[Objectives]To investigate the acute toxicity and hepatoprotective effect of Jinchuan formula plum wine extract on mice,determine its safety range,and evaluate its hepatoprotective effect.[Methods]The median lethal do...[Objectives]To investigate the acute toxicity and hepatoprotective effect of Jinchuan formula plum wine extract on mice,determine its safety range,and evaluate its hepatoprotective effect.[Methods]The median lethal dose(LD_(50))was determined by acute toxicity test with the toxic reaction and mortality of mice as indexes.Sixty Kunming mice were randomly divided into 6 groups:normal control group,model group(ConA-induced liver injury model),Jinchuan formula plum wine high,medium and low dose groups(1.0,0.5,0.25 g/kg)and silybin group(0.1 g/kg).The levels of ALT,AST,LDH in serum and TG,VLDL in liver were measured.After HE staining,the pathological changes of liver tissue in mice were observed,and the liver protective effect of Jinchuan formula plum wine extract was analyzed and evaluated.[Results]LD_(50)was 11.18 g/kg,and the 95%confidence limit of LD_(50)was 10.31-12.05 g/kg.The high-dose group of Jinchuan formula plum wine extract could significantly reduce the serum ALT and AST activities of ConA-induced liver injury mice(P<0.05).[Conclusions]Jinchuan formula plum wine extract is relatively safe,and also has a protective effect on liver injury.展开更多

Applications of nanotechnology in agriculture will result in the development of efficient and potential approaches towards the management of insect pests.The toxicity effects of four essential oils peppermint,thyme,ca...Applications of nanotechnology in agriculture will result in the development of efficient and potential approaches towards the management of insect pests.The toxicity effects of four essential oils peppermint,thyme,camphor and sage oils were tested against the fourth instar larvae of Agrotis ipsilon to select the most effective essential oil to be converted to the nano form.According to the results obtained,peppermint oil was the most toxic compound,which has been used in the present investigation compared with neem oil.The toxicity of bulk and nano-formulations of neem and pepper mint oils were tested against 2nd and 4th instar larvae of A.ipsilon under laboratory conditions of 25±2°C&65-70%R.H.relative humidity The results show that the LC50 value(the concentration used which kill 50%of the tested individuals)of loaded neem or pepper mint were lower(0.62 and 36.47 ppm)compared with neem or pepper mint oil nano-emulsion and bulk neem for the second larval instar.The different formulations of neem are more potent than in case of peppermint oil,as LC50 and LC90 values were significantly lower.The same trend was found concerning the 4th larval instar.Age of treated larvae had a detrimental effect on the response to the compounds tested.It was noticed that the younger larvae were much more sensitive to the prepared compounds compared to the older ones.The least LC50 value for loaded neem nano-emulsion was 6.68 ppm compared with the highest value for bulk neem oil(16.68 ppm).Also,LC90 values followed the same trend as in case ofLC50.Again,the toxicity of loaded peppermint oil had the most insecticidal activity as expressed by the lowest LC50 value(51.9 ppm)with more insecticidal effect than the bulk(125.43 ppm)or nano-emulsion(85.43 ppm).The present results indicated that these novel systems could be used in integrated pest management program for A.ipsilon control.展开更多

Methods: CCK8 method was used to detect the cell survival rate of H9C2 cardiomyocytes treated with different concentrations of H2O2. Normal cultured cells were used as the blank control group, and the cells were treated with 200 μmol/L H2O2 for 24 h. An oxidative stress injury model was constructed as the model group. The cells were pretreated with 1%, 0.1% and 0.01% camellia oil for 24 h, and then H2O2 was added for 24 h as the experimental group. The β-galactosidase senescence staining assay, mitochondrial membrane potential assay, EdU cell proliferation staining assay and scratch assay were used to observe the changes of cell senescence, mitochondrial membrane potential, proliferation, apoptosis and migration in each group. The superoxide dismutase (SOD) activity, lactate dehydrogenase (LDH) activity, and malondialdehyde (MDA) content of the cells in each group were detected by using the kit. Results: The cell viability of H9C2 cardiomyocytes treated with different concentrations of H2O2 was inhibited and positively correlated with the concentration of H2O2 (P<0.01). Compared with the blank control group, the positive rate of cell senescence, MDA content and LDH activity increased in the H2O2 model group (P<0.01);mitochondrial membrane potential, cellular value-added rate, migration rate and SOD activity decreased (P<0.01). Compared with the H2O2 model group, the positive rate of cellular senescence (P<0.01 or P<0.05), MDA content and LDH activity decreased (P< 0.01 or P<0.05);mitochondrial membrane potential increased, cell proliferation rate and migration rate increased (P<0.01 or P<0.05) in the experimental group. Conclusion: Camellia oil can significantly inhibit oxidative stress injury in H9C2 cells and exert cardiomyocyte protective effects.展开更多

Methods: The rats were assigned to eight groups, seven rats in each, and were injected intraperitoneally with vehicle, a single dose of ethane dimethane sulfonate (EDS) (75 mg/kg bodyweight), vit E (100 mg/kg bodyweight) or EDS + vit E for 3-7 days. Thereafter, the rats were weighed, anaesthetized with ether and killed by cervical dislocation. The left testis weights were recorded and the relative testis weights were calculated. The left testes were processed for routine paraffin embedding. Three right testes from each group were taken randomly and then processed for routine electron microscopy. Tissue sections were examined using light and electron microscopy, and were scored for histopathological changes. Results: Vit E coadministration did not prevent the bodyweight loss on days 3 and 7. However, vit E administration prevented the EDS-induced testicular-weight loss in rats that received vit E for 3 days but not 7 days. The relative testis weight was higher on day 3 (instead of on day 7) than other groups. Nevertheless, the testis histology was not markedly protected by vit E in the EDS-treated rats. Detailed microscopic assessment showed few Leydig cells and abundant fibroblast-like cells indicating only some protection. Conclusion: Vit E cotreatment showed partial protective effects on the testicular weight and testicular histology in rats that received EDS.展开更多

Then, urinary N- acetyl-β-D- glucosaminidase (NAG ), uri -nary cadmium and bloocl cadmium were measured. The accumulation of cadmium in the kid-ney cortex, content of renal calmodulin, hemoglobin and the ultrastructural damage of proxi-mal convoluted tubules of rats were examined three weeks after the last administration. Re-sults indicated that the calcium antagonists partly protected against toxic effects induced bycadmium in different manners. These data provide further evidence for the new hypothesisthat the cross effect of cadmium and calcium in calmodulin regulated systems may be responsi-ble for the mechanism of cadmium intoxication. The effects of chromium and tin on survival, growth, carbon fixation, nitrate reduction, ammonia assimilation, and nitrogenase activity of a N_2-fixing cyanobacterium, Anabaena doliolum, and their amelioration by synt...The effects of chromium and tin on survival, growth, carbon fixation, nitrate reduction, ammonia assimilation, and nitrogenase activity of a N_2-fixing cyanobacterium, Anabaena doliolum, and their amelioration by synthetic and natural complexans, viz., EDTA, nitrilotriacetic acid (NTA), pyridine dicarboxylic acid (PDA), and citrate, have been studied. Chromium proved to be much more toxic than tin, as it inhibited growth yield (49%), carbon fixation (53%), and nitrate reductase (79%), glutamine synthetase (30%), and nitrogenase activities (77%) at its sublethal concentration, whereas tin induced less inhibition of growth yield (42%), carbon fixation (50%). and nitrate reductase (66%), glutamine synthetase (32.4%), and nitrogenase activities (70%). Despite its inhibitory effects at 10μml^(-1), EDTA supplementation in metal-spiked medium counteracted the toxicity of chromium and tin more significantly than NTA, PDA, and citrate. When supplemented with LD_(50) of Cr, EDTA protected growth, carbon fixation, NR, GS. and Noase, respectively, by 32.6, 50.0, 33.3. 17.7. and 65.4%. However, EDTA-induced restoration of the above parameters at a sublethal concentration of tin was only 30.2, 50.0,28.1, 27.7, and 61.5%, respectively. Although NTA and citrate at 10/μgml^(-1) each were stimulatory to various processes of test cyanobacterium, they were comparatively less effective than EDTA in the amelioration of metal toxicity. On the basis of these observations, a generalized hierarchical sequence of protective efficiency of synthetic and natural cornplexing ligands may be given as EDTA > NTA > citrate > PDA. It seems plausible that the toxicity of various heavy metals may be regulated by a large array of organic complexing agents of the aquatic environment because they possess various metal binding sites. In the present study, the chemical composition, antinociceptive effect and acute toxicity of essential oils(EOs) of Asarum heterotropoides Fr. Schmidt var. mandshuricum(Maxim.) Kitag.(AHM), A. sieboldii Miq. var...In the present study, the chemical composition, antinociceptive effect and acute toxicity of essential oils(EOs) of Asarum heterotropoides Fr. Schmidt var. mandshuricum(Maxim.) Kitag.(AHM), A. sieboldii Miq. var. seoulense Nakai(ASS) and A. himalaicum Hook. f. et Thoms. ex Klotzsch.(AH) were comparatively evaluated. A total of 55 compounds were identified in EOs of AHM, ASS and AH by GC-MS. Methyleugenol(20.16%–62.89%), safrole(2.67%–32.42%), 3,5-dimethoxytoluene(2.00%–18.59%) and eucarvone(1.52%–19.16%) were the major constituents of EO of AHM, and methyleugenol(48.35%–61.06%), eucarvone(11.13%–13.93%) and elemicin(4.79%–11.14%) were the major constituents of EO of ASS. The EO of AH was different from that of AHM and ASS, in which patchouli alcohol(27.42%–51.95%) and elemicin(13.11%–42.23%) were found in a greater amount. Moreover, the antinociceptive effect of EOs of AHM(5.5, 11.0, 16.5 μL/kg) and AH(2.0, 4.0, 6.0 μL/kg) was comparatively assayed in acetic acid-induced writhing, hot plate and formalin tests. The results indicated a weak central, but potent peripheral antinociceptive effect of EO of AHM, and more potent central and peripheral antinociceptive effect of EO of AH. The LD50 of the EOs of AHM and AH were 1.7 and 7.7 mL/kg, respectively. These findings suggest that EOs of AHM and AH possess evident antinociceptive activity and are probably safe within the range of its clinical doses. However, their chemical compositions are quite different. Therefore, AH can be clinically used as an herbal medicinal product with broad analgesic effects, but should not be confused with AHM and ASS used in traditional Chinese medicine.展开更多

It was found that abundant toxic gas (H2S) is generated in thermal recovery of heavy oil. The H2S gas is mainly formed during thermochemical sulfate reduction (TSR) occurring in oil reservoirs or the thermal decomposition of sulfocompounds (TDS) in crude oil. H2S generation is controlled by thermal recovery time, temperature and the injected chemical compounds. The quantity of SO4^2- in the injected compounds is the most influencing factor for the rate of TSR reaction. Therefore, for prevention of H2S formation, periodic and effective monitoring should be undertaken and adequate H2S absorbent should also be provided during thermal recovery of heavy oil. The result suggests that great efforts should be made to reduce the SO4^2- source in heavy oil recovery, so as to restrain H2S generation in reservoirs. In situ burning or desulfurizer adsorption are suggested to reduce H2S levels. Prediction and prevention of H2S are important in heavy oil production. This will minimize environmental and human health risks, as well as equipment corrosion.展开更多

Clarke (Apiaceae) is o...Plant secondary metabolites play an important role in plant-insect interactions and therefore such compounds may have insecticidal or antifeedant activity against insects. Carum copticum C. B. Clarke (Apiaceae) is one of these plants that have medicinal effects on humans. The chemical composition of the essential oil from dry seeds of C. copticum was studied by gas chromatography (GC) and gas chromatography mass spectrometry (GC-MS). Thymol (41.34%),α-terpinolene (17.46%) and ρ-cymene (11.76%) were found to be the major constituents of the oil. In fumigant toxicity tests with the essential oil against adults of Sitophilus oryzae (L.) and Tribolium castaneum (Herbst) at 27 ±1℃ and 60% ± 5% RH, it was observed that S. oryzae (LC50 = 0.91 μL/L) were significantly susceptible than T. castaneum (LC50 = 33.14 μL/L). The mortalities of the insect species reached 100% at concentrations higher than 185.2 μL/L and 12-h exposure time. The findings indicate the strong insecticidal activity of C. copticum oil and its potential role as a fumigant for storedproduct insects.展开更多

Methods: CCK8 method was used to detect the cell survival rate of H9C2 cardiomyocytes treated with different concentrations of H2O2. Normal cultured cells were used as the blank control group, and the cells were treated with 200 μmol/L H2O2 for 24 h. An oxidative stress injury model was constructed as the model group. The cells were pretreated with 1%, 0.1% and 0.01% camellia oil for 24 h, and then H2O2 was added for 24 h as the experimental group. The β-galactosidase senescence staining assay, mitochondrial membrane potential assay, EdU cell proliferation staining assay and scratch assay were used to observe the changes of cell senescence, mitochondrial membrane potential, proliferation, apoptosis and migration in each group. The superoxide dismutase (SOD) activity, lactate dehydrogenase (LDH) activity, and malondialdehyde (MDA) content of the cells in each group were detected by using the kit. Results: The cell viability of H9C2 cardiomyocytes treated with different concentrations of H2O2 was inhibited and positively correlated with the concentration of H2O2 (P<0.01). Compared with the blank control group, the positive rate of cell senescence, MDA content and LDH activity increased in the H2O2 model group (P<0.01);mitochondrial membrane potential, cellular value-added rate, migration rate and SOD activity decreased (P<0.01). Compared with the H2O2 model group, the positive rate of cellular senescence (P<0.01 or P<0.05), MDA content and LDH activity decreased (P< 0.01 or P<0.05);mitochondrial membrane potential increased, cell proliferation rate and migration rate increased (P<0.01 or P<0.05) in the experimental group. Conclusion: Camellia oil can significantly inhibit oxidative stress injury in H9C2 cells and exert cardiomyocyte protective effects.
文摘Aim: To evaluate the protective/ameliorative effects of vitamin E (vit E) on ethane dimethane sulfonate (EDS) induced testicular toxicity in rats. Methods: The rats were assigned to eight groups, seven rats in each, and were injected intraperitoneally with vehicle, a single dose of ethane dimethane sulfonate (EDS) (75 mg/kg bodyweight), vit E (100 mg/kg bodyweight) or EDS + vit E for 3-7 days. Thereafter, the rats were weighed, anaesthetized with ether and killed by cervical dislocation. The left testis weights were recorded and the relative testis weights were calculated. The left testes were processed for routine paraffin embedding. Three right testes from each group were taken randomly and then processed for routine electron microscopy. Tissue sections were examined using light and electron microscopy, and were scored for histopathological changes. Results: Vit E coadministration did not prevent the bodyweight loss on days 3 and 7. However, vit E administration prevented the EDS-induced testicular-weight loss in rats that received vit E for 3 days but not 7 days. The relative testis weight was higher on day 3 (instead of on day 7) than other groups. Nevertheless, the testis histology was not markedly protected by vit E in the EDS-treated rats. Detailed microscopic assessment showed few Leydig cells and abundant fibroblast-like cells indicating only some protection. Conclusion: Vit E cotreatment showed partial protective effects on the testicular weight and testicular histology in rats that received EDS.

文摘Protective effects of calcium antagonists, chlorpromazine (CPZ) and nimodepine (NI-MO), on cadmium-induced toxicity were investigated. After giving CdCl2 (0. 44mg Cd/kg,ip), CPZ (5mg/kg, ip) or NIMO (8mg/kg, po) were administered every day to Sprague-Dawley (S. D. ) rats for a week. Then, urinary N- acetyl-β-D- glucosaminidase (NAG ), uri -nary cadmium and bloocl cadmium were measured. The accumulation of cadmium in the kid-ney cortex, content of renal calmodulin, hemoglobin and the ultrastructural damage of proxi-mal convoluted tubules of rats were examined three weeks after the last administration. Re-sults indicated that the calcium antagonists partly protected against toxic effects induced bycadmium in different manners. These data provide further evidence for the new hypothesisthat the cross effect of cadmium and calcium in calmodulin regulated systems may be responsi-ble for the mechanism of cadmium intoxication. 文摘The effects of chromium and tin on survival, growth, carbon fixation, nitrate reduction, ammonia assimilation, and nitrogenase activity of a N_2-fixing cyanobacterium, Anabaena doliolum, and their amelioration by synthetic and natural complexans, viz., EDTA, nitrilotriacetic acid (NTA), pyridine dicarboxylic acid (PDA), and citrate, have been studied. Chromium proved to be much more toxic than tin, as it inhibited growth yield (49%), carbon fixation (53%), and nitrate reductase (79%), glutamine synthetase (30%), and nitrogenase activities (77%) at its sublethal concentration, whereas tin induced less inhibition of growth yield (42%), carbon fixation (50%). and nitrate reductase (66%), glutamine synthetase (32.4%), and nitrogenase activities (70%). Despite its inhibitory effects at 10μml^(-1), EDTA supplementation in metal-spiked medium counteracted the toxicity of chromium and tin more significantly than NTA, PDA, and citrate. When supplemented with LD_(50) of Cr, EDTA protected growth, carbon fixation, NR, GS. and Noase, respectively, by 32.6, 50.0, 33.3. 17.7. and 65.4%. However, EDTA-induced restoration of the above parameters at a sublethal concentration of tin was only 30.2, 50.0,28.1, 27.7, and 61.5%, respectively. Although NTA and citrate at 10/μgml^(-1) each were stimulatory to various processes of test cyanobacterium, they were comparatively less effective than EDTA in the amelioration of metal toxicity. On the basis of these observations, a generalized hierarchical sequence of protective efficiency of synthetic and natural cornplexing ligands may be given as EDTA > NTA > citrate > PDA. It seems plausible that the toxicity of various heavy metals may be regulated by a large array of organic complexing agents of the aquatic environment because they possess various metal binding sites. 文摘An Ayurvedic medicine, Liv-52 has been studied as a prophylactic against beryllium induced toxicity in rats. Administration of beryllium per se caused severe degenerative and necrotic changes in liver, kidney, lungs and spleen. Its administration reduced glycogen content, activity of alkaline phosphatase and adenosine triphosphatase in these oreans. 文摘Introduction Lightning can cause great destruction. According to the record in the northern China, more than 150 lightning strokes occur on a power system annually. High voltage (millions of volts) and large current (hundreds of -kA) associated with lightning, will damage property and even kill people and livestocks. It will be very dangerous if a lightning stroke strikes oil depot of power plant or substation. In addition, the high voltage induced by the electrostatic charge generated due to oil flowing will probably cause damage unless the electrification has been limited and drained off.

文摘T-2 toxin is one of the most widespread and toxic fungal toxins in food and feed.It can cause gastrointestinal toxicity,hepatotoxicity,immunotoxicity,reproductive toxicity,neurotoxicity,and nephrotoxicity in humans and animals.T-2 toxin is physicochemically stable and does not readily degrade during food and feed processing.Therefore,suppressing T-2 toxin-induced organ toxicity through antidotes is an urgent issue.Protective agents against the organ toxicity of T-2 toxin have been recorded widely in the literature,but these protective agents and their molecular mechanisms of detoxification have not been comprehensively summarized.In this review,we provide an overview of the various protective agents to T-2 toxin and the molecular mechanisms underlying the detoxification effects.Targeting appropriate targets to antagonize T-2 toxin toxicity is also an important option.This review will provide essential guidance and strategies for the better application and development of T-2 toxin antidotes specific for organ toxicity in the future.

文摘In the present study, the chemical composition, antinociceptive effect and acute toxicity of essential oils(EOs) of Asarum heterotropoides Fr. Schmidt var. mandshuricum(Maxim.) Kitag.(AHM), A. sieboldii Miq. var. seoulense Nakai(ASS) and A. himalaicum Hook. f. et Thoms. ex Klotzsch.(AH) were comparatively evaluated. A total of 55 compounds were identified in EOs of AHM, ASS and AH by GC-MS. Methyleugenol(20.16%–62.89%), safrole(2.67%–32.42%), 3,5-dimethoxytoluene(2.00%–18.59%) and eucarvone(1.52%–19.16%) were the major constituents of EO of AHM, and methyleugenol(48.35%–61.06%), eucarvone(11.13%–13.93%) and elemicin(4.79%–11.14%) were the major constituents of EO of ASS. The EO of AH was different from that of AHM and ASS, in which patchouli alcohol(27.42%–51.95%) and elemicin(13.11%–42.23%) were found in a greater amount. Moreover, the antinociceptive effect of EOs of AHM(5.5, 11.0, 16.5 μL/kg) and AH(2.0, 4.0, 6.0 μL/kg) was comparatively assayed in acetic acid-induced writhing, hot plate and formalin tests. The results indicated a weak central, but potent peripheral antinociceptive effect of EO of AHM, and more potent central and peripheral antinociceptive effect of EO of AH. The LD50 of the EOs of AHM and AH were 1.7 and 7.7 mL/kg, respectively. These findings suggest that EOs of AHM and AH possess evident antinociceptive activity and are probably safe within the range of its clinical doses. However, their chemical compositions are quite different. Therefore, AH can be clinically used as an herbal medicinal product with broad analgesic effects, but should not be confused with AHM and ASS used in traditional Chinese medicine.

文摘The distribution and treatment of harmful gas (H2S) in the Liaohe Oilfield, Northeast China, were investigated in this study. It was found that abundant toxic gas (H2S) is generated in thermal recovery of heavy oil. The H2S gas is mainly formed during thermochemical sulfate reduction (TSR) occurring in oil reservoirs or the thermal decomposition of sulfocompounds (TDS) in crude oil. H2S generation is controlled by thermal recovery time, temperature and the injected chemical compounds. The quantity of SO4^2- in the injected compounds is the most influencing factor for the rate of TSR reaction. Therefore, for prevention of H2S formation, periodic and effective monitoring should be undertaken and adequate H2S absorbent should also be provided during thermal recovery of heavy oil. The result suggests that great efforts should be made to reduce the SO4^2- source in heavy oil recovery, so as to restrain H2S generation in reservoirs. In situ burning or desulfurizer adsorption are suggested to reduce H2S levels. Prediction and prevention of H2S are important in heavy oil production. This will minimize environmental and human health risks, as well as equipment corrosion.
文摘Plant secondary metabolites play an important role in plant-insect interactions and therefore such compounds may have insecticidal or antifeedant activity against insects. Carum copticum C. B. Clarke (Apiaceae) is one of these plants that have medicinal effects on humans. The chemical composition of the essential oil from dry seeds of C. copticum was studied by gas chromatography (GC) and gas chromatography mass spectrometry (GC-MS). Thymol (41.34%),α-terpinolene (17.46%) and ρ-cymene (11.76%) were found to be the major constituents of the oil. In fumigant toxicity tests with the essential oil against adults of Sitophilus oryzae (L.) and Tribolium castaneum (Herbst) at 27 ±1℃ and 60% ± 5% RH, it was observed that S. oryzae (LC50 = 0.91 μL/L) were significantly susceptible than T. castaneum (LC50 = 33.14 μL/L). The mortalities of the insect species reached 100% at concentrations higher than 185.2 μL/L and 12-h exposure time. The findings indicate the strong insecticidal activity of C. copticum oil and its potential role as a fumigant for storedproduct insects.
