miR-10b通过激活KLF4/Notch-1/STAT3信号途径加速腹主动脉瘤的进展

目的:miR-10b促进动脉瘤的进展但其机制未明确,本实验通过检测miR-10b对KLF4/Notch-1/STAT3途径的影响阐明其机理。方法:双荧光素酶实验验证miR-10b的靶基因。取20只apoE-/-小鼠构建腹主动脉瘤模型,其中10只尾静脉注射miR-10b过表达的... 目的:miR-10b促进动脉瘤的进展但其机制未明确,本实验通过检测miR-10b对KLF4/Notch-1/STAT3途径的影响阐明其机理。方法:双荧光素酶实验验证miR-10b的靶基因。取20只apoE-/-小鼠构建腹主动脉瘤模型,其中10只尾静脉注射miR-10b过表达的慢病毒,分别记为模型组和miR-10b组。另取10只apoE-/-小鼠作为假手术组,28 d后解剖小鼠腹主动脉行病理学染色、Western-blot实验。将血管紧张素Ⅱ(AngⅡ)作用后的THP-1巨噬细胞根据是否转染miR-10b mimic及其Nc对照、是否添加抑制剂分组:(1)Nc组;(2)miR-10b mimic组;(3)Nc+KLF4抑制组;(4)miR-10b mimic+KLF4抑制组;(5)Nc+Notch-1抑制组;(6)miR-10b mimic+Notch-1抑制组;(7)Nc+STAT3抑制组;(8)miR-10b mimic+STAT3抑制组,检测细胞内蛋白水平。结果:检测双荧光素酶活性得知miR-10b可靶向作用于基因KLF4。小鼠血管病理实验发现,与正常小鼠相比,动脉瘤模型小鼠的血管明显增粗、组织结构异常,miR-10b可加重血管病变;CD68+巨噬细胞、蛋白Notch-1、p-STAT3、MMP-2、MMP-9在假手术组、模型组与miR-10b逐渐增多,而α-SMA、蛋白KLF4逐渐减少。(1)、(2)两组细胞间,后者KLF4表达较少、而Notch-1、p-STAT3、MMP-2、MMP-9表达显著偏多;KLF4抑制剂抑制KLF4表达,而Notch-1显著增多;Notch-1被抑制而减少表达,p-STAT3也随之减少;p-STAT3被抑制表达,MMP-2与MMP-9表达量也明显降低。结论:miR-10b通过作用于KLF4/Notch-1/STAT3信号途径,引起动脉瘤组织中MMP-2、MMP-9表达增加,从而加速动脉瘤的进程。 展开更多

白细胞介素10工程化修饰人脐带间充质干细胞优效治疗炎症性肠病

背景:间充质干细胞来源广泛、易体外增殖,并可分泌一系列免疫调节因子抑制炎症、促进组织修复再生,广泛应用于各种疾病治疗研究。但是对于多种疾病,间充质干细胞的治疗效果有限。针对疾病特定发病机制或者干预靶点进行工程化修饰间充质... 背景:间充质干细胞来源广泛、易体外增殖,并可分泌一系列免疫调节因子抑制炎症、促进组织修复再生,广泛应用于各种疾病治疗研究。但是对于多种疾病,间充质干细胞的治疗效果有限。针对疾病特定发病机制或者干预靶点进行工程化修饰间充质干细胞是未来细胞治疗的重要发展方向。目的:白细胞介素10是一种典型的抗炎细胞因子,有助于调节免疫反应并诱导巨噬细胞向抗炎表型极化,该研究探讨白细胞介素10基因工程化修饰人脐带间充质干细胞对炎症性肠病的治疗效果。方法:通过电转染建立稳定过表达人白细胞介素10基因的人脐带间充质干细胞,并基于细胞治疗产品标准筛选出临床级细胞。给予C57BL/6J小鼠自由饮用5%葡聚糖硫酸钠盐水溶液建立急性结肠炎模型,分别在造模前1 d(尾静脉途径)与造模后第4天(腹腔途径)注射空质粒转染的人脐带间充质干细胞或过表达人白细胞介素10基因的人脐带间充质干细胞(1×10~6个/只)。在造模后第6天取结肠组织进行苏木精-伊红染色评估组织学变化,免疫荧光染色检测增殖细胞核抗原与CD31的表达。结果与结论:构建的稳定过表达白细胞介素10的工程化人脐带间充质干细胞,满足临床级人脐带间充质干细胞质量标准;人脐带间充质干细胞对小鼠急性结肠炎具有修复效果,过表达白细胞介素10基因的人脐带间充质干细胞的治疗效果更加优效,更显著地抑制急性结肠炎小鼠的体质量下降(P<0.05)、结肠缩短(P<0.05)以及结肠组织损伤(P<0.05);过表达白细胞介素10基因人脐带间充质干细胞组结肠组织切片中增殖细胞核抗原阳性细胞与CD31阳性细胞显著多于人脐带间充质干细胞组,表明过表达白细胞介素10基因的人脐带间充质干细胞可能通过显著促进肠组织细胞增殖和血管再生进而促进结肠组织修复。 展开更多

Optimal extent of lymphadenectomy improves prognosis and guides adjuvant chemotherapy in esophageal cancer: A propensity scorematched analysis

BACKGROUND The optimal extent of lymphadenectomy in esophageal squamous cell carcinoma(ESCC)patients remained debatable.AIM To explore the ideal number of cleared lymph nodes in ESCC patients undergoing upfront surgery.METHODS In this retrospective,propensity score-matched study,we included 1042 ESCC patients who underwent esophagectomy from November 2008 and October 2019.Patients who underwent neoadjuvant therapy were excluded.We collected pa-tients’clinicopathological features and information regarding lymph nodes,in-cluding the total number of resected lymph nodes(NRLN),and pathologically diagnosed positive lymph nodes(RPLN).SPSS and R software were used for statistical analysis.RESULTS Among the included 1042 patients,two cohorts:≤21(n=664)and>21 NRLN(n=378)were identified.The final prognostic model included four variables:T stage,N,venous thrombus,and the number of removed lymph nodes.Among them,NRLN>21 was determined as an independent prognosticator after surgery for esophageal cancer(hazards regression=0.66,95%confidence interval:0.50-0.87,P=0.004).A nomogram was created based on the regression coefficients of the variables in the final model.In the training cohort,the predictive model dis-played an uncorrected five-year overall survival C-index of 0.659,with a bootstrap-corrected C-index of 0.654.In the subgroup analysis,adjuvant chemotherapy was beneficial in the subgroup with NRLN>21 and RPLN≤0.16 and NRLN≤21 and RPLN>0.16.CONCLUSION NRLN>21 was an independent prognostic factor after ESCC surgery.The combination of NRLN and RPLN may provide a reference for adjuvant chemotherapy use in potential beneficiaries. 展开更多

A Wnt10a-Notch signaling axis controls Hertwig’s epithelial root sheath cell behaviors during root furcation patterning

Human with bi-allelic WNT10A mutations and epithelial Wnt10a knockout mice present enlarged pulp chamber and apical displacement of the root furcation of multi-rooted teeth,known as taurodontism;thus,indicating the critical role of Wnt10a in tooth root morphogenesis.However,the endogenous mechanism by which epithelial Wnt10a regulates Hertwig’s epithelial root sheath(HERS)cellular behaviors and contributes to root furcation patterning remains unclear.In this study,we found that HERS in the presumptive root furcating region failed to elongate at an appropriate horizontal level in K14-Cre;Wnt10a^(fl/fl)mice from post-natal day 0.5(PN0.5)to PN4.5.EdU assays and immunofluorescent staining of cyclin D1 revealed significantly decreased proliferation activity of inner enamel epithelial(IEE)cells of HERS in K14-Cre;Wnt10a^(fl/fl)mice at PN2.5 and PN3.5.Immunofluorescent staining of E-Cadherin and acetyl-α-Tubulin demonstrated that the IEE cells of HERS tended to divide perpendicularly to the horizontal plane,which impaired the horizontal extension of HERS in the presumptive root furcating region of K14-Cre;Wnt10a^(fl/fl)mice.RNA-seq and immunofluorescence showed that the expressions of Jag1 and Notch2 were downregulated in IEE cells of HERS in K14-Cre;Wnt10a^(fl/fl)mice.Furthermore,after activation of Notch signaling in K14-Cre;Wnt10a^(fl/fl)molars by Notch2 adenovirus and kidney capsule grafts,the root furcation defect was partially rescued.Taken together,our study demonstrates that an epithelial Wnt10a-Notch signaling axis is crucial for modulating HERS cell proper proliferation and horizontal-oriented division during tooth root furcation morphogenesis. 展开更多

Background:Hand,foot and mouth disease(HFMD)is a common infectious disease caused by viral infection by a variety of enteroviruses,with coxsackievirus A 10(CA10)having become more prevalent in recent years.Methods:In this study,models of CA10 infection were established in 7-day-old Institute of Cancer Research(ICR)mice by intraperitoneal injection to analyze the pathogenicity of the virus.RNA sequencing analysis was used to screen the differentially expressed genes(DEGs)after CA10 infection.Coxsackievirus A 16(CA16)and enterovirus 71(EV71)infections were also compared with CA10.Results:After CA10 virus infection,the mice showed paralysis of the hind limbs at 3 days post infection and weight loss at 5 days post infection.We observed viral replication in various tissues and severe inflammatory cell infiltration in skeletal muscle.The RNA-sequencing analysis showed that the DEGs in blood,muscle,thymus and spleen showed heterogeneity after CA10 infection and the most upregulated DEGs in muscle were enriched in immune-related pathways.Compared with CA16 and EV71 infection,CA10 may have an inhibitory effect on T helper(Th)cell differentiation and cell growth.Additionally,the common DEGs in the three viruses were most enriched in the immune system response,including the Toll-l ike receptor pathway and the nucleotide-binding and oligomerization domain(NOD)-l ike pathway.Conclusions:Our findings revealed a group of genes that coordinate in response to CA10 infection,which increases our understanding of the pathological mechanism of HFMD. 展开更多

Objective:Lung squamous cell carcinoma(LUSC)is associated with a low survival rate.Evidence suggests that bone morphogenetic proteins(BMPs)and their receptors(BMPRs)play crucial roles in tumorigenesis and progression.However,a comprehensive analysis of their role in LUSC is lacking.Our study aimed to explore the relationship between BMPs/BMPRs expression levels and the tumorigenesis and prognosis of LUSC.Methods:The“R/Limma”package was utilized to analyze the differential expression characteristics of BMPs/BMPRs in LUSC,using data from TCGA,GTEx,and GEO databases.Concurrently,the“survminer”packages were employed to investigate their prognostic value and correlation with clinical features in LUSC.The core gene associated with LUSC progression was further explored through weighted gene correlation network analysis(WGCNA).LASSO analysis was conducted to construct a prognostic risk model for LUSC.Clinical specimens were examined by immunohistochemical analysis to confirm the diagnostic value in LUSC.Furthermore,based on the tumor immune estimation resource database and tumor-immune system interaction database,the role of the core gene in the tumor microenvironment of LUSC was explored.Results:GDF10 had a significant correlation only with the pathological T stage of LUSC,and the protein expression level of GDF10 decreased with the tumorigenesis of LUSC.A prognostic risk model was constructed with GDF10 as the core gene and 5 hub genes(HRASLS,HIST1H2BH,FLRT3,CHEK2,and ALPL)for LUSC.GDF10 showed a significant positive correlation with immune cell infiltration and immune checkpoint expression.Conclusion:GDF10 might serve as a diagnostic biomarker reflecting the tumorigenesis of LUSC and regulating the tumor immune microenvironment to guide more effective treatment for LUSC. 展开更多

Objective:The aim of this study was to examine angiotensin converting enzyme(ACE)insertion/deletion,alpha adducin,and interleukin-10(IL-10)gene polymorphisms(GPs)in terms of both idiopathic sudden sensorineural hearing loss(ISSNHL)risk and their potential prognostic effects.Methods:The study group consisted of 70 patients and the control group consisted of 50 patients.Venous blood samples were analyzed for relevant GPs via kompetitive allele-specific polymerase chain reaction.Age,sex,affected side,tinnitus,and vertiginous symptom status,number of days between symptom onset and hospital admission,pure tone audiometry results at admission and after treatment were included in the study.Data were compared statistically.Results:The D allele of ACE insertion/deletion GP was significantly more frequent in patients with ISSNHL than in the control group(p=0.032).II genotype was associated with a reduced risk of ISSNHL(p=0.036).The amount of hearing loss was significantly higher in patients with the TT genotype(p=0.027)and T allele of the IL-10 GP(p=0.035)than in the patients without this allele.Severe hearing loss was a poor prognostic factor(p=0.008).Conclusions:The D allele of ACE insertion/deletion GP may be involved in the ISSNHL etiology.Due to the association of this allele with occlusive vascular pathologies,ischemia is believed to be a common pathway in the etiopathogenesis of ISSNHL. 展开更多

BACKGROUND Secondary hemophagocytic lymphohistiocytosis(sHLH)triggered by Salmonella enterica serovar Typhimurium is rare in pediatric patients.There is no consensus on how to treat S.typhimurium-triggered sHLH.CASE SUMMARY A 9-year-old boy with intermittent fever for 3 d presented to our hospital with positive results for S.typhimurium,human rhinovirus,and Mycoplasma pneumoniae infections.At the time of admission to our institution,the patient’s T helper 1/T helper 2 cytokine levels were 326 pg/mL for interleukin 6(IL-6),9.1 pg/mL for IL-10,and 246.7 pg/mL for interferon-gamma(IFN-γ),for which the ratio of IL-10 to IFN-γwas 0.04.In this study,the patient received meropenem,linezolid,and cefoperazone/sulbactam in combination with high-dose methylprednisolone therapy(10 mg/kg/d for 3 d)and antishock supportive treatment twice.After careful evaluation,this patient did not receive HLH chemotherapy and recovered well.CONCLUSION S.Typhimurium infection-triggered sHLH patient had a ratio of IL-10 to IFN-γ≤1.33,an IL-10 concentration≤10.0 pg/mL,and/or an IFN-γconcentration≤225 pg/mL at admission.Early antimicrobial and supportive treatment was sufficient,and the HLH-94/2004 protocol was not necessary under these conditions. 展开更多

评价 C6H10S2O联合Nano-CaCO3对铅染毒小鼠的海马学习记忆功能产生的影响,以指导铅染毒相关问题的处理。方法 选择齐齐哈尔医学院动物实验中心提供的20-30g 健康雌性 KM 小鼠,健康清洁级,分为正常对照组、模型对照组、C6H10S2O 组、Nano... 评价 C6H10S2O联合Nano-CaCO3对铅染毒小鼠的海马学习记忆功能产生的影响,以指导铅染毒相关问题的处理。方法 选择齐齐哈尔医学院动物实验中心提供的20-30g 健康雌性 KM 小鼠,健康清洁级,分为正常对照组、模型对照组、C6H10S2O 组、Nano-CaCO3组、联合治疗组,其中后四组均为铅染毒模型。五组的饲养条件相同,方法 /剂量建立铅染毒模型,提供对应治疗方法,通过评估体质量与血液中铅元素含量、NO含量和ACHE活力、水迷宫、新物体识别测量的结果,判断治疗效果。结果 与空白对照组比较,模型对照组的体质量显著降低(P<0.05),血铅含量显著增加(P<0.05);与模型对照组比较,C6H10S2O组和联合治疗组小鼠的体质量显著提高(P<0.05),而血铅含量仅在联合治疗组显著减少(P<0.05);其他三组的体质量和血铅含量均无统计学差异(P>0.05)。与模型对照组比较,联合治疗组的跨越隐匿平台次数和平台象限游泳路程显著增加(P<0.05);与Nano-CaCO3组比较,联合治疗组的跨越隐匿平台次数和平台象限游泳路程也显著增加(P<0.05)。五组在NO含量和ACHE活力、认知指数方面的对比,差异均不存在统计学意义(P>0.05)。结论 C6H10S2O联合Nano-CaCO3可以减少铅含量,提高体质量,对空间学习能力有一定影响,但对认知能力、学习记忆能力不存在显著影响。 展开更多

In this paper,we perform the detailed modeling for the light curves(LCs)of PTF 10iuv which is a calcium-rich(Ca-rich)supernova(SN)to constrain the physical properties of its ejecta and the energy sources,as well as the explosion mechanism.We find that the^(56)Ni model and the56Ni plus circumstellar interaction model fail to explain the LCs,while the four-element(^(56)Ni,^(48)Cr,^(52)Fe,and^(44)Ti)model can account for the LCs.The ejecta mass of PTF10iuv derived by the model(1.52_(-0.25)^(+0.34)M_(⊙))is consistent with that of the merger of a sub-Chandrasekhar mass white dwarf.The early-time LCs were mainly powered by^(56)Ni whose mass is~0.03 M_(⊙),while the contributions of^(48)Cr and^(52)Fe can be neglected.The derived^(44)Ti mass(~0.25 M_(⊙))is~1.8 times the upper limit of the derived^(44)Ti mass of Ca-rich SN 2005E.We suggest that subtracting the contributions of the host-galaxy,which are unknown,and including the flux from other long-lived elements(e.g.,^(57)Co,^(55)Fe,^(60)Co)can reduce the amount of^(44)Ti,and that this value can be regarded as an upper limit. 展开更多

Background:S100A8 is a member of the S100 protein family and plays a pivotal role in regulating inflammation and tumor progression.This study aimed to comprehensively assess the expression patterns and functional roles of S100A8 in glioma progression.Methods:Glioma tissues were collected from 98 patients who underwent surgical treatment at Fudan University Shanghai Cancer Center.S100A8 expression in glioma tissues was analyzed using immunohistochemistry(IHC)to establish its correlation with clinicopathological features in patients.The expression and prognostic effect of S100A8 in glioma were analyzed using TCGA and CGGA public databases.Then,we investigated the role of S100A8 in glioma through a series of in vivo and in vitro experiments including Transwell,wound healing,CCK8,and intracranial tumor models.Subsequently,bioinformatics analysis,single-cell sequencing and coimmunopre-cipitation(Co-IP)were used to explore the underlying mechanism.Results:S100A8 was upregulated in gliomas compared to paracancerous tissues,and this phenotype was sig-nificantly correlated with poor prognosis.Subgroup analysis showed that S100A8 expression was higher in the high-grade glioma(HGG)group than that in the low-grade glioma(LGG)group.S100A8 overexpression in glioma cell lines promoted cell proliferation,migration and invasion,while silencing S100A8 reversed these effects.In vivo experiments showed that S100A8 knockdown can significantly reduce the tumor burden of glioma cells.Notably,S100A8 was observed to stimulate microglial M2 polarization by interacting with TLR4,which subse-quently induced NF-𝜅B signaling and IL-10 secretion within the tumor microenvironment.Conclusions:S100A8 promotes tumor progression by inducing phenotypic polarization of microglia through the TLR4/IL-10 signaling pathway in glioma.It might represent a therapeutic target for further basic research or clinical management of glioma. 展开更多

Retinitis pigmentosa is a group of inherited diseases that lead to retinal degeneration and photoreceptor cell death.However,there is no effective treatment for retinitis pigmentosa caused by PDE6B mutation.Adeno-associated virus(AAV)-mediated gene therapy is a promising strategy for treating retinitis pigmentosa.The aim of this study was to explore the molecular mechanisms by which AAV2-PDE6B rescues retinal function.To do this,we injected retinal degeneration 10(rd10)mice subretinally with AAV2-PDE6B and assessed the therapeutic effects on retinal function and structure using dark-and light-adapted electroretinogram,optical coherence tomography,and immunofluorescence.Data-independent acquisition-mass spectrometry-based proteomic analysis was conducted to investigate protein expression levels and pathway enrichment,and the results from this analysis were verified by real-time polymerase chain reaction and western blotting.AAV2-PDE6B injection significantly upregulated PDE6βexpression,preserved electroretinogram responses,and preserved outer nuclear layer thickness in rd10 mice.Differentially expressed proteins between wild-type and rd10 mice were closely related to visual perception,and treating rd10 mice with AAV2-PDE6B restored differentially expressed protein expression to levels similar to those seen in wild-type mice.Kyoto Encyclopedia of Genes and Genome analysis showed that the differentially expressed proteins whose expression was most significantly altered by AAV2-PDE6B injection were enriched in phototransduction pathways.Furthermore,the phototransductionrelated proteins Pde6α,Rom1,Rho,Aldh1a1,and Rbp1 exhibited opposite expression patterns in rd10 mice with or without AAV2-PDE6B treatment.Finally,Bax/Bcl-2,p-ERK/ERK,and p-c-Fos/c-Fos expression levels decreased in rd10 mice following AAV2-PDE6B treatment.Our data suggest that AAV2-PDE6B-mediated gene therapy promotes phototransduction and inhibits apoptosis by inhibiting the ERK signaling pathway and upregulating Bcl-2/Bax expression in retinitis pigmentosa. 展开更多

目的探讨中药复方清化饮对大鼠慢性萎缩性胃炎(CAG)胃黏膜病理与血清白细胞介素-10(IL-10)、一氧化氮(NO)、胃泌素(GAS)及血浆胃动素(MTL)的影响。方法将53只Wistar大鼠随机分为空白组8只和CAG造模组45只,予建立病证结合CAG大鼠模型。... 目的探讨中药复方清化饮对大鼠慢性萎缩性胃炎(CAG)胃黏膜病理与血清白细胞介素-10(IL-10)、一氧化氮(NO)、胃泌素(GAS)及血浆胃动素(MTL)的影响。方法将53只Wistar大鼠随机分为空白组8只和CAG造模组45只,予建立病证结合CAG大鼠模型。抽检大鼠确认造模成功后,将CAG造模组余下40只大鼠随机分为模型组、维酶素组、中药低、中、高剂量组各8只。予相应药物干预30d后处死大鼠,腹主动脉取血并留取胃组织,分析各组大鼠胃黏膜病理与血清IL-10、NO、GAS及血浆MTL浓度。结果与空白组比较,模型组大鼠见不同程度的胃黏膜萎缩,血清IL-10、GAS浓度降低,血清NO、血浆MTL含量升高(P<0.01)。与模型组比较,清化饮可不同程度改善胃黏膜病理情况,升高血清IL-10、GAS浓度,降低血清NO、血浆MTL含量(P<0.05或P<0.01),且高剂量组疗效更为显著(P<0.01)。结论中药复方清化饮可调控血清IL-10、NO、GAS及血浆MTL的表达失衡,有效逆转CAG大鼠胃黏膜病变。 展开更多

目的:构建幽门螺杆菌(Helicobacter pylori,Hp)感染C57BL/6小鼠动物模型,检测NOD1/NF-κB信号通路在小鼠胃组织的激活情况,研究其在Hp感染引起的炎症反应中的作用。方法:6-8周龄C57BL/6小鼠随机分为对照组和不同浓度Hp感染组。每48 ... 目的:构建幽门螺杆菌(Helicobacter pylori,Hp)感染C57BL/6小鼠动物模型,检测NOD1/NF-κB信号通路在小鼠胃组织的激活情况,研究其在Hp感染引起的炎症反应中的作用。方法:6-8周龄C57BL/6小鼠随机分为对照组和不同浓度Hp感染组。每48 h分别以PBS和Hp灌胃1次,共5次。并以不同时间点处死小鼠,HE染色法鉴定感染小鼠胃组织炎症程度的变化;RT-PCR检测小鼠胃组织中NOD1、RIP2的表达情况;ELISA检测血清中β干扰素(IFN-β)和趋化因子10(IP-10)的浓度;Western blot检测胃组织中p65的核转位情况。结果:与对照组相比,Hp感染后胃组织腺体减少、萎缩,肌层间质有炎症细胞浸润;血清中IP-10和IFN-β含量均有一定程度的增高,16周达到高峰;胃组织中NOD1 mRNA表达水平以及细胞核中p65含量在24-120 h时间段均显著高于对照组(P〈0.05),48 h后达到峰值,随后下降;RIP2 mRNA 48 h达到高峰,72 h后开始下降,但120 h时表达量又会升高。结论:Hp感染能激活NOD1/NF-κB信号通路,并能诱导IFN-β和IP-10的产生。 展开更多

目的探讨rno-miR-30b-5p在葡萄膜炎中对白细胞介素10(interleukin-10,IL-10)和Toll样受体4(toll-like receptor 4,TLR4)表达的调控作用。方法构建IL-10、TLR4基因的3’端非编码区域(3’UTR)荧光素酶质粒载体以及结合位点突变质粒载体,将... 目的探讨rno-miR-30b-5p在葡萄膜炎中对白细胞介素10(interleukin-10,IL-10)和Toll样受体4(toll-like receptor 4,TLR4)表达的调控作用。方法构建IL-10、TLR4基因的3’端非编码区域(3’UTR)荧光素酶质粒载体以及结合位点突变质粒载体,将rnomiR-30b-5p模拟物(mimic)与构建好的报告基因载体共转染293T细胞,通过报告基因相对荧光值的表达改变检测rno-miR-30b-5p对IL-10、TLR4表达的调控作用;制备实验性自身免疫性葡萄膜炎(experimental autoimmune uveitis,EAU)模型,通过注射视网膜光感受器间维生素A类结合蛋白乳糜液,在造模后12 d分离EAU大鼠的脾脏和淋巴结,实时荧光定量PCR检测rno-miR-30b-5p和IL-10、TLR4基因的表达情况;ELISA检测IL-10和TLR4蛋白的表达情况。结果双荧光素酶报告基因表达结果表明,经rno-miR-30b-5p mimic干预后IL-10、TLR4野生型的报告荧光有明显的下调作用,对其预测靶位点进行突变后,突变型载体中的报告荧光也存在明显的下调作用,与阴性对照相比,差异均有统计学意义(均为P<0.01)。实时荧光定量PCR检测结果发现rno-miR-30b-5p在EAU大鼠脾脏和淋巴结中的表达均较对照组明显降低(均为P<0.01),而IL-10 mRNA、TLR4 mRNA表达明显升高(均为P<0.01);ELISA检测发现IL-10、TLR4蛋白表达升高,与对照组相比,差异均有统计学意义(均为P<0.05)。结论rno-miR-30b-5p对带有IL-10、TLR4基因片段3’UTR基因表达的调控作用可能不是通过预测到的位点起作用而是通过其他的调控结合位点发挥作用;rno-miR-30b-5p在EAU大鼠脾脏和淋巴结的下调表达导致IL-10和TLR4的表达水平升高,进而调控葡萄膜炎的发展。本研究为进一步探索microRNA在葡萄膜炎发生发展进程中的调控作用、治疗葡萄膜炎奠定了基础。 展开更多
