Notopterygium Incisum Extract Promotes Apoptosis by Preventing the Degradation of BIM in Colorectal Cancer

Objective Colorectal cancer(CRC),a prevalent malignancy worldwide,has prompted extensive research into anticancer drugs.Traditional Chinese medicinal materials offer promising avenues for cancer management due to their diverse pharmacological activities.This study investigated the effects of Notopterygium incisum,a traditional Chinese medicine named Qianghuo(QH),on CRC cells and the underlying mechanism.Methods The sulforhodamine B assay and colony formation assay were employed to assess the effect of QH extract on the proliferation of CRC cell lines HCT116 and Caco-2.Propidium iodide(PI)staining was utilized to detect cell cycle progression,and PE Annexin V staining to detect apoptosis.Western blotting was conducted to examine the levels of apoptotic proteins,including B-cell lymphoma 2-interacting mediator of cell death(BIM),B-cell lymphoma 2(Bcl-2),Bcl-2-associated X protein(BAX)and cleaved caspase-3,as well as BIM stability after treatment with the protein synthesis inhibitor cycloheximide.The expression of BAX was suppressed using lentivirus-mediated shRNA to validate the involvement of the BIM/BAX axis in QH-induced apoptosis.The in vivo effects of QH extract on tumor growth were observed using a xenograft model.Lastly,APC^(Min+)mice were used to study the effects of QH extract on primary intestinal tumors.Results QH extract exhibited significant in vitro anti-CRC activities evidenced by the inhibition of cell proliferation,perturbation of cell cycle progression,and induction of apoptosis.Mechanistically,QH extract significantly increased the stability of BIM proteins,which undergo rapid degradation under unstressed conditions.Knockdown of BAX,the downstream effector of BIM,significantly rescued QH-induced apoptosis.Furthermore,the in vitro effect of QH extract was recapitulated in vivo.QH extract significantly inhibited the tumor growth of HCT116 xenografts in nude mice and decreased the number of intestinal polyps in the APC^(Min+)mice.Conclusion QH extract promotes the apoptosis of CRC cells by preventing the degradation of BIM. 展开更多

Methods The total essentia... Aim To analyse the chemical constituents of the essential oil extracted from the rhizome and root of Notopterygium incisum Ting ex H.T. Chang, providing scientific basis for quality control. Methods The total essential oil was extracted by water-steam distillation and separated by capillary gas chromatography (GC). The components were determined by normalization method, and identified by GC-MS. Results GC-MS exhibited 242 peaks and 83 compounds were identified, accounting for 75.77% of the total essential oil. Conclusion In the total essential oil of the rhizome and root of N. incisum, monoterpenes and sesquiterpenes accounted for 13.63% and 67.93%, respectively, in which ( 1S)-β-pinene ( 1.67% ), 3-carene ( 1.05% ), limonene ( 1.22% ), and 1S-endo-bornyl acetate ( 1.68% ) as the monoterpenes and its derivatives, and ( + ) -β-elemene (6.78%), sativene (1.54%), α-caryophyllene (2.64%), germacrene D (1.67%), eudesma-4 ( 14 ), ll-diene (2.36%), α-selinene (2.42%), δ-cadinene ( 1.55% ), 3,7,11-trimethyl-2,6,10-dodecatrien-1-ol (1.03%), ( + )-elemol (5.18%), (-)-spathulenol (1.40%), guaiol (3.81%), dehydroxy-isocalamendiol ( 1.06% ), γ-eudesmol ( 1.05% ), α-eudesmol (7.97%), bulnesol (3.09%), and carotol (2. 30% ) as the sesquiterpenes and its derivatives were main components. Chang(Apiaceae) is an endangered perennial herb in China. The lack of transcriptomic and genomic resources for N. incisum greatly hinders studies of its population genetics and conservation. In this study, we employed RNA-seq technology to characterize transcriptomes for the flowers, leaves, and stems of this endangered herb. A total of 56 million clean reads were assembled into 120,716 unigenes with an N50 length of 850 bp. Among these unigenes, 70,245(58.19%) were successfully annotated and 65,965(54.64%) were identified as coding sequences based on their similarities with sequences in public databases. We identified 21 unigenes that had significant relationships with cold tolerance in N. incisum according to gene ontology(GO) annotation analysis. In addition, 13,149 simple sequence repeats(SSRs) and 85,681 single nucleotide polymorphisms were detected as potential molecular genetic markers. Ninety-six primer pairs of SSRs were randomly selected to validate their amplification efficiency and polymorphism. Nineteen SSR loci exhibited polymorphism in three natural populations of N. incisum. These results provide valuable resources to facilitate future functional genomics and conservation genetics studies of N. incisum. 展开更多

The research for the Intelligent Reflecting Surface(IRS)which has the advantages of cost and energy efficiency has been studied.Channel capacity can be effectively increased by appropriately setting the phase value of IRS elements according to the channel conditions.However,the problem of obtaining an appropriate phase value of IRs is difficult to solve due to the non-convex problem.This paper proposes an iterative algorithm for the alternating optimal solution in the Single User Multiple-Input-Multiple-Output(SU-MIMO)systems.The proposed iterative algorithm finds an alternating optimal solution that is the phase value of IRS one by one.The results show that the proposed method has better performance than that of the randomized IRS systems.The number of iterations for maximizing the performance of the proposed algorithm depends on the channel state between the IRS and the receiver. 展开更多

文章基于递归残差的逆序特征和隔离检测研究了回归模型多参数变点的检测方法。首先,构建带有变点的回归模型,考虑到多元正向CUSUM检验能防止协变量均值与偏移量正交时损失功效,但其变点检测效果并不理想的情况,引入修正的检验统计量BCU... 文章基于递归残差的逆序特征和隔离检测研究了回归模型多参数变点的检测方法。首先,构建带有变点的回归模型,考虑到多元正向CUSUM检验能防止协变量均值与偏移量正交时损失功效,但其变点检测效果并不理想的情况,引入修正的检验统计量BCUSUM。其次,结合快速高效的隔离检测技术,提出MCPDP算法用于估计变点数目及位置。最后,模拟结果表明,所提出的方法能较好地控制检验水平,有更高的功效;评价结果显示,MCPDP算法在变点估计性能方面表现较优;实例分析表明,交通流变点符合实际交通情况,验证了该方法的有效性,且所构建的模型可以作为交通参数确定性经验关系的一种修正。 展开更多

Slope stability analysis is a classical mechanical problem in geotechnical engineering and engineering geology.It is of great significance to study the stability evolution of expansive soil slopes for engineering construction in expansive soil areas.Most of the existing studies evaluate the slope stability by analyzing the limit equilibrium state of the slope,and the analysis method for the stability evolution considering the damage softening of the shear zone is lacking.In this study,the large deformation shear mechanical behavior of expansive soil was investigated by ring shear test.The damage softening characteristic of expansive soil in the shear zone was analyzed,and a shear damage model reflecting the damage softening behavior of expansive soil was derived based on the damage theory.Finally,by skillfully combining the vector sum method and the shear damage model,an analysis method for the stability evolution of the expansive soil slope considering the shear zone damage softening was proposed.The results show that the shear zone subjected to large displacement shear deformation exhibits an obvious damage softening phenomenon.The damage variable equation based on the logistic function can be well used to describe the shear damage characteristics of expansive soil,and the proposed shear damage model is in good agreement with the ring shear test results.The vector sum method considering the damage softening behavior of the shear zone can be well applied to analyze the stability evolution characteristics of the expansive soil slope.The stability factor of the expansive soil slope decreases with the increase of shear displacement,showing an obvious progressive failure behavior. 展开更多

目前单一模型预测精度存在难以随着功率波动保持最优的问题,为提高并网系统运行的稳定性和电网的节能调度,文中提出了一种基于k-sums分层聚类的动态学习组合光伏短期功率预测方法。利用k-sums算法经过分段聚类,将天气类型分为晴天A 1、... 目前单一模型预测精度存在难以随着功率波动保持最优的问题,为提高并网系统运行的稳定性和电网的节能调度,文中提出了一种基于k-sums分层聚类的动态学习组合光伏短期功率预测方法。利用k-sums算法经过分段聚类,将天气类型分为晴天A 1、多云A 2、阴雨天B。通过TCN(Temporal Convolutional Network)提取数据的时序特征,并结合GRU(Gate Recurrent Unit)建立融合提取时序特征模块的改进GRU结构,以达到对时序特征敏感的效果。将改进GRU结构与SVM(Support Vector Machine)动态组合,使用Elastic Net算法输出最佳权重值叠加得到最终预测值。文中采用江苏某地区的光伏发电功率数据及对应的气象数据对所提方法进行验证,结果表明动态组合学习模型的MAE(Mean Absolute Error)为1.888,RMSE(Root Mean Squared Error)为2.403。 展开更多

It is well established that Nash equilibrium exists within the framework of mixed strategies in strategic-form non-cooperative games. However, finding the Nash equilibrium generally belongs to the class of problems known as PPAD (Polynomial Parity Argument on Directed graphs), for which no polynomial-time solution methods are known, even for two-player games. This paper demonstrates that in fixed-sum two-player games (including zero-sum games), the Nash equilibrium forms a convex set, and has a unique expected payoff. Furthermore, these equilibria are Pareto optimal. Additionally, it is shown that the Nash equilibrium of fixed-sum two-player games can theoretically be found in polynomial time using the principal-dual interior point method, a solution method of linear programming. 展开更多

A new multi-target filtering algorithm, termed as the Gaussian sum probability hypothesis density (GSPHD) filter, is proposed for nonlinear non-Gaussian tracking models. Provided that the initial prior intensity of the states is Gaussian or can be identified as a Gaussian sum, the analytical results of the algorithm show that the posterior intensity at any subsequent time step remains a Gaussian sum under the assumption that the state noise, the measurement noise, target spawn intensity, new target birth intensity, target survival probability, and detection probability are all Gaussian sums. The analysis also shows that the existing Gaussian mixture probability hypothesis density (GMPHD) filter, which is unsuitable for handling the non-Gaussian noise cases, is no more than a special case of the proposed algorithm, which fills the shortage of incapability of treating non-Gaussian noise. 目的探讨槐耳清膏对源自人乳腺癌细胞系SUM-159的干细胞是否具有抑制或者杀伤作用。方法以人乳腺癌细胞系SUM-159为细胞模型,以0、1、10、100mg/mL的槐耳清膏溶液处理细胞,观察槐耳清膏给药前后细胞形态学的变化;应用CCK-8法检测IC50值... 目的探讨槐耳清膏对源自人乳腺癌细胞系SUM-159的干细胞是否具有抑制或者杀伤作用。方法以人乳腺癌细胞系SUM-159为细胞模型,以0、1、10、100mg/mL的槐耳清膏溶液处理细胞,观察槐耳清膏给药前后细胞形态学的变化;应用CCK-8法检测IC50值,观察给药前后细胞克隆形成能力和成球能力的变化;流式细胞术检测给药前后ALDH1high细胞比例的变化。结果CCK-8检测结果显示:槐耳清膏对SUM-159细胞系的IC50值约为10mg/mL。以浓度为0、1、10mg/mL的槐耳清膏作用于SUM-159细胞,发现随着浓度的增加,克隆形成的菌落数目逐渐减少(P<0.05),成球数目和体积也逐渐降低(P<0.05)。10mg/mL槐耳清膏作用该细胞后,流式细胞术检测结果显示,ALDH1high细胞的比例由给药前的2.8%降到1.5%;1μg/mL紫杉醇作用该细胞后,ALDH1high细胞的比例由给药前的2.8%升到9.1%。结论槐耳清膏对人乳腺癌细胞系SUM-159细胞的克隆形成和成球能力具有明显的抑制作用,同时能够降低SUM-159细胞中ALDH1high细胞的比例,提示槐耳清膏可能对乳腺癌干细胞具有一定的抑制或杀伤作用。 展开更多

在三相交流级联系统中,源模块以及负载模块之间的相互作用将导致整个系统性能下降,甚至有可能导致系统不稳定,尤其是在含有恒功率负载的系统中表现尤为明显。通过分析系统的小信号模型可以看出,模块之间由于相互耦合而产生的稳定性问题... 在三相交流级联系统中,源模块以及负载模块之间的相互作用将导致整个系统性能下降,甚至有可能导致系统不稳定,尤其是在含有恒功率负载的系统中表现尤为明显。通过分析系统的小信号模型可以看出,模块之间由于相互耦合而产生的稳定性问题和子模块的端口阻抗以及导纳有着直接关系。因此,基于子模块端口阻抗以及导纳的稳定性判据是分析系统级稳定性问题的有效方法。目前,基于子模块阻抗以及导纳的稳定性判据已经广泛应用于直流系统。但是,由于交流系统和直流系统在系统特性上的内在差异,目前广泛应用于直流系统的稳定性判据无法直接使用于交流系统。根据交流系统的系统特性,提出一种基于端口阻抗以及导纳矩阵的G-范数和sum-范数的稳定性判据。且根据矩阵计算的特点,对所提出的稳定性判据做进一步改进,以减少判据的保守性。最终,将所提判据和现有的范数判据进行保守性的比较,可以看出,所提判据的保守性最小,更利于实际应用。 展开更多

铅易切削钢生产的关键技术是控制铅的收得率,铅在钢液中的均匀性,铅蒸汽对环境的污染、铸坯表面质量及顺行浇铸。从铅的控制和提高可浇铸性两方面研究了SUM24HSL钢(%:≤0.11C、1.0~1.5Mn、≤0.05Si、0.04~0.09P、0.26~0.42S、0.25~0... 铅易切削钢生产的关键技术是控制铅的收得率,铅在钢液中的均匀性,铅蒸汽对环境的污染、铸坯表面质量及顺行浇铸。从铅的控制和提高可浇铸性两方面研究了SUM24HSL钢(%:≤0.11C、1.0~1.5Mn、≤0.05Si、0.04~0.09P、0.26~0.42S、0.25~0.32Pb)的100 t EAF-LF-CC工艺。结果表明,在1 620~1 640℃加铅3.5~4.0 kg/t,铅粒度2~3 mm,加入速度50~60 kg/min,氩气流量420~480 L/min,铅的收得率为75%。匹配结晶器振动参数,合适的结晶器保护渣、浇铸温度和保护浇铸技术可保证连铸顺行和轧材冶金质量。 展开更多
