Expanding the Boundaries of the Chemical Space of DNA Methyltransferase 1 Modulators

DNA methyltransferase 1 (DNMT1), one of the main epigenetic targets, is involved in the duplication of the DNA methylation pattern during replication, and it is essential for proper mammalian development. Small molecule DNMT1 modulators are attractive for biochemical epigenetic studies and have the potential to become drugs. So far, more than five hundred small molecules have been reported as DNMT1 inhibitors. However, only a limited number of DNMT1 activators have been disclosed because, at least in part, DNMT1 activators are typically regarded as negative data in virtual screening campaigns or optimization projects. This manuscript aims to report the chemical structures and biological activity of small molecules that increase the enzymatic activity of DNMT1. Results of the biochemical experimental assays are discussed. It was found that small molecule activators have a large variety of chemical scaffolds but share pharmacophore features. Visual analysis of the chemical space and multiverse based on molecular fingertips supported that activators are structurally diverse. This is the first report of eight small molecules that increase the enzymatic activity of DNMT1 by more than 400% in an enzymatic-based assay. The outcome warrants further investigation of the epigenetic activity of the compounds in a counter-screen assay, e.g., cell-based and in vivo context. 展开更多

We consider the Hyperverse as a collection of multiverses in a (4 + 1)-dimensional spacetime with gravitational constant G. Multiverses in our model are bouquets of thin shells (with synchronized intrinsic times). If gis the gravitational constant of a shell Sand εits thickness, then G~εg. The physical universe is supposed to be one of those thin shells inside the local bouquet called Local Multiverse. Other remarkable objects of the Hyperverse are supposed to be black holes, black lenses, black rings and (generalized) Black Saturns. In addition, Schwarzschild-de Sitter multiversal nurseries can be hidden inside those Black Saturns, leading to their Bousso-Hawking nucleation. It also suggests that black holes in our physical universe might harbor embedded (2 + 1)-dimensional multiverses. This is compatible with outstanding ideas and results of Bekenstein, Hawking-Vaz and Corda about “black holes as atoms” and the condensation of matter on “apparent horizons”. It allows us to formulate conjecture 12.1 about the origin of the Local Multiverse. As an alternative model, we examine spacetime warping of our universe by external universes. It gives data for the accelerated expansion and the cosmological constant Λ, which are in agreement with observation, thus opening a possibility for verification of the multiverse model. 展开更多

This work examined Bertrand Russell’s main arguments against the existence of God and his strongly negative opinion on the historic role of religions.These arguments were:(1)The idea that God is a First Cause is invalid because the cosmos doesn’t have to have a beginning;(2)If there were reasons for God to issue the observed natural laws and not others then God Himself was just an intermediary of older laws;(3)Considering all the defects of the world one cannot consider God as an omnipotent designer;(4)If God created both right and wrong then it is no longer a significant statement that God is good;And(5)most people believe in God just because they have been taught from early infancy to do it.The criticisms of cosmological neuroscience were as follows:(1)The particular cosmic wave that carries our Universe could have a beginning;(2)Although there were reasons for the natural laws which God issued,it cannot diminish the divine creativity of this act;(3)Although the world is indeed full of defects,they are as much parts of the cosmic order as its splendors,due to their common roots in the Law of Coexistence in Diversity;(4)Although right and wrong may ultimately be both in God’s blueprint,He is still benevolent due to the Law of Divine-Evil Asymmetry permitting wrong only under the dominance of right;And(5)although many believe in God only because they were taught to do so and hope it provides safety,this is an oversimplification unable to explain the genuine Faith of others.The devastating judgement of Russel that religions have been just“a source of untold misery to the human race”was also examined.This brief article could not detail all the positive social changes with which the religions of goodness(e.g.,Abrahamic religions)enriched history,such as introducing the respect for human life.But the paper did remind the reader of the God-inspired work of Lao Tzu,Rumi,Michelangelo,the Bach-interpreting Leonard Bernstein,Mother Teresa,Martin Luther King Jr.,George Harrison and others including the architects of the Borobudur temple and astronauts like Frank Borman who read from the Bible while flying around the Moon.The paper ended with the central thought of Bertrand Russell that“The good life is one inspired by love and guided by knowledge”,to emphasize that if committed to their Conscience atheists like him can live as moral lives as the men and women of Faith—the difference is being nothing else than sensing or not the Soul that permeates the cosmos. 展开更多

Density also has the same effect as gravity because gravity attracts matter thereby increasing matter density. The laws of Physics and all the constants of nature such as the Speed of light, Planck’s constant, the Gravitational constant, and so on are a function of the dimension of the space they are in since the vacuum energy density of each higher dimension is greater. A DMVOCC-MVDA (distributed multiversion optimistic concurrency control with multiversion dynamic adjustment) protocol was presented to process mobile distributed real-time transaction in mobile broadcast environments. At the mobile hosts, all transactions perform local pre-validation. The local pre-validation process is carried out against the committed transactions at the server in the last broadcast cycle. Transactions that survive in local pre-validation must be submitted to the server for local final validation. The new protocol eliminates conflicts between mobile read-only and mobile update transactions, and resolves data conflicts flexibly by using multiversion dynamic adjustment of serialization order to avoid unnecessary restarts of transactions. Mobile read-only transactions can be committed with no-blocking, and respond time of mobile read-only transactions is greatly shortened. The tolerance of mobile transactions of disconnections from the broadcast channel is increased. In global validation mobile distributed transactions have to do check to ensure distributed serializability in all participants. The simulation results show that the new concurrency control protocol proposed offers better performance than other protocols in terms of miss rate, restart rate, commit rate. Under high work load (think time is ls) the miss rate of DMVOCC-MVDA is only 14.6%, is significantly lower than that of other protocols. The restart rate of DMVOCC-MVDA is only 32.3%, showing that DMVOCC-MVDA can effectively reduce the restart rate of mobile transactions. And the commit rate of DMVOCC-MVDA is up to 61.2%, which is obviously higher than that of other protocols. 展开更多

Other pieces of evidence are mentioned as well. The use of the general scientific principle of the physical reality of imaginary numbers allowed to prove that the relativistic formulas in the existing version of the special relativity theory are wrong, as well as to deny the principle of non-exceeding the speed of light and the hypothesis of monoverse. Corrected relativistic formulas are given and on their basis a theory of hidden Multiverse is proposed, parallel universes of which are mutually invisible. Using data obtained by the spacecrafts WMAP and Planck, it is proved that the hidden Multiverse has a quaternion structure, exists in the six-dimensional space, and forms Hyperverse together with other Multiverses. It is shown that the parallel universes of the hidden Multiverse are continuously drifting in multidimensional space, touch each other in many places and are even partially immersed in each other, forming portals. It is alleged that the search for extraterrestrial life in deep space is devoid of meaning, as you can even walk to the adjacent parallel universes through portals, which are anomalous zones on Earth. Moreover, extraterrestrial super civilizations have already found us long time ago and are located on Earth. The attempts to contact these super civilizations are meaningless as well, because such contacts are possible only at their wish. Consequently, to make the contacts possible, we need to convince super civilizations that human civilization is of interest to them and that it has a sufficiently high intelligence. Such explanation of dark matter and dark energy phenomenon in astrophysics has become possible only after proving of the principle of physical reality of imaginary numbers discovered in mathematics 500 years ago. And the principle of physical reality of imaginary numbers has made the postulate of light speed non-exceedance in the special theory of relativity unnecessary. Therefore,?all relativistic formulas in this theory should be adjusted. As follows from the adjusted relativistic formulas, we live in Multiverse rather than in Monoverse. All parallel universes of the Multiverse are mutually invisible, therefore Multiverse is called hidden. WMAP and Planck data analysis has showed that the hidden Multiverse has a quaternion structure and contains 20?-?22 parallel universes, of which 5?-?6 are adjacent to our universe, and others are further. The most complicated problems to solve in science are multidisciplinary problems, which include the problem of explaining of physical reality and physical essenceofimaginary numbers discovered more than five hundred years ago. However, the authors of the special relativity theory created more than a hundred years ago, were trying to unprovenly claim that they have solved this problem and that the imaginary numbers are not physically real with a principle of non-exceeding the speed of light. And physical experiments MINOS at the Tevatron collider and OPERA at the Large Hadron Collider were even seem to confirm such an assertion, since they could not refute the principle of non-exceeding the speed of light. However, since mathematics is the universal language of all exact sciences, the principle of the physical reality of imaginary numbers was still proven, but proven otherwise—with the theoretical and experimental studies of the vibrational processes in linear electric circuits that can be repeated and verified in any radioelectronic and electrical laboratory. And that is why they are definitely reliable and conclusive. Using the very general scientific principle of the physical reality of imaginary numbers allowed to correct the relativistic formula of special relativity theory and on this basis to cre-ate a theory of hidden Multiverse containing more than two mutually invisible parallel universes. The nature of their invisibility is explained. It is proven that the existence of these unseen parallel universes explains the phenomenon of dark matter and dark energy. Besides, edges of the hidden Multiverse are connected to other (from one to four) Multiverses, which are observable neither by electromagnetic nor by gravitational manifestations. The Multiverse described contains four matter-antimatter pairs, annihilation of which is prevented by relative spatial position of the universes. The experimental proof of existence of the hidden Multiverse is explained to be the phenomenon of dark matter and dark energy that correspond to other invisible parallel universes, except ours, included in the hidden Multiverse. General scientific principle of physical reality of imaginary numbers, refuting some of the statements of the existing version of the special theory of relativity, is a physical and mathematical foundation of the outlined conception of the hidden Multiverse. The article presents relativistic formulas of the theory of special relativity adjusted in accordance with the principle. Our third spatial dimension (3-D) is currently in the process of building the fourth spatial dimension. Dark Energy is being used to create higher dimensions of space from the first to the nth, and it is also responsible for the formation and the rotation of the fourth dimension from the point singularities of the vacuum space of the third dimension. To start the creation up to the nth dimension, <i>n</i>! zero dimensional singularities arise simultaneously from the <i>n</i> = 0 level of the void. Once the creation of an n dimensional ellipsoidal matter Universe has been completed, the outward Dark Energy force ends, and then the inward force of gravity takes over to crush the n dimensional Universe into the center of the nth dimensional singularity of the void. The system is now in a non-equilibrium state because the entropy of the system has attained its lowest possible value. To bring the system back to equilibrium there will arise out of the nth dimensional void an antimatter Universe which will reverse the process to deposit all the Dark Energy back into the original levels of the void so that the net change in entropy of the system becomes zero. Dark Matter is the matter that exists in the fourth spatial dimension as 4-D matter spewed there through the Black Hole singularities of our third dimension. It has Energy in the form of photons in quantum levels “n”. This Photon (Dark) Energy is used to create the Quantum of the physical Universe with different quantum numbers n<sup>*</sup> and the “n + 1” dimensions of the Matter and Antimatter Multiverses by creating space, time, mass, and charge. The quantum numbers n<sup>*</sup> of the physical Universe can range up to very large values compared to the quantum levels “n” of the void because they serve a different purpose which is to connect Quantum Mechanics to Classical Mechanics which does not exist within the void. Photons have a wave-particle dual nature in the Quantum of the physical Universe which is non-existent in the void. Photons exist in spin-up and spin-down states in the Quantum of the void. The four Fundamental Forces of nature exist only in the physical Universe.
