渤海湾盆地南堡凹陷2号构造带具有多洼、多层系供烃的特征。为了揭示其复杂的油气来源,开展色谱-质谱等地化测试,分析原油地化特征,将原油分为3类,并确定每类原油的来源。结果表明:(1)Ⅰ类原油主要分布于西构造带沙三段储层中,原油中C27... 渤海湾盆地南堡凹陷2号构造带具有多洼、多层系供烃的特征。为了揭示其复杂的油气来源,开展色谱-质谱等地化测试,分析原油地化特征,将原油分为3类,并确定每类原油的来源。结果表明:(1)Ⅰ类原油主要分布于西构造带沙三段储层中,原油中C27与C29规则甾烷相对含量接近,孕甾烷参数((孕甾烷+升孕甾烷)/规则甾烷)和甾烷异构化参数(C29甾烷20S/(20R+20S)、C29甾烷ββ/(αα+ββ))较高,具有水生生物和高等植物共同输入特征,原油成熟度高,来源于沙三段烃源岩;(2)Ⅱ类原油主要分布于西构造带东二段储层中,原油中C29规则甾烷含量具明显优势,孕甾烷参数和甾烷异构化参数均低,原油母质中高等植物具有明显优势,原油成熟度整体偏低,来源于沙一段—东三段烃源岩;(3)Ⅲ类原油主要分布于东构造带东三段和西构造带东一段和明化镇组储层中,原油中C29规则甾烷具有一定优势,孕甾烷参数较低,甾烷异构化参数较高,以高等植物输入为主,原油成熟度较高,为3套烃源岩的混合来源;(4)原油4-/1-MDBT和Ts/Tm指数具有沿断层向浅部层系减小的趋势,且原油甲基菲换算的镜质体反射率与下覆烃源岩镜质体反射率一致,表明研究区原油具有原地供烃的特点。本研究成果为研究区的浅层精细勘探及深层勘探提供理论指导。 展开更多

视网膜静脉阻塞患者血清FABP4,VWF和LCN-2水平表达与继发黄斑水肿程度及预后的关系研究

目的 探究视网膜静脉阻塞(retinal vein obstruction,RVO)患者血清脂肪细胞型脂肪酸结合蛋白(fatty acid-binding protein 4,FABP4)、血管性假血友病因子(von willebrand factor,VWF)、脂质运载蛋白2(lipocalin-2,LCN-2)水平表达与继发... 目的 探究视网膜静脉阻塞(retinal vein obstruction,RVO)患者血清脂肪细胞型脂肪酸结合蛋白(fatty acid-binding protein 4,FABP4)、血管性假血友病因子(von willebrand factor,VWF)、脂质运载蛋白2(lipocalin-2,LCN-2)水平表达与继发黄斑水肿(macular edema,ME)程度及预后的关系研究。方法 选取2021年1月~2023年8月来西北大学附属第一医院治疗的98例确诊的RVO继发ME患者为观察组,根据黄斑水肿程度分为重度组(n=46)、中度组(n=32)和轻度组(n=20),根据预后情况分为预后不良组(n=36)和预后良好组(n=62);另选取98例单纯RVO患者为对照组。比较各组血清FABP4,VWF,LCN-2水平;多因素Logistic回归分析RVO继发ME患者发生预后不良的影响因素;ROC曲线分析血清FABP4,VWF和LCN-2水平对RVO继发ME患者发生预后不良的预测价值。结果 与对照组相比,观察组血清FABP4(35.24±4.43 ng/ml vs 23.19±3.69 ng/ml),VWF(2.67±0.67 ng/ml vs1.48±0.38 ng/ml),LCN-2(112.53±12.74 mg/L vs 85.49±9.86 mg/L)水平明显升高,差异具有统计学意义(t=20.690,15.294,16.616,均P<0.05);随病情逐渐发展,轻、中、重度组血清FABP4(33.38±4.29 ng/ml,35.79±4.49 ng/ml,38.64±4.65 ng/ml),VWF(2.36±0.59 ng/ml,2.72±0.66 ng/ml,3.29±0.87 ng/ml),LCN-2(105.84±12.24 mg/L,114.61±12.88 mg/L,124.59±13.69 mg/L)水平逐渐升高,差异具有统计学意义(F=10.194,13.294,15.703,均P<0.05);与预后良好组相比,预后不良组血清FABP4(39.64±4.59 ng/ml vs 32.69±4.34 ng/ml),VWF(3.26±0.87ng/ml vs 2.33±0.55 ng/ml),LCN-2(124.56±13.42 mg/L vs 105.54±12.34 mg/L)水平明显升高,差异具有统计学意义(t=7.482,6.487,7.122,均P<0.05));血清FABP4(OR=2.236,95%CI:1.109~4.510),VWF(OR=2.069,95%CI:1.044~4.100),LCN-2(OR=1.875,95%CI:1.015~3.463)为RVO继发ME患者发生预后不良的独立危险因素(均P<0.05);血清FABP4,VWF,LCN-2水平预测患者发生预后不良的AUC(95%CI)分别为0.817(0.726~0.888),0.791(0.697~0.867)和0.798(0.705~0.872),三者联合预测的AUC为0.932(0.863~0.973),三者联合预测优于各因子单独预测(Z=2.034,2.375,2.385,P=0.042,0.018,0.017)。结论 RVO继发ME患者血清FABP4,VWF和LCN-2水平均上调,且均为患者发生预后不良的独立危险因素,三者联合检测对RVO继发ME患者发生预后不良具有更高的预测价值。 展开更多

AIM: To investigate the killing efficiency of a recombinant plasmid containing a thymidine kinase (TK) domain insert driven by the vascular endothelial growth factor receptor 2 (VEGFR2) promoter (KDR) on vascular endothelial cells.METHODS: The KDR-TK fragment was extracted from pBluescript Ⅱ KDR-TK plasmid by enzymatic digestion with Xho I and Sal I. The enhanced green fluorescence protein (EGFP) carrier was extracted from pEGFP by the same procedure. The KDR-TK was inserted into the pEGFP carrier to construct pEGFP-KDR-TK. Using ultrasound irradiation and microbubble, pEGFP-KDR-TK was transferred into human umbilical vein endothelial cells (HUVECs). The transient infection rate was estimated by green fluorescent protein (GFP) expression. Transfected HUVECs, non-transfected HUVECs, and HepG2 cells were cultured in the presence of different concentrations of ganciclovir (GCV), and the killing efficacy of HSV-TK/GCV was analyzed by 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyl tetrazolium bromide (MTT) assay. RESULTS: The recombinant pEGFP-KDR-TK was successfully constructed by inserting the KDR-TK fragment into the pEGFP carrier. Transfected HUVECs showed cytoplasmic green fluorescence, and the transient transfection rate was about 20.3%. Pools of G418-resistant cells exhibited a higher sensitivity to theprodrug/GCV compared to non-transfected HUVECs or non-transfected HepG2 cells, respectively. CONCLUSION: KDR promoter and the suicide gene/prodrug system mediated by diagnostic ultrasound combined with microbubble can significantly kill HUVECs. Such therapy may present a novel and attractive approach to target gene therapy on tumor vessels. 展开更多

目的:探究邻苯二甲酸二(2-乙基己基)酯(di-2-ethylhexyl phthalate,DEHP)诱导的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)的氧化损伤,以及白藜芦醇对该损伤的修复作用。方法:通过CCK8实验筛选DEHP的损伤浓度和... 目的:探究邻苯二甲酸二(2-乙基己基)酯(di-2-ethylhexyl phthalate,DEHP)诱导的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVEC)的氧化损伤,以及白藜芦醇对该损伤的修复作用。方法:通过CCK8实验筛选DEHP的损伤浓度和白藜芦醇的修复浓度;化学分光光度比色法检测HUVEC丙二醛和超氧化物歧化酶(SOD)的不同表达情况;活性氧荧光探针检测不同处理组HUVEC内活性氧水平;JC-1试剂盒检测不同组HUVEC线粒体膜电位;CCK8实验、Transwell实验、划痕愈合实验观察HUVEC增殖、迁移能力;细胞流式实验、蛋白质印迹实验观察HUVEC的凋亡情况和凋亡相关蛋白的表达。结果:80μmol/L DEHP作用HUVEC 24 h后,细胞增殖能力明显降低(P<0.01),40μmol/L白藜芦醇对细胞无明显毒性;损伤组细胞的丙二醛水平和SOD活性较对照组均明显上升(P均<0.01),修复组丙二醛水平和SOD活性较损伤组均明显下降(P均<0.05);损伤组细胞的活性氧表达较对照组明显上升(P<0.01),修复组较损伤组明显下降(P<0.01);损伤组细胞的线粒体膜电位较对照组明显下降(P<0.01),修复组较损伤组明显上升(P<0.01);损伤组细胞的增殖、迁移、成管能力较对照组明显下降(P均<0.05),修复组较损伤组明显上升(P均<0.05);损伤组细胞的凋亡水平较对照组明显上升(P<0.05),修复组较损伤组明显下降(P<0.05)。结论:DEHP能够通过氧化应激途径抑制HUVEC的增殖、迁移和成管能力,并诱导其凋亡,而白藜芦醇可以有效修复该种损伤。 展开更多

目的通过自制小鼠尾静脉留置针,为小鼠^(13)N-NH_(3)·H_(2)O心肌灌注显像提供实用、快捷、成功率高的实验工具,并总结归纳出一套流畅、高效的实验流程。方法实验组和对照组分别使用本实验室自制的鼠尾静脉留置针及人外周浅静脉一... 目的通过自制小鼠尾静脉留置针,为小鼠^(13)N-NH_(3)·H_(2)O心肌灌注显像提供实用、快捷、成功率高的实验工具,并总结归纳出一套流畅、高效的实验流程。方法实验组和对照组分别使用本实验室自制的鼠尾静脉留置针及人外周浅静脉一次性使用静脉留置针(洁瑞26G)为小鼠行尾静脉留置针穿刺,经留置针推注^(13)N-NH_(3)·H_(2)O,配合Micro-PET动态采集,记录图像采集的成功率及图像质量并评价两组注射的优缺点。结果两组实验在穿刺成功率、穿刺时间、药物残留量、图像质量及小鼠尾部放射性药物残留量5方面差异均具有统计学意义(P<0.05)。综合比较,自制留置针制作及操作简便、穿刺成功率高、对鼠尾静脉损伤小,且在图像采集成功率及质量方面均占优势。结论自制鼠尾静脉留置针联合优化的实验流程,能显著提高鼠尾静脉穿刺成功率和图像质量,很好地应用于小鼠^(13)N-NH_(3)·H_(2)O心肌灌注Micro-PET显像中。 展开更多

目的 探究急性下肢深静脉血栓形成(DVT)患者置管溶栓前后D-二聚体(D-D)、凝血酶原片段1+2(F1+2)、P选择素(P-selectin)变化及临床意义。方法 选取2020年3月至2022年3月云南省第三人民医院186例急性DVT患者作为研究对象,均行置管溶栓术,... 目的 探究急性下肢深静脉血栓形成(DVT)患者置管溶栓前后D-二聚体(D-D)、凝血酶原片段1+2(F1+2)、P选择素(P-selectin)变化及临床意义。方法 选取2020年3月至2022年3月云南省第三人民医院186例急性DVT患者作为研究对象,均行置管溶栓术,术后12个月以门诊形式进行随访,4例失访,共182例完成术后随访。根据术后12个月是否发生血栓后综合征(PTS)分为PTS组(n=27)、非PTS组(n=155),比较2组一般资料及溶栓前后血浆D-D、F1+2、P-selectin表达,Logistic分析PTS发生影响因素,受试者工作特征曲线(ROC)、曲线下面积(AUC)分析血浆D-D、F1+2、P-selectin预测PTS发生价值,采用相对危险度(RR)分析不同血浆D-D、F1+2、P-selectin表达对PTS的影响。结果 PTS组年龄、BMI、静脉通畅评分及溶栓后1周、1个月血浆D-D、F1+2、P-selectin表达高于非PTS组(P <0.05);Logistic显示:BMI及溶栓后1周、1个月血浆D-D、F1+2、P-selectin是急性DVT患者发生PTS的影响因素(P <0.05);ROC曲线,溶栓后1个月血浆D-D、F1+2、P-selectin联合预测PTS效能明显优于溶栓后1周D-D、F1+2、P-selectin联合预测效能;溶栓后1个月血浆D-D、F1+2、P-selectin高表达患者发生PTS风险是低表达的4.211、2.550、3.189倍。结论 急性DVT患者置管溶栓后血浆D-D、F1+2、P-selectin表达升高,其联合预测患者发生PTS具有一定预测效能。 展开更多

目的探讨创伤性骨折(TF)患者血清微小RNA-374a-5p(miR-374a-5p)、骨形态发生蛋白2(BMP2)水平与凝血功能指标及术后深静脉血栓(DVT)形成的关系。方法选择2023年1-12月在该院诊治的204例TF患者,根据术后是否发生DVT将患者分为非DVT组和DV... 目的探讨创伤性骨折(TF)患者血清微小RNA-374a-5p(miR-374a-5p)、骨形态发生蛋白2(BMP2)水平与凝血功能指标及术后深静脉血栓(DVT)形成的关系。方法选择2023年1-12月在该院诊治的204例TF患者,根据术后是否发生DVT将患者分为非DVT组和DVT组。采用酶联免疫吸附试验(ELISA)检测血清BMP2水平,采用实时荧光定量PCR(qRT-PCR)检测血清miR-374a-5p水平,采用全自动凝血仪及配套试剂检测抗凝血酶Ⅲ(ATⅢ)、纤维蛋白原(Fib)、凝血酶原时间(PT)、活化部分凝血活酶时间(APTT)、凝血酶时间(TT)、D-二聚体(D-D)。通过多因素Logistic回归分析TF患者术后发生DVT的影响因素;在StarBase网站上分析miR-374a-5p与BMP2的关系;采用Pearson相关分析miR-374a-5p与BMP2水平的相关性及二者与凝血功能指标的相关性;采用受试者工作特征(ROC)曲线分析miR-374a-5p与BMP2水平对TF患者术后发生DVT的预测价值,比较曲线下面积(AUC)的差异。结果DVT组重度骨折的比例及miR-374a-5p、D-D、Fib水平均高于非DVT组(P<0.05),BMP2、ATⅢ水平低于非DVT组(P<0.05)。miR-374a-5p水平与ATⅢ水平呈负相关(r=-0.414,P<0.05),与D-D、Fib水平均呈正相关(r=0.424、0.427,P<0.05)。BMP2水平与ATⅢ水平呈正相关(r=0.423,P<0.05),与D-D、Fib水平均呈负相关(r=-0.416、-0.441,P<0.05)。miR-374a-5p、D-D水平升高是TF患者术后发生DVT的独立危险因素(P<0.05),BMP2、ATⅢ水平升高是TF患者术后发生DVT的独立保护因素(P<0.05)。miR-374a-5p、BMP2二者之间存在结合位点。DVT组血清miR-374a-5p水平与BMP2水平呈负相关(r=-0.449,P<0.001)。血清miR-374a-5p、BMP2单项及联合检测预测TF患者术后发生DVT的AUC分别为0.830(95%CI:0.771~0.879)、0.805(95%CI:0.744~0.857)、0.943(95%CI:0.902~0.971),2项联合检测预测的AUC大于miR-374a-5p、BMP2单项检测预测的AUC(Z=4.511、5.039,均P<0.001)。结论TF术后发生DVT的患者中血清miR-374a-5p、BMP2水平异常,2项指标之间存在结合位点,且均与凝血功能指标ATⅢ、D-D、Fib相关;miR-374a-5p、BMP2联合检测对TF患者术后发生DVT具有较高的预测价值。 展开更多

This study was carried out explore the mechanism underlying the inhibition of platelet activation by kelp fucoidans in deep venous thrombosis(DVT)mouse.In the control and sham mice,the walls of deep vein were regular and smooth with intact intima,myometrium and adventitia.The blood vessel was wrapped with the tissue and there was no thrombosis in the lumen.In the DVT model,the wall was uneven with thicken intima,myometrium and adventitia.After treated with fucoidans LF1 and LF2,the thrombus was dissolved and the blood vessel was recanalized.Compared with the control group,the ROS content,ET-1 and VWF content and the expression of PKC-βand NF-κB in the model were significantly higher(P<0.05);these levels were significantly reduced following treatments with LF2 and LF1.Compared with H_(2)O_(2)treated-HUVECs,combined LF1 and LF2 treatment resulted in significant decrease in the expression of PKC-β,NF-κB,VWF and TM protein(P<0.05).It is clear that LF1 and LF2 reduces DVT-induced ET-1,VWF and TM expressions and production of ROS,thus inhibiting the activation of PKC-β/NF-κB signal pathway and the activation of coagulation system and ultimately reducing the formation of venous thrombus. 展开更多

Changes of Ca^2+ activated potassium channels and cellular proliferation in autogenous vein grafts

Methods: Contraction of venous ring was measured by means of perfusion in vitro. The intimal rabbits proliferation of vascular and proliferation of cultured smooth muscle cells(vascular smooth muscle cells, VSMCs)were observed by the means of computerised image analysis and MTT method respectively. Furthermore, whole cell mode of patch clamp was used to record KCa of VSMCs isolated from autogenous vein grafts. Results: One week after transplantation there were no significant differences of contraction and intimal relative thickness between autogenous vein grafts and control. Contraction and intimal relative thickness of autogenous vein graft were significantly increased 2 weeks after transplantation (P<0.05, n=8 vs control), and they was more enhanced 4 weeks after vein transplantation (P<0.01, n=8 vs control).TEA(blocker of Ca2+ activated potassium channels)increased MTT A490 nm value of VSMCs from femoral vein in a dose dependent manner(P<0.05, n=8). KCa current density was significantly attenuated in VSMCs from autogenous vein grafts (1-4) week after transplantation(P<0.05, n=5).Conclusion: KCa is inhibited in autogenous vein graft, which account for vasospasm and intimal proliferation. 展开更多

目的探讨内镜下组织胶“三明治”联合套扎术治疗2型胃底静脉曲张(GOV2)的临床疗效。方法选取2020年5月至2021年5月赣州市第五人民医院接诊的50例GOV2患者作为研究对象,根据治疗方法不同分为研究组与对照组,每组25例。研究组采用内镜下... 目的探讨内镜下组织胶“三明治”联合套扎术治疗2型胃底静脉曲张(GOV2)的临床疗效。方法选取2020年5月至2021年5月赣州市第五人民医院接诊的50例GOV2患者作为研究对象,根据治疗方法不同分为研究组与对照组,每组25例。研究组采用内镜下组织胶“三明治”联合套扎术,对照组采用内镜下组织胶+聚桂醇“三明治”夹心法。比较两组临床疗效、治疗前后门静脉血流动力学指标、术后并发症发生情况。结果两组治疗总有效率比较差异无统计学意义。治疗后,两组门静脉内径(PVD)、门静脉血流量(PBF)、胃左静脉(LGV)及脾静脉内径(SVD)均大于治疗前,门静脉血流速度(PVV)快于治疗前,门静脉压(PVP)低于治疗前,且研究组PVD、PBF、LGV及SVD均大于对照组,PVV快于对照组,PVP低于对照组,差异有统计学意义(P<0.05)。研究组术后并发症发生率低于对照组,差异有统计学意义(P<0.05)。结论内镜下组织胶“三明治”联合套扎术可有效改善GOV2患者血流动力学变化,减少术后并发症发生,值得临床推广应用。 展开更多

目的探讨2型糖尿病老年患者发生下肢深静脉血栓(LDVT)的危险因素及应对策略。方法收集2021年1月至2022年12月首都医科大学宣武医院收治的69例2型糖尿病老年LDVT患者的临床资料为LDVT组,根据LDVT组患者性别与年龄,采用倾向匹配法按1:1比... 目的探讨2型糖尿病老年患者发生下肢深静脉血栓(LDVT)的危险因素及应对策略。方法收集2021年1月至2022年12月首都医科大学宣武医院收治的69例2型糖尿病老年LDVT患者的临床资料为LDVT组,根据LDVT组患者性别与年龄,采用倾向匹配法按1:1比例选取同期收治的69例无LDVT患者为NLDVT组。收集两组患者的一般资料、不良生活习惯、相关合并症、相关临床指标,分析2型糖尿病老年患者发生LDVT的影响因素,据此制订应对策略。结果LDVT组患者下肢手术和/或创伤史、未规范用药、合并下肢周围血管病变、长期卧床比例及总胆固醇(TC)、糖化血红蛋白(HbA1c)、D-二聚体(D-D)水平均高于NLDVT组患者,差异均有统计学意义(P<0.05)。多因素分析结果显示,下肢手术和/或创伤史、未规范用药、合并下肢周围血管病变、长期卧床及TC≥6.64 mmol/L、Hb A1c≥10.31%、D-D≥523.67μg/L均为2型糖尿病老年患者发生LDVT的危险因素(OR>1,P<0.05)。结论未规范用药、下肢手术和/或创伤史、合并下肢周围血管病变、长期卧床、TC≥6.64 mmol/L、HbA1c≥10.31%、D-D≥523.67μg/L均是2型糖尿病老年患者发生LDVT的危险因素,2型糖尿病老年患者需要重视遵医规范化治疗,通过良好的控制血糖、血脂、血压水平实现改善凝血功能和预防LDVT的目标,叮嘱患者定期到院检查,完善相关指标的检测,从而降低2型糖尿病患者LDVT的发生风险。 展开更多

内源性H_(2)S、TAT、sPECAM-1对老年髋部骨折围术期下肢深静脉血栓形成的预测价值研究

目的探讨内源性硫化氢(endogenous hydrogen sulfidem,H_(2)S)、血浆凝血酶-抗凝血酶复合物(thrombin/antithrombin complex,TAT)及可溶性血小板内皮细胞黏附分子-1(soluble platelet endothelial cell adhesion molecule 1,sPECAM-1)... 目的探讨内源性硫化氢(endogenous hydrogen sulfidem,H_(2)S)、血浆凝血酶-抗凝血酶复合物(thrombin/antithrombin complex,TAT)及可溶性血小板内皮细胞黏附分子-1(soluble platelet endothelial cell adhesion molecule 1,sPECAM-1)对老年髋部骨折围术期下肢深静脉血栓形成(deep vein thrombosis,DVT)的预测价值。方法选取2018年11月至2019年7月北京积水潭医院急诊科住院接受手术治疗的新发老年髋部骨折患者187例,随机抽取120例患者(建模集),采用多因素logistic回归方程分析建模集患者DVT的影响因素,并建立预测模型;将剩余67例患者纳入验证集,进行预测模型的内部验证。结果建模集120例患者中男39例(32.50%),女81例(67.50%);平均年龄(78.6±7.1)岁;BMI为(22.5±4.2)kg/m^(2)。多因素logisitic回归分析显示,BMI、发病至就诊的时间及TAT越高,H_(2)S及sPECAM-1越低的患者,越容易发生DVT(P<0.05)。建立的DVT风险预测模型为:logit(P)=0.130×BMI+0.010×发病至就诊的时间-0.098×H_(2)S+0.039×TAT-0.322×sPECAM-1-3.581。预测模型的ROC曲线的AUC为0.844(95%CI:0.774~0.915,P=0.000)。应用验证集数据对模型进行内部验证结果显示验证模型的ROC曲线的AUC为0.816(95%CI:0.713~0.919,P=0.000)。结论基于BMI、受伤至入院时间、TAT、H_(2)S和sPECAM-1等因素建立的模型对于预测老年髋部骨折患者围术期DVT的发生具有良好的筛查能力。 展开更多

We first report discovery of the spinel-garnet-orthopyroxene granulite with pure CO2 fluid inclusions from the Fuyun region of the late Paleozoic Altay orogenic belt in Central Asia, NW China. The rock is characterized by an assemblage of garnet, orthopyroxene, spinel, cordierite, biotite, plagioclase and quartz. Symplectites of orthopyroxene and spinel, and orthopyroxene and cordierite indicate decompression under UHT conditions. Mineral chemistry shows that the or-thopyroxenes have high XMg and A12O3 contents (up to 9.23 wt%). Biotites are enriched in TiO2 and XMg and are stable under granulite facies conditions. The garnet and quartz from the rock carry monophase fluid inclusions which show peak melting temperatures of around -56.7℃, indicating a pure CO2 species being presented during the ultrahigh-T metamorphism in the Altay orogenic belt. The inclusions homogenize into a liquid phase at temperatures around 15.3-23.8℃ translating into CO2 densities of the order of 0.86-0.88 g/cm3. Based on preliminary mineral paragenesis, reaction textures and petrogenetic grid considerations, we infer that the rock was subjected to UHT conditions. The CO2-rich fluids were trapped during exhumation along a clockwise P-T path following isothermal decompression under UHT conditions. 展开更多

Leaf veins play an important role in plant growth and development,and the bundle sheath(BS)is believed to greatly improve the photosynthetic efficiency of C_(4) plants.The OBV mutation in tomato(Solanum lycopersicum)results in dark veins and has been used widely in processing tomato varieties.However,physiological performance has difficulty explaining fitness in production.In this study,we confirmed that this mutation was caused by both the increased chlorophyll content and the absence of bundle sheath extension(BSE)in the veins.Using genome-wide association analysis and map-based cloning,we revealed that OBV encoded a C_(2)H_(2) L domain class transcription factor.It was localized in the nucleus and presented cell type-specific gene expression in the leaf veins.Furthermore,we verified the gene function by generating CRISPR/Cas9 knockout and overexpression mutants of the tomato gene.RNA sequencing analysis revealed that OBV was involved in regulating chloroplast development and photosynthesis,which greatly supported the change in chlorophyll content by mutation.Taken together,these findings demonstrated that OBV affected the growth and development of tomato by regulating chloroplast development in leaf veins.This study also provides a solid foundation to further decipher the mechanism of BSEs and to understand the evolution of photosynthesis in land plants. 展开更多

Four different varieties of charnockitic rocks, with different modes of formation, from the Mesoproterozoic Natal belt are described and new C isotope data presented. Excellent coastal exposures in a number of quarries and river sections make this part of the Natal belt a good location for observing charnockitic field relationships. Whereas there has been much debate on genesis of charnockites and the use of the term charnockite, it is generally recognized that the stabilization of orthopyroxene relative to biotite in granitoid rocks is a function of low aH2O (-- high CO2), high temperature, and composition (especially Fe/(Fe +Mg)). From the Natal belt exposures, it is evident that syn-emplacement, magmatic crystallization of charnockite can arise from mantle-derived differentiated melts that are inherently hot and dry (as in the Oribi Gorge granites and Munster enderbite), as well as from wet granitic melts that have been affected through interaction with dry country rock to produce localized charnockitic marginal facies in plutons (as in the Portobello Granite). Two varieties of post-emplacement sub-solidus charnock- ites are also evident. These include charnockitic aureoles developed in leucocratic, biotite, garnet granite adjacent to cross-cutting enderbitic veins that are attributed to metamorphic-metasomatic processes (as in the Nicholson's Point granite, a part of the Margate Granite Suite), as well as nebulous, patchy charnocki- tic veins in the Margate Granite that are attributed to anatectic metamorphic processes under low-aH2O fluid conditions during a metamorphic event. These varieties of charnockite show that the required physical conditions of their genesis can be achieved through a number of geological processes, providing some important implications for the classification of charnockites, and for the interpretation of charnock- ite genesis in areas where poor exposure obscures field relationships. 展开更多

The Wulasigou Cu-Pb-Zn deposit,located 15 km northwest of Altay city in Xinjiang,is one of many Cu-Pb-Zn polymetallic deposits in the Devonian Kelan volcanic-sedimentary basin in southern Altaids.Two mineralizing periods can be distinguished:the marine volcanic sedimentary PbZn mineralization period,and the metamorphic hydrothermal Cu mineralization period,which is further divided into an early bedded foliated quartz vein stage(Q1) and a late sulfide-quartz vein stage(Q2) crosscutting the foliation.Four types of fluid inclusions were recognized in the Q1 and Q2 quartz from the east orebodies of the Wulasigou deposit:H2O-CO2 inclusions,carbonic fluid inclusions,aqueous fluid inclusions,and daughter mineral-bearing fluid inclusions.Microthermometric studies show that solid CO2 melting temperatures(T(m,CO2)) of H2O-CO2 inclusions in Ql are from-62.3℃ to-58.5C,clathrate melting temperatures(T(m,clath)l) are from 0.5 C to 7.5 C,partial homogenization temperatures(T(h,CO2)) vary from 3.3℃ to 25.9℃(to liquid),and the total homogenization temperatures(T(h,tot)) vary from 285℃ to 378℃,with the salinities being 4.9%-15.1%NaCl eqv.and the CO2-phase densities being 0.50-0.86 g/cm-3.H2O-CO2 inclusions in Q2 have T(m,CO2) from-61.9℃ to-56.9℃,T(m,clath)from 1.3℃ to 9.5℃,T(h,CO2) from 3.4℃ to 28.7℃(to liquid),and T(h,tot) from 242℃ to 388℃,with the salinities being 1.0%-15.5%NaCl eqv.and the CO2-phase densities being 0.48-0.89 g/cm-3.The minimum trapping pressures of fluid inclusions in Q1 and Q2 are estimated to be 260-360 MPa and180-370 MPa,respectively.The δ-(34)S values of pyrite from the volcanic sedimentary period vary from2.3‰ to 2.8‰(CDT),and those from the sulfide-quartz veins fall in a narrow range of-1.9‰ to 2.6‰(CDT).The δD values of fluid inclusions in Q2 range from-121.0‰ to-100.8‰(SMOW),and theδ-(18)O(H2O) values calculated from δ-(18)O of quartz range from-0.2‰ to 8.3‰(SMOW).The δD-δ-(18)O(H2O)data are close to the magmatic and metamorphic fields.The fluid inclusion and stable isotope data documented in this study indicate that the vein-type copper mineralization in the Wulasigou Pb-Zn-Cu deposit took place in an orogenic-metamorphic enviroment. 展开更多

Objective:This study aimed to analyze the effects of transcatheter arterial embolization(TAE)combined with portal venous embolization(PVE)on the expression of MMP-2 in residual VX2 liver tumor tissues,liver function and non-embolic lobe regeneration.Methods:A total of 72 rabbits were randomly divided into Sham,TAE,PVE and TAE+PVE groups(n=18/group).The tissue samples from each group were taken at 6 h,3 days and 7 days after interventional operation,respectively.MMP-2 expression was detected by immunohistochemistry,Real-time PCR,and Western-blotting.The main indicators(such as AST,ATL,and TBIL)of liver function and the volume of non-embolized hepatic lobes were measured in each group after operation.One-way ANOVA and Kruskal-wallis method were used for statistical analysis.Results:The expression of MMP-2 mRNA and protein remained the highest in the Sham group,and the expression of MMP-2 mRNA and protein in TAE,PVE and TAE+PVE groups were successively increased,and the expression of MMP-2 in TAE+PVE group was always significantly higher than TAE group.The AST and ALT levels in each group on day 7 after operation showed a significant declination,and all groups have recovered to the preoperative baseline level and TBIL has a slight fluctuation in each group after operation with no statistical difference.On day 7 after operation,the increasing volume of non-embolized liver lobes in TAE+PVE group showed a more significant effect than those in PVE group,but there was no statistical significance(37.62±1.54 ml VS 36.18±1.15 ml,P=0.881),and its volume was significantly higher than those in the sham group(27.03±1.11 ml).Conclusion:TAE+PVE is considered to be an efficient and safe approach for treating rabbit VX2 liver transplantation tumor,but the expression of MMP-2 increased fastest after TAE+PVE,which might promote tumor cell invasion and metastasis. 展开更多
