OBJECTIVE: To observe the relationship betwe...BACKGROUND: Glutathione, as an in vivo free radical scavenger, plays an important role in the anti-oxidation defense mechanism in patients with acute cerebral infarction. OBJECTIVE: To observe the relationship between the levels of glutathione (GSH) and malonaldehyde (MDA), the metabolite of lipid peroxidation, in erythrocyte and deformation index of erythrocyte in patients at various periods following acute cerebral infarction. DESIGN: Controlled observation.SETTING: Qilu Hospital of Shandong University and Institute of Cerebrovascular Disease of Qingdao Medical College. PARTICIPANTS: We chose 98 inpatients with acute cerebral infarction from Department of Neurology, Qilu Hospital of Shandong University from January to December 2000 , serving as cerebral infarction group, including 50 male and 48 female, with mean age of (62±7)years. There were 23 cases found on the 1^st day after onset; 25 cases on the 3^rd day after onset; 25 cases on the 7^th day after onset; 25 cases on the 14th days after onset, and they were all confirmed by craniocerebral CT or MRI. Another 30 homeochronous inpatients with neurosis, cervical syndrome, lumbar intervertebral disc herniation and motor neuron disease were chosen as control group, including 20 male and 10 female, with mean age of (52±8)years . There was no significant difference in age and gender distribution between two groups (P 〉 0.05). Patients in the two groups were informed of detected index.METHODS:①Ulnar venous blood was chosen from the patients who were fasted on the 1^st, 3^rd,7^th and 14^th days after onset. Deformation index of erythrocyte was measured with BL88-CKX laser diffraction erythrocyte deformeter and photographing was performed. GSH level in erythrocyte was measured with DTNB assay introduced by Beu-tler. MDA level in erythrocyte was measured with modified thiobarbituric acid colorimetric method.②At each sample collecting , according to the criteria accepted by the Fourth National Conference of Cerebrovascular Disease, intergrant of neurologic impairment 〈 15 points was regarded as mild (n=46), 15 to 30 points as moderate (n=40)and, 〉 30 points as severe (n=12). ③ t test was used to compare data between two groups , and linear correlation analysis was used in relationship analysis among indexes. MAIN OUTCOME MEASURES :① Comparison of erythrocyte GSH and MDA levels and deformation index of erythrocyte at various periods between patients with acute cerebral infarction and controls. ②Correlation of erythrocyte GSH level with erythrocyte MDA level and with deformation index of erythrocyte in patients with cerebral infarction . ③ Relationship between erythrocyte GSH level and severity of disease in patients with acute cerebral infarction. RESULTS: Totally 98 patients with acute cerebral infarction and 30 controls all entered the stage of result analysis. ① Erythrocyte GSH level and deformation index of erythrocyte were lower on the 1^st, 3^rd, 7^th and 14^th days after onset in cerebral infarction group than in control group (P 〈 0.05-0.01), and erythrocyte MDA level was significantly higher in cerebral infarction group than in control group (P 〈 0.05-0.01). The three indexes changed most significantly on day 3 after onset in patients, and began to recover or decrease on day 7 after onset and inclined to be normal on day 14 after onset. ② Erythrocyte GSH level was significantly negatively correlated with erythrocyte MDA level in patients with acute cerebral infarction on the 1^st, 3^rd, 7^th and 14^th days after onset (r=-0.534, -0.713, -0.645, -0.656, respectively, P 〈 0.05-0.01 ) ,and significantly positively correlated with erythrocyte deformation index (t-=0.502, 0.560, 0.455, 0.504, respectively, P 〈 0.05). ③Erythrocyte GSH level was significantly lower in moderate or severe patients with acute cerebral infarction than in mild patients[(0.215±0.088),(0.192±0.102), (0.281±0.090) g/L, P〈 0.05]. CONCLUSION:①Erythrocyte GSH and MDA levels and deformation index of erythrocyte change significantly on the 3^rd day following acute cerebral infarction. Index detection results gradually tended to be normal on from the 7^th day to 14^th days day after onset. ② Change of erythrocyte GSH level can reflect the severity of disease of patients with acute cerebral infarction.③Decrease of erythrocyte GSH level in patients with acute cerebral infarction is one of reasons that result in the decrease of deformation ability of erythrocyte.展开更多

[Method] The high fat and cholesterol diet were adopted to create the hyperlipemia model of rabbit. The rabbits were randomly and equally divided into 5 groups which were NC( normal control)group, HLM (high lipid model) group, LFPE, MFPE and HFPE group (low, middle, high dosage of FPE were 0.17 g/kg, 0.5 g/kg and 1.5 g/kg respectively). The contents of TC, TG, LDL-C, HDL-C, MDA and the activity of SOD in serum were detected respectively. [ Result] The three FPE groups could decrease the contents of TC, TG, LDL-C, MDA significantly (P〈0.01) compared with these of HLM group, and the contents of HDL-C (P〈0.01) and the activity of SOD (P〈0.05 ) in serum was increased significantly. METHODS: The SAP rat models were randomly divided into the model control gr...AIM: To observe the therapeutic efficacy of Baicalin in rats with severe acute pancreatitis (SAP) and explore its therapeutic mechanisms. METHODS: The SAP rat models were randomly divided into the model control group, Baicalin treatment group, octreotide treatment group and sham operation group. All groups were randomly subdivided into 3 h, 6 h and 12 h groups with 15 rats in each group. The survival, ascites volume and pathological changes of pancreas in all rats were observed at different time points after operation. The plasma amylase content and serum TNF-α, IL-6, malonaldehyde (MDA) and PLA2 contents were also determined. RESULTS: The survival was not obviously different between the treated groups, and was significantly higher in treated groups at 12 h compared to the model control group (P 〈 0.05, 15 vs 10). The ascites/body weight ratio at 3 h and 6 h was significantly lower in Baicalin treatment group compared to the model control group and octreotide treatment group (P 〈 0.05, 1.00 vs 2.02 and 1.43 and P 〈 0.001, 2.29 (1.21) vs 2.70 (0.80) and 2.08 (2.21), respectively). The contents of amylase, TNF-α, IL-6, MDA and PLA2 were significantly lower in the treated groups than in the model control group (P 〈 0.05, 4342 vs 5303, 5058 vs 6272 in amylase, P 〈 0.01, 21.90 vs 36.30, 23.80 vs 39.70, 36 vs 54.35 in MDA and 56.25 vs 76.10 in PIA2, or P 〈 0.001, 65.10 and 47.60 vs 92.15 in TNF-α, 3.03 vs 5.44, 2.88 vs 6.82, 2.83 vs 5.36 in IL-6, respectively). The pathological scores of pancreas in the treated groups were significantly lower than that in the model control group (P 〈 0.05, 9.00 vs 10.05, 6.00 vs 9.00, 8.00 vs 10.05), but no marked difference was found between the treated groups. CONCLUSION: The Baicalin injection has significant therapeutic effects on SAP rats, its effects are similar to those of octreotide. The Baicalin injection is also cheap and has a big application range, quite hopefully to be used in clinical treatment of SAP.展开更多

However, changes of homocysteine (Hey) as markers of cardiovascular and eerebrovascular disease associated with OSAHS and their mechanism have not been elucidated so far. This study aims to investigate the changes of both serum Hcy and oxidative stress and their possible links with OSAHS in elderly patients. Methods Based on polysomnogram (PSG) and age, 83 patients with OSAHS were recruited and divided into elderly-OSAHS (n=32) and non-elderly OSAHS groups (n=51). Fifty two subjects without OSAHS were divided into elderly control (n=29) and non-elderly control groups (n-23). A total of 135 subjects were included in the present study. All subjects were recorded for PSG variables and the contents of homocysteine (Hcy), malonaldehyde (MDA), and glutathione (GSH) which were detected after sleep. Serum homocysteine was measured by cyclophorase. MDA and GSH were measured by speetrophotometer. Results (1) The serum levels of Hcy showed significant difference among the four groups (P〈0.05). The concentrations of Hcy in elderly OSAHS patients were higher than in other groups, while those in the elderly control group were higher than in the non-elderly control; the concentrations in the non-elderly OSAHS group were higher than in the non-elderly control. (2) The concentrations of MDA and GSH changed at an equal pace with Hcy in the four groups. (3) Multielement linearity regression analysis indicated a statistically significant relationship between Hcy concentration and age, MDA, GSH, and apnea hypopnea index (AHI). Conclusions (1) The concentrations of Hcy and oxidative stress have increased with advancing age. (2) The concentrations of Hcy and oxidative stress have further increased in the elderly patients with OSAHS. (3) Oxidative stress might cause high-level serum Hcy in the elderly patients with OSAHS.展开更多

Whether this process is relat...Our previous studies revealed that etomidate, a non-barbiturate intravenous anesthetic agent, has protective effects on retinal ganglion cells within 7 days after optic nerve transection. Whether this process is related to anti-oxidative stress is not clear. To reveal its mechanism, we established the optic nerve transection injury model by transecting 1 mm behind the left eyeball of adult male Sprague-Dawley rats. The rats received an intraperitoneal injection of etomidate(4 mg/kg) once per day for 7 days. The results showed that etomidate significantly enhanced the number of retinal ganglion cells retrogradely labeled with Fluorogold at 7 days after optic nerve transection. Etomidate also significantly reduced the levels of nitric oxide and malonaldehyde in the retina and increased the level of glutathione at 12 hours after optic nerve transection. Thus, etomidate can protect retinal ganglion cells after optic nerve transection in adult rats by activating an anti-oxidative stress response. Huanshaodan, a Chinese traditional preparation for kidney-replenishing essence, was used to be the preparation for reinforcing renal deficiency and preventing aging for aged people. OBJECTIVE: To observe the effects of Huanshaodan on swimming durance and the abilities of catalase (CAT) in serum and monoamine oxidase-B (MAO-B) in brain tissue as well as in vitro anti-oxidative ability of aging mouse. DESIGN: A controlled animal experiment. SETTING: College of Basic Medicine, Hunan University of Traditional Chinese Medicine. MATERIALS: Fifty-four healthy NIH mice, aged 18 months old, of either gender, weighing (48.9±5.4) g, and one SD male rat, aged 16 months old, weighing 51.7 g, were provided by Animal Experimental Center, Hunan University of Traditional Chinese Medicine. Thirty NIH mice were randomly chosen for swimming test, and divided into experimental group and control group, with 15 in each; The other 24 NIH mice were used for enzyme activity assay, and also divided into experimental group and control group, with 12 in each. SD rat was used for in vitro anti-oxidative ability test, Huanshaodan water decoction was composed of Cheqianzi, Wuweizi, Huaishan, Danggui, Huangbai, Shudi, Baizhi, Niuxi, Baishen, Tusizi, Buguzhi, Roucongrong and Heshouwu 13 Chinese herbs. METHODS: This study was carried out in the Second Laboratory, Department of Biochemistry, Hunan University of Traditional Chinese Medicine in June 2006. Swimming and enzyme activity assay: Mice in the two experimental groups were intragastrically administrated with l0 μL/g Huanshaodan water decoction. Mice in the two control groups were intragastrically administrated with the same amount of normal saline. All the mice were intragastrically administrated for 5 days, and they were free to access to medicine in the other 2 days in a week. Each mouse was administrated for 8 weeks. MAIN OUTCOME MEASURES: ① Forty days after administration, mice in the experimental group and control group for swimming test were loaded at tails and allowed to swim in the water-tank. Swimming durance was recorded. ② Following the method of Chen Qi, the activities of CAT in serum and MAO-B in brain tissue as well as the inhibitory rate of each medicine on malonaldehyde (MDA) content in the in vitro rat hepatic tissue were determined; Meanwhile, the inhibitory rate of different doses of Tusizi liquid to MDA content in the rat hepatic tissue in vitro was also assayed. RESULTS: Fifty-four NIH mice and one SD rat were recruited in this experiment. Three mice died in the swimming test, and all the other animals were involved in the final analysis. ① Swimming durance of mice in the experimental group was significantly longer than that in the control group ( t =7.502, P 〈 0.01 ) . The activity of CAT in serum of mice in the experimental group was significantly higher than that in the control group ( t =13.307, P 〈 0.01 ) . ② The activity of MAO-B in brain tissue of mice in the experimental group was significantly lower than that in the control group (t =l3.27, P 〈 0.01 ) . ③The inhibitory rate of Cheqianzi, Wuweizi, Huaishan, Danggui, Huangbai, Shudi, Baizhi, Niuxi, Baishen, Tusizi, Buguzhi, Roucongrong and Heshouwu 13 Chinese herbs of Huanshaodan to MDA in the rat hepatic tissue in vitro was - 62.9, - 95.1, - 34.9, - 65.1, - 99.1, - 87.2, - 94.1, - 20.0, - 67.0, - 83.7, - 91.0, - 98.4, - 93.0, respectively. ④ The inhibitory rate of low to high dose of Tusizi liquid to MDA content in the rat hepatic tissue in vitro was - 3.41, - 18.1, - 26.6, - 83.7, respectively. CONCLUSION: Huanshaodan enhances swimming endurance, anti-oxygen free radical and anti-oxidativeabilities, and thus, it can delay aging.展开更多
BACKGROUND: This study was undertaken to observe the concentration of SP-A/B and the pulmonary surfactant in the lung tissue of rats with acute lung injury/acute respiratory distress syndrome caused by paraquat poison...BACKGROUND: This study was undertaken to observe the concentration of SP-A/B and the pulmonary surfactant in the lung tissue of rats with acute lung injury/acute respiratory distress syndrome caused by paraquat poisoning after the treatment of metabolic antioxidant-lipoic acid and whether its influence was related to TNF-α.METHODS: Sixty-six male Sprage-Dawley rats were randomly divided into three groups: normal control group(NS group), 6 rats; paraquat poisoning group(PQ group), 30 rats; and paraquat+lipoic acid treatment group(LA group), 30 rats. The rats in the PQ and LA groups were subdivided into 3-, 6-, 12-, 24-, 48-hour subgroups, with 6 rats in each group. After the rats were sacrificed, lung tissue from the same part was taken from the rats. After HE staining, histological changes were observed in the tissue under a light microscope. Lung tissue was also taken to test the levels of superoxide dismutase(SOD) and malondialdehyde(MDA). Whole blood(0.8 mL) without anticoagulant was drawn from the tail vein of rats for the determination of the TNF-α level. The total RNA of the lung tissue was collected, and the Rt-PCR method was used to measure the levels of SP-A and SP-B mRNA.RESULTS: HE staining showed that histopathological changes were milder in the LA group than in the PQ group. There were significant differences in MDA and SOD levels between different intervals both in intergroups and intragroups except the 3-hour subgroup(P<0.01). Likewise, the significant differences in the levels of TNF-α were also present between the three groups and between different intervals(P<0.01). The significant differences in SP-A mRNA and SP-B mRNA amplification ratio were seen between the three groups at the same intervals(P<0.01), but the differences between different intervals in the PQ group were statistically significant(P<0.05). The differences between different intervals in the LA group were statistically significant(P<0.01).CONCLUSION: Lipoic acid in acute paraquat poisoning could diminish lung tissue damage by regulating directly tumor necrosis factor and indirectly the content of pulmonary surfactant so as to reduce pulmonary edema, improve lung compliance, and finally protect lung tissues.展开更多

However, little has been documented on its effects on zooplankton grazing. In this study, the authors investigated the effects of photosynthetically active radiation (PAR, 400–700 nm), ultraviolet-A (UV-A, 320–400 nm) and ultraviolet-B (UV-B, 280–320 nm) radiation on grazing, mortality and lipids oxidation of the copepod Acartia pacifica collected from the Xiamen Bay. After 30 min of the exposures, the copepod was fed in darkness with the diatom Phaeodactylum tricornutum at two cell concentrations (2.5 × 10 4 and 2.5 × 10 5 cells/ml). At the low cell concentration, the individuals pre-exposed to PAR (218.0 W/m 2 )+UV-A (48.2 W/m 2 ) or PAR+UV-A+UV-B (2.1 W/m 2 ) showed suppressed clearance and grazing activities compared with those receiving PAR alone, by 22.7% and 17.1% for clearance and by 22.6% and 5.5% for grazing rates, respectively. However, the suppression on clearance and grazing became indistinctive at the high food concentration. Exposures to UV-A and UV-B led to increased lipid oxidation and higher mortality, furthermore, the mortality linearly increased with enhanced oxidation of lipid.展开更多
Based on previous studies that have shown flavonoids from the stems and leaves of Scutellaria baicalensis Georgi are neuroprotective agents in a naturally senile, D-galactose, aging in vivo model, as well as an in vit...Based on previous studies that have shown flavonoids from the stems and leaves of Scutellaria baicalensis Georgi are neuroprotective agents in a naturally senile, D-galactose, aging in vivo model, as well as an in vitro model of oxidative/hypoxic injury, we established a cerebral ischemia/reperfusion model in rats by middle cerebral artery occlusion. The light/electron microscopic observations found significant neuropathological changes including neuron loss or swelling and rough endoplasmic reticulum injury. Moreover, the activities of lactate dehydrogenase Na+-K+-ATPase, Ca2+-ATPase and superoxide dismutase were significantly lowered, and the levels of malonaldehyde increased. In addition, the memory of rats worsened. However, treatment with flavonoids from Scutellaria baicalensis Georgi (35, 70 and 140 mg/kg) for 13 days dramatically improved the above abnormal changes. These results suggest that the ability of flavonoids from Scutellaria baicalensis Georgi in attenuating cerebral functional and morphological consequences after cerebral ischemia/reperfusion may be beneficial for the treatment of ischemic brain disease.展开更多

This is manifested by hypomnesia and disordered metabolism of free radicals. Studies have reported that Longyanshen polysaccharides have the function of antioxidation and improved brain memory. OBJECTIVE: To observe the effects of Longyanshen polysaccharides on free radical metabolism in brain tissue to verify the anti-aging mechanisms in senescence accelerated-prone (SAMP8) mice. DESIGN, TIME AND SETTING: The randomized, controlled, biochemical experiment was performed in the Department of Pharmacology and Scientific Experimental Center of Guangxi Medical University (China) from September 2005 to January 2008. MATERIALS: Forty SAMP8 mice were randomized into four groups: SAMP8 control group, as well as low-, mid-, and high-dose polysaccharide, with 10 mice in each group. Ten senescence accelerated-resistantprone (SAMR 1) mice served as the normal control group. Longyanshen polysaccharides, extracted from the medical plant Longyanshen, were supplied by the Department of Pharmacology, Guangxi Medical University Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malonaldehyde (MDA), nitric oxide (NO), and total protein test kitwere purchased from Nanjing Jiancheng Bioengineering Institute (China). METHODS: SAMP8 mice were used to establish a dementia animal model. SAMP8 and SAMR1 control mice were administered 30 mL/kg saline. The low-, middle-, and high-dose polysaccharide groups were administered 45, 90, and 180 mg/kg Longyanshen polysaccharides, respectively. Each group was treated by intragastric administration, once daily, for 50 continuous days. MAIN OUTCOME MEASURES: One hour after the last administration, mouse brain tissues were collected, and retro orbital blood sampling was performed. Spectrophotometry was used to measure SOD and GSH-Px activity, as well as MDA and NO concentration in sera and brains of SAMP8 mice. RESULTS: SOD and GSH-Px activity decreased significantly, and MDA and NO concentration increased significantly, in SAMP8 control group brain tissues, compared with the SAMP1 control group (P 〈 0.05). Compared with the SAMP8 control group, Longyanshen polysaccharide-treated groups exhibited enhanced SOD and GSH-Px activity, as well as decreased MDA and NO concentration, in serum and brain tissue (P 〈 0.05). Longyanshen polysaccharides exerted a similar effect on SOD, GSH-Px, MDA, and NO concentrations in serum and brain tissues of SAMP8 mice. CONCLUSION: Longyanshen polysaccharides scavenged free radicals effectively, reduced NO concentration and ameliorated NO toxicity, thereby influenced aging and stress, as well as improving memory capacity in the brain.展开更多

Malonaldehyde(MDA) is determined as an index for lipid peroxidation both in vitro...The study aims to investigate the effects of(-)-Linarinic acid(LA) and one of its derivatives(LAd) on brain injury induced by ischemia. Malonaldehyde(MDA) is determined as an index for lipid peroxidation both in vitro and vivo. Mice were pre-treated with LA and LAd for 3 d.Thereafter, they were induced to have incomplete cerebral ischemia with both bilateral carotid artery occlusion and hypotension(BCAOH). In the first part of the in vivo experiment, mice were divided into four groups: sham(control), ischemia, ischemia + LA(200 mg/kg, i.g.) and ischemia + LAd(200 mg/kg, i.g.). In the second part, the dose-response of LAd was investigated at 100, 200 and 400 mg/kg i.g., respectively. A modified neurological severity score was developed for evaluating behavioral deficits of the mice with ischemia. Brains of the mice were excised in order to determinate MDA after ischemia for 6 h. Survival time, survival rate, neurological injury score and MDA level in brains were observed. Results were: 1) The data in vitro showed that both LA and LAd could inhibit the generation of MDA. IC50 values obtained by Probit analysis were 2.9 mM for LAd and 4.88 mM for LA;2) BCAOH could significantly shorten the survival span, reduce the survival rate and cause neurological deficits,which were associated with high level of lipid hydroperoxide production in cerebral tissues;3) LAd decreased lipid peroxidation and improved the neurological outcome more than LA.It is concluded that LAd offers a better neuroprotection than LA against brain damage caused by cerebral ischemia.展开更多
Cerium oxide nanoparticles(CeO_(2) NPs)are likely to have dramatic impacts on plant performances,yet the effects of CeO_(2) NPs on seed germination and seedling growth have not been fully explored.In this study,the se...Cerium oxide nanoparticles(CeO_(2) NPs)are likely to have dramatic impacts on plant performances,yet the effects of CeO_(2) NPs on seed germination and seedling growth have not been fully explored.In this study,the seed germination and seedling growth of subshrub species Vitex negundo under different concentrations of CeO_(2) NPs(low-1 mg/L,medium-100 mg/L,high-500 mg/L)have been discussed.Results showed that:(1)The seed germination rate reduces by 11.25%and 2.5%under the low and medium concentrations of CeO_(2) NPs,respectively,but increased by 7.08%under the high concentration;(2)CeO_(2) NPs had significant effects on the growth traits(root length,shoot height and biomass)of seedlings,being the highest under the medium concentration and the lowest under the highest concentration;(3)The superoxide dismutase activity was the maximum(355.91 U/g),but the protein concentration was the minimum(3.85 ug/mL)under the high concentration of CeO_(2) NPs.Our results indicated that the effects of CeO_(2) NPs on seed germination and seedling growth are concentration-dependency,i.e.,low and medium concentrations inhibited while high concentration promoted seed germination,however,seedling growth showed opposite responses.Therefore,appropriate CeO_(2) NPs concentrations are beneficial to the seed germination and seedling growth of Vitex negundo and improve the physiological performance of seedlings and enhance their adaptability to environmental adversity.展开更多

AIM: To observe the effect of erythropoietin receptor antibody (EpoRA) on oxygen-induced retinal neovascularization. METHODS: C57BL / 63 mice, newly born 7 days, were exposed in high oxygen for 5 days and then placed ...AIM: To observe the effect of erythropoietin receptor antibody (EpoRA) on oxygen-induced retinal neovascularization. METHODS: C57BL / 63 mice, newly born 7 days, were exposed in high oxygen for 5 days and then placed in normal air for another 5 days, thus the animal models of retinal neovascularization were made. Experimental animals were allocated into 3 groups: normal, experimental and therapeutic. The normal group was fed in the normal environment. Into the vitreous cavity of mice in the therapeutic group were injected 2 mu L of EpoRA for 5 successive days. And the experimental group was injected the same amount of normal saline. Mice were sacrificed 17 days after birth and their eyeballs were removed for detection of malonaldehyde (MDA) content in the retina and by HE staining endothelial cells were counted the breaking through internal limiting membrane. RESULTS: In the experimental group, MDA content in the retina was 25.11 +/- 3.46 mu mol/g, which was obviously less than those in the normal group (5.34 +/- 1.79 mu mol/g, P<0.01) and those in the therapeutic group (12.04 +/- 1.91 mu mol/g). Pathological sections showed the nuclear number of the endothelial cells breaking through internal limiting membrane was 0.7 +/- 0.2 in normal group, and 46.2 +/- 6.5 in high oxygen induced experimental group. In the therapeutic group injected with EpoRA, it was lowered to 24.0 +/- 5.0(P < 0.01). CONCLUSION: EpoRA can effectively inhibit oxygen-induced neovascularization in retina of mouse by reducing oxidative damage.展开更多

Methods: A rabbit model for knee joint OA was established by the classic Huith's method. The OA model rabbits were randomized into 5 groups: the model control group, the positive control group treated with glucosamine sulfate, and the three BSHXD treated groups treated respectively with low, moderate, and high doses of BSHXD. In addition, a normal control group and a sham-operated group were set up. Experimental animals were sacrificed after a 7-week treatment, and pathological changes in cartilaginous tissue were estimated using the Mankin criteria. Hydroxyproline (Hyp) and malonaldehyde (MDA) contents in blood serum and urine, as well as superoxide dismutase (SOD) activity and nitric oxide (NO) content in blood serum and knee joint synovial homogenates were detected. Results: Mankin scoring showed insignificant statistical differences between the various treatment groups (P〉0.05), but all were better than the model control group (P〈0.05). Serum and udnary contents of Hyp and MDA as well as serum and synovial levels of NO were significantly lower, but the SOD activity in blood serum and synovial tissue was higher in the BSHXD treated groups than in the model group (P〈0.01); the effect of BSHXD was dose-dependent to some extent. Coaclasloa: The modified BSHXD shows an effect of improving cartilage metabolism in experimental rabbits with OA, and possesses osteo-chondric protective effects in antagonizing peroxidation injury.展开更多
文摘BACKGROUND: Glutathione, as an in vivo free radical scavenger, plays an important role in the anti-oxidation defense mechanism in patients with acute cerebral infarction. OBJECTIVE: To observe the relationship between the levels of glutathione (GSH) and malonaldehyde (MDA), the metabolite of lipid peroxidation, in erythrocyte and deformation index of erythrocyte in patients at various periods following acute cerebral infarction. DESIGN: Controlled observation.SETTING: Qilu Hospital of Shandong University and Institute of Cerebrovascular Disease of Qingdao Medical College. PARTICIPANTS: We chose 98 inpatients with acute cerebral infarction from Department of Neurology, Qilu Hospital of Shandong University from January to December 2000 , serving as cerebral infarction group, including 50 male and 48 female, with mean age of (62±7)years. There were 23 cases found on the 1^st day after onset; 25 cases on the 3^rd day after onset; 25 cases on the 7^th day after onset; 25 cases on the 14th days after onset, and they were all confirmed by craniocerebral CT or MRI. Another 30 homeochronous inpatients with neurosis, cervical syndrome, lumbar intervertebral disc herniation and motor neuron disease were chosen as control group, including 20 male and 10 female, with mean age of (52±8)years . There was no significant difference in age and gender distribution between two groups (P 〉 0.05). Patients in the two groups were informed of detected index.METHODS:①Ulnar venous blood was chosen from the patients who were fasted on the 1^st, 3^rd,7^th and 14^th days after onset. Deformation index of erythrocyte was measured with BL88-CKX laser diffraction erythrocyte deformeter and photographing was performed. GSH level in erythrocyte was measured with DTNB assay introduced by Beu-tler. MDA level in erythrocyte was measured with modified thiobarbituric acid colorimetric method.②At each sample collecting , according to the criteria accepted by the Fourth National Conference of Cerebrovascular Disease, intergrant of neurologic impairment 〈 15 points was regarded as mild (n=46), 15 to 30 points as moderate (n=40)and, 〉 30 points as severe (n=12). ③ t test was used to compare data between two groups , and linear correlation analysis was used in relationship analysis among indexes. MAIN OUTCOME MEASURES :① Comparison of erythrocyte GSH and MDA levels and deformation index of erythrocyte at various periods between patients with acute cerebral infarction and controls. ②Correlation of erythrocyte GSH level with erythrocyte MDA level and with deformation index of erythrocyte in patients with cerebral infarction . ③ Relationship between erythrocyte GSH level and severity of disease in patients with acute cerebral infarction. RESULTS: Totally 98 patients with acute cerebral infarction and 30 controls all entered the stage of result analysis. ① Erythrocyte GSH level and deformation index of erythrocyte were lower on the 1^st, 3^rd, 7^th and 14^th days after onset in cerebral infarction group than in control group (P 〈 0.05-0.01), and erythrocyte MDA level was significantly higher in cerebral infarction group than in control group (P 〈 0.05-0.01). The three indexes changed most significantly on day 3 after onset in patients, and began to recover or decrease on day 7 after onset and inclined to be normal on day 14 after onset. ② Erythrocyte GSH level was significantly negatively correlated with erythrocyte MDA level in patients with acute cerebral infarction on the 1^st, 3^rd, 7^th and 14^th days after onset (r=-0.534, -0.713, -0.645, -0.656, respectively, P 〈 0.05-0.01 ) ,and significantly positively correlated with erythrocyte deformation index (t-=0.502, 0.560, 0.455, 0.504, respectively, P 〈 0.05). ③Erythrocyte GSH level was significantly lower in moderate or severe patients with acute cerebral infarction than in mild patients[(0.215±0.088),(0.192±0.102), (0.281±0.090) g/L, P〈 0.05]. CONCLUSION:①Erythrocyte GSH and MDA levels and deformation index of erythrocyte change significantly on the 3^rd day following acute cerebral infarction. Index detection results gradually tended to be normal on from the 7^th day to 14^th days day after onset. ② Change of erythrocyte GSH level can reflect the severity of disease of patients with acute cerebral infarction.③Decrease of erythrocyte GSH level in patients with acute cerebral infarction is one of reasons that result in the decrease of deformation ability of erythrocyte.

文摘[Objective]To explore an effective method for evaluating drought resistance of Yixing lily and provide a reference for selecting drought-resistant Yixing lily varieties. [Method]Through artificial simulation of drought stress environment,the changes of physiological indexes,such as content of proline,malonaldehyde,and reducing sugar in Yixing lily leaf and bulb were observed,when soil water content changed. [Result]The response to drought stress of the Yixing lily leaf was greater than that of the bulb,and the content of the proline and malonaldehyde was higher than that of the reducing sugar. [Conclusion]The leaves as an appropriate material for evaluation and analysis on drought resistance can better reflect the physiological responses of Yixing lily to drought stress. And the content of the malonaldehyde and that of proline are preferable indexes.

文摘[Objective] The preventive and therapeutic effects of Folium perillae extract (FPE) on hyperlipemia induced by feeding rabbit with high fat forage were observed. [Method] The high fat and cholesterol diet were adopted to create the hyperlipemia model of rabbit. The rabbits were randomly and equally divided into 5 groups which were NC( normal control)group, HLM (high lipid model) group, LFPE, MFPE and HFPE group (low, middle, high dosage of FPE were 0.17 g/kg, 0.5 g/kg and 1.5 g/kg respectively). The contents of TC, TG, LDL-C, HDL-C, MDA and the activity of SOD in serum were detected respectively. [ Result] The three FPE groups could decrease the contents of TC, TG, LDL-C, MDA significantly (P〈0.01) compared with these of HLM group, and the contents of HDL-C (P〈0.01) and the activity of SOD (P〈0.05 ) in serum was increased significantly. [Conclusion] FPE could regulate blood-fat and counteract lipid peroxidation.

文摘AIM: To observe the therapeutic efficacy of Baicalin in rats with severe acute pancreatitis (SAP) and explore its therapeutic mechanisms. METHODS: The SAP rat models were randomly divided into the model control group, Baicalin treatment group, octreotide treatment group and sham operation group. All groups were randomly subdivided into 3 h, 6 h and 12 h groups with 15 rats in each group. The survival, ascites volume and pathological changes of pancreas in all rats were observed at different time points after operation. The plasma amylase content and serum TNF-α, IL-6, malonaldehyde (MDA) and PLA2 contents were also determined. RESULTS: The survival was not obviously different between the treated groups, and was significantly higher in treated groups at 12 h compared to the model control group (P 〈 0.05, 15 vs 10). The ascites/body weight ratio at 3 h and 6 h was significantly lower in Baicalin treatment group compared to the model control group and octreotide treatment group (P 〈 0.05, 1.00 vs 2.02 and 1.43 and P 〈 0.001, 2.29 (1.21) vs 2.70 (0.80) and 2.08 (2.21), respectively). The contents of amylase, TNF-α, IL-6, MDA and PLA2 were significantly lower in the treated groups than in the model control group (P 〈 0.05, 4342 vs 5303, 5058 vs 6272 in amylase, P 〈 0.01, 21.90 vs 36.30, 23.80 vs 39.70, 36 vs 54.35 in MDA and 56.25 vs 76.10 in PIA2, or P 〈 0.001, 65.10 and 47.60 vs 92.15 in TNF-α, 3.03 vs 5.44, 2.88 vs 6.82, 2.83 vs 5.36 in IL-6, respectively). The pathological scores of pancreas in the treated groups were significantly lower than that in the model control group (P 〈 0.05, 9.00 vs 10.05, 6.00 vs 9.00, 8.00 vs 10.05), but no marked difference was found between the treated groups. CONCLUSION: The Baicalin injection has significant therapeutic effects on SAP rats, its effects are similar to those of octreotide. The Baicalin injection is also cheap and has a big application range, quite hopefully to be used in clinical treatment of SAP.

文摘Objective Elderly patients with obstructive sleep apnea/hypopnea syndrome (OSAHS) has a higher risk of cardiovascular and cerebrovascular disease. However, changes of homocysteine (Hey) as markers of cardiovascular and eerebrovascular disease associated with OSAHS and their mechanism have not been elucidated so far. This study aims to investigate the changes of both serum Hcy and oxidative stress and their possible links with OSAHS in elderly patients. Methods Based on polysomnogram (PSG) and age, 83 patients with OSAHS were recruited and divided into elderly-OSAHS (n=32) and non-elderly OSAHS groups (n=51). Fifty two subjects without OSAHS were divided into elderly control (n=29) and non-elderly control groups (n-23). A total of 135 subjects were included in the present study. All subjects were recorded for PSG variables and the contents of homocysteine (Hcy), malonaldehyde (MDA), and glutathione (GSH) which were detected after sleep. Serum homocysteine was measured by cyclophorase. MDA and GSH were measured by speetrophotometer. Results (1) The serum levels of Hcy showed significant difference among the four groups (P〈0.05). The concentrations of Hcy in elderly OSAHS patients were higher than in other groups, while those in the elderly control group were higher than in the non-elderly control; the concentrations in the non-elderly OSAHS group were higher than in the non-elderly control. (2) The concentrations of MDA and GSH changed at an equal pace with Hcy in the four groups. (3) Multielement linearity regression analysis indicated a statistically significant relationship between Hcy concentration and age, MDA, GSH, and apnea hypopnea index (AHI). Conclusions (1) The concentrations of Hcy and oxidative stress have increased with advancing age. (2) The concentrations of Hcy and oxidative stress have further increased in the elderly patients with OSAHS. (3) Oxidative stress might cause high-level serum Hcy in the elderly patients with OSAHS.

文摘Our previous studies revealed that etomidate, a non-barbiturate intravenous anesthetic agent, has protective effects on retinal ganglion cells within 7 days after optic nerve transection. Whether this process is related to anti-oxidative stress is not clear. To reveal its mechanism, we established the optic nerve transection injury model by transecting 1 mm behind the left eyeball of adult male Sprague-Dawley rats. The rats received an intraperitoneal injection of etomidate(4 mg/kg) once per day for 7 days. The results showed that etomidate significantly enhanced the number of retinal ganglion cells retrogradely labeled with Fluorogold at 7 days after optic nerve transection. Etomidate also significantly reduced the levels of nitric oxide and malonaldehyde in the retina and increased the level of glutathione at 12 hours after optic nerve transection. Thus, etomidate can protect retinal ganglion cells after optic nerve transection in adult rats by activating an anti-oxidative stress response. The study was approved by the Animal Ethics Committee at Air Force Medical University, China(approval No. 20180305) on March 5, 2018.
文摘In the theory of traditional Chinese medicine, aging is mainly thought renal deficiency caused renal failure, mainly involving decline of kidney-Yang and deficiency of kidney-essence. Huanshaodan, a Chinese traditional preparation for kidney-replenishing essence, was used to be the preparation for reinforcing renal deficiency and preventing aging for aged people. OBJECTIVE: To observe the effects of Huanshaodan on swimming durance and the abilities of catalase (CAT) in serum and monoamine oxidase-B (MAO-B) in brain tissue as well as in vitro anti-oxidative ability of aging mouse. DESIGN: A controlled animal experiment. SETTING: College of Basic Medicine, Hunan University of Traditional Chinese Medicine. MATERIALS: Fifty-four healthy NIH mice, aged 18 months old, of either gender, weighing (48.9±5.4) g, and one SD male rat, aged 16 months old, weighing 51.7 g, were provided by Animal Experimental Center, Hunan University of Traditional Chinese Medicine. Thirty NIH mice were randomly chosen for swimming test, and divided into experimental group and control group, with 15 in each; The other 24 NIH mice were used for enzyme activity assay, and also divided into experimental group and control group, with 12 in each. SD rat was used for in vitro anti-oxidative ability test, Huanshaodan water decoction was composed of Cheqianzi, Wuweizi, Huaishan, Danggui, Huangbai, Shudi, Baizhi, Niuxi, Baishen, Tusizi, Buguzhi, Roucongrong and Heshouwu 13 Chinese herbs. METHODS: This study was carried out in the Second Laboratory, Department of Biochemistry, Hunan University of Traditional Chinese Medicine in June 2006. Swimming and enzyme activity assay: Mice in the two experimental groups were intragastrically administrated with l0 μL/g Huanshaodan water decoction. Mice in the two control groups were intragastrically administrated with the same amount of normal saline. All the mice were intragastrically administrated for 5 days, and they were free to access to medicine in the other 2 days in a week. Each mouse was administrated for 8 weeks. MAIN OUTCOME MEASURES: ① Forty days after administration, mice in the experimental group and control group for swimming test were loaded at tails and allowed to swim in the water-tank. Swimming durance was recorded. ② Following the method of Chen Qi, the activities of CAT in serum and MAO-B in brain tissue as well as the inhibitory rate of each medicine on malonaldehyde (MDA) content in the in vitro rat hepatic tissue were determined; Meanwhile, the inhibitory rate of different doses of Tusizi liquid to MDA content in the rat hepatic tissue in vitro was also assayed. RESULTS: Fifty-four NIH mice and one SD rat were recruited in this experiment. Three mice died in the swimming test, and all the other animals were involved in the final analysis. ① Swimming durance of mice in the experimental group was significantly longer than that in the control group ( t =7.502, P 〈 0.01 ) . The activity of CAT in serum of mice in the experimental group was significantly higher than that in the control group ( t =13.307, P 〈 0.01 ) . ② The activity of MAO-B in brain tissue of mice in the experimental group was significantly lower than that in the control group (t =l3.27, P 〈 0.01 ) . ③The inhibitory rate of Cheqianzi, Wuweizi, Huaishan, Danggui, Huangbai, Shudi, Baizhi, Niuxi, Baishen, Tusizi, Buguzhi, Roucongrong and Heshouwu 13 Chinese herbs of Huanshaodan to MDA in the rat hepatic tissue in vitro was - 62.9, - 95.1, - 34.9, - 65.1, - 99.1, - 87.2, - 94.1, - 20.0, - 67.0, - 83.7, - 91.0, - 98.4, - 93.0, respectively. ④ The inhibitory rate of low to high dose of Tusizi liquid to MDA content in the rat hepatic tissue in vitro was - 3.41, - 18.1, - 26.6, - 83.7, respectively. CONCLUSION: Huanshaodan enhances swimming endurance, anti-oxygen free radical and anti-oxidativeabilities, and thus, it can delay aging.

文摘BACKGROUND: This study was undertaken to observe the concentration of SP-A/B and the pulmonary surfactant in the lung tissue of rats with acute lung injury/acute respiratory distress syndrome caused by paraquat poisoning after the treatment of metabolic antioxidant-lipoic acid and whether its influence was related to TNF-α.METHODS: Sixty-six male Sprage-Dawley rats were randomly divided into three groups: normal control group(NS group), 6 rats; paraquat poisoning group(PQ group), 30 rats; and paraquat+lipoic acid treatment group(LA group), 30 rats. The rats in the PQ and LA groups were subdivided into 3-, 6-, 12-, 24-, 48-hour subgroups, with 6 rats in each group. After the rats were sacrificed, lung tissue from the same part was taken from the rats. After HE staining, histological changes were observed in the tissue under a light microscope. Lung tissue was also taken to test the levels of superoxide dismutase(SOD) and malondialdehyde(MDA). Whole blood(0.8 mL) without anticoagulant was drawn from the tail vein of rats for the determination of the TNF-α level. The total RNA of the lung tissue was collected, and the Rt-PCR method was used to measure the levels of SP-A and SP-B mRNA.RESULTS: HE staining showed that histopathological changes were milder in the LA group than in the PQ group. There were significant differences in MDA and SOD levels between different intervals both in intergroups and intragroups except the 3-hour subgroup(P<0.01). Likewise, the significant differences in the levels of TNF-α were also present between the three groups and between different intervals(P<0.01). The significant differences in SP-A mRNA and SP-B mRNA amplification ratio were seen between the three groups at the same intervals(P<0.01), but the differences between different intervals in the PQ group were statistically significant(P<0.05). The differences between different intervals in the LA group were statistically significant(P<0.01).CONCLUSION: Lipoic acid in acute paraquat poisoning could diminish lung tissue damage by regulating directly tumor necrosis factor and indirectly the content of pulmonary surfactant so as to reduce pulmonary edema, improve lung compliance, and finally protect lung tissues.

文摘UV radiation is known to affect aquatic primary producers and their grazers. However, little has been documented on its effects on zooplankton grazing. In this study, the authors investigated the effects of photosynthetically active radiation (PAR, 400–700 nm), ultraviolet-A (UV-A, 320–400 nm) and ultraviolet-B (UV-B, 280–320 nm) radiation on grazing, mortality and lipids oxidation of the copepod Acartia pacifica collected from the Xiamen Bay. After 30 min of the exposures, the copepod was fed in darkness with the diatom Phaeodactylum tricornutum at two cell concentrations (2.5 × 10 4 and 2.5 × 10 5 cells/ml). At the low cell concentration, the individuals pre-exposed to PAR (218.0 W/m 2 )+UV-A (48.2 W/m 2 ) or PAR+UV-A+UV-B (2.1 W/m 2 ) showed suppressed clearance and grazing activities compared with those receiving PAR alone, by 22.7% and 17.1% for clearance and by 22.6% and 5.5% for grazing rates, respectively. However, the suppression on clearance and grazing became indistinctive at the high food concentration. Exposures to UV-A and UV-B led to increased lipid oxidation and higher mortality, furthermore, the mortality linearly increased with enhanced oxidation of lipid.

文摘Based on previous studies that have shown flavonoids from the stems and leaves of Scutellaria baicalensis Georgi are neuroprotective agents in a naturally senile, D-galactose, aging in vivo model, as well as an in vitro model of oxidative/hypoxic injury, we established a cerebral ischemia/reperfusion model in rats by middle cerebral artery occlusion. The light/electron microscopic observations found significant neuropathological changes including neuron loss or swelling and rough endoplasmic reticulum injury. Moreover, the activities of lactate dehydrogenase Na+-K+-ATPase, Ca2+-ATPase and superoxide dismutase were significantly lowered, and the levels of malonaldehyde increased. In addition, the memory of rats worsened. However, treatment with flavonoids from Scutellaria baicalensis Georgi (35, 70 and 140 mg/kg) for 13 days dramatically improved the above abnormal changes. These results suggest that the ability of flavonoids from Scutellaria baicalensis Georgi in attenuating cerebral functional and morphological consequences after cerebral ischemia/reperfusion may be beneficial for the treatment of ischemic brain disease.

文摘BACKGROUND: Along with aging, antioxidase activity decreases and oxygen-derived free radicals greatly accumulate, resulting in cellular senescence, or even cell death. This is manifested by hypomnesia and disordered metabolism of free radicals. Studies have reported that Longyanshen polysaccharides have the function of antioxidation and improved brain memory. OBJECTIVE: To observe the effects of Longyanshen polysaccharides on free radical metabolism in brain tissue to verify the anti-aging mechanisms in senescence accelerated-prone (SAMP8) mice. DESIGN, TIME AND SETTING: The randomized, controlled, biochemical experiment was performed in the Department of Pharmacology and Scientific Experimental Center of Guangxi Medical University (China) from September 2005 to January 2008. MATERIALS: Forty SAMP8 mice were randomized into four groups: SAMP8 control group, as well as low-, mid-, and high-dose polysaccharide, with 10 mice in each group. Ten senescence accelerated-resistantprone (SAMR 1) mice served as the normal control group. Longyanshen polysaccharides, extracted from the medical plant Longyanshen, were supplied by the Department of Pharmacology, Guangxi Medical University Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), malonaldehyde (MDA), nitric oxide (NO), and total protein test kitwere purchased from Nanjing Jiancheng Bioengineering Institute (China). METHODS: SAMP8 mice were used to establish a dementia animal model. SAMP8 and SAMR1 control mice were administered 30 mL/kg saline. The low-, middle-, and high-dose polysaccharide groups were administered 45, 90, and 180 mg/kg Longyanshen polysaccharides, respectively. Each group was treated by intragastric administration, once daily, for 50 continuous days. MAIN OUTCOME MEASURES: One hour after the last administration, mouse brain tissues were collected, and retro orbital blood sampling was performed. Spectrophotometry was used to measure SOD and GSH-Px activity, as well as MDA and NO concentration in sera and brains of SAMP8 mice. RESULTS: SOD and GSH-Px activity decreased significantly, and MDA and NO concentration increased significantly, in SAMP8 control group brain tissues, compared with the SAMP1 control group (P 〈 0.05). Compared with the SAMP8 control group, Longyanshen polysaccharide-treated groups exhibited enhanced SOD and GSH-Px activity, as well as decreased MDA and NO concentration, in serum and brain tissue (P 〈 0.05). Longyanshen polysaccharides exerted a similar effect on SOD, GSH-Px, MDA, and NO concentrations in serum and brain tissues of SAMP8 mice. CONCLUSION: Longyanshen polysaccharides scavenged free radicals effectively, reduced NO concentration and ameliorated NO toxicity, thereby influenced aging and stress, as well as improving memory capacity in the brain.

文摘The study aims to investigate the effects of(-)-Linarinic acid(LA) and one of its derivatives(LAd) on brain injury induced by ischemia. Malonaldehyde(MDA) is determined as an index for lipid peroxidation both in vitro and vivo. Mice were pre-treated with LA and LAd for 3 d.Thereafter, they were induced to have incomplete cerebral ischemia with both bilateral carotid artery occlusion and hypotension(BCAOH). In the first part of the in vivo experiment, mice were divided into four groups: sham(control), ischemia, ischemia + LA(200 mg/kg, i.g.) and ischemia + LAd(200 mg/kg, i.g.). In the second part, the dose-response of LAd was investigated at 100, 200 and 400 mg/kg i.g., respectively. A modified neurological severity score was developed for evaluating behavioral deficits of the mice with ischemia. Brains of the mice were excised in order to determinate MDA after ischemia for 6 h. Survival time, survival rate, neurological injury score and MDA level in brains were observed. Results were: 1) The data in vitro showed that both LA and LAd could inhibit the generation of MDA. IC50 values obtained by Probit analysis were 2.9 mM for LAd and 4.88 mM for LA;2) BCAOH could significantly shorten the survival span, reduce the survival rate and cause neurological deficits,which were associated with high level of lipid hydroperoxide production in cerebral tissues;3) LAd decreased lipid peroxidation and improved the neurological outcome more than LA.It is concluded that LAd offers a better neuroprotection than LA against brain damage caused by cerebral ischemia.

文摘Cerium oxide nanoparticles(CeO_(2) NPs)are likely to have dramatic impacts on plant performances,yet the effects of CeO_(2) NPs on seed germination and seedling growth have not been fully explored.In this study,the seed germination and seedling growth of subshrub species Vitex negundo under different concentrations of CeO_(2) NPs(low-1 mg/L,medium-100 mg/L,high-500 mg/L)have been discussed.Results showed that:(1)The seed germination rate reduces by 11.25%and 2.5%under the low and medium concentrations of CeO_(2) NPs,respectively,but increased by 7.08%under the high concentration;(2)CeO_(2) NPs had significant effects on the growth traits(root length,shoot height and biomass)of seedlings,being the highest under the medium concentration and the lowest under the highest concentration;(3)The superoxide dismutase activity was the maximum(355.91 U/g),but the protein concentration was the minimum(3.85 ug/mL)under the high concentration of CeO_(2) NPs.Our results indicated that the effects of CeO_(2) NPs on seed germination and seedling growth are concentration-dependency,i.e.,low and medium concentrations inhibited while high concentration promoted seed germination,however,seedling growth showed opposite responses.Therefore,appropriate CeO_(2) NPs concentrations are beneficial to the seed germination and seedling growth of Vitex negundo and improve the physiological performance of seedlings and enhance their adaptability to environmental adversity.

文摘AIM: To observe the effect of erythropoietin receptor antibody (EpoRA) on oxygen-induced retinal neovascularization. METHODS: C57BL / 63 mice, newly born 7 days, were exposed in high oxygen for 5 days and then placed in normal air for another 5 days, thus the animal models of retinal neovascularization were made. Experimental animals were allocated into 3 groups: normal, experimental and therapeutic. The normal group was fed in the normal environment. Into the vitreous cavity of mice in the therapeutic group were injected 2 mu L of EpoRA for 5 successive days. And the experimental group was injected the same amount of normal saline. Mice were sacrificed 17 days after birth and their eyeballs were removed for detection of malonaldehyde (MDA) content in the retina and by HE staining endothelial cells were counted the breaking through internal limiting membrane. RESULTS: In the experimental group, MDA content in the retina was 25.11 +/- 3.46 mu mol/g, which was obviously less than those in the normal group (5.34 +/- 1.79 mu mol/g, P<0.01) and those in the therapeutic group (12.04 +/- 1.91 mu mol/g). Pathological sections showed the nuclear number of the endothelial cells breaking through internal limiting membrane was 0.7 +/- 0.2 in normal group, and 46.2 +/- 6.5 in high oxygen induced experimental group. In the therapeutic group injected with EpoRA, it was lowered to 24.0 +/- 5.0(P < 0.01). CONCLUSION: EpoRA can effectively inhibit oxygen-induced neovascularization in retina of mouse by reducing oxidative damage.

文摘Objective: To observe the metabolic, regulatory and anti-oxidative effects of modified Bushen Huoxue Decoction (BSHXD, 补肾活血方), a Chinese herbal medicine for Kidney (Shen)-reinforcement and bloodactivation, on an osteoarthritis (OA) rabbit model. Methods: A rabbit model for knee joint OA was established by the classic Huith's method. The OA model rabbits were randomized into 5 groups: the model control group, the positive control group treated with glucosamine sulfate, and the three BSHXD treated groups treated respectively with low, moderate, and high doses of BSHXD. In addition, a normal control group and a sham-operated group were set up. Experimental animals were sacrificed after a 7-week treatment, and pathological changes in cartilaginous tissue were estimated using the Mankin criteria. Hydroxyproline (Hyp) and malonaldehyde (MDA) contents in blood serum and urine, as well as superoxide dismutase (SOD) activity and nitric oxide (NO) content in blood serum and knee joint synovial homogenates were detected. Results: Mankin scoring showed insignificant statistical differences between the various treatment groups (P〉0.05), but all were better than the model control group (P〈0.05). Serum and udnary contents of Hyp and MDA as well as serum and synovial levels of NO were significantly lower, but the SOD activity in blood serum and synovial tissue was higher in the BSHXD treated groups than in the model group (P〈0.01); the effect of BSHXD was dose-dependent to some extent. Coaclasloa: The modified BSHXD shows an effect of improving cartilage metabolism in experimental rabbits with OA, and possesses osteo-chondric protective effects in antagonizing peroxidation injury.
