Background The poultry industry needs effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE)caused by Clostridium perfringens.Methods The aim of this study was to investigate the effects of d...Background The poultry industry needs effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE)caused by Clostridium perfringens.Methods The aim of this study was to investigate the effects of dietary supplementation with Macleaya cordata extract(MCE)on the immune function and gut microbiota of broilers with NE.A total of 2881-day-old broiler chicks were randomly assigned to a 2×2 factorial arrangement with two concentrations of dietary MCE supplementation(0 or 350 mg/kg of diet)and two disease challenge statuses(control or NE).Results The results revealed that NE significantly increased the feed conversion rate(FCR),mortality,intestinal lesion score,the levels of IL-1β,IL-17 and IFN-γ/IL-4 in serum and IL-17/IL-10 in the jejunal mucosa,m RNA levels of TLR2,IFN-γand p Ig R in the jejunum,and Clostridium perfringens concentrations in the cecum.NE significantly decreased the body weight(BW),body weight gain(BWG),jejunal villus height,V/C,m RNA level of AMPK-α1 in jejunum,IL-4 level in the jejunal mucosa and lactic acid bacteria abundance in the cecum.MCE significantly increased BW,BWG,jejunal villus height,V/C,m RNA levels of occludin,ZO-1 and AMPK-α1 in the jejunum,the levels of Ig A and Ig G in serum and IL-10 in the jejunal mucosa and m RNA levels of NF-κB,IL-10 and MHC-II in the jejunum.Additionally,MCE significantly decreased the FCR,mortality,intestinal lesion score,jejunal crypt depth,the levels of IFN-γand IL-17 in serum and IL-17/IL-10 in the jejunal mucosa,Clostridium perfringens concentrations in the cecum,and m RNA levels of IL-17/IL-10 in the jejunum.Moreover,NE significantly increased the abundance of bacteria that are associated with inflammation,obesity and depression(Alistipes,Barnesiella,Intestinimonas,RF39 and UCG-005)and significantly decreased the abundance of short-chain fatty acid(SCFA)-producing bacteria(Anaerotruncus,Butyricicoccus and Bacteroides)in the cecum.MCE significantly increased the abundance of SCFA-producing bacteria(Streptococcus,Ruminococcus_torques_group and Lachnospiraceae_NK4A136_group)and significantly reduced the abundance of bacteria that are associated with inflammation and obesity(Alistipes,Barnesiella and UCG-010)in the cecum.In the cecum of broilers with NE,the relative abundance of Barnesiella and Alistipes was higher and that of Lachnoclostridium and Shuttleworthia was lower.Interestingly,these trends were reversed by the addition of MCE to the diet.Spearman correlation analysis showed that Barnesiella and Alistipes were associated with enhanced intestinal inflammation and inhibited growth performance,whereas Lachnoclostridium and Shuttleworthia were associated with anti-inflammatory effects.Conclusions MCE ameliorated the loss of growth performance in broiler chickens with NE,probably by regulating the intestinal barrier,immune function,and gut microbiota.展开更多
Background:This study aimed to investigate whether the combination of Macleaya cordata extract(MCE)and Bacil-lus could improve the laying performance and health of laying hens better.Methods:A total of 36029-week-old ...Background:This study aimed to investigate whether the combination of Macleaya cordata extract(MCE)and Bacil-lus could improve the laying performance and health of laying hens better.Methods:A total of 36029-week-old Jingbai laying hens were randomly divided into 4 treatments:control group(basal diet),MCE group(basal diet+MCE),Probiotics Bacillus Compound(PBC)group(basal diet+compound Bacil-lus),MCE+PBC group(basal diet+MCE+compound Bacillus).The feeding experiment lasted for 42 d.Results:The results showed that the laying rate and the average daily egg mass in the MCE+PBC group were significantly higher than those in the control group(P<0.05)and better than the MCE and PBC group.Combina-tion of MCE and Bacillus significantly increased the content of follicle-stimulating hormone(FSH)in the serum and up-regulated the expression of related hormone receptor gene(estrogen receptor-β,FSHR and luteinizing hormone/choriogonadotropin receptor)in the ovary of laying hens(P<0.05).In the MCE+PBC group,the mRNA expressions of zonula occluden-1,Occludin and mucin-2 in jejunum was increased and the intestinal epithelial barrier detected by transmission electron microscopy was enhanced compared with the control group(P<0.05).In addition,compared with the control group,combination of MCE and Bacillus significantly increased the total antioxidant capacity and catalase activity(P<0.05),and down-regulated the mRNA expressions of inflammation-related genes(interleukin-1βand tumor necrosis factor-α)as well as apoptosis-related genes(Caspase 3,Caspase 8 and P53)(P<0.05).The concen-tration of acetic acid and butyric acid in the cecum content of laying hens in the MCE+PBC group was significantly increased compared with the control group(P<0.05).Conclusions:Collectively,dietary supplementation of 600μg/kg MCE and 5×108 CFU/kg compound Bacillus can improve laying performance by improving microbiota to enhance antioxidant capacity and intestinal barrier,regulate reproductive hormones and the concentration of cecal short-chain fatty acids of laying hens,and the combined effect of MCE and Bacillus is better than that of single supplementation.展开更多

The optimization conditions of ...A reliable ultrasound-assisted extraction (UAE) method combined with HPLC-UV for quantification of eight active alkaloids in fruits of Macleaya cordata (Willd) R. Br. was developed. The optimization conditions of UAE were obtained by using Box-Behnken design of response surface methodology. Chromatography was carried out using a Kromasil C18 column by gradient elution with 0.1% phosphoric acid aqueous solution for HPLC-UV. All calibration curves showed good linear correlation coefficients (R^2〉0.999 6) and recoveries (from 97.3% to 104.9%) were acceptable. 1,1-diphenyl-2-picrylhydrazyl (DPPH) method was employed to test the antioxidant activity of the extract from the samples. The proposed method was successfully applied to quantifying eight components in nine samples of M.cordata, and significant variations of alkaloid contents and antioxidant aetivity of the samples from different habitats were demonstrated. It presents a powerful proof for the selection of the best sources to extract eight kinds of alkaloids.展开更多

The effects of the consumption of the appetizing ingredient M. cordata extract preparation (MCEP;trade name Sangrovit®) added to the feed at 50 and 100 ppm on the wellbeing and growth in the late gestation and lactation phases of sows and the resulting piglets prior to weaning was evaluated in preliminary and confirmatory feeding trials. Small increases in sow feed intake and piglet body weights were found in the preliminary trials. In the confirmatory field trial, low increases in sow feed intake with Sangrovit®resulted in decreased sow backfat loss and increased (P < 0.05) sow body condition scores. There was a positive effect on piglet growth, with significantly (P < 0.05) increased litter weight gain and litter weight at weaning. The results of these field trials show that the addition of Sangrovit®to sow feed during late gestation and lactation has positive effects on both the sows and the resulting litters.展开更多

[Method] A total of 120 1-day-old laying chicks with the body we...[Objective] The paper was to study the effects of Macleaya cordata extracts on growth performance and immune function of immunosuppressed laying chicks. [Method] A total of 120 1-day-old laying chicks with the body weight of (38.76±1.12) g were randomly divided into 3 groups with 40 replicates per treatment and 1 chick per replicate according to the principle of consistent body weight and half male and half female,namely control group, cyclophosphamide (CTX) group and M. cordata group. The trial lasted 21 d. The chicks in control group and CTX group were fed with basal diet, and those in M. cordata group were fed with the basal diet supplemented with 40 mg/kg M. cordata extract. On the 8^(th), 9^(th), 10^(th) and 17^(th) day of the trial, the chicks in CTX group and M. cordata group were intraperitoneally injected with 80 mg/kg BW CTX, and those in control group were intraperitoneally injected with equal dose of normal saline. [Result] (1) Compared with control group, the average daily gain (ADG) and average daily feed intake (ADFI) of chicks in CTX group reduced significantly;the feed gain ratio (F/G) increased significantly;the white blood cell count (WBC) decreased significantly;the alanine transaminase (ALT) and aspertate aminotransferase (AST) levels in the blood increased significant -ly;the total protein (TP) and globulin (GLOB) content in the blood decreased significantly;the serum malondialdehyde (MDA) content increased significantly;and the contents of immunoglobulin G (IgG), immunoglobulin M (IgM), interleukin-1 β (IL-1β) and interleukin-10 (IL-10) in the serum decreased significantly. (2)Compared with CTX group, the ADG and ADFI of chicks in M. cordata group increased significantly;the F/G decreased significantly;the WBC, red blood cell count (RBC), hemoglobin (HGB), hematokrit (HCT) and mean corpuscular hemoglobin (MCH) increased sig -nificantly;the contents of ALT, AST and MDA in the serum decreased significantly;the contents of TP, GLOB, IgG, Ig M, IL -1β, interleukin-6(IL-6) and IL-10 in the serum increased significantly;and the bursa of fabricius index increased significantly. (3) Compared with control group, the ADG and ADFI of chicks in M. cordata group increased significantly, and the contents of HGB and HCT in the blood increased significantly. [Conclusion] CTX injection will reduce the growth performance, antioxidant and immune function of laying chicks. A total of 128 weaned piglets were r...The aim of this study was to assess the effects of Sangrovit?, a standardized preparation of Macleaya cordata extract (MCE), on the health status and performance in weaned piglets. A total of 128 weaned piglets were randomly divided into four groups (n = 32, 16/sex/group) and fed either a control feed (T1) or the control feed supplemented with 100 mg/kg (T2), 500 mg/kg (T3), or 1000 mg/kg (T4) Sangrovit? (resulting in 0, 3.5, 17.5, 35 mg MCE/kg feed, respectively) for ninety days. The parameters for growth and health status were evaluated during the trial and blood was collected on Day 0 and Day 91 for hematology and biochemistry analysis. After the animals were sacrificed, the weight of the major organs and tissues was measured and histopathological examination on the organs from 8 animals in each group (4/sex/group) was performed. The results showed no statistically significant differences in live weight, feed intake, and average daily weight gain between the treatment and control groups. The feed containing 100, 500, or 1000 mg/kg Sangrovit? was well tolerated by piglets, with no adverse effects noted during the feeding period or in the histopathological results. The majority of the hematological and blood biochemistry parameters displayed no significant changes between the control and treatment groups. Compared to the control group, some hematological parameters including MCV, platelets concentration, and APTT were modified in some but not all treatment groups. Some significant changes in the blood biochemistry parameters were found in the treatment groups including levels in bile acid, haptoglobin, total serum protein, and GTT, as well as creatine kinase activity, but those changes were not toxicologically significant because no observed changes were seen in other similar biomarkers. In sum, this study demonstrated that the addition of Sangrovit? to swine feed at up to 1000 mg/kg does not result in any toxicological effects to the health status and growth of weaned piglets.展开更多

A total of tw...The aim of this study was to assess the effects of Sangrovit®, a standardized preparation of Macleaya cordata extract (MCE), on the health status and retained residues in growing-finishing swine. A total of twelve growing-finishing swine (n = 6 for each group) were randomly divided into two groups and fed either a control feed or the control feed supplemented with 100 mg/kg Sangrovit®(3.5 mg/kg MCE) for 28 days. The parameters for growth and health status were evaluated during the trial and after which the animals were slaughtered. Residual levels of MCE in swine organs and tissues were determined by measuring sanguinarine and chelerythrine levels by LC-MS/MS. The results showed no statistically significant differences in live weight, feed intake and average daily gain between the treatment and control groups. The feed supplemented with 100 mg/kg Sangrovit®was well tolerated by the swine, with no adverse effects noted during the feeding period or in the necropsy results. Residue analysis indicated that levels of sanguinarine or chelerythrine were under the limit of detection in all the examined tissues and organs from the treated swine. This study demonstrates that Sangrovit®, a standardized preparation of MCE when fed to growing-finishing swine for 28 days, at the level of 100 mg/kg in feed, does not result in sanguinarine or chelerythrine residues in the organs or tissues.展开更多

Layer chickens (N = ...The objective of this study was to assess the effects of Sangrovit?, a standardized preparation of Macleaya cordata extract (MCE), on the health status and egg laying parameters of layer chickens. Layer chickens (N = 360) were randomly divided into four groups (90 birds/treatment, 10 birds/pen) and fed either a standard basal feed (T1) or a basal diet that was supplemented with 100 mg/kg (T2), 500 mg/kg (T3) or 1000 mg/kg (T4) Sangrovit? (providing 0, 3.7, 18.5, and 37.0 mg MCE/kg feed, respectively) for 56 consecutive days. Live Weight (LW), Average Daily Feed Intake (ADFI) and the Feed Conversion Ratios (FCR) were calculated during the study, and biochemical and hematological endpoints were obtained at the end of the study (Day 56). Eggs were analyzed for the isoquinoline alkaloids sanguinarine and chelerythrine. No statistically significant (P > 0.05) differences were found between control and treatment groups for LW, ADFI and FCR. There was a significant increase in the % laying in groups T3 and T4 for the study overall, but no significant differences in egg size during the study. Blood biochemical analyses showed a near-significant trend for decreased bilirubin in the T2 and T4 groups, but this was not dose-dependent and not considered treatment-related. The percent hemoglobin was significantly decreased in the high dose group, but was not considered treatment-related as it was not a dose-dependent effect. No treatment-related changes were found after necropsy of the selected organs. No quantifiable sanguinarine or chelerythrine was found in the eggs after 56 days administration of the MCE preparation to the hens. The results of this study show that consumption of a standardized MCE preparation at up to 1000 mg/kg feed in laying hens had no adverse effect on the hen or eggs, and no residual sanguinarine or chelerythrine was transferred to the eggs.展开更多

Macleaya cordata extract(MCE)is a potential replacement for antibiotics.In the current study,effects of MCE on the gastrointestinal health and humoral responses of host animals were explored.A total of 30 weanling goa...Macleaya cordata extract(MCE)is a potential replacement for antibiotics.In the current study,effects of MCE on the gastrointestinal health and humoral responses of host animals were explored.A total of 30 weanling goats with similar body weight of 9.15±1.36 kg were randomly allocated into three groups(n=10 per group):control group(CON group,fed with a basal diet),antibiotic group(Abx group,fed with the basal diet supplemented with 0.18 g/d vancomycin and 0.36 g/d neomycin),and MCE group(fed with the basal diet supplemented with 5 g/d MCE),for three weeks.Results showed that antibiotic addition decreased the height and area of rumen papillae,ruminal mucosa Toll-like receptor 8(TLR8),interleukin-8(IL-8)and interleukin-1β(IL-1β)gene relative expression levels and microbial diversity,altered the volatile fatty acid(VFA)profile in the rumen,and increased monocytes amount and CD4+T cells percentage in the peripheral blood(P<0.05)compared to CON group.MCE addition increased the average daily gain,ileal villus height,villus height/crypt depth,and immunoglobulin M(IgM)content in the peripheral blood(P<0.05)compared to the CON.Additionally,MCE addition decreased the proportion of isobutyric acid in the chyme of the ileum(P=0.005)compared to the CON group.These results suggest that antibiotic supplementation may suppress the epithelial state and microbial diversity and fermentation in goats,but stimulate cellular response to maintain the growth performance of goats.MCE administration improved the epithelial state and humoral response to promote the growth performance in goats.展开更多

Macleaya cordata is a medicinal plant species from the Papaveraceae family, providing a safe res...The overuse of antibiotics in animal agriculture and medicine has caused a series of potential threats to public health. Macleaya cordata is a medicinal plant species from the Papaveraceae family, providing a safe resource for the manufacture of antimicrobial feed additive for livestock. The active constituents from M. cordata are known to include benzylisoquinoline alkaloids (BIAs) such as sanguinarine (SAN) and chelerythrine (CHE), but their metabolic pathways have yet to be studied in this non-model plant. The active biosynthesis of SAN and CHE in M. cordata was first examined and confirmed by feeding ^13C-labeled tyrosine. To gain further insights, we de novo sequenced the whole genome of M. cordata, the first to be sequenced from the Papaveraceae family. The M. cordata genome covering 378 Mb encodes 22,328 predicted protein-coding genes with 43.5% being transposable elements. As a member of basal eudicot, M. cordata genome lacks the paleohexaploidy event that occurred in almost all eudicots. From the genomics data, a complete set of 16 metabolic genes for SAN and CHE biosynthesis was retrieved, and 14 of their biochemical activities were validated. These genomics and metabolic data show the conserved BIA metabolic pathways in M. cordata and provide the knowledge foundation for future productions of SAN and CHE by crop improvement or microbial pathway reconstruction.展开更多

Macleaya cordata, a traditional Chinese medicine, is usually used as a natural additive in livestock because of its anti-microbial, anti-fungal, anti-inflammatory, and pesticidal activity. In this work, we studied whether M. cordata helps regulate the growth-promoting effect of CTC on broiler chickens. It is demonstrated that M. cordata improves the growth-promoting effect of CTC on growth per- formance indices of broiler chickens, such as survival rate, daily weight, and feed to weight rate. M. cordata also delays the maximum of CTC residues in plasma. It may depend on the higher values of operational taxonomic unit (OTU) and the indices of o diversity driven by simultaneous use of CTC and M. cordata.展开更多

ICR mice were randomly distributed into one of the fol...This study examines the effects of dietary Macleaya cordata extract(MCE) on bacterial burden and resistance to enterotoxigenic Escherichia coli(ETEC) in ICR mice. ICR mice were randomly distributed into one of the following groups:(i) basal diet;(ii)basal diet with 200 mg kg–1 MCE;(iii) basal diet challenged with ETEC;and(iv) basal diet with 200 mg kg–1 MCE and challenged with ETEC. Following a 7-day period of pre-treatment, CTRL-ETEC and MCE-ETEC mice were subjected to oral infection using 5×108 E. coli SEC 470. The results showed dietary 200 mg kg–1 MCE markedly reduced intestinal ETEC burden(P<0.05) and the disease-associated mortality was significantly alleviated in the MCE treated group(P<0.05). In addition,dietary MCE markedly alleviated ETEC-induced oxidative stress, evidenced by the lowered methane dicarboxylic aldehyde(MDA) abundance and enhanced activities of catalase and glutathione peroxidase(P<0.05). Furthermore, MCE mice exhibited higher immune activity, which might have further mediated ETEC infection. These results indicate MCE plays a preventative role with respect to ETEC infection. Future research should aim to develop MCE as a therapeutic approach to the promotion of intestinal health and a safeguard against ETEC infection.展开更多

文摘Background The poultry industry needs effective antibiotic alternatives to control outbreaks of necrotic enteritis(NE)caused by Clostridium perfringens.Methods The aim of this study was to investigate the effects of dietary supplementation with Macleaya cordata extract(MCE)on the immune function and gut microbiota of broilers with NE.A total of 2881-day-old broiler chicks were randomly assigned to a 2×2 factorial arrangement with two concentrations of dietary MCE supplementation(0 or 350 mg/kg of diet)and two disease challenge statuses(control or NE).Results The results revealed that NE significantly increased the feed conversion rate(FCR),mortality,intestinal lesion score,the levels of IL-1β,IL-17 and IFN-γ/IL-4 in serum and IL-17/IL-10 in the jejunal mucosa,m RNA levels of TLR2,IFN-γand p Ig R in the jejunum,and Clostridium perfringens concentrations in the cecum.NE significantly decreased the body weight(BW),body weight gain(BWG),jejunal villus height,V/C,m RNA level of AMPK-α1 in jejunum,IL-4 level in the jejunal mucosa and lactic acid bacteria abundance in the cecum.MCE significantly increased BW,BWG,jejunal villus height,V/C,m RNA levels of occludin,ZO-1 and AMPK-α1 in the jejunum,the levels of Ig A and Ig G in serum and IL-10 in the jejunal mucosa and m RNA levels of NF-κB,IL-10 and MHC-II in the jejunum.Additionally,MCE significantly decreased the FCR,mortality,intestinal lesion score,jejunal crypt depth,the levels of IFN-γand IL-17 in serum and IL-17/IL-10 in the jejunal mucosa,Clostridium perfringens concentrations in the cecum,and m RNA levels of IL-17/IL-10 in the jejunum.Moreover,NE significantly increased the abundance of bacteria that are associated with inflammation,obesity and depression(Alistipes,Barnesiella,Intestinimonas,RF39 and UCG-005)and significantly decreased the abundance of short-chain fatty acid(SCFA)-producing bacteria(Anaerotruncus,Butyricicoccus and Bacteroides)in the cecum.MCE significantly increased the abundance of SCFA-producing bacteria(Streptococcus,Ruminococcus_torques_group and Lachnospiraceae_NK4A136_group)and significantly reduced the abundance of bacteria that are associated with inflammation and obesity(Alistipes,Barnesiella and UCG-010)in the cecum.In the cecum of broilers with NE,the relative abundance of Barnesiella and Alistipes was higher and that of Lachnoclostridium and Shuttleworthia was lower.Interestingly,these trends were reversed by the addition of MCE to the diet.Spearman correlation analysis showed that Barnesiella and Alistipes were associated with enhanced intestinal inflammation and inhibited growth performance,whereas Lachnoclostridium and Shuttleworthia were associated with anti-inflammatory effects.Conclusions MCE ameliorated the loss of growth performance in broiler chickens with NE,probably by regulating the intestinal barrier,immune function,and gut microbiota.

文摘Background:This study aimed to investigate whether the combination of Macleaya cordata extract(MCE)and Bacil-lus could improve the laying performance and health of laying hens better.Methods:A total of 36029-week-old Jingbai laying hens were randomly divided into 4 treatments:control group(basal diet),MCE group(basal diet+MCE),Probiotics Bacillus Compound(PBC)group(basal diet+compound Bacil-lus),MCE+PBC group(basal diet+MCE+compound Bacillus).The feeding experiment lasted for 42 d.Results:The results showed that the laying rate and the average daily egg mass in the MCE+PBC group were significantly higher than those in the control group(P<0.05)and better than the MCE and PBC group.Combina-tion of MCE and Bacillus significantly increased the content of follicle-stimulating hormone(FSH)in the serum and up-regulated the expression of related hormone receptor gene(estrogen receptor-β,FSHR and luteinizing hormone/choriogonadotropin receptor)in the ovary of laying hens(P<0.05).In the MCE+PBC group,the mRNA expressions of zonula occluden-1,Occludin and mucin-2 in jejunum was increased and the intestinal epithelial barrier detected by transmission electron microscopy was enhanced compared with the control group(P<0.05).In addition,compared with the control group,combination of MCE and Bacillus significantly increased the total antioxidant capacity and catalase activity(P<0.05),and down-regulated the mRNA expressions of inflammation-related genes(interleukin-1βand tumor necrosis factor-α)as well as apoptosis-related genes(Caspase 3,Caspase 8 and P53)(P<0.05).The concen-tration of acetic acid and butyric acid in the cecum content of laying hens in the MCE+PBC group was significantly increased compared with the control group(P<0.05).Conclusions:Collectively,dietary supplementation of 600μg/kg MCE and 5×108 CFU/kg compound Bacillus can improve laying performance by improving microbiota to enhance antioxidant capacity and intestinal barrier,regulate reproductive hormones and the concentration of cecal short-chain fatty acids of laying hens,and the combined effect of MCE and Bacillus is better than that of single supplementation.

文摘A reliable ultrasound-assisted extraction (UAE) method combined with HPLC-UV for quantification of eight active alkaloids in fruits of Macleaya cordata (Willd) R. Br. was developed. The optimization conditions of UAE were obtained by using Box-Behnken design of response surface methodology. Chromatography was carried out using a Kromasil C18 column by gradient elution with 0.1% phosphoric acid aqueous solution for HPLC-UV. All calibration curves showed good linear correlation coefficients (R^2〉0.999 6) and recoveries (from 97.3% to 104.9%) were acceptable. 1,1-diphenyl-2-picrylhydrazyl (DPPH) method was employed to test the antioxidant activity of the extract from the samples. The proposed method was successfully applied to quantifying eight components in nine samples of M.cordata, and significant variations of alkaloid contents and antioxidant aetivity of the samples from different habitats were demonstrated. It presents a powerful proof for the selection of the best sources to extract eight kinds of alkaloids.
文摘Consistent feed intake during the stress of gestation and lactation in the sow may help maintain the wellbeing of the sow and resulting litter. The effects of the consumption of the appetizing ingredient M. cordata extract preparation (MCEP;trade name Sangrovit®) added to the feed at 50 and 100 ppm on the wellbeing and growth in the late gestation and lactation phases of sows and the resulting piglets prior to weaning was evaluated in preliminary and confirmatory feeding trials. Small increases in sow feed intake and piglet body weights were found in the preliminary trials. In the confirmatory field trial, low increases in sow feed intake with Sangrovit®resulted in decreased sow backfat loss and increased (P < 0.05) sow body condition scores. There was a positive effect on piglet growth, with significantly (P < 0.05) increased litter weight gain and litter weight at weaning. The results of these field trials show that the addition of Sangrovit®to sow feed during late gestation and lactation has positive effects on both the sows and the resulting litters.
文摘[Objective] The paper was to study the effects of Macleaya cordata extracts on growth performance and immune function of immunosuppressed laying chicks. [Method] A total of 120 1-day-old laying chicks with the body weight of (38.76±1.12) g were randomly divided into 3 groups with 40 replicates per treatment and 1 chick per replicate according to the principle of consistent body weight and half male and half female,namely control group, cyclophosphamide (CTX) group and M. cordata group. The trial lasted 21 d. The chicks in control group and CTX group were fed with basal diet, and those in M. cordata group were fed with the basal diet supplemented with 40 mg/kg M. cordata extract. On the 8^(th), 9^(th), 10^(th) and 17^(th) day of the trial, the chicks in CTX group and M. cordata group were intraperitoneally injected with 80 mg/kg BW CTX, and those in control group were intraperitoneally injected with equal dose of normal saline. [Result] (1) Compared with control group, the average daily gain (ADG) and average daily feed intake (ADFI) of chicks in CTX group reduced significantly;the feed gain ratio (F/G) increased significantly;the white blood cell count (WBC) decreased significantly;the alanine transaminase (ALT) and aspertate aminotransferase (AST) levels in the blood increased significant -ly;the total protein (TP) and globulin (GLOB) content in the blood decreased significantly;the serum malondialdehyde (MDA) content increased significantly;and the contents of immunoglobulin G (IgG), immunoglobulin M (IgM), interleukin-1 β (IL-1β) and interleukin-10 (IL-10) in the serum decreased significantly. (2)Compared with CTX group, the ADG and ADFI of chicks in M. cordata group increased significantly;the F/G decreased significantly;the WBC, red blood cell count (RBC), hemoglobin (HGB), hematokrit (HCT) and mean corpuscular hemoglobin (MCH) increased sig -nificantly;the contents of ALT, AST and MDA in the serum decreased significantly;the contents of TP, GLOB, IgG, Ig M, IL -1β, interleukin-6(IL-6) and IL-10 in the serum increased significantly;and the bursa of fabricius index increased significantly. (3) Compared with control group, the ADG and ADFI of chicks in M. cordata group increased significantly, and the contents of HGB and HCT in the blood increased significantly. [Conclusion] CTX injection will reduce the growth performance, antioxidant and immune function of laying chicks. The addition of M. cordata extract could restore the growth performance, immune function and antioxidant function impaired by CTX.
文摘The aim of this study was to assess the effects of Sangrovit?, a standardized preparation of Macleaya cordata extract (MCE), on the health status and performance in weaned piglets. A total of 128 weaned piglets were randomly divided into four groups (n = 32, 16/sex/group) and fed either a control feed (T1) or the control feed supplemented with 100 mg/kg (T2), 500 mg/kg (T3), or 1000 mg/kg (T4) Sangrovit? (resulting in 0, 3.5, 17.5, 35 mg MCE/kg feed, respectively) for ninety days. The parameters for growth and health status were evaluated during the trial and blood was collected on Day 0 and Day 91 for hematology and biochemistry analysis. After the animals were sacrificed, the weight of the major organs and tissues was measured and histopathological examination on the organs from 8 animals in each group (4/sex/group) was performed. The results showed no statistically significant differences in live weight, feed intake, and average daily weight gain between the treatment and control groups. The feed containing 100, 500, or 1000 mg/kg Sangrovit? was well tolerated by piglets, with no adverse effects noted during the feeding period or in the histopathological results. The majority of the hematological and blood biochemistry parameters displayed no significant changes between the control and treatment groups. Compared to the control group, some hematological parameters including MCV, platelets concentration, and APTT were modified in some but not all treatment groups. Some significant changes in the blood biochemistry parameters were found in the treatment groups including levels in bile acid, haptoglobin, total serum protein, and GTT, as well as creatine kinase activity, but those changes were not toxicologically significant because no observed changes were seen in other similar biomarkers. In sum, this study demonstrated that the addition of Sangrovit? to swine feed at up to 1000 mg/kg does not result in any toxicological effects to the health status and growth of weaned piglets.
文摘The aim of this study was to assess the effects of Sangrovit®, a standardized preparation of Macleaya cordata extract (MCE), on the health status and retained residues in growing-finishing swine. A total of twelve growing-finishing swine (n = 6 for each group) were randomly divided into two groups and fed either a control feed or the control feed supplemented with 100 mg/kg Sangrovit®(3.5 mg/kg MCE) for 28 days. The parameters for growth and health status were evaluated during the trial and after which the animals were slaughtered. Residual levels of MCE in swine organs and tissues were determined by measuring sanguinarine and chelerythrine levels by LC-MS/MS. The results showed no statistically significant differences in live weight, feed intake and average daily gain between the treatment and control groups. The feed supplemented with 100 mg/kg Sangrovit®was well tolerated by the swine, with no adverse effects noted during the feeding period or in the necropsy results. Residue analysis indicated that levels of sanguinarine or chelerythrine were under the limit of detection in all the examined tissues and organs from the treated swine. This study demonstrates that Sangrovit®, a standardized preparation of MCE when fed to growing-finishing swine for 28 days, at the level of 100 mg/kg in feed, does not result in sanguinarine or chelerythrine residues in the organs or tissues.
文摘The objective of this study was to assess the effects of Sangrovit?, a standardized preparation of Macleaya cordata extract (MCE), on the health status and egg laying parameters of layer chickens. Layer chickens (N = 360) were randomly divided into four groups (90 birds/treatment, 10 birds/pen) and fed either a standard basal feed (T1) or a basal diet that was supplemented with 100 mg/kg (T2), 500 mg/kg (T3) or 1000 mg/kg (T4) Sangrovit? (providing 0, 3.7, 18.5, and 37.0 mg MCE/kg feed, respectively) for 56 consecutive days. Live Weight (LW), Average Daily Feed Intake (ADFI) and the Feed Conversion Ratios (FCR) were calculated during the study, and biochemical and hematological endpoints were obtained at the end of the study (Day 56). Eggs were analyzed for the isoquinoline alkaloids sanguinarine and chelerythrine. No statistically significant (P > 0.05) differences were found between control and treatment groups for LW, ADFI and FCR. There was a significant increase in the % laying in groups T3 and T4 for the study overall, but no significant differences in egg size during the study. Blood biochemical analyses showed a near-significant trend for decreased bilirubin in the T2 and T4 groups, but this was not dose-dependent and not considered treatment-related. The percent hemoglobin was significantly decreased in the high dose group, but was not considered treatment-related as it was not a dose-dependent effect. No treatment-related changes were found after necropsy of the selected organs. No quantifiable sanguinarine or chelerythrine was found in the eggs after 56 days administration of the MCE preparation to the hens. The results of this study show that consumption of a standardized MCE preparation at up to 1000 mg/kg feed in laying hens had no adverse effect on the hen or eggs, and no residual sanguinarine or chelerythrine was transferred to the eggs.

文摘[Objectives]To explore the stability of sanguinarine and chelerythrine in Macleaya cordata(Willd.)R.Br.[Methods]The solubility and stability of sanguinarine and chelerythrine in seven solvents were measured by HPLC.Besides,the effects of water quality,light source,oxidant,temperature,and pH on stability were investigated.[Results]The solubility and stability of sanguinarine and chelerythrine in methanol and ethanol are good;the stability of sanguinarine and chelerythrine in distilled water and rainwater is not affected by light and is very stable,but they are unstable in tap water whether they are protected from light or not;oxidants have a great influence on the stability of sanguinarine and chelerythrine;sanguinarine and chelerythrine are stable at room temperature lower than 54℃;sanguinarine is stable in pH 2.5-7.0,while chelerythrine is stable in pH 2.5-8.0.[Conclusions]The good stability of sanguinarine and chelerythrine under specific conditions shows that they have broad development prospects and value.

文摘Macleaya cordata extract(MCE)is a potential replacement for antibiotics.In the current study,effects of MCE on the gastrointestinal health and humoral responses of host animals were explored.A total of 30 weanling goats with similar body weight of 9.15±1.36 kg were randomly allocated into three groups(n=10 per group):control group(CON group,fed with a basal diet),antibiotic group(Abx group,fed with the basal diet supplemented with 0.18 g/d vancomycin and 0.36 g/d neomycin),and MCE group(fed with the basal diet supplemented with 5 g/d MCE),for three weeks.Results showed that antibiotic addition decreased the height and area of rumen papillae,ruminal mucosa Toll-like receptor 8(TLR8),interleukin-8(IL-8)and interleukin-1β(IL-1β)gene relative expression levels and microbial diversity,altered the volatile fatty acid(VFA)profile in the rumen,and increased monocytes amount and CD4+T cells percentage in the peripheral blood(P<0.05)compared to CON group.MCE addition increased the average daily gain,ileal villus height,villus height/crypt depth,and immunoglobulin M(IgM)content in the peripheral blood(P<0.05)compared to the CON.Additionally,MCE addition decreased the proportion of isobutyric acid in the chyme of the ileum(P=0.005)compared to the CON group.These results suggest that antibiotic supplementation may suppress the epithelial state and microbial diversity and fermentation in goats,but stimulate cellular response to maintain the growth performance of goats.MCE administration improved the epithelial state and humoral response to promote the growth performance in goats.

文摘The overuse of antibiotics in animal agriculture and medicine has caused a series of potential threats to public health. Macleaya cordata is a medicinal plant species from the Papaveraceae family, providing a safe resource for the manufacture of antimicrobial feed additive for livestock. The active constituents from M. cordata are known to include benzylisoquinoline alkaloids (BIAs) such as sanguinarine (SAN) and chelerythrine (CHE), but their metabolic pathways have yet to be studied in this non-model plant. The active biosynthesis of SAN and CHE in M. cordata was first examined and confirmed by feeding ^13C-labeled tyrosine. To gain further insights, we de novo sequenced the whole genome of M. cordata, the first to be sequenced from the Papaveraceae family. The M. cordata genome covering 378 Mb encodes 22,328 predicted protein-coding genes with 43.5% being transposable elements. As a member of basal eudicot, M. cordata genome lacks the paleohexaploidy event that occurred in almost all eudicots. From the genomics data, a complete set of 16 metabolic genes for SAN and CHE biosynthesis was retrieved, and 14 of their biochemical activities were validated. These genomics and metabolic data show the conserved BIA metabolic pathways in M. cordata and provide the knowledge foundation for future productions of SAN and CHE by crop improvement or microbial pathway reconstruction.

文摘Chlortetracycline (CTC), one kind of common antibiotic for prevention and treatment of various diseases, also exhibits good performance in accelerating the growth of livestock. Macleaya cordata, a traditional Chinese medicine, is usually used as a natural additive in livestock because of its anti-microbial, anti-fungal, anti-inflammatory, and pesticidal activity. In this work, we studied whether M. cordata helps regulate the growth-promoting effect of CTC on broiler chickens. It is demonstrated that M. cordata improves the growth-promoting effect of CTC on growth per- formance indices of broiler chickens, such as survival rate, daily weight, and feed to weight rate. M. cordata also delays the maximum of CTC residues in plasma. It may depend on the higher values of operational taxonomic unit (OTU) and the indices of o diversity driven by simultaneous use of CTC and M. cordata.

文摘This study aimed to investigate the ability of chelerythrine chloride(CHE),the main active ingredient of Macleaya cordata,to induce apoptosis in human gastric cancer BGC-823 cells.The results demonstrate that CHE inhibits cell proliferation in a time-and dose-dependent manner with accompanying S phase arrest.It also induces apoptosis by a mechanism involving a reduction in the mitochondrial mem brane potential,the release of cytochrome c,activation of caspase 3 and cleavage of poly-ADP-ribose polymerase.In addition,CHE-induced apoptosis is accompanied by down-regulation of Bcl-x1 and Bcl-2 proteins with no change in the levels of Bax proteins.Taken together,the results support the development of CHE as a potentially useful anticancer drug for the treatment of gastric cancer.

文摘This study examines the effects of dietary Macleaya cordata extract(MCE) on bacterial burden and resistance to enterotoxigenic Escherichia coli(ETEC) in ICR mice. ICR mice were randomly distributed into one of the following groups:(i) basal diet;(ii)basal diet with 200 mg kg–1 MCE;(iii) basal diet challenged with ETEC;and(iv) basal diet with 200 mg kg–1 MCE and challenged with ETEC. Following a 7-day period of pre-treatment, CTRL-ETEC and MCE-ETEC mice were subjected to oral infection using 5×108 E. coli SEC 470. The results showed dietary 200 mg kg–1 MCE markedly reduced intestinal ETEC burden(P<0.05) and the disease-associated mortality was significantly alleviated in the MCE treated group(P<0.05). In addition,dietary MCE markedly alleviated ETEC-induced oxidative stress, evidenced by the lowered methane dicarboxylic aldehyde(MDA) abundance and enhanced activities of catalase and glutathione peroxidase(P<0.05). Furthermore, MCE mice exhibited higher immune activity, which might have further mediated ETEC infection. These results indicate MCE plays a preventative role with respect to ETEC infection. Future research should aim to develop MCE as a therapeutic approach to the promotion of intestinal health and a safeguard against ETEC infection.
