋鳕精子结构产生了显著影响,细胞膜破裂、线粒体肿胀变形,鞭毛脱落、断裂。 展开更多

The relationship betaren SGR and ration levels was a decelerating curve. Stepwise mpgression was used to derelop the predictive models for the specific growth rates. Maintenance rations and optimum ratlons both increased with increased temperature. The relationship between maintenance. optimum rations and body welght wds influenced by temperuture. Conversion efficiencies increased with ration from zero at maintenance ration to a peak at the optimum ration, then declined with further inceased ration. 展开更多

The sea area surrounding Baishishan Island has abundant fishery resources because of its plentiful food and rocky reefs that provide refuge from predators.However,overfishing has occurred in many areas and some species’local populations need to be replenished through stocking programs.This case study,using Sparus microcephalus as the test species,designed and analyzed detailed stocking strategies to improve the survival ratio and control the active range of the released fishes.Controlled variables included release location,release time,and tidal level.Two stocking strategies were designed,executed,and evaluated for comparison.A traditional stocking method was applied to the high flow area of Baishishan Island in Xiangshan Bay,East China Sea.The released fry were transported by boat and released to the sea using buckets.Released fish were traced using plastic tags.The average distance of the 19 recaptured fry from the release point was 7.6 km with 52.9%mortality rate.The distance at recapture and mortality rate among recaptured individuals showed that the traditional releasing method was not successful.An in situ stocking method was used in a low tide area and natural process was imitated to decrease the stress response and promote physiological adaptation.The purpose of this method was to prolong the release process applying temporary net cages at the release location,which resulted in a regional distribution of the released fish and possibly improving the food availability.A new Sr+tagging technique was adopted to evaluate the release effect and quantify comparative release data.Only 1 fish among the 354 caught was found farther than 1 km from the release point in the following 6 months.The results validated the promotion of the new stocking technique. 展开更多

Rodents with prevailing subterranean activity usually play an important role in the ecosystems of which they are a part due to the combined effect of herbivory and soil perturbation.This is the case for the giant root-rat Tachyoryctes macrocephalus endemic to the Afroalpine ecosystem of the Bale Mountains,Ethiopia.We studied the impact of root-rats on various ecosystem features within a 3.5-ha study locality dominated by Alchemilla pasture,which represents an optimal habitat for this species,in 2 periods of a year.The root-rats altered plant species composition,reducing the dominant forb,Alchemilla abyssinica,while enhancing Salvia merjame and a few other species,and reduced vegetation cover,but not the fresh plant biomass.Where burrows were abandoned by root-rats,other rodents took them over and A.abyssinica increased again.Root-rat burrowing created small-scale heterogeneity in soil compactness due to the backfilling of some unused burrow segments.Less compacted soil tended to be rich in nutrients,including carbon,nitrogen and phosphorus,which likely affected the plant growth on sites where the vegetation has been reduced as a result of root-rat foraging and burrowing. 展开更多

驯化野生大头鳕(Gadus macrocephalus Tilesius)作为亲鱼,通过自然受精和人工授精两种方法获得受精卵,自然受精卵通过虹吸法收集。受精卵孵化条件为水温6~8℃,盐度32~33,pH7.8~8.2,微充气。在奥林巴斯SZ61解剖镜下对大头鳕的胚胎发育... 驯化野生大头鳕(Gadus macrocephalus Tilesius)作为亲鱼,通过自然受精和人工授精两种方法获得受精卵,自然受精卵通过虹吸法收集。受精卵孵化条件为水温6~8℃,盐度32~33,pH7.8~8.2,微充气。在奥林巴斯SZ61解剖镜下对大头鳕的胚胎发育进行了观察,并使用CCD图像传感器拍照,描述了各发育时期的发育时序和形态特征。大头鳕受精卵为圆球形端黄卵,弱黏性,无油球,卵膜上具龟裂结构,卵子直径0.95~1.12mm,n=30,卵黄均匀透明。类似于大部分硬骨鱼类,其胚胎发育可分为胚盘形成期、卵裂期、桑椹期、囊胚期、原肠期、神经胚期、器官发生期、尾芽期、肌肉效应期。在水温6~8℃,盐度32~33的条件下,受精卵历时约336h完成胚胎发育过程,孵化出膜。在大头鳕胚胎发育过程中,具有4个不同于其他硬骨鱼类的地方:(1)发现早期分裂球具有不规则现象;(2)很多胚胎发育过程不出现柯氏囊,有柯氏囊的胚胎仅占10%左右,且出现晚,消失快;(3)胚胎孵化为仔鱼与发育进程关联性不大,孵化的仔鱼有3种主要形态;(4)胚胎各个时期都有较高死亡率,以原肠期最为严重。 展开更多

通过对江鳕(LotalataLinnaeus)和大头鳕(GadusmacrocephalusTilexius)的形态学比较研究发现,江鳕和大头鳕都有胆囊、幽门盲囊,肝脏较大。脊柱和附肢骨骼种类相同,区别仅在于骨块的数量和嵴的发达程度上,尾都是等尾型。

The complex life histories of demersal fishes are artificially separated into multiple stages along with changes in morphology and habitat.It is worth exploring whether the phenotypes expressed earlier and later during the life cycle are related or decoupled.The life stages of first year Pacific cod(Gadus macrocephalus)were tracked over different hatch years and regions to test whether the early life history had a long-lasting effect on subsequent growth.We further explored the contribution of growth in the early and subsequent life history stages to body size at the end of each stage.In addition to the accessory growth centre and the first annual ring,the other two checks on the otolith possibly related to settlement and entering deeper waters were identified in 75 Pacific cod individuals.The direct and indirect relationships among the life history stages was interpreted based on path analysis.The results showed that growth prior to the formation of the accessory growth centre had a significant effect on the absolute growth of the fish before and after settlement and migration to deep water.However,there was no or moderate evidence that early growth affected the body size at each stage,which was mainly regulated by growth during the stage.This study supports the lasting effect of early growth and clarifies that it affects size mainly by indirectly regulating staged growth.Quantifying the phenotype relationships and identifying the internal mechanisms form the basis for assessing population dynamics and understanding the processes behind the changes. 展开更多
