Type 1 diabetes mellitus(T1DM) lacks insulin secretion due to autoimmune deficiency of pancreaticβ-cells.Protecting pancreatic islets and enhancing insulin secretion has been therapeutic approaches.Mannogalactoglucan is the main type of polysaccharide from natural mushroom, which has potential medicinal prospects.Nevertheless, the antidiabetic property of mannogalactoglucan in T1DM has not been fully elucidated. In this study,we obtained the neutral fraction of alkali-soluble Armillaria mellea polysaccharide(AAMP-N) with the structure of mannogalactoglucan from the fruiting body of A.mellea and investigated the potential therapeutic value of AAMP-N in T1DM.We demonstrated that AAMP-N lowered blood glucose and improved diabetes symptoms in T1DM mice.AAMP-N activated unfolded protein response(UPR) signaling pathway to maintain ER protein folding homeostasis and promote insulin secretion in vivo.Besides that,AAMP-N promoted insulin synthesis via upregulating the expression of transcription factors, increased Ca^(2+) signals to stimulate intracellular insulin secretory vesicle transport via activating calcium/calmodulin-dependent kinase Ⅱ(CamkⅡ) and cAMP/PKA signals, and enhanced insulin secretory vesicle fusion with the plasma membrane via vesicle-associated membrane protein 2(VAMP2).Collectively,these studies demonstrated that the therapeutic potential of AAMP-N on pancreatic islets function, indicating that mannogalactoglucan could be natural nutraceutical used for the treatment of T1DM. 展开更多

The molecular mechanisms of energy status related to the umami taste of postharvest shiitake mushrooms during spore release remain poorly understood. In this study, the variations of energy status and umami taste of mushrooms were measured at 25℃.At 24 h storage, slight spore prints of mushrooms were first pictured, respiration peaked.Significant ATP decrease and ADP increase were also observed as the initiation of postharvest senescence(P<0.05).Meanwhile,the activities of phosphohexose isomerase, succinate dehydrogenase,glucose-6-phosphate dehydrogenase and cytochrome c oxidase and the contents of umami nucleotides and amino acids were maintained at higher levels in mushrooms with spore release.Notably, the equivalent umami concentration(EUC)was strongly correlated with energy levels(R=0.80).Fifteen related gene expression levels in the energy metabolism pathway were downregulated.LecpdP1 and LeAK were significantly expressed in the conversion of ATP into AMP and played key roles in connecting the energy state and umami level. These results provided valuable insights on the umami taste associated with energy metabolism mechanism during postharvest mushroom spore release. 展开更多

The isolated secondary metabolites from 39 edible mushrooms are reported, among which 107 compounds were active,61 demonstrated antitubercular activities with IC_(50) range of 0.2-50μg/mL and 46 manifested antimalarial effects with IC_(50) range of 0.061-36μg/mL.While more than 2000 strains of edible mushrooms are identified, this review shows the paucity of research in these rich organisms featuring a vital culinary ingredient worldwide.A thorough search was conducted on basidiomycetes to discuss the chemistry and biology of the isolated compounds, structure activity relationships(SAR)as well as the cytotoxicity profiles of,primarily, the active anti-plasmodial and antitubercular molecules. With a safe cellular profile, lanostane triterpenoids were found to be the only molecules with combined activities against both diseases.SAR correlations reviewed here indicated the significance of 3β-and 7α-hydroxylation in the anti-tuberculosis activity and the terminal unsaturated moiety between C-4 and C-28 in the antimalarial activity in the same terpene skeleton. This review will attract the attention of medicinal chemists, and food scientists to optimize and rationalize the use of mushrooms both as unexploited sources of novel molecules and as nutraceuticals to treat two of the deadliest infectious diseases, malaria, and tuberculosis. 展开更多

The application of molecular tools for the identification of strains, populations and species has become a common practice in a variety of applied and basic investigations in many parts of the world. However, standardization of such applications varies widely among organisms and scientific fields. In this mini-review,the author provides a brief introduction to one of the most prominent effort for species identification-the international barcode of life(iBOL) project, discusses the features of fungal diversity including the proposed fungal barcode DNA fragment-the intergenic spacer regions(ITS) of the nuclear ribosomal RNA gene clusters, and illustrates the potential promises and problems of using ITS for barcoding and for analyzing the phylogeographic pattern of the wild gourmet mushroom Tricholoma matsutake species complex. The analyses show that identification to species level is often insufficient for practical applications and that sequences from multiple genes in combination with critical morphological and physiological evaluations are needed to identify strains, populations and species. 展开更多

The mushrooms are highly regarded as one of the most nutritious foods across the globe but also recognized bio-accumulators of heavy metals. The nature and level of industrial activities are continually changing and affecting the environment adversely. The mushrooms are not an exception and may inevitably have heavy metal contaminations. In this vein, this study aimed to determine heavy metal (Cu, Ni, Co, Zn, Pb, and Cd) uptake levels in wild edible mushrooms from the sites with different economic activities. The wild mushrooms considered for this study included Tente (Amanita Zambiana), Ichikolowa (Termitomyces Titaniscus), and Kabansa (Lactarius Tataniscus). The analysis of heavy metal concentration was carried out using atomic absorption spectrometry (AAS). For the selected mushrooms, concentration ranges (mg·kg-1 total dry weight (dw)) of 46.90 - 141.80 for Cu, 0.10 - 6.60 for Cd, 1.10 - 2.00 for Pb, 19.00 - 38.90 for Zn, 1.00 - 3.40 for Ni, and 44.80 - 79.70 for Co were obtained. However, for the respective soil samples, concentration ranges (mg·kg-1·dw) of 51.00 - 279.40 for Cu, 1.00 - 99.50 for Cd, 8.00 - 10.00 for Pb, 22.80 - 209.10 for Zn, 9.00 - 33.70 for Ni, and 60.00 - 111.90 for Co were obtained. To a certain degree, the concentrations reflected the impact of diversity in the surrounding activities. This study discovered that for the selected mushrooms, the contamination level of cadmium, cobalt, nickel, and copper exceeded the World Health Organization (WHO)/FAO (Food and Agriculture Organization) recommended limits. Although some minor aberrations from the prescribed limits were also observed in the case of copper and nickel. Further, the data established that the heavy metal concentrations in respective soils are not the sole determinant of concentrations in mushrooms. Thus, these findings merit attention as, in some cases, the extent of contamination has exceeded the WHO permissible limit, and it may pose a health risk to consumers. 展开更多

Eleven different wild-edible mushroom species growing in the Black Sea region of Turkey were analysed for their metal content. Specimens of mushrooms were gathered in Trabzon, Giresun and Ordu and analyzed for 31 minerals, four of which (Be, Sb, Te and Ti) were not detected. Whereas some minerals including Ag, As, Cd, La, Mo, Pb, Se, Y and Zr were detected in just a few mushroom species, another 18 minerals were found in all 11 species. All metal concentrations were expressed on a dry weight basis (d.w.). With regard to nutritionally important amounts of essential and trace minerals, the metal content (μg/g d.w.) of mushroom samples ranged from 21,800-39,800 for K, 2590-14,000 for P, 268-1600 for Ca, 561-1210 for Mg, 74-829 for Fe, 11.2-321 for Cu, 36.2-241 for Zn, 14.1-76.5 for Mn and 0.13-2.85 for Co. Small amounts of toxic metals such as As, Cd and Pb were found in all 11 mushroom species. L. laccata contained a large amount of As (145 μg/g d.w.). These results show that the investigated mushrooms can be a useful component for human diets because of their high content of many essential minerals and trace elements and low content of toxic metals. 展开更多

However, their antioxidant and whether their values changing with drying processes are not known. This study establishes their values and the effect of sun drying on their quality and quantity. Methanolic extracts of five Cantharellus and two Afrocantharellus species were investigated. The investigation used (1,1-diphenyl-2-picrylhydrazyl radical) DPPH as a substrate to determine radical scavenging abilities. Further quantitative analyses for carotenoids, flavonoid, total phenolic compounds and vitamin C content were done using spectrophotometric assay at 515 nm. Comparatively, fresh mushroom had higher vitamin C and Lycopene contents than their dry counterparts with β-carotene. Flavonoid and phenolic compounds were higher in dry forms than those in their fresh counterparts. Radical scavenging ability also varied with fresh forms depicting higher activity compared to dry forms. The variation was low in Afrocantharellus species (0.8% and 0.99%) while it was higher in Cantharellus species (24.12% and 7.52%). The highest ability was observed in fresh forms of Cantharellus pseudocibarius (EC50 50 = 0.4 mg/ml) was from dry form of Cantharellus rufopunctatus. This shows that studied mushrooms are rich in antioxidants and their quality and quantities are affected differently in both forms. Both dry and fresh mushrooms are potential sources of natural antioxidants. The portrayed high scavenging ability by these mushrooms envisages promoting them into potential nutraceutics. 展开更多

In Guizhou Province, the area of rocky desertification land is 3.023 8 million hm^2, accounting for 17.16% of total area of land in the province. Rocky desertification is the most important ecological problem that restricts the economic and social development in Guizhou, so the primary task of ecological construction in Guizhou Province is to curb rocky desertification. How to effectively, efficiently and persistently use rocky desertification land is not only the key to the improvement of ecological environment, but also an urgent problem solved for local people's survival and anti-poverty. Due to having developed roots, growing fast, high yield of grass, strong resistance to drought, and high regeneration capacity, Pennisetum sinese Roxb has become a new method to control rocky desertification. How to use P. sinese Roxb efficiently becomes the key to the ecological control of rocky desertification by P. sinese Roxb. In this study, the ecological agriculture development model of "P. sinese Roxb-rocky desertification control-edible mushrooms" was studied, and the effect of cultivating Pleurotus eryngii with P. sinese Roxb in rocky desertification land was analyzed. Moreover, an outlook for the application of P. sinese Roxb in rocky desertification areas of Guizhou was given. 展开更多

Oyster mushrooms of genus Pleurotus are well known as valuable edible mushrooms which are broadly cultivated in the world as well as wildly propagated in forests of the temperate climate zones including Russia. Indoor cultivation conditions adapted for a laboratory with non-specialized facilities were suggested for effective solid-state cultivation and fruit bodies’ production of Pleurotus species. Seven of 9 Pleurotus species of different origin produced fruit bodies successfully. The best fructification was shown for species P. sajor-caju, P. ostreatus, and P. pulmonarius with fruit body yields of 74%, 72%, and 61% by dry substrate weight in the first flushing cycle. Fruit bodies appeared in five to 10-day flushes. “Spawn run” stage was completed within 17 to 24 days, the longest colonization stage (26 days) being for the pink oyster, P. djamor. The cultivation parameters proposed in this study can be employed with ease for laboratory and “home” cultivation of oyster mushrooms. The species and strains’ identification was confirmed by restriction analysis of ITS region of rRNA gene cluster. Molecular barcodes based on restriction enzyme (AluI, and BsuRI) profiles of ITS sequences were shown to be applicable for molecular genotyping of Pleurotus species of different origin. 展开更多

The ethnological knowledge of mushrooms despite its millennial existence and its empirical documentation are more recent phenomena. In Africa, the knowledge of their historical uses as food, medicine, source of income and small scale businesses, and the sociological impacts (myth, culture and spirituality) are apparently threatened due to slow ethnomycology research drive. The poor identification and documentation of edible and medicinal species of mushrooms in many developing nations have created some degrees of inconsistencies in their usages relative to folk medicine practice, food and mythological beliefs. Their relevance in modern day pharmaceutics and nutraceuticals is a product of human experimentation over time.Factors that may be anthropogenic, ethnographic, ethnoecological/environmental have been implicated in mushrooms underutilization and under-exploration of mushrooms in West Africa. Ethnomycological literatures on West Africa are scant, random, are limited in scope and fraught with taxonomic inconsistencies. This paper is based on extant ethnomycology treatise and aims at representing an integrative knowledge of useful mushrooms of West Africa and their uses vis-a-vis indigent cultures. 展开更多

This paper presents the status of mushroom cultivation in Malawi. This is a developing country located in southeastern Africa between latitudes 9°25' South and 17°08' South and longitudes 33° East and 36°East. Almost all the mushroom cultivators in the country are growing Pleurotus ostreatus. This species is most preferred because of its easiness to cultivate using the low-cost cultivation method being practiced in the country. On average, the annual P. ostreatus production is estimated at 240 kg per grower. Mushroom cultivators are selling their produce at prices ranging from MK800 (USD2.04) to MK2000 (USD5.10) per kg. At present, there are four institutions that are producing spawn namely Bunda College (Lilongwe University of Agriculture and Natural resources), Bvumbwe Agricultural Research Station, Natural Resources College and the Biology Department at Chancellor College, University of Malawi. Currently, a total of about 1307 bottles (330 ml each) of P. ostreatus spawn are sold by these four spawn producers per month. Mushroom cultivation is not that popular in Malawi. This may be, partly, attributed to lack of know-how and awareness on the economic, nutritive and medicinal benefits of cultivated mushrooms. Some of the major supermarkets do sell Agaricus bisporus mushrooms which are imported from the Republic of South Africa. They also sell Pleurotus ostreatus sourced from local cultivators. 展开更多
