The invasion of etched dentinal tubules(DTs)by external substances induces dentin hypersensitivity(DH).The deep and compact occlusion of DTs is highly desirable for treating DH but still challenging due to the limited penetrability and mineralization capacities of most current desensitizers.Matrix vesicles(MVs)participate in the regulation of ectopic mineralization.Herein,ectopic MV analogs are prepared by employing natural cell membranes to endow mineral precursors with natural biointerfaces and integrated biofunctions for stimulating dentin remineralization.The analogs quickly access DTs(>20μm)in only 5 min and further penetrate deep into the interior of DTs(an extraordinary~200μm)in 7 days.Both in vitro and in vivo studies confirm that the DTs are efficiently sealed by the newly formed minerals(>50μm)with excellent resistance to wear and acid erosion,which is significantly deeper than most reported values.After repair,the microhardness of the damaged dentin can be recovered to those of healthy dentin.For the first time,cell membrane coating nanotechnology is used as a facile and efficient therapy for in-depth remineralization of DTs in treating DH with thorough and long-term effects,which provides insights into their potential for hard tissue repair. 展开更多

The bio- inspired strategy of triggered release of Ca^2+ , Pi, F from prosomal compartments was used to induce rapid formation of FHA for potential dental use : Calcium and inorganic phosphate salts were separately loaded into temperature-sensitive liposome composed of 90% DPPC and 10% DMPC. When heated from the room temperature to 35- 37 ℃ , the entrapped calcium, inorganic phosphate and fl^rin released from the liposome,A mixture of Ca- Pi and F-loaded liposome formed apatite. The mineral was indicated by a rapid drop in suspension PH from 7.4 approximately 6. This was illustrated by applying a liposome suspension warmed human dentin and enamel surface which resulted in deposition of apatite onto the tissue substrates. 展开更多

As the quality of life improves,people pay more and more attention to health.They are concerned about the causes of diseases,and seek better treatments.The most common diseases are biomineralized diseases,four different kinds of typical examples among which are selected to elaborate their mechanisms and existing treatments.Whether it is tooth and bone in physiological mineralization or cartilage and blood vessel in pathological mineralization,they are all related to matrix vesicle(MV)-mediated biomineralization.MV-mediated biomineralization is the initial stage of biomineralization and the nucleation site mediating collagen mineralization.Definition,composition,biogenesis,and action mechanism of MVs are refined and expounded,especially a novel biomineralization pathway similar to exosome(EX)origin.Four differences are summarized to distinguish MVs and EXs.A series of treatments using MVs to solve biomineralized diseases such as tooth and bone defects,osteoarthritis and atherosclerosis are proposed,and the experimental extraction steps of MVs are summarized. 展开更多
