According to structure function and lightning damage of a cable car, a feasible method of lightning strike risk evaluation for a cable car was put forward based on the evaluation model and evaluation method in the standard IEC62305-2. According to the difference between common buildings and cable cars, problems of height non-uniformity of equivalent section caused by inclination of the cable car and diversity of lightning activity regularity caused by the large area were resolved, and expected annual average frequency of lightning strike was calculated using three dimensional graphic approach and regional lightning characteristic analysis. Based on different types of damage process and loss consequences, according to interception effect against lightning invasion of the lightning protection measures and the method of probability selection proposed in the standard, the probability of casu- alty caused by direct lightning strike in a cable car and a waiting area as well as probabilities of casualty caused by failure of electronic information systems were cal- culated. 展开更多

香蕉枯萎病是由尖孢镰刀菌古巴转化型(Fusarium oxysporum f. sp. cubense, Foc)引起的香蕉毁灭性土传病害,其中4号生理小种(Foc4)能感染几乎所有的香蕉品系,危害最严重。carS基因通过调控下游car结构基因参与调控镰刀菌类胡萝卜素的生... 香蕉枯萎病是由尖孢镰刀菌古巴转化型(Fusarium oxysporum f. sp. cubense, Foc)引起的香蕉毁灭性土传病害,其中4号生理小种(Foc4)能感染几乎所有的香蕉品系,危害最严重。carS基因通过调控下游car结构基因参与调控镰刀菌类胡萝卜素的生物合成,本研究克隆鉴定了Foc4carS基因(FOIG_05085),Foc4carS蛋白具有典型的RING-finger蛋白结构域。利用分割标记法(Split-marker PCR)获得Foc4carS基因的融合片段,同时构建含有Foc4carS基因sgRNA591序列的pUC-fFuCas9-HTBNLS-hph-Foc4carS基因编辑载体,通过PEG介导的原生质体转化获得该基因的敲除突变体、回补突变体以及基因编辑敲除体,并对敲除和回补突变体的生物学特性和致病力进行分析。结果显示:ΔFoc4carS突变体的菌落直径、产孢量和致病力等生物学表型与野生菌株Foc4无显著差异,而ΔFoc4carS突变体菌落颜色呈深橙色,Foc4carS基因的缺失影响了次生代谢产物类胡萝卜素的生物合成;基因编辑的ΔFoc4carS(HDR)突变体不论是再生筛选板还是继代后的PDA平板,其菌落均出现典型的深橙色,表明Foc4carS可作为内源报告基因,在香蕉枯萎菌Foc4中进行基因质粒型CRISPR/Cas9编辑可行。 展开更多

Mixed adenoneuroendocrine carcinoma(MANEC)is a malignant tumor with adenocarcinoma and neuroendocrine components,with≥30%of each component required.MANEC of the ampulla is rare.To the best of our knowledge,only 15 cases of MANEC of the ampulla have been reported in the English-language literature.Here,we report two cases of MANEC of the ampulla in two womenaged 43 and 60 years,which was confirmed by histology after pancreaticoduodenectomy.These tumors contained neuroendocrine and adenocarcinoma components.The neuroendocrine components were positive for chromogranin A(Cg A),synaptophysin(Syn)and CD56 by immunostaining.The adenocarcinoma components were negative for Cg A,Syn and CD56.Both cases were T3N0M0(StageⅢA).They survived for 15 and 20 mo after surgery,respectively.A brief discussion about the histopathological features,clinical behavior and treatment of MANEC of ampulla,and review of the relevant literature are presented. 展开更多

Finally, numerical simulations are carried out for the stability of a high speed passenger car operating on straight and large radius curved tracks. CAR-T细胞治疗后复发/难治性多发性骨髓瘤患者的临床特征

目的分析嵌合抗原受体T(chimeric antigen receptor,CAR-T)细胞治疗后复发/难治多发性骨髓瘤(relapsed/refractory multiple myeloma,RRMM)患者的临床特征,初步探索此类患者未来可能的治疗策略。方法以2020年1月至2024年6月在首都医科... 目的分析嵌合抗原受体T(chimeric antigen receptor,CAR-T)细胞治疗后复发/难治多发性骨髓瘤(relapsed/refractory multiple myeloma,RRMM)患者的临床特征,初步探索此类患者未来可能的治疗策略。方法以2020年1月至2024年6月在首都医科大学附属北京朝阳医院住院治疗的RRMM患者为研究对象,分析CAR-T细胞治疗后复发的临床特征及生存情况。结果共收集14例符合条件的RRMM患者,均接受靶向BCMA的CAR-T细胞治疗,中位无进展生存时间为17个月(范围:3~47个月)。疾病复发进展时,1例仅表现为髓外复发,2例继发浆细胞白血病,11例为髓内复发;7例检查细胞遗传学,其中4例患者存在1个高危基因,1例患者存在2个高危基因。首次挽救治疗中,常用药物包括达雷妥尤单抗(6例)、泊马度胺(5例)、卡非佐米(3例),3例患者入组临床试验。13例患者可评价疗效,总有效率为61.5%(8/13),其中完全缓解率为30.8%(4/13),非常好的部分缓解率为23.1%(3/13)。中位随访时间为15个月(范围:1~32个月),中位无进展生存时间为7个月。截至末次随访,死亡6例,中位总生存期为32个月;1例患者出现第二肿瘤,类型为肺癌。结论接受CAR-T细胞治疗后RRMM患者常见多类药物耐药,挽救治疗疗效及预后不佳。 展开更多

How to simulate interior aerodynamic noise accurately is an important question of a car interior noise reduction. The unsteady aerodynamic pressure on body surfaces is proved to be the key effect factor of car interior aerodynamic noise control in high frequency on high speed. In this paper, a detail statistical energy analysis (SEA) model is built. And the vibra-acoustic power inputs are loaded on the model for the valid result of car interior noise analysis. The model is the solid foundation for further optimization on car interior noise control. After the most sensitive subsystems for the power contribution to car interior noise are pointed by SEA comprehensive analysis, the sound pressure level of car interior aerodynamic noise can be reduced by improving their sound and damping characteristics. The further vehicle testing results show that it is available to improve the interior acoustic performance by using detailed SEA model, which comprised by more than 80 subsystems, with the unsteady aerodynamic pressure calculation on body surfaces and the materials improvement of sound/damping properties. It is able to acquire more than 2 dB reduction on the central frequency in the spectrum over 800 Hz. The proposed optimization method can be looked as a reference of car interior aerodynamic noise control by the detail SEA model integrated unsteady computational fluid dynamics (CFD) and sensitivity analysis of acoustic contribution. 展开更多

目的:评价靶向B细胞成熟抗原(B cell maturation antigen,BCMA)嵌合抗原受体T细胞(chimeric antigen receptor-T cell,CAR-T)治疗复发/难治性多发性骨髓瘤(relapsed/refractory multiple myeloma,R/R MM)的长期疗效和安全性。方法:回顾... 目的:评价靶向B细胞成熟抗原(B cell maturation antigen,BCMA)嵌合抗原受体T细胞(chimeric antigen receptor-T cell,CAR-T)治疗复发/难治性多发性骨髓瘤(relapsed/refractory multiple myeloma,R/R MM)的长期疗效和安全性。方法:回顾性分析2018年7月至2023年7月在南昌大学第一附属医院接受BCMA CAR-T细胞治疗20例R/R MM患者的临床资料,随访日期截至2023年12月31日。应用Kaplan-Meier生存分析评估患者总生存(overall survival,OS)率和无进展生存(progression-free survival,PFS)率,并统计相关不良反应。结果:20例R/R MM患者,既往中位治疗线数为3(2~6)线,客观缓解率(objective response rate,ORR)为75%,完全缓解(complete response,CR)率为50%;中位随访时间29个月,中位PFS为26个月。10例CR的患者中,5例在末次随访时仍处于缓解状态,缓解持续时间最短为6个月,最长48个月。亚组分析中,髓外浸润、17p缺失遗传学异常和肿瘤高负荷患者PFS显著更差(P<0.05)。细胞因子释放综合征(cytokine release syndrome,CRS)是CAR-T细胞治疗最常见的不良反应,发生率为90%,3~4级CRS的发生率为35%;远期不良反应少,未发生CAR-T细胞治疗相关死亡。结论:BCMA CAR-T细胞是当前R/R MM治疗的有效方案,不良反应可控。髓外浸润和肿瘤高负荷的患者治疗有效,但持久反应欠佳,如何进一步巩固和维持患者的疗效,值得进一步设计前瞻性的临床研究并探究其差异性。 展开更多

预后营养指数联合CAR、Alb对腹膜透析患者远期预后的预测价值

目的 评估预后营养指数(PNI)联合C反应蛋白/白蛋白比值(CAR)、血清白蛋白(Alb)对腹膜透析(PD)患者远期预后的预测价值。方法 分析纳入2019年1月至2024年1月在自贡市第一人民医院肾病内科接受PD治疗的终末期肾病患者102例,根据5年随访期... 目的 评估预后营养指数(PNI)联合C反应蛋白/白蛋白比值(CAR)、血清白蛋白(Alb)对腹膜透析(PD)患者远期预后的预测价值。方法 分析纳入2019年1月至2024年1月在自贡市第一人民医院肾病内科接受PD治疗的终末期肾病患者102例,根据5年随访期间PD患者生存结局分为死亡组(n=33)和生存组(n=69)。采用单因素及多因素Logistic回归分析影响PD患者远期预后的独立危险因素。绘制受试者工作特征(ROC)曲线评估PNI联合CAR、Alb对PD患者生存结局的预测价值。结果 与生存组相比,死亡组患者合并糖尿病、心血管疾病及腹膜炎并发症的比例较高,差异有统计学意义(χ^(2)=0.947,4.846,5.840);肾小球滤过率(eGFR)、PNI、Alb水平降低,差异有统计学意义(t=9.495,6.068,5.422);血清肌酐(SCR)、尿酸(UA)及CAR水平升高,差异有统计学意义(t=5.369,4.581,5.048),差异均有统计学意义(P<0.05)。Logistic回归分析显示,患者合并糖尿病、心血管疾病、CAR是PD患者死亡的独立危险因素(P<0.05),eGFR、PNI、Alb是PD患者死亡的独立保护因素(P<0.05)。ROC曲线分析显示,PNI、CAR、Alb联合预测的曲线下面积分别为0.928,显著高于单独预测(P<0.05)。结论 PNI、Alb水平降低及CAR升高是PD患者死亡的独立影响因素,三者均可为PD患者长期预后生存结局提供参考,且联合预测价值更高。 展开更多

Laboratory testing of a shuttle car canopy air curtain for respirable coal mine dust control 被引量:4

Canopy air curtain (CAC) technology has been developed by the National Institute for Occupational Safety and Health (NIOSH) for use on continuous miners and subsequently roof bolting machines in underground coal mines to protect operators of these machines from overexposure to respirable coal mine dust. The next logical progression is to develop a CAC for shuttle cars to protect operators from the same overexposures. NIOSH awarded a contract to Marshall University and J.H. Fletcher to develop the shuttle car CAC. NIOSH conducted laboratory testing to determine the dust control efficiency of the shuttle car CAC. Testing was conducted on two different cab configurations: a center drive similar to that on a Joy 10SC32AA cab model and an end drive similar to that on a Joy 10SC32AB cab model. Three different ventilation velocities were tested-0.61, 2.0, 4.3 rrds (120, 400, and 850 fpm). The lowest, 0.61 m/s (120 fpm), represented the ventilation velocity encountered during loading by the continuous miner, while the 4.3 m/s (850 fpm) velocity represented ventilation velocity airflow over the shuttle car while tramming against ventilation airflow. Test results showed an average of the dust control efficiencies ranging from 74 to 83% for 0.61 m/s (120 fpm), 39%-43% for 2.0 m/s (400 fpm), and 6%-16% for 4.3 m/s (850 fpm). Incorporating an airflow spoiler to the shuttle car CAC design and placing the CAC so that it is located 22.86 cm (9 in.) forward of the operator improved the dust control efficiency to 51%-55% for 4.3 m/s (850 fpm) with minimal impact on dust control efficiencies for lower ventilation velocities. These laboratory tests demonstrate that the newly developed shuttle car CAC has the potential to successfully protect shuttle car operators from coal mine respirable dust overexposures. 展开更多

目的:探讨IL-7的诱导表达对靶向磷脂酰肌醇蛋白聚糖3(GPC3)嵌合抗原受体基因修饰T淋巴细胞(CAR-T细胞)的增殖和体外抗肿瘤活性的影响。方法:通过无缝克隆将GPC3 CAR序列片段插入GV400载体的Bam HⅠ/Eco RⅠ位置,构建第二代CAR慢病毒载体... 目的:探讨IL-7的诱导表达对靶向磷脂酰肌醇蛋白聚糖3(GPC3)嵌合抗原受体基因修饰T淋巴细胞(CAR-T细胞)的增殖和体外抗肿瘤活性的影响。方法:通过无缝克隆将GPC3 CAR序列片段插入GV400载体的Bam HⅠ/Eco RⅠ位置,构建第二代CAR慢病毒载体GPC3-BBZ及GPC3-BBZ-NFAT-IL-7,以293T细胞包装相应的慢病毒载体后,感染人T细胞制备CAR-T细胞。实验分为未转导T细胞(NT)组、GPC3-BBZ CAR-T细胞组、GPC3-BBZ-NFAT-IL-7 CAR-T细胞组。采用流式细胞术检测各组CAR-T细胞中CAR的表达水平,qPCR法检测经GPC3蛋白激活的CAR-T细胞中IL-7 m RNA的表达水平,细胞计数法检测CAR-T细胞在GPC3抗原刺激下的增殖能力,ELISA检测CAR-T细胞在受到肿瘤细胞刺激后IL-7、IFN-γ和TNF-α的分泌水平。应用实时细胞分析(RTCA)技术检测CAR-T细胞对人肝癌Huh-7细胞的杀伤作用。结果:成功构建慢病毒载体GPC3-BBZ和GPC3-BBZ-NFAT-IL-7,制备出靶向GPC3的CAR-T细胞。经GPC3抗原激活后,GPC3-BBZ-NFAT-IL-7 CAR-T细胞可有效表达IL-7 mRNA(P<0.01),其表现出更强的增殖能力(P<0.05)。与GPC3-BBZ CAR-T细胞相比,GPC3-BBZ-NFAT-IL-7 CAR-T细胞与GPC3阳性靶细胞Huh-7细胞共培养后,分泌更高水平的IL-7、IFN-γ和TNF-α(P<0.01或P<0.001)。RTCA结果显示,GPC3-BBZ-NFAT-IL-7 CAR-T细胞对GPC3阳性Huh-7细胞的杀伤活性显著高于GPC3-BBZ CAR-T细胞(P<0.05)。结论:成功制备可诱导表达IL-7的靶向GPC3的CAR-T细胞,IL-7的诱导表达增强靶向GPC3 CAR-T细胞的免疫活性,在体外展现出较强的肿瘤细胞杀伤能力。 展开更多

This paper proposes a novel collision post structure designed to improve the crashworthiness of subway cab cars.The structure provides two innovative features:1)a simpler connection between the post and the car roof,which gives a more reasonable load transfer path to reduce the stress concentration at the joint;and 2)a stiffness induction design that provides an ideal deformation model to protect the safe space of the cab cars.The novel collision post structure was evaluated with finite element analysis,and a prototype cab car was mechanically tested.The results demonstrate that the deformation response was stable and agreed well with the expected ideal mode.The maximum load was 874.17 kN and the responses remained well above the elastic design load of 334 kN as required by the design specification.In addition,there was no significant tearing failure during the whole test process.Therefore,the novel collision post structure proposed has met the requirements specified in new standard to improve the crashworthiness of subway cab cars.Finally,the energy absorption efficiency and light weight design highlights were also summarized and discussed. 展开更多

A K-nearest neighbor (K-NN) based nonparametric regression model was proposed to predict travel speed for Beijing expressway. By using the historical traffic data collected from the detectors in Beijing expressways, a specically designed database was developed via the processes including data filtering, wavelet analysis and clustering. The relativity based weighted Euclidean distance was used as the distance metric to identify the K groups of nearest data series. Then, a K-NN nonparametric regression model was built to predict the average travel speeds up to 6 min into the future. Several randomly selected travel speed data series, collected from the floating car data (FCD) system, were used to validate the model. The results indicate that using the FCD, the model can predict average travel speeds with an accuracy of above 90%, and hence is feasible and effective. 展开更多

Recently, new tech- nologies, such as cell phone tracking, license plate matching, automatic vehicle identifications and video detection, are employed for this purpose. In this study, the data collected by TRANSMIT readers, Bluetooth sensors, and INRIX are assessed by comparing each to the "ground truth" travel times collected by probe vehicles carrying GPS-based naviga- tion devices. Travel times of probe vehicles traveling on the study segment of 1-287 in New Jersey were collected in 2009. Statistical measures, such as standard deviation, average absolute speed error, and speed error bias, were used to make an in-depth analysis. The accuracy of each travel time estimation method is analyzed. The data collected by Bluetooth sensors and the TRANSMIT readers seem more consistent with the ground true data, and slightly outperform the data reported by 1NRIX. This study established a procedure for analyzing the accuracy of floating car data (FCD) collected by different technologies. 展开更多

目的分离纯化碧根果致敏原Car i 1,并对其结构进行表征鉴定。方法以新鲜碧根果果仁为原料,通过粉碎、脱脂、浸提、粗分级、凝胶过滤层析,对碧根果致敏原蛋白Car i 1进行分离纯化。结合十二烷基硫酸钠-聚丙烯酰胺凝胶电泳、液相色谱-串... 目的分离纯化碧根果致敏原Car i 1,并对其结构进行表征鉴定。方法以新鲜碧根果果仁为原料,通过粉碎、脱脂、浸提、粗分级、凝胶过滤层析,对碧根果致敏原蛋白Car i 1进行分离纯化。结合十二烷基硫酸钠-聚丙烯酰胺凝胶电泳、液相色谱-串联质谱法和免疫印迹法3种方法对Cari1进行鉴定,并通过圆二色谱仪与紫外分光光度计表征其二、三级结构。结果本方法纯化获得碧根果致敏原Cari1,单轮制备量可达5 mg以上,且纯度大于95%,蛋白质高级结构未被破坏,能够被全部3名碧根果过敏患者的血清准确识别。结论该纯化方法技术路线简单、设备要求低且单次制备量高,总得率可达65%,操作便捷,为碧根果致敏原Car i 1的相关研究奠定了物质基础。 展开更多

Using data from the Economic Advisory Center of the State Information Center(SIC), we examined the spatial patterns of car sales in China at the prefectural level in 2012. We first analyzed the spatial distributions of car sales of different kinds of automakers(foreign automakers, Sino-foreign joint automakers, and Chinese automakers), and then identified spatial clusters using the local Moran's indexes. Location quotient analysis was applied to examine the relative advantage of each type of automaker in the local markets. To explain the variations of car sales across cities, we collected several socioeconomic variables and conducted regression analyses. Further, factor analysis was used to extract independent variables to avoid the problem of multicollinearity. By incorporating a spatial lag or spatial error in the models, we calibrated our spatial regression models to address the spatial dependence problem. The analytical results show that car sales varied significantly across cities in China, and most of the cities with higher car sales were the developed cities. Different automakers exhibit diverse spatial patterns in terms of car sales volume, spatial clusters, and location quotients. The scale and incomes factor were extracted and verified as the two most significant and positive factors that shape the spatial distributions of car sales, and together with the spatial effect, explained most of the variations of car sales across cities. 展开更多

It is well recognized that one cause of chronic liver disease and hepatocellular carcinoma(HCC)is alcohol consumption.Research in Italy and the United States concludes that the most common cause of HCC(responsible for 32%to 45%of HCC)is alcohol.It has recently been shown that a significant relationship between alcohol intake,metabolic changes,and hepatitis virus infection does exist.Alcohol may be a factor in the development of HCC via direct(genotoxic)and indirect mechanisms(cirrhosis).There is only one way of diagnosing HCC,which is early identification through surveillance,when curative treatments become possible.After stopping alcohol intake the risk of liver cancer decreases by 6%to 7%a year,and an estimated time period of 23 years is also needed.Therefore,surveillance is also important in former drinkers and,in our opinion,independently from the presence of compensated cirrhosis.In cases of very early stage(VES)and early stage with portal hypertension,liver transplantation is the optimal option;and in cases of associated disease,percutaneous ethanol injections,radiofrequency and microwave ablation are the ideal treatments.Despite the possibility of detecting microvascular invasion with HR,several studies and some randomized controlled trials revealed that overall survival and DSF rates in patients with VES HCC are much the same after ablation and HR.Therefore,ablation can be regarded as a firstline choice for patients with VES HCC.It is important to emphasize that the choice of treatment should be weighed carefully in the context of a multidisciplinary cancer team. 展开更多

Hepatocellular carcinoma(HCC) is the fifth most common cancer and the second leading cause of cancer-related deaths worldwide. Although the prognosis of patients with HCC is generally poor, the5-year survival rate is > 70% if patients are diagnosed at an early stage. However, early diagnosis of HCC is complicated by the coexistence of inflammation and cirrhosis. Thus, novel biomarkers for the early diagnosis of HCC are required. Currently, the diagnosis of HCC without pathological correlation is achieved by analyzing serum α.fetoprotein levels combined with imaging techniques. Advances in genomics and proteomics platforms and biomarker assay techniques over the last decade have resulted in the identification of numerous novel biomarkers and have improved the diagnosis of HCC. The most promising biomarkers,such as glypican-3, osteopontin, Golgi protein-73 and nucleic acids including microRNAs, are most likely to become clinically validated in the near future. These biomarkers are not only useful for early diagnosis of HCC, but also provide insight into the mechanisms driving oncogenesis. In addition, such molecular insight creates the basis for the development of potentially more effective treatment strategies. Lung cancer is a common malignancy in the world; however symptomatic colonic metastasis from primary lung cancer is rare. A 64-year-old man was originally found poorly differentiated squamous cell carcinoma of right lung and received right lower lobectomy and lymph node dissection. Three years later, the patient presented to our emergency room with the symptom of upper abdominal pain and weight loss. Abdominal palpation and computed tomography scan of the abdomen revealed a large mass measuring 7.6 cm &#x000d7; 8.5 cm in the ascending colon. Colonoscopy and biopsy revealed poorly differentiated squamous cell carcinoma with similar morphological pattern to that of the previous lung cancer. Chemotherapy was given and the patient died 5 mo later. Lung cancer metastatic to the colon confers a poor prognosis: overall survival ranged from 5 wk to 1 year, with a median survival of 3 mo after the diagnosis of the colonic metastasis. 展开更多
