Objective:To evaluate the supplementation effects of vitamin E,vitamin C,superoxide dismutase(SOD),and catalase to diluents on bull cryopreserved epididymal sperm.Methods:Sperm were retrieved from 20 bull testes and w...Objective:To evaluate the supplementation effects of vitamin E,vitamin C,superoxide dismutase(SOD),and catalase to diluents on bull cryopreserved epididymal sperm.Methods:Sperm were retrieved from 20 bull testes and were then supplemented with 0.1 mM vitamin E,5.0 mM vitamin C,100.0 IU/mL SOD,and 100.0μg/mL catalase alone,or in a combination.The control treatment contained no addition.After supplementation,samples were frozen and stored in liquid nitrogen.The sperm parameters including motility,progressive motility,viability,acrosome integrity,plasma membrane integrity,kinematics and DNA damage were evaluated following the thawing process.Results:Vitamin E alone significantly increased the parameters of acrosome and membrane integrity compared to the control treatment(P<0.05).While compared to the control treatment,vitamin C had no improvement effect on sperm characteristics except for membrane integrity.Treatment of vitamin E+vitamin C had a significant improvement in total motility,progressive motility,viability,membrane and acrosome integrity compared to the control and other treatments(P<0.05).Compared to the control treatment,addition of SOD or catalase alone significantly improved the percentages of total motility,progressive motility,viability,membrane and acrosome integrity(P<0.05).Furthermore,SOD+catalase significantly increased total motility,progressive motility,viability,acrosome and membrane integrity characteristics compared to the catalase treatment(P<0.05).Vitamin E alone,vitamin E+vitamin C,and SOD in diluents decreased DNA damages and thereby improved the rate of intact sperm heads.Conclusions:Addition of 100.0 IU/mL SOD alone and 0.1 mM vitamin E+5.0 mM vitamin C,and also 5.0 mM vitamin C+100μg/mL catalase in a combination improves the quality of cryopreserved bull epididymal sperm and could be used for cryopreservation.展开更多
Evidences show that the storage period greatly affects the quality of yogurt.In this study,three types of yogurt:control yogurt(CY),non-hydrolyzed potato powder yogurt(PPY)and enzymatically hydrolyzed potato powder yo...Evidences show that the storage period greatly affects the quality of yogurt.In this study,three types of yogurt:control yogurt(CY),non-hydrolyzed potato powder yogurt(PPY)and enzymatically hydrolyzed potato powder yogurt(EHPPY)were prepared at 42℃ for 5 h and stored for 28 days at 4℃.The yogurts were evaluated for quality characteristics at different storage periods.Negligible differences in pH values,titratable acidities and viable counts were detected in all three types of yogurt during storage.However,compared to other yogurts,EHPPY exhibited desirable water holding capacity,throughout the storage period.Apart from this,sensory properties and antioxidant activities(2-diphenyl-1-picryl-hydrazyl(DPPH)free radical scavenging activity and ferric reducing antioxidant power(FRAP))of EHPPY were also significantly improved during the storage period.Furthermore,the storage(G’)and loss(G”)modulus of PPY,EHPPY were lower than CY at 4℃ while a hysteresis loop was shown by all yogurts at the temperature range of 4-50℃ indicating higher G’(elasticity)than G”(viscosity).Based on our findings,EHPP could be an important functional ingredient in improving the quality and storage stability of yogurt for its production at an industrial level.展开更多

As many of them are known to be antioxidants and can prevent several chronic and degenerative diseases in humans, they are a part of a healthy diet. However, these antioxidants can act as prooxidants under high phenolic concentration, high pH, or in the presence of transition metal ions such as Cu<sup>2+</sup> or Fe<sup>3+</sup>, producing reactive oxygen species (ROS) including hydroxyl radicals resulting in oxidative stress and cell toxicity. While this can lead to pathogenesis including the development of various types of cancers, elevated levels of ROS are beneficial to kill malignant cells and foodborne pathogens to improve food safety. Thus, the dual nature of phenolic compounds allows them to act as antioxidants and prooxidants. Abiotic stresses,including drought,have been found to affect the growth and medicinal quality of numerous herbs.The proposed study aims to study the effects of different drought regimes on the metabolic profile,growth...Abiotic stresses,including drought,have been found to affect the growth and medicinal quality of numerous herbs.The proposed study aims to study the effects of different drought regimes on the metabolic profile,growth,ecophysiology,cellular antioxidants,and antioxidant potential of Nigella sativa(Black cumin)leaf.Forty-day-old seedlings of N.sativa were exposed to three regimes of drought(control,moderate and high)for a week.UPLCMS/MS metabolic profile of the leaf reveals the presence of more than a hundred metabolites belonging to anthocyanins,chalcones,dihydro flavonoids,flavonoids,flavanols,flavones,flavonoid carbonoside,isoflavones,etc.Drought was found to alter the contents of identified metabolites.Drought stress-induced oxidative stress and increased production of hydrogen peroxide and superoxide anions.Physiological changes,activities of antioxidant enzymes,contents of antioxidants,and proline were significantly high under drought to protect against the low water regimes.Furthermore,stressed leaf extract had higher antioxidant potential.Thus,N.sativa leaf bears multiple metabolic pathways and can tolerate a higher degree of drought or osmotic stress.展开更多

Fishes were sampled, acclimatized and kept treated or untreated with lead (9.43 mg/L) under observation for 4 days. At day 4, respiratory burst activity, lipid peroxidation activity and superoxide dismutase level increased significantly in treated group as compared to the control. On the contrary, catalase, glutathione S-transferase, glutathione peroxidase, glutathione reductase and reduced glutathione activity decreased on treatment with lead. These results suggest that heavy metal like lead induces oxidative stress, influences the antioxidant defense system and may lead to physiological disorders rendering the health and survival of exposed fish to a compromised state.展开更多

This work was undertaken at Saint Peter TB specialized hospital and TekleHaimanot health center from March 2012 to May 2013.Aim: To determine changes in Non-Enzymatic Antioxidants and level of oxidative stress of tuberculosis Patients before and after taking anti tuberculosis treatment.Materials and Methods: In this comparative cross sectional study, a total of 210 individuals including: newly diagnosed TB patients as group-I (n = 70), TB patients who completed treatment as group-II (n = 70), and healthy volunteers as group-III (n = 70) were enrolled. Different methods were used to determine the parameters;vit-C (HPLC method), lipid peroxidation (thiobarbuituric acid method), and bilirubin (Colorimetric assay). Results: Vitamin-C (Vit-C) and of group-I showed a significant reduction (p < 0.001) as compared with both group-II and group-III whereas Malondialdehyde (MDA) level was increased. However, the total and direct bilirubin was not different among the groups. In group-III, there was a positive correlation between BMI and serum Vit-C (r = -0.305, p = 0.010). Vit-C showed a negative correlation with serum MDA in all the groups with values (r = -0.265, p = 0.027), (r = -0.389, p = 0.001) and (r = -0.375, p = 0.001) for group-I, group-II and group-III respectively. In addition to this Vit-C was negatively correlated with serum UA (r = -0.285, p = 0.017) in group-I. Conclusion: The findings of the current study suggest that the amount of Vit-C in the newly diagnosed TB patients and those who finished their treatment is much lower than the healthy volunteers. In contrast to this, the MDA value was significantly higher both in the newly diagnosed TB patients and TB patients who completed treatment than in healthy volunteers suggesting higher degree of oxidative stress.展开更多

Phlorizin(PHL)is a natural compound with strong antioxidant properties mainly found in apples.In this paper,the interaction mechanism of PHL with pepsin and trypsin was comparatively evaluated by computer simulation,f...Phlorizin(PHL)is a natural compound with strong antioxidant properties mainly found in apples.In this paper,the interaction mechanism of PHL with pepsin and trypsin was comparatively evaluated by computer simulation,fluorescence spectra,circular dichroism(CD),and Fourier transform infrared(FT-IR)spectra at a molecular level.Fluorescence spectra showed that PHL quenches the pepsin/trypsin by static quenching.Thermodynamic parameters indicated that PHL binds to pepsin mainly through hydrogen bonds and van der Waals forces,and that of trypsin was electrostatic forces.The ground state complexes PHL and protease have a moderate affinity of 105 L/mol PHL binds more strongly to trypsin than to pepsin.CD and FT-IR spectra results showed that pepsin/trypsin decreased theβ-sheet content and slightly changed its secondary structure upon PHL.These experimental results are mutually verified with the predicted computer-aid simulation results.Upon PHL and trypsin binding,the antioxidant capacity of PHL was elevated.Nevertheless,the antioxidant capacity of PHL was decreased after binding to pepsin.This work elucidates the binding of PHL binding mechanisms to pepsin/trypsin and provides useful information for the digestion of PHL to improve the application of PHL in food processing.展开更多

It also aimed to identify phenolic acids present in the extracts. The 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and ferric thiocyanate lipid peroxidation antioxidant assays were performed. High performance liquid chromatography was used to identify phenolic acids. There was no solvent effect on TPC nor on scavenging activities, and inhibition of lipid peroxidation (p > 0.05) among solvent extracts. On the other hand, 1:1:3 water: acetone: methanol extract (10.22 mg GAE/g sample) had significantly higher reducing potential than 50% ethanol extract (EE) (9.259 mg GAE/g sample) (p < 0.05);but EE was not significantly different from 80% methanol extract (9.781 mg GAE/g sample) (p > 0.05). Phenolic fraction designated as fraction 4 had the highest antioxidant activity (p < 0.05) with 69.49% ABTS scavenging activity and FRAP reducing potential, 22.26 mg of GAE/g sample. For nutritional quality, the parameters meas...The aim of the present study was to evaluate the nutritional quality of green and purple eggplant, their antioxidant activity and their nutritional efficacy on Wistar rats. For nutritional quality, the parameters measured are dry matter, protein, lipid, ash, carbohydrate, iron, phosphorus, calcium, magnesium and energy content. For antioxidant activity, the parameters measured are 50% DDPH free radical inhibition concentration and total polyphenol content. Nutritional efficacy was evaluated in rats fed the control diet and in rats fed the three treated diets containing eggplant meal obtained by replacing 5%, 10%, and 15% of the control diet. The parameters measured are the amount consumed, the weight of the animal and target organs, and hematologic parameters. The results of the nutritional analysis show the following values: 13.31% protein, 2.66% lipids, 0.84% calcium, 0.12% magnesium, 0.43% phosphorus for the Solanum aethiopicum L. species and 13.47% protein, 3.66% lipids, 0.36% calcium, 0.22% magnesium, 0.35% phosphorus for the Solanum melongena L. species. In terms of antioxidant activity, we obtained DDPH inhibition percentages of 40.28 mg/ml for Solanum aethiopicum L. and 12.42 mg/ml for Solanum melongena L., respectively. Finally, hematologic and anthropometric tests showed that for the different diets used, weight loss and an increase in hematologic parameters were observed in the rats tested. This study showed that eggplant has interesting nutritional characteristics and antioxidant activity, contributing to an increase in weight and anthropometric parameters.展开更多
Drought stress is a major factor affecting plant growth and crop yield production.Plant extracts as natural biostimulants hold great potential to strengthen plants to overcome drought impacts.To explore the effect of ...Drought stress is a major factor affecting plant growth and crop yield production.Plant extracts as natural biostimulants hold great potential to strengthen plants to overcome drought impacts.To explore the effect of Polygonum minus extract(PME)in enhancing drought tolerance in plants,a study was set up in a glasshouse environment using 10 different treatment combinations.PME foliar application were designed in CRD and effects were closely observed related to the growth,physiology,and antioxidant system changes in maize(Zea mays L.)under well-watered and drought conditions.The seaweed extract(SWE)was used as a comparison.Plants subjected to drought stress exhibited a significant reduction in fresh weight,dry weight,relative water content(RWC),and soluble sugar,but they stimulated the phenolic,flavonoid,proline,glutathione(GSH),malondialdehyde(MDA)and antioxidant enzyme(catalase,CAT;peroxidase,POD;superoxide dismutase,SOD)activities.Foliar application of PME improved fresh and dry weight(FW:33.1%~41.4%;DW:48.0%~43.1%),chlorophyll content(Chl b:87.9%~100.76%),soluble sugar(23.6%~49.3%),and soluble protein(48.6%~56.9%)as well as antioxidant enzyme activities(CAT and POD)compared to CK under drought conditions.while decreasing the level of MDA.Notably,the mitigating effect of PME application with high concentration was more effective than those of SWE.Our study reveals that PME could alleviate drought stress by regulating osmoprotectant content and antioxidant defense system and can be used as an economical and environmentally friendly biostimulants for promoting maize growth under drought stress.展开更多

Cold stress(CS) is one of...This study assessed the influence of exogenous ME in the mitigation of cold damage in pepper seedlings. Melatonin(ME) is a dynamic molecule that helps plants cope with stress in several ways. Cold stress(CS) is one of the most important environmental factors that restrict plant growth and yield. Pepper(Capsicum annuum L.) is a valuable commercial crop, highly sensitive to CS. Thus, identifying an efficient strategy to mitigate cold damage is critical for long-term pepper production. For this purpose, the roots of pepper seedlings were pretreated with ME(5 μmol · L^(-1)) and exposed to CS for 7 d. The results indicated that CS suppressed pepper growth, hampered photosynthetic capacity, and damaged root architecture in pepper plants. In contrast, the production of reactive oxygen species(ROS), malondialdehyde(MDA), electrolyte leakage(EL), proline, and soluble sugars were enhanced in plants under CS. ME(5 μmol · L^(-1)) pretreatment reduced the negative effects of CS by recovering plant growth, root traits, gas exchange elements, and pigment molecules compared to CS control treatment. Furthermore, ME application efficiently reduced oxidative stress markers [hydrogen peroxide(H_(2)O_(2)), superoxide ion(O_(2)^(·-)), EL, and MDA] while increasing proline and soluble sugar content in pepper leaves. ME application combined with CS further increased antioxidant enzymes and related gene expression. Collectively, our results confirmed the mitigating potential of ME supplementation for CS by maintaining pepper seedling growth,improving the photosynthesis apparatus, regulating pigments, and osmolyte content.展开更多
Bone collagen hydrolysates(peptides)derived from byproduct of animal product processing have been used to produce commercially valuable products due to their potential antioxidant activity.Maillard glycosylated reacti...Bone collagen hydrolysates(peptides)derived from byproduct of animal product processing have been used to produce commercially valuable products due to their potential antioxidant activity.Maillard glycosylated reaction is considered as a promising method to enhance the antioxidant activity of peptides.Hence,this research aims at investigating the Maillard glycosylation activity and antioxidant activity of bone collagen hydrolysates from different sources.In this study,3 glycosylated bone collagen hydrolysates were prepared and characterized,and cytotoxicity and antioxidant activity were analyzed and evaluated.The free amino groups loss,browning intensity,and fluorescence intensity of G-Cbcp(glycosylated chicken bone collagen hydrolysates(peptides))were the heaviest,followed by G-Pbcp(glycosylated porcine bone collagen hydrolysates(peptides))and G-Bbcp(glycosylated bovine bone collagen hydrolysates(peptides)).The results of amino acid analysis showed that amino acid composition of different bone collagen hydrolysates was significantly different and the amino acid decreased to different degrees after Maillard glycosylated reaction,which may lead to differences in Maillard glycosylated reaction activity.Furthermore,the 3 glycosylated hydrolysates showed no significant cytotoxicity.The results showed that glycosylation process significantly increased the antioxidant activity of bone collagen hydrolysates,and G-Cbcp showed the strongest antioxidant activity,followed by G-Pbcp and G-Bbcp.Therefore,compared with the bone collagen hydrolysates,3 glycosylated hydrolysates showed significant characteristic and structural changes,and higher antioxidant activity.展开更多

For this purpose, sequential extraction using solvents of increasing polarity was essential. The various extracts obtained underwent phytochemical and biochemical analyses. Phytochemical screening revealed the presence of flavonoids, alkaloids, tannins, polyphenols, anthocyanins and steroids/terpenes. Quantitative analysis of total polyphenols, total flavonoids and total anthocyanins yielded the following results: total flavonoids (0.803 ± 0029 mg EQ/100g P for acetone extract of roots and 0.871 ± 0.401 mg EQ/100g P for methanol extract of leaves);total polyphenols (23.298 ± 12.68 mg EAG/100g P for acetone extract of roots and 24.69 ± 0.49 401 mg EAG/100g P for methanol extract of leaves);total monomeric anthocyanins (44.697 ± 0.939 mg EC3G/100g P and 16.699 ± 0.193 mg EC3G/100g P respectively for acetone and methanol extracts of stem bark). Results showed that both chlorophyll and carotenoid contents significantly(p<0.05) decreased with increasing Mn treatment levels(0, 0.5, 1, 2, 4, and 8 mg/L) in hydroponics. The concentrations of malondialdehyde(MDA) and hydrogen peroxide(H_2O_2) in the root and shoot of P. hydropiper were accumulated under Mn stress. Meanwhile, the anti-oxidative functions of several important enzymes, including superoxide dismutase(SOD), catalase(CAT), ascorbate peroxidase(APX) and peroxidase(POD) in plants were stimulated by Mn spike in leaves and roots, especially at low Mn stress; while sulfhydryl group(—SH) and glutathion(GSH) were likely involved in Mn detoxification of P. hydropiper under high Mn stress.展开更多
This study aimed to investigate the dose-effect of iron on growth performance,antioxidant function.intestinal morphology,and mRNA expression of jejunal tight junction protein in 1-to21-d-old yellow-feathered broilers....This study aimed to investigate the dose-effect of iron on growth performance,antioxidant function.intestinal morphology,and mRNA expression of jejunal tight junction protein in 1-to21-d-old yellow-feathered broilers.A total of 7201-d-old yellow-feathered maleb roilers were allocated to 9 treatments with 8 replicate cages of 10 birds per cage.The dietary treatments were consisted of a basal diet(contained 79.6 mg Fe kg^(-1))supplemented with 0,20,40,60,80,160,320,640,and 1,280 mg Fe kg^(-1)in the form of FeSO_(4)·7H_(2)O.Compared with the birds in the control group,birds supplemented with 20mg Fe kg^(-1)had higher average daily gain(ADG)(P<0.0001).Adding 640 and 1,280 mg Fe kg^(-1)significantly decreased ADG(P<0.0001)and average daily feed intake(ADFI)(P<0.0001)compared with supplementation of 20mg Fe kg^(-1).Malondialdehyde(MDA)concentration in plasma and duodenum increased linearly(P<0.0001),but MDA concentration in liver and jejunum increased linearly(P<0.05)or quadratically(P<0.05)with increased dietary Fe concentration.The villus height(VH)in duodenum and jejunum,and the ratio of villus height to crypt depth(V/C)in duodenum decreased linearly(P?0.05)as dietary Feincreased.As dietary Fe increased,the jejunal relative mRNA abundance of claudin-1 decreased linearly(P=0.001),but the jejunal relative mRNA abundance of zona occludens-1(ZO-1)and occludin decreased linearly(P?0.05)or quadratically(P?0.05).Compared with the supplementation of 20 mg Fe kg^(-1),the supplementation of640 mg Fe kg^(-1)or higher increased(P?0.05)MDA concentrations in plasma,duodenum,and jejunum,decreased VH in the duodenum and jejunum,and the addition of 1,280 mg Fe kg^(-1)reduced(P?0.05)the jejunal tight junction protein(claudin-1,ZO-1,occludin)mRNA abundance.In summary,640 mg of supplemental Fe kg^(-1)or greater was associated with decreased growth performance,increased oxidative stress,disrupted intestinal morphology,and reduced mRNA expression of jejunal tight junction protein.展开更多
Enzymatic hydrolysis of proteins can enhance their emulsifying properties and antioxidant activities.However,the problem related to the hydrolysis of proteins was the generation of the bitter taste.Recently,high hydro...Enzymatic hydrolysis of proteins can enhance their emulsifying properties and antioxidant activities.However,the problem related to the hydrolysis of proteins was the generation of the bitter taste.Recently,high hydrostatic pressure(HHP)treatment has attracted much interest and has been used in several studies on protein modification.Hence,the study aimed to investigate the effects of enzymatic hydrolysis by Corolase PP under different pressure treatments(0.1,100,200,and 300 MPa for 1-5 h at 50℃)on the emulsifying property,antioxidant activity,and bitterness of soybean protein isolate hydrolysate(SPIH).As observed,the hydrolysate obtained at 200 MPa for 4 h had the highest emulsifying activity index(47.49 m^(2)/g)and emulsifying stability index(92.98%),and it had higher antioxidant activities(44.77%DPPH free radical scavenging activity,31.12%superoxide anion radical scavenging activity,and 61.50%copper ion chelating activity).At the same time,the enhancement of emulsion stability was related to the increase of zeta potential and the decrease of mean particle size.In addition,the hydrolysate obtained at 200 MPa for 4 h had a lower bitterness value and showed better palatability.This study has a broad application prospect in developing food ingredients and healthy foods.展开更多
Ellagic acid(EA)is a natural antioxidant,widely present in a lot of forms’soft fruits,nuts,and other plant tissues,and helpful for promoting human health;however,its protective effect on postharvest fruit and improvi...Ellagic acid(EA)is a natural antioxidant,widely present in a lot of forms’soft fruits,nuts,and other plant tissues,and helpful for promoting human health;however,its protective effect on postharvest fruit and improving the quality index of postharvest fruit have rarely been studied.In this experiment,the strawberries were soaked in 0,100,200,300,400,and 500 mg L^(−1) EA,respectively,and the influential EA on fruit quality and the antioxidant system of strawberries were studied.Compared with the control,EA treatment can reduce the browning degree and rotting rate of strawberry fruit during storage and augment the soluble solid content(SSC).EA treatment can also increase the content of related stuff and enzyme activity in antioxidant systems;the gene expression level of polyphenol oxidase(PPO)in strawberries treated with EA was always down-regulated,correspondingly,the expression of other antioxidant enzyme genes was enhanced.Among the strawberry fruits treated with EA of different concentrations,300 mg L^(−1) EA had the best effect in the process of strawberry preservation.The results suggested that the proper concentration of exogenous EA at 300 mg L−1 could maintain strawberries’quality and enhance the antioxidant system by improving the activities of antioxidative enzymes and the ascorbateglutathione(AsA-GSH)cycle during storage.展开更多
Background:Artemisia vulgaris,a medicinal aromatic plant,is widely used as a food item,tonic pharmaceutical,and cosmetic industry additive owing to its antibacterial,antihypertensive,hepatoprotective,antioxidant,and a...Background:Artemisia vulgaris,a medicinal aromatic plant,is widely used as a food item,tonic pharmaceutical,and cosmetic industry additive owing to its antibacterial,antihypertensive,hepatoprotective,antioxidant,and antispasmodic properties.But the effect of different geographic locations on the chemical composition and bioactivities of its extracts is unclear.Methods:Biological activities of essential oils and ethanol extracts of three varieties of Artemisia vulgaris leaves,which are grown in Shanxi province China,were studied.Results:Gas chromatography-mass spectrometry analysis revealed that the main components of essential oils were terpenes and ketones.Essential oils and ethanol extract of Artemisia vulgaris leaves possessed good antioxidant activities,and their half maximal inhibitory concentrations determined using 1,1-diphenyl-2-picrylhydrazyl and 2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate)assays were 57.0 and 22.9μg/mL,respectively.The essential oils also exhibited remarkable antibacterial activity against three foodborne pathogenic bacterial strains.The ethanol extract presented a high anticancer activity against the MGC-803 human gastric cancer cell line.Conclusion:These biological activities were well correlated with the composition of the extract and EOs,which in turn is affected by the genetic composition of Artemisia vulgaris and geographic location and diverse climatic condition under which it is grown.These findings demonstrate the remarkable potential of Artemisia vulgaris as a valuable source of antioxidant,antibacterial,and anticancer agents.展开更多
文摘Objective:To evaluate the supplementation effects of vitamin E,vitamin C,superoxide dismutase(SOD),and catalase to diluents on bull cryopreserved epididymal sperm.Methods:Sperm were retrieved from 20 bull testes and were then supplemented with 0.1 mM vitamin E,5.0 mM vitamin C,100.0 IU/mL SOD,and 100.0μg/mL catalase alone,or in a combination.The control treatment contained no addition.After supplementation,samples were frozen and stored in liquid nitrogen.The sperm parameters including motility,progressive motility,viability,acrosome integrity,plasma membrane integrity,kinematics and DNA damage were evaluated following the thawing process.Results:Vitamin E alone significantly increased the parameters of acrosome and membrane integrity compared to the control treatment(P<0.05).While compared to the control treatment,vitamin C had no improvement effect on sperm characteristics except for membrane integrity.Treatment of vitamin E+vitamin C had a significant improvement in total motility,progressive motility,viability,membrane and acrosome integrity compared to the control and other treatments(P<0.05).Compared to the control treatment,addition of SOD or catalase alone significantly improved the percentages of total motility,progressive motility,viability,membrane and acrosome integrity(P<0.05).Furthermore,SOD+catalase significantly increased total motility,progressive motility,viability,acrosome and membrane integrity characteristics compared to the catalase treatment(P<0.05).Vitamin E alone,vitamin E+vitamin C,and SOD in diluents decreased DNA damages and thereby improved the rate of intact sperm heads.Conclusions:Addition of 100.0 IU/mL SOD alone and 0.1 mM vitamin E+5.0 mM vitamin C,and also 5.0 mM vitamin C+100μg/mL catalase in a combination improves the quality of cryopreserved bull epididymal sperm and could be used for cryopreservation.

文摘Evidences show that the storage period greatly affects the quality of yogurt.In this study,three types of yogurt:control yogurt(CY),non-hydrolyzed potato powder yogurt(PPY)and enzymatically hydrolyzed potato powder yogurt(EHPPY)were prepared at 42℃ for 5 h and stored for 28 days at 4℃.The yogurts were evaluated for quality characteristics at different storage periods.Negligible differences in pH values,titratable acidities and viable counts were detected in all three types of yogurt during storage.However,compared to other yogurts,EHPPY exhibited desirable water holding capacity,throughout the storage period.Apart from this,sensory properties and antioxidant activities(2-diphenyl-1-picryl-hydrazyl(DPPH)free radical scavenging activity and ferric reducing antioxidant power(FRAP))of EHPPY were also significantly improved during the storage period.Furthermore,the storage(G’)and loss(G”)modulus of PPY,EHPPY were lower than CY at 4℃ while a hysteresis loop was shown by all yogurts at the temperature range of 4-50℃ indicating higher G’(elasticity)than G”(viscosity).Based on our findings,EHPP could be an important functional ingredient in improving the quality and storage stability of yogurt for its production at an industrial level.
文摘Natural phenolic compounds are secondary metabolites found in a wide range of plants including food crops. As many of them are known to be antioxidants and can prevent several chronic and degenerative diseases in humans, they are a part of a healthy diet. However, these antioxidants can act as prooxidants under high phenolic concentration, high pH, or in the presence of transition metal ions such as Cu<sup>2+</sup> or Fe<sup>3+</sup>, producing reactive oxygen species (ROS) including hydroxyl radicals resulting in oxidative stress and cell toxicity. While this can lead to pathogenesis including the development of various types of cancers, elevated levels of ROS are beneficial to kill malignant cells and foodborne pathogens to improve food safety. Thus, the dual nature of phenolic compounds allows them to act as antioxidants and prooxidants. Similarly, depending on the level of prooxidant activity, ROS can induce either pathogenesis or serve as a potential agent to kill malignant cells and foodborne pathogens.

文摘Abiotic stresses,including drought,have been found to affect the growth and medicinal quality of numerous herbs.The proposed study aims to study the effects of different drought regimes on the metabolic profile,growth,ecophysiology,cellular antioxidants,and antioxidant potential of Nigella sativa(Black cumin)leaf.Forty-day-old seedlings of N.sativa were exposed to three regimes of drought(control,moderate and high)for a week.UPLCMS/MS metabolic profile of the leaf reveals the presence of more than a hundred metabolites belonging to anthocyanins,chalcones,dihydro flavonoids,flavonoids,flavanols,flavones,flavonoid carbonoside,isoflavones,etc.Drought was found to alter the contents of identified metabolites.Drought stress-induced oxidative stress and increased production of hydrogen peroxide and superoxide anions.Physiological changes,activities of antioxidant enzymes,contents of antioxidants,and proline were significantly high under drought to protect against the low water regimes.Furthermore,stressed leaf extract had higher antioxidant potential.Thus,N.sativa leaf bears multiple metabolic pathways and can tolerate a higher degree of drought or osmotic stress.
文摘The aim of this study is to assess the adverse effects of lead, a well-documented non-essential element that occurs naturally in the environment, on Channa punctatus, in relation with ROS production and oxidative stress. Fishes were sampled, acclimatized and kept treated or untreated with lead (9.43 mg/L) under observation for 4 days. At day 4, respiratory burst activity, lipid peroxidation activity and superoxide dismutase level increased significantly in treated group as compared to the control. On the contrary, catalase, glutathione S-transferase, glutathione peroxidase, glutathione reductase and reduced glutathione activity decreased on treatment with lead. These results suggest that heavy metal like lead induces oxidative stress, influences the antioxidant defense system and may lead to physiological disorders rendering the health and survival of exposed fish to a compromised state.
文摘Introduction: Non-enzymatic antioxidants are good scavengers of free radicals preventing their overproduction there by reducing the level of oxidative stress. This work was undertaken at Saint Peter TB specialized hospital and TekleHaimanot health center from March 2012 to May 2013.Aim: To determine changes in Non-Enzymatic Antioxidants and level of oxidative stress of tuberculosis Patients before and after taking anti tuberculosis treatment.Materials and Methods: In this comparative cross sectional study, a total of 210 individuals including: newly diagnosed TB patients as group-I (n = 70), TB patients who completed treatment as group-II (n = 70), and healthy volunteers as group-III (n = 70) were enrolled. Different methods were used to determine the parameters;vit-C (HPLC method), lipid peroxidation (thiobarbuituric acid method), and bilirubin (Colorimetric assay). Results: Vitamin-C (Vit-C) and of group-I showed a significant reduction (p < 0.001) as compared with both group-II and group-III whereas Malondialdehyde (MDA) level was increased. However, the total and direct bilirubin was not different among the groups. In group-III, there was a positive correlation between BMI and serum Vit-C (r = -0.305, p = 0.010). Vit-C showed a negative correlation with serum MDA in all the groups with values (r = -0.265, p = 0.027), (r = -0.389, p = 0.001) and (r = -0.375, p = 0.001) for group-I, group-II and group-III respectively. In addition to this Vit-C was negatively correlated with serum UA (r = -0.285, p = 0.017) in group-I. Conclusion: The findings of the current study suggest that the amount of Vit-C in the newly diagnosed TB patients and those who finished their treatment is much lower than the healthy volunteers. In contrast to this, the MDA value was significantly higher both in the newly diagnosed TB patients and TB patients who completed treatment than in healthy volunteers suggesting higher degree of oxidative stress.
文摘Betalains are natural coloring pigments with betalamic acid as the core structure of all subclasses.Besides their coloring properties,betalains exhibit various biological activities,including antioxidant and anti-inflammatory properties,which are highly imperative.Further in-vivo studies reported that betalains protect various body organs,leading to health enhancement.Body organs,including the heart,liver,kidney,lung,etc.,are important for a healthy life.However,these organs can be affected or damaged by various stress factors,toxicants,and harmful substances.Recent studies have claimed that betalains could protect all vital organs of the body through antioxidant and anti-inflammatory mechanisms.This review article described the in-vivo antioxidant and anti-inflammatory activities of betalains in various cell-line or animal models.A comprehensive discussion has been provided on the mechanism of action of betalains in protecting various body organs,including cardio-protective effect,hepato-protective ability,renal protection capacity,repro-protective ability,neuro-protective effect,lung protection,and gut protection ability.Finally,future research directions and conclusions have been outlined.

文摘Phlorizin(PHL)is a natural compound with strong antioxidant properties mainly found in apples.In this paper,the interaction mechanism of PHL with pepsin and trypsin was comparatively evaluated by computer simulation,fluorescence spectra,circular dichroism(CD),and Fourier transform infrared(FT-IR)spectra at a molecular level.Fluorescence spectra showed that PHL quenches the pepsin/trypsin by static quenching.Thermodynamic parameters indicated that PHL binds to pepsin mainly through hydrogen bonds and van der Waals forces,and that of trypsin was electrostatic forces.The ground state complexes PHL and protease have a moderate affinity of 105 L/mol PHL binds more strongly to trypsin than to pepsin.CD and FT-IR spectra results showed that pepsin/trypsin decreased theβ-sheet content and slightly changed its secondary structure upon PHL.These experimental results are mutually verified with the predicted computer-aid simulation results.Upon PHL and trypsin binding,the antioxidant capacity of PHL was elevated.Nevertheless,the antioxidant capacity of PHL was decreased after binding to pepsin.This work elucidates the binding of PHL binding mechanisms to pepsin/trypsin and provides useful information for the digestion of PHL to improve the application of PHL in food processing.
文摘This study aimed to compare the total phenolic content (TPC) and antioxidant activities of A. rigidula extracts. It also aimed to identify phenolic acids present in the extracts. The 2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,2-diphenyl-1-picrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), and ferric thiocyanate lipid peroxidation antioxidant assays were performed. High performance liquid chromatography was used to identify phenolic acids. There was no solvent effect on TPC nor on scavenging activities, and inhibition of lipid peroxidation (p > 0.05) among solvent extracts. On the other hand, 1:1:3 water: acetone: methanol extract (10.22 mg GAE/g sample) had significantly higher reducing potential than 50% ethanol extract (EE) (9.259 mg GAE/g sample) (p < 0.05);but EE was not significantly different from 80% methanol extract (9.781 mg GAE/g sample) (p > 0.05). Phenolic fraction designated as fraction 4 had the highest antioxidant activity (p < 0.05) with 69.49% ABTS scavenging activity and FRAP reducing potential, 22.26 mg of GAE/g sample. DPPH scavenging activities of fractions 4 (55.59%) and 5 (55.64%) were significantly higher than the other fractions (p A. rigidula extracts contain gallic, caffeic, vanillic, p-coumaric, salicylic acids and vanillin.
文摘The aim of the present study was to evaluate the nutritional quality of green and purple eggplant, their antioxidant activity and their nutritional efficacy on Wistar rats. For nutritional quality, the parameters measured are dry matter, protein, lipid, ash, carbohydrate, iron, phosphorus, calcium, magnesium and energy content. For antioxidant activity, the parameters measured are 50% DDPH free radical inhibition concentration and total polyphenol content. Nutritional efficacy was evaluated in rats fed the control diet and in rats fed the three treated diets containing eggplant meal obtained by replacing 5%, 10%, and 15% of the control diet. The parameters measured are the amount consumed, the weight of the animal and target organs, and hematologic parameters. The results of the nutritional analysis show the following values: 13.31% protein, 2.66% lipids, 0.84% calcium, 0.12% magnesium, 0.43% phosphorus for the Solanum aethiopicum L. species and 13.47% protein, 3.66% lipids, 0.36% calcium, 0.22% magnesium, 0.35% phosphorus for the Solanum melongena L. species. In terms of antioxidant activity, we obtained DDPH inhibition percentages of 40.28 mg/ml for Solanum aethiopicum L. and 12.42 mg/ml for Solanum melongena L., respectively. Finally, hematologic and anthropometric tests showed that for the different diets used, weight loss and an increase in hematologic parameters were observed in the rats tested. This study showed that eggplant has interesting nutritional characteristics and antioxidant activity, contributing to an increase in weight and anthropometric parameters.

文摘Drought stress is a major factor affecting plant growth and crop yield production.Plant extracts as natural biostimulants hold great potential to strengthen plants to overcome drought impacts.To explore the effect of Polygonum minus extract(PME)in enhancing drought tolerance in plants,a study was set up in a glasshouse environment using 10 different treatment combinations.PME foliar application were designed in CRD and effects were closely observed related to the growth,physiology,and antioxidant system changes in maize(Zea mays L.)under well-watered and drought conditions.The seaweed extract(SWE)was used as a comparison.Plants subjected to drought stress exhibited a significant reduction in fresh weight,dry weight,relative water content(RWC),and soluble sugar,but they stimulated the phenolic,flavonoid,proline,glutathione(GSH),malondialdehyde(MDA)and antioxidant enzyme(catalase,CAT;peroxidase,POD;superoxide dismutase,SOD)activities.Foliar application of PME improved fresh and dry weight(FW:33.1%~41.4%;DW:48.0%~43.1%),chlorophyll content(Chl b:87.9%~100.76%),soluble sugar(23.6%~49.3%),and soluble protein(48.6%~56.9%)as well as antioxidant enzyme activities(CAT and POD)compared to CK under drought conditions.while decreasing the level of MDA.Notably,the mitigating effect of PME application with high concentration was more effective than those of SWE.Our study reveals that PME could alleviate drought stress by regulating osmoprotectant content and antioxidant defense system and can be used as an economical and environmentally friendly biostimulants for promoting maize growth under drought stress.

文摘Plant extracts from natural sources are an excellent choice for food additives and natural antioxidants.In this study,the active components of Tulipa edulis were extracted and analysed,and their antioxidant capacity was measured.Then,the crude extract mixture was separated and purified using a Sephadex LH-20 gel,and the antioxidant activity of the purified products was determined.Human umbilical vein endothelial human umbilical vein endothelial cells(HUVEC)cells were treated with 35 mmol/L glucose to construct a model of oxidative stress.Then,the cells were treated with the active component to observe whether the products of T.edulis could have a good protective effect on HUVEC cells induced by glucose.Transcriptome analysis was also performed on HUVEC cells after same treatment to explore the possible mechanism of the component F2 protecting HUVEC cells from oxidative stress induced by high glucose.The results showed that component F2 obtained from T.edulis has strong antioxidant activity.Moreover,F2 can play a strong antioxidant protective role in HUVEC cells.Meanwhile,the gene expression of heme oxygenase 1(HO-1),γ-glutamyl cysteine ligase catalytic subunit(GCLC)and NAD(P)H quinone oxidoreductase-1(NQO1)in HUVEC cells was up-regulated after treated with F2.This study provides reference value for the further development and application of T.edulis and the d evelopment of functional food.

文摘This study assessed the influence of exogenous ME in the mitigation of cold damage in pepper seedlings. Melatonin(ME) is a dynamic molecule that helps plants cope with stress in several ways. Cold stress(CS) is one of the most important environmental factors that restrict plant growth and yield. Pepper(Capsicum annuum L.) is a valuable commercial crop, highly sensitive to CS. Thus, identifying an efficient strategy to mitigate cold damage is critical for long-term pepper production. For this purpose, the roots of pepper seedlings were pretreated with ME(5 μmol · L^(-1)) and exposed to CS for 7 d. The results indicated that CS suppressed pepper growth, hampered photosynthetic capacity, and damaged root architecture in pepper plants. In contrast, the production of reactive oxygen species(ROS), malondialdehyde(MDA), electrolyte leakage(EL), proline, and soluble sugars were enhanced in plants under CS. ME(5 μmol · L^(-1)) pretreatment reduced the negative effects of CS by recovering plant growth, root traits, gas exchange elements, and pigment molecules compared to CS control treatment. Furthermore, ME application efficiently reduced oxidative stress markers [hydrogen peroxide(H_(2)O_(2)), superoxide ion(O_(2)^(·-)), EL, and MDA] while increasing proline and soluble sugar content in pepper leaves. ME application combined with CS further increased antioxidant enzymes and related gene expression. Collectively, our results confirmed the mitigating potential of ME supplementation for CS by maintaining pepper seedling growth,improving the photosynthesis apparatus, regulating pigments, and osmolyte content.

文摘Bone collagen hydrolysates(peptides)derived from byproduct of animal product processing have been used to produce commercially valuable products due to their potential antioxidant activity.Maillard glycosylated reaction is considered as a promising method to enhance the antioxidant activity of peptides.Hence,this research aims at investigating the Maillard glycosylation activity and antioxidant activity of bone collagen hydrolysates from different sources.In this study,3 glycosylated bone collagen hydrolysates were prepared and characterized,and cytotoxicity and antioxidant activity were analyzed and evaluated.The free amino groups loss,browning intensity,and fluorescence intensity of G-Cbcp(glycosylated chicken bone collagen hydrolysates(peptides))were the heaviest,followed by G-Pbcp(glycosylated porcine bone collagen hydrolysates(peptides))and G-Bbcp(glycosylated bovine bone collagen hydrolysates(peptides)).The results of amino acid analysis showed that amino acid composition of different bone collagen hydrolysates was significantly different and the amino acid decreased to different degrees after Maillard glycosylated reaction,which may lead to differences in Maillard glycosylated reaction activity.Furthermore,the 3 glycosylated hydrolysates showed no significant cytotoxicity.The results showed that glycosylation process significantly increased the antioxidant activity of bone collagen hydrolysates,and G-Cbcp showed the strongest antioxidant activity,followed by G-Pbcp and G-Bbcp.Therefore,compared with the bone collagen hydrolysates,3 glycosylated hydrolysates showed significant characteristic and structural changes,and higher antioxidant activity.
文摘The aim of the present work is to assess the value of Detarium Senegalense by determining the content of total phenols, total flavonoids and total anthocyanins, and by evaluating the free radical scavenging activity of Detarium Senegalense extracts. For this purpose, sequential extraction using solvents of increasing polarity was essential. The various extracts obtained underwent phytochemical and biochemical analyses. Phytochemical screening revealed the presence of flavonoids, alkaloids, tannins, polyphenols, anthocyanins and steroids/terpenes. Quantitative analysis of total polyphenols, total flavonoids and total anthocyanins yielded the following results: total flavonoids (0.803 ± 0029 mg EQ/100g P for acetone extract of roots and 0.871 ± 0.401 mg EQ/100g P for methanol extract of leaves);total polyphenols (23.298 ± 12.68 mg EAG/100g P for acetone extract of roots and 24.69 ± 0.49 401 mg EAG/100g P for methanol extract of leaves);total monomeric anthocyanins (44.697 ± 0.939 mg EC3G/100g P and 16.699 ± 0.193 mg EC3G/100g P respectively for acetone and methanol extracts of stem bark). DPPH free radical scavenging activity was 1.674 ± 0.023 mg/mL for the acetone extract and 0.934 ± 0.24 mg/mL for the methanol extract of roots. .

文摘The response of enzyme and non-enzymatic antioxidants of Mn hyperaccumuator, Polygonum hydropiper(P. hydropiper), to Mn stress was studied using hydroponics culture experiments to explore the mechanism of Mn tolerance in this species. Results showed that both chlorophyll and carotenoid contents significantly(p<0.05) decreased with increasing Mn treatment levels(0, 0.5, 1, 2, 4, and 8 mg/L) in hydroponics. The concentrations of malondialdehyde(MDA) and hydrogen peroxide(H_2O_2) in the root and shoot of P. hydropiper were accumulated under Mn stress. Meanwhile, the anti-oxidative functions of several important enzymes, including superoxide dismutase(SOD), catalase(CAT), ascorbate peroxidase(APX) and peroxidase(POD) in plants were stimulated by Mn spike in leaves and roots, especially at low Mn stress; while sulfhydryl group(—SH) and glutathion(GSH) were likely involved in Mn detoxification of P. hydropiper under high Mn stress.

文摘This study aimed to investigate the dose-effect of iron on growth performance,antioxidant function.intestinal morphology,and mRNA expression of jejunal tight junction protein in 1-to21-d-old yellow-feathered broilers.A total of 7201-d-old yellow-feathered maleb roilers were allocated to 9 treatments with 8 replicate cages of 10 birds per cage.The dietary treatments were consisted of a basal diet(contained 79.6 mg Fe kg^(-1))supplemented with 0,20,40,60,80,160,320,640,and 1,280 mg Fe kg^(-1)in the form of FeSO_(4)·7H_(2)O.Compared with the birds in the control group,birds supplemented with 20mg Fe kg^(-1)had higher average daily gain(ADG)(P<0.0001).Adding 640 and 1,280 mg Fe kg^(-1)significantly decreased ADG(P<0.0001)and average daily feed intake(ADFI)(P<0.0001)compared with supplementation of 20mg Fe kg^(-1).Malondialdehyde(MDA)concentration in plasma and duodenum increased linearly(P<0.0001),but MDA concentration in liver and jejunum increased linearly(P<0.05)or quadratically(P<0.05)with increased dietary Fe concentration.The villus height(VH)in duodenum and jejunum,and the ratio of villus height to crypt depth(V/C)in duodenum decreased linearly(P?0.05)as dietary Feincreased.As dietary Fe increased,the jejunal relative mRNA abundance of claudin-1 decreased linearly(P=0.001),but the jejunal relative mRNA abundance of zona occludens-1(ZO-1)and occludin decreased linearly(P?0.05)or quadratically(P?0.05).Compared with the supplementation of 20 mg Fe kg^(-1),the supplementation of640 mg Fe kg^(-1)or higher increased(P?0.05)MDA concentrations in plasma,duodenum,and jejunum,decreased VH in the duodenum and jejunum,and the addition of 1,280 mg Fe kg^(-1)reduced(P?0.05)the jejunal tight junction protein(claudin-1,ZO-1,occludin)mRNA abundance.In summary,640 mg of supplemental Fe kg^(-1)or greater was associated with decreased growth performance,increased oxidative stress,disrupted intestinal morphology,and reduced mRNA expression of jejunal tight junction protein.

文摘Enzymatic hydrolysis of proteins can enhance their emulsifying properties and antioxidant activities.However,the problem related to the hydrolysis of proteins was the generation of the bitter taste.Recently,high hydrostatic pressure(HHP)treatment has attracted much interest and has been used in several studies on protein modification.Hence,the study aimed to investigate the effects of enzymatic hydrolysis by Corolase PP under different pressure treatments(0.1,100,200,and 300 MPa for 1-5 h at 50℃)on the emulsifying property,antioxidant activity,and bitterness of soybean protein isolate hydrolysate(SPIH).As observed,the hydrolysate obtained at 200 MPa for 4 h had the highest emulsifying activity index(47.49 m^(2)/g)and emulsifying stability index(92.98%),and it had higher antioxidant activities(44.77%DPPH free radical scavenging activity,31.12%superoxide anion radical scavenging activity,and 61.50%copper ion chelating activity).At the same time,the enhancement of emulsion stability was related to the increase of zeta potential and the decrease of mean particle size.In addition,the hydrolysate obtained at 200 MPa for 4 h had a lower bitterness value and showed better palatability.This study has a broad application prospect in developing food ingredients and healthy foods.

文摘Ellagic acid(EA)is a natural antioxidant,widely present in a lot of forms’soft fruits,nuts,and other plant tissues,and helpful for promoting human health;however,its protective effect on postharvest fruit and improving the quality index of postharvest fruit have rarely been studied.In this experiment,the strawberries were soaked in 0,100,200,300,400,and 500 mg L^(−1) EA,respectively,and the influential EA on fruit quality and the antioxidant system of strawberries were studied.Compared with the control,EA treatment can reduce the browning degree and rotting rate of strawberry fruit during storage and augment the soluble solid content(SSC).EA treatment can also increase the content of related stuff and enzyme activity in antioxidant systems;the gene expression level of polyphenol oxidase(PPO)in strawberries treated with EA was always down-regulated,correspondingly,the expression of other antioxidant enzyme genes was enhanced.Among the strawberry fruits treated with EA of different concentrations,300 mg L^(−1) EA had the best effect in the process of strawberry preservation.The results suggested that the proper concentration of exogenous EA at 300 mg L−1 could maintain strawberries’quality and enhance the antioxidant system by improving the activities of antioxidative enzymes and the ascorbateglutathione(AsA-GSH)cycle during storage.

文摘Background:Artemisia vulgaris,a medicinal aromatic plant,is widely used as a food item,tonic pharmaceutical,and cosmetic industry additive owing to its antibacterial,antihypertensive,hepatoprotective,antioxidant,and antispasmodic properties.But the effect of different geographic locations on the chemical composition and bioactivities of its extracts is unclear.Methods:Biological activities of essential oils and ethanol extracts of three varieties of Artemisia vulgaris leaves,which are grown in Shanxi province China,were studied.Results:Gas chromatography-mass spectrometry analysis revealed that the main components of essential oils were terpenes and ketones.Essential oils and ethanol extract of Artemisia vulgaris leaves possessed good antioxidant activities,and their half maximal inhibitory concentrations determined using 1,1-diphenyl-2-picrylhydrazyl and 2,2’-azinobis-(3-ethylbenzthiazoline-6-sulphonate)assays were 57.0 and 22.9μg/mL,respectively.The essential oils also exhibited remarkable antibacterial activity against three foodborne pathogenic bacterial strains.The ethanol extract presented a high anticancer activity against the MGC-803 human gastric cancer cell line.Conclusion:These biological activities were well correlated with the composition of the extract and EOs,which in turn is affected by the genetic composition of Artemisia vulgaris and geographic location and diverse climatic condition under which it is grown.These findings demonstrate the remarkable potential of Artemisia vulgaris as a valuable source of antioxidant,antibacterial,and anticancer agents.
