Background Liver fibrosis and hepatocellular carcinogenesis secondary to liver fibrosis are serious liver diseases with no effective treatments.Mori fructus aqueous extracts(MFAEs)have served as successful treatments for many types of liver injury including fibrosis although the molecular mechanisms are unknown at present.Purpose To investigate the effect of MFAEs in alleviating acute and chronic liver injury and tried to decipher the underlying mechanism.Methods and results Mice were divided into 5 groups(n ps:contro=8)for acute(groups:control,0.3%CCl_(4),bifendate(BD),100 and 200 mg/kg MFAEs,7 d)and chronic(groul,10%CCl_(4),BD,100 and 200 mg/kg MFAEs,4 weeks)liver injury study.Each mouse was injected intraperitoneally with 10μL/g corn oil containing CCl_(4)expect the control group.HepG2 cells were used in vitro study.Eighteen communal components were identified by UPLC-LTQOrbitrap-MS.We utilized a mouse model for acute and chronic liver injury using CCl_(4)and MFAEs administration effectively blocked fibrosis and significantly inhibited inflammation in the liver.MFAEs activated the nuclear factor erythroid derived 2 like 2/heme oxygenase 1(Nrf2/HO-1)pathway and promoted the synthesis of the antioxidants glutathione(GSH),superoxidedismutase(SOD)and glutathione peroxidase(GSH-Px)that resulted in reduced levels of CCl_(4)-induced oxidative stress molecules including reactive oxygen species.These extracts administered to mice also inhibited ferroptosis in the liver by regulating the expression of Acyl-CoA synthetase long chain family member 4(ACSL4),solute carrier family 7 member 11(SLC7A11)and glutathione peroxidase 4(GPX4),thus reducing the occurrence of liver fibrosis.Both in vivo and in vitro tests indicated that the mechanism of MFAEs protection against liver fibrosis was linked to activation of Nrf2 signaling.These effects were blocked in vitro by the addition of a specific Nrf2 inhibitor.Conclusion MFAEs inhibited oxidative stress,ferroptosis and inflammation of the liver by activating Nrf2 signal pathway and provided a significant protective effect against CCl_(4)-induced liver fibrosis. 展开更多

[Objectives]In order to clarify the regulatory effects of insect hormones on the expression of BmSuc1 and provide a reference for further analysis of the function and expression regulation mechanism of BmSuc1,this study explored the expression profiles of BmSuc1 in different tissues and periods of silkworm larvae and the expression changes of BmSuc1 after treatment with exogenous hormones.[Methods]By using the real-time fluorescence quantitative PCR technique,the expression characteristics of BmSuc1 were detected in different periods,different tissues and after treatment with exogenous hormones during the development of silkworm larvae.The expression of BmSuc1 and 20E receptor gene USP was detected after RNA interference with double-stranded RNA(dsRNA)of USP.[Results]The relative expression of BmSuc1 gene in the midgut was the highest,followed by the silk glands,epidermis and hemolymph.However,there was much lower or almost no expression in other tissues.In addition,the BmSuc1 expression profile exhibited a pulse-like pattern in silkworm larvae.The expression level of BmSuc1 was higher at each instar stage before molting,late fifth instar before cocooning and prepupal stage.Silkworm larvae at day 2 of the fifth instar were treated with 20-hydroxyecdysone(20E)and juvenile hormone(JH).It was found that the expression of BmSuc1 was extremely significantly higher at 12 and 18 h after 20E treatment than the control group injected with 0.1%dimethyl sulfoxide(DMSO)(P<0.01,the same below).But there were no significant difference in BmSuc1 expression between the JH treatment and the control group during the measurement time range(P>0.05).The dsRNA of USP was synthesized in vitro and injected into silkworm larvae at day 3 of the fifth instar.It was showed that the USP relative expression was extremely significantly down-regulated at 24 and 36 h after injection,which indicated that dsRNA interference was successful.RNAi of USP would block 20E signal transduction,and the expression of BmSuc1 was inhibited and significantly down-regulated at 24 and 36 h after injection of dsRNA of USP(P<0.05).[Conclusions]The BmSuc1 expression peaks appeared in the molting of silkworm larvae and the metamorphosis of larvae to pupae,which suggests that BmSuc1 may be involved in the metamorphic development process of silkworms.Treatment with exogenous ecdysone 20E can activate the expression of BmSuc1,but blocking the 20E signal transduction pathway may suppress expression of BmSuc1.It indicates that BmSuc1 as a downstream target gene in the 20E signal transduction pathway is directly or indirectly regulated by 20E signals. 展开更多

目的分析比较不同Mori分型颅内段椎动脉重度狭窄(SIVAS)的颅内、颅外段椎动脉彩色多普勒血流成像(CDFI)特点与血流动力学参数的差异。方法回顾性连续纳入2017年1月至2018年6月因症状性单侧SIVAS住院治疗的患者共122例(122支),通过CT血... 目的分析比较不同Mori分型颅内段椎动脉重度狭窄(SIVAS)的颅内、颅外段椎动脉彩色多普勒血流成像(CDFI)特点与血流动力学参数的差异。方法回顾性连续纳入2017年1月至2018年6月因症状性单侧SIVAS住院治疗的患者共122例(122支),通过CT血管成像或DSA影像,按照Mori分型的标准分为MoriA、B、C型,分别检测椎动脉狭窄段的CDFI血流特征、狭窄段与狭窄近段的收缩期峰值流速(PSV)、舒张期末流速(EDV)、平均流速(MFV)、血管搏动指数(PI)、阻力指数(RI)并进行分析比较。结果122例(122支)患者,MoriA型46例(46支),MoriB型55例(55支),MoriC型21例(21支)。不同Mori分型间患者年龄、性别、病变侧别和脑血管病危险因素如高血压病、糖尿病、高血脂、吸烟以及后循环供血区脑梗死发生率方面差异均无统计学意义(均P>0.05);MoriC型组平均病变长度[(16.0±6.3)mm]明显高于MoriB型[(7.0±1.6)mm]和MoriA型[(3.4±0.8)mm],差异有统计学意义(F=145.297,P<0.01)。MoriA型和B型颅内经颅彩色多普勒超声检测表现为狭窄段局限性血流充盈不全,血流束变细,呈“五彩相间”血流信号;MoriC型中12支(57.1%)椎动脉显示间断性血流成像或血流中断征。MoriC型椎动脉颅内段狭窄处及颅外段的PSV、EDV、MFV均明显低于A型和B型[颅内段:分别为34(8,198)cm/s比231(206,296)、234(205,294)cm/s,13(0,88)cm/s比107(85,143)、115(91,152)cm/s,19(3,128)cm/s比142(127,193)、155(129,196)cm/s;颅外段:分别为38(34,45)cm/s比50(41,58)、47(41,56)cm/s,8(7,13)cm/s比18(14,20)、17(12,24)cm/s,19(16,23)cm/s比28(23,33)、26(22,35)cm/s],PI值、RI值明显高于A型和B型[分别为0.98(0.78,1.26)比0.87(0.74,1.02)、0.78(0.71,0.90),0.75(0.68,0.82)比0.64(0.61,0.69)、0.64(0.59,0.71)],差异均有统计学意义(均P<0.017),而MoriA型与B型间各血流动力学参数差异均无统计学意义(均P>0.017),MoriC型病变中74.1%(15支)未达到重度狭窄的血流速度标准。结论SIVAS的MoriC型病变的局部彩色多普勒显像特点、颅内及颅外段血流动力学参数与MoriA型和B型病变存在差异,可表现为颅外段低速高阻和颅内段血流信号不连续的征象。 展开更多

In this study,silkworm strain T6,tolerant to fluoride,and silkworm strain 733xin,highly sensitive to fluoride,were used to construct the near-isogenic lines.300 random primers were used in RAPD amplification to DNAs of these lines.A molecular marker named S207 was found linked to the fluoride tolerance gene.Examination to F 2 segregated individuals of the above lines verified that this molecular marker was reliable.Subsequently,the molecular marker was cloned into a T vector (pUCm-T) for sequencing.Comparing with sequences available in the GenBank showed that this molecular marker was novel.We plan to convert it into a SCAR marker to facilitate establishment of a molecular marker assisted breeding system. 展开更多

[Objective] The aim of this study was to investigate the efficient technique of artificial insemination for silkworm. [Method] Sperms were extracted from bursa copulatrix of female moths mated for 30 min through extruding and centrifugal method, and then the semen was injected into other virgin moths with trypsinase. [ Result] A high-effective collection technology of spermatids from silkworm was established successfully, 50 μl semen could be collected by only one person in each hour. The survival rate of spermatids was over 80% in vito after collected from bursa copulatrix, while the obtained semen was quite pure and the average fertilization rate of silkworm was 76,5%. [ Conclusion] The establishment of high-effective semen extraction technique of silkworm provides the technical basis for studies on other related techniques for silkworm sperm. 展开更多

车铣复合机床与传统的五轴联动机床在结构上有区别,所控制的轴数超过5个。因此,在开发后置处理过程当中,与传统的五轴机床有较大区别,需要根据不同的功能来分析不同的运动链。文章主要以1500S车铣复合机床为例,详细介绍该类机床后置处... 车铣复合机床与传统的五轴联动机床在结构上有区别,所控制的轴数超过5个。因此,在开发后置处理过程当中,与传统的五轴机床有较大区别,需要根据不同的功能来分析不同的运动链。文章主要以1500S车铣复合机床为例,详细介绍该类机床后置处理开发过程。首先建立MORI SEIKI车铣数控机床的运动坐标系,根据运动链分析及坐标变换,推导出MORI SEIKI机床的转角计算公式和坐标变换公式,分析了回转角在各个象限的取值;利用JAVA语言开发出MORI SEIKI 1500S数控机床的专用后置处理软件,并通过某叶片仿真加工验证了该后置处理算法的正确性。该研究不仅为用户解决了软件使用问题,也为车铣复合类多轴机床提供了后置处理开发思路。 展开更多

[ Objective] The experiment aimed to clone Paramyosin/mini-Paramyosin (PM/mPM) gene to analyze the relations between it and moving behaviors. [Method] PCR method and RACE technology were used to obtain whole cDNA of PM and some cDNA of mPM of Bombyx moil. By comparing wgs of Bombyx mori, the genomic sequence of PM/mPM of Bombyx mori was obtained, and, their genome structure was determined. [ Result] PM/mPM genes consisted of 17 exons and 16 intrens. By the use of selective promoters, The gene sequence encoded PM and mPM, while PM and mPM shared the last 6 exons. The cluster analysis between PM of Bombyx mori and PM of other invertebrate animals demonstrated that the relation between Bombyx mori and Bombyx mandarina was closest and the relation between Bombyx mori and Drosophila melanogaster was farthest in Insecta. [ Conclusion] There was no point mutation which could influence flight in amino acid sequence of PM of Bombyx mori and Bombyx mandarina, so the difference of flight capacity of Bombyx mori and Bombyx mandarina might be regulated by other mechanism. 展开更多

目的比较Mori A型颅内椎基底动脉重度狭窄血管内支架成形术与单纯药物治疗疗效上的差异。方法回顾性分析122例Mori A型颅内椎基底动脉重度狭窄病人的临床资料,按处理方法不同分为药物组和支架组,主要终点事件定义为缺血性卒中、短暂性... 目的比较Mori A型颅内椎基底动脉重度狭窄血管内支架成形术与单纯药物治疗疗效上的差异。方法回顾性分析122例Mori A型颅内椎基底动脉重度狭窄病人的临床资料,按处理方法不同分为药物组和支架组,主要终点事件定义为缺血性卒中、短暂性脑缺血发作(TIA)或死亡。比较两组间围手术期及1年内主要终点事件及mRS评分的差异。结果两组30 d内主要终点事件发生率(5.8%vs 13.2%)差异无统计学意义(P=0.272)。1年随访期内支架组累积无终点事件发生率明显高于药物组(92.8%vs 74.2%,P<0.05),支架组改良RANKIN量表(mRS)评分≤2分病人比例明显高于药物组(92.8%vs75.5%,P<0.01)。结论对于Mori A型颅内椎基底动脉重度狭窄病人,支架置入治疗安全、有效,1年内预防再缺血效果优于单纯药物治疗。 展开更多

[Objective] The aim was to investigate the expression level of recombinant gene P1-2A3C of FMDV Asia I in different Bombyx mori varieties,so as to screen out the B.mori varieties suitable for the foreign gene expression.[Method] The recombinant B.mori baculovirus rBmNPV(P1-2A3C) are injected into pupae of original species and hybrids of B.mori respectively.Then,the expression of antigen is detected by ELISA method and compared.[Result] The expression level of P1-2A3C significantly differed in different B.mori varieties,in which,the hybrids of Qiufeng×TQ78 and Qiufeng×Sijiaojian could be considered as the effective bioreactors for high-level expression of foreign genes.[Conclusion] This study provided the basis for breeding special B.mori varieties which can highly express target protein of Asia I FMDV. 展开更多

在注浆加固工程中,为了避免空间变异性导致的支护结构受力过大而失稳,提出合理的注浆加固体刚度及强度预测方法十分必要.本文利用分形理论描述颗粒土类型的隧道围岩的孔隙特征,基于迂曲度经验公式建立了地层体孔隙率和面孔隙率的转换关... 在注浆加固工程中,为了避免空间变异性导致的支护结构受力过大而失稳,提出合理的注浆加固体刚度及强度预测方法十分必要.本文利用分形理论描述颗粒土类型的隧道围岩的孔隙特征,基于迂曲度经验公式建立了地层体孔隙率和面孔隙率的转换关系.以体孔隙率为变量,结合Mori-Tanaka方法,提出新的注浆加固体的宏观刚度预测方法.利用能量法求解连通孔隙不同屈曲半波分布形态下的临界载荷作为加固体的单轴抗压强度,从而建立了新的注浆加固体的强度预测理论模型.以玉京山隧道为工程算例,将本文方法在FLAC3D中进行二次开发,通过对不同的地层单元随机生成孔隙率,并利用本文方法计算并赋予地层单元不同的物理力学性质,实现了围岩空间变异性的模拟,数值模拟和现场监测得到的隧道洞身水平收敛的最大绝对误差仅为8 mm.数值模拟结果表明,对玉京山隧道回填区域进行注浆加固可以降低50%~90%的围岩位移,极大地降低围岩不良区域对应的支护结构弯矩,降低支护结构扭矩至0,并将支护结构变形控制在10 mm以内,但注浆加固难以改变灰岩和回填体之间的载荷分配规律. 展开更多

目的分析和探讨白内障超声乳化联合Mori Upright房角镜直视下分离房角治疗原发性闭角型青光眼的疗效。方方法在2019年1—12月于兰州大学第二医院接受治疗的原发性闭角型青光眼患者中,随机收集60例患者(60只眼)作为本研究的研究对象和主... 目的分析和探讨白内障超声乳化联合Mori Upright房角镜直视下分离房角治疗原发性闭角型青光眼的疗效。方方法在2019年1—12月于兰州大学第二医院接受治疗的原发性闭角型青光眼患者中,随机收集60例患者(60只眼)作为本研究的研究对象和主体,将其按照"动态随机分组法"分为对照组和观察组,每组30例(30只眼)。对照组采用超声乳化联合传统前房角分离术,观察组采用超声乳化联合Mori Upright房角镜直视下前房角分离术,对比2组患者术后的最佳矫正视力(BCVA)、眼压(IOP)、前房深度(ACD)、角膜内皮细胞密度(ECD)、术后并发症发生率。结果观察组BCVA、ACD、ECD高于对照组(P<0.05),IOP以及术后并发症发生率低于对照组(P<0.05)。结论超声乳化联合Mori Upright房角镜下分离房角治疗原发性闭角型青光眼的疗效显著,安全性高,适合在临床开展和普及。 展开更多

[Objective] The paper was to explore the regularity between heat shock protein expression and the healthiness changes of Bombyx moil materials. [Method] The representative heat shock protein gene Bmhsp24.3 was screened by bioinfor- matic analysis method, and carried out real-time PCR expression analysis. [Result] The target gene Bmhsp24.3 expressed in different B. mori materials, but the expres- sion level in different materials significantly varied. The relative expression level of the gene had different degrees of changes under different rearing conditions. With the increase of rearing temperature, the gene expression was upregulated. The ma- terials with better healthiness had remarkable increase in expression of target gene, while the materials with poorer healthiness had less increase in expression of target gene. The expression difference of target gene Bmhsp24.3 was exactly consistent with the healthiness of breeds. [Conclusion] The healthiness of materials had rela- tionship with expression of target gene Bmhsp24.3. the higher the expression of tar- get gene Bmhsp24.3 was, the better the healthiness of materials was; conversely, the lower the expression of target gene Bmhsp24.3 was, the poorer the healthiness of materials was. 展开更多

[Objective] This study aimed to develop a new Bombyx mori variety Shuhuang No.1 and investigate its characteristics.[Method] The new variety was developed through cross breeding and pedigree separation.Its characteristics were identified by laboratory tests and rural promotion practices in Sichuan Province.[Result]The cocoon produced by the new variety Shuhuang No.1 was golden-yellow and radiantly beautiful.Shuhuang No.1 was strong and easy to rear,uniform in development.The male and female ones could be distinguished by their larval color and markings.The cocoon yield per 10 000 the 4th instar larvae reached 20.30 kg; the length of a cocoon filament was about 1 115.0 m,and a non-broken filament was 882.95 mm long,accounting for 79.15% of the total length.The raw silk ratio of fresh cocoon was 18.39%,and the neatness was 91.25 point.The main economic features of Shuhuang No.1 were better than those of the control.[Conclusion] The new variety had been approved by Sichuan Silkworm Evaluation Commission.It can be promoted in parent silkworm rearing areas Sichuan,Chongqing,Yangtze River basin. 展开更多
