Receptor tyrosine kinase-like orphan receptor 1(ROR1)is a member of the type I receptor tyrosine kinase family.ROR1 is pivotal in embryonic development and cancer,and serves as a biomarker and therapeutic target.It has soluble and membrane-bound subtypes,with the latter highly expressed in tumors.ROR1 is conserved throughout evolution and may play a role in the development of gastrointestinal cancer through multiple signaling pathways and molecular mechanisms.Studies suggest that overexpression of ROR1 may increase tumor invasiveness and metastasis.Additionally,ROR1 may regulate the cell cycle,stem cell characteristics,and interact with other signaling pathways to affect cancer progression.This review explores the structure,expression and role of ROR1 in the development of gastrointestinal cancers.It discusses current antitumor strategies,outlining challenges and prospects for treatment. 展开更多

Objective:To explore the effect of Bushen Huoxue Decoction(补肾活血饮,BHD) on the orphan receptor(Nurr1) and tyrosine hydroxylase(TH) in the brain of rats with Parkinson's disease(PD).Methods:One hundred and twenty SD rats were divided into 100 in the model group and 20 in the normal control group,fifty-eight SD rats from the model group,established into PD model successfully by injuring their substantia nigra (SSN) with 6-hydroxydopamine,were divided equally into the model group and the test group,and they were treated with saline and BHD,respectively,for eight successive weeks.The change in the rats' behavior before and after treatment was observed by counting the cycles of rotation induced by apomorphine injection; the pathology of neurons,level of Nurr1 mRNA expression,and amount of TH positive cells in SSN were observed after treatment.Results:The rats' behavior was improved in the tested group significantly,the rotation cycle after treatment being 84.0±20.0 cycles/40 min,which was significantly lower than that in the model group(377.0±62.3 cycles/40 min,P〈0.01).Besides,the Nurr1 mRNA expression and TH positive cell in the test group were 0.97±0.15 and 49.40±14.72,respectively,which were significantly higher than those in the model group,0.22±0.03 and 5.45±2.58,respectively(all P〈0.01).Conclusion:BHD could treat PD by enhancing the Nurr1 mRNA expression,increasing the TH content in brain,and promoting the repairing of injured neuron in cerebral SSN. 展开更多

Expression and cellular localization of orphan receptor TR2 mRNA in relation to germ cell apoptosis in cryptorchid testes of rat and rhesus monkey have been studied by using in situ hybridization and in situ 3’-end labeling of DNA fragments (T躈EL). The results show that: (i) TR2 mRNA is specifically expressed in the germ cells, mainly in the spermatocytes, round and elongated spermatids. The expression level of TR2 mRNA varies with the seminiferous cycle. (ii) in the rat cryptorchid testes on days 3 and 5 after the surgery, the germ cells began to undergo apoptosis with no evident decrease in TR2 mRNA level. On day 7.5, however, most germ cells underwent apoptosis, while the expression level of TR2 mRNA declined markedly, and TR2 mRNA was rarely expressed on day 10 thereafter. (iii) On days 15 and 20 of the cryptorchid testes of rhesus monkey, TR2 mRNA was only expressed in a few of primary spermatocytes and the mRNA was almost undetectable on days 30, 45, 60. The cellular localization and expression of TR2 mRNA in rhesus monkey testis were investigated by the methods of in situ and Northern hybridization using the digoxigenin labeled in vitro transcripted cRNA probe from a TR2 cDNA fragment template and α-<sup>32</sup>P-dCTP labeled cDNA probe from the same TR2 cDNA fragment. It is demonstrated that TR2 mRNA was localized specifically in the germ cells, and was predominantly expressed in the meiotic more advanced cells. The expression level was various in different seminiferous tubules, and there was stronger expression in the tubules where spermatogenesis was vigorously going on. The expression level of TR2 mRNA was low in the immature rhesus monkey testis and increased dramatically in the adult and varied with a seminiferous cycle. These data suggest for the first time that orphan receptor TR2 may have an important function in regulating spermatogenesis at later stages of germ cell development in rhesus monkey. 展开更多

By in situ hybridization, the localization of orphan receptor TR3 mRNA has been observed in early developmental follicles. TR3 mRNA is first expressed in the ovarian interstitial cells on day 2 after birth, and then in granulosa cells (GC) in primary follicles on day 4. The expression level of TR3 mRNA in GC increases following the follicular development. Its higher expression can be observed in the outer layer of GC and inner layer of theca cells (TC) on day 6, where the cells present active proliferation and differentiation. The expression of TR3 is in an increasing manner until the large antral follicles on day 30. The mRNA is only expressed in the healthy, but not atretic follicles in adult rat ovaries. Injection of epidermal growth factor (EGF) has dramatically enhanced its expression in the early stage of developmental follicles. It is therefore suggested that TR3 may play a role in regulating growth and differentiation of ovarian somatic cells in the early stage, and its expression is regulated 展开更多

Objective:This study aimed to investigate the role of receptor tyrosine kinase-like orphan receptor 2(ROR2)in triple-negative breast cancer(TNBC).Methods:ROR2 expression in primary TNBC and metastatic TNBC tissues was analyzed by immunohistochemical staining and PCR.ROR2 expression in TNBC cell lines was detected by PCR and Western blot analysis.The migration,invasion and chemosensitivity of TNBC cells with overexpression or knockdown of ROR2 were examined.Results:ROR2 expression was high in metastatic TNBC tissues.ROR2 knockdown suppressed the migration,invasion and chemoresistance of TNBC cells.ROR2 overexpression in MDA-MB-435 cells promoted the migration,invasion,and chemoresistance.Moreover,ROR2 knockdown in HC1599 and MDA-MB-435 adriamycin-resistant cells enhanced chemosensitivity to adriamycin.ROR2 could activate PI3K/AKT/mTOR signaling in TNBC cells.Conclusion:ROR2 is upregulated and promotes metastatic phenotypes of TNBC by activating PI3K/AKT/mTOR signaling. 展开更多

牙周组织SOCS6、RORA表达水平对牙周炎患者的预后价值

目的探讨细胞因子信号转导抑制因子6(SOCS6)和维甲酸受体相关孤核受体A(RORA)在牙周炎患者牙周组织中的表达,并分析二者对患者的预后价值。方法选取2021年7月至2023年1月在济南市中医医院接受治疗的224例牙周炎患者作为研究组,另选取22... 目的探讨细胞因子信号转导抑制因子6(SOCS6)和维甲酸受体相关孤核受体A(RORA)在牙周炎患者牙周组织中的表达,并分析二者对患者的预后价值。方法选取2021年7月至2023年1月在济南市中医医院接受治疗的224例牙周炎患者作为研究组,另选取224例到济南市中医医院门诊因正畸治疗需要拔除健康牙齿的患者作为对照组。收集其牙周组织并采用实时荧光定量PCR检测牙周组织中SOCS6、RORA表达水平。对牙周炎患者随访3个月,根据随访结果将其分为预后良好组(148例)与预后不良组(76例)。采用多因素Logistic回归分析牙周炎患者预后不良的影响因素,采用受试者工作特征(ROC)曲线分析牙周组织SOCS6、RORA表达水平对牙周炎患者预后不良的预测价值。结果与对照组比较,研究组牙周组织SOCS6、RORA表达水平均降低(P<0.05)。224例牙周炎患者均完成随访,预后不良发生率为33.93%(76/224)。预后不良组牙周组织SOCS6、RORA表达水平均低于预后良好组(P<0.05)。多因素Logistic回归分析显示,患牙位于下颌、患牙为前牙、患牙最深探诊深度、牙周炎严重程度为重度、根形态异常、牙槽骨高度、平均附着丧失均是导致牙周炎患者预后不良的危险因素(P<0.05),牙周组织SOCS6、RORA表达水平均是保护因素(P<0.05)。牙周组织SOCS6、RORA单独预测牙周炎患者预后不良的曲线下面积(AUC)分别为0.833、0.829,二者联合预测的AUC(0.914)高于SOCS6与RORA单独预测(Z=3.659、3.408,P<0.001)。结论牙周炎预后不良患者牙周组织SOCS6、RORA表达水平均降低,二者均对牙周炎患者预后不良具有一定预测效能,且二者联合预测的效果更优。 展开更多

目的 探讨慢性牙周炎(CP)患者血清维甲酸受体相关孤儿受体A(RORA)的表达水平及其与辅助性T细胞17(Th17)/调节性T细胞(Treg)平衡的相关性。方法 选取2019年1月—2021年12月于本院进行检查并接受治疗的90例CP患者作为观察组,并选择同期在... 目的 探讨慢性牙周炎(CP)患者血清维甲酸受体相关孤儿受体A(RORA)的表达水平及其与辅助性T细胞17(Th17)/调节性T细胞(Treg)平衡的相关性。方法 选取2019年1月—2021年12月于本院进行检查并接受治疗的90例CP患者作为观察组,并选择同期在本院检查的牙周健康志愿者100例作为对照组,比较两组的年龄、性别、体质指数(BMI)及合并高血压、冠心病、糖尿病等一般临床资料;采用实时荧光定量PCR(qRT-PCR)检测两组血清中RORA的相对表达水平;采用流式细胞术检测两组Th17、Treg细胞水平;采用酶联免疫吸附(ELISA)法检测两组血清中转化生长因子-β(TGF-β)、白细胞介素-10(IL-10)、白细胞介素-17(IL-17)及白细胞介素-23(IL-23)的水平;采用Pearson法分析CP患者血清RORA与TGF-β、IL-10、IL-23、IL-23及Th17/Treg细胞比例之间的相关性;Logistic回归分析影响CP发生的因素。结果 两组年龄、性别、BMI及合并高血压、冠心病、糖尿病等指标比较,差异均无统计学意义(P>0.05)。观察组患者血清中Th17、IL-17、IL-23水平高于对照组,RORA、Treg、TGF-β、IL-10水平低于对照组,且Th17/Treg细胞比例高于对照组(P<0.05)。Pearson法分析结果显示,CP患者血清中RORA与TGF-β、IL-10呈正相关(P<0.05),与IL-17、IL-23及Th17/Treg细胞比例呈负相关(P<0.05)。Logistic回归分析结果显示,RORA、TGF-β、IL-10、IL-17、IL-23及Th17/Treg是CP发生的影响因素(P<0.05)。结论 CP患者血清中RORA低表达,Th17/Treg细胞比例失衡,RORA的表达与Th17/Treg细胞比例之间呈负相关,RORA可能通过调节Th17/Treg细胞比例影响CP的发生。 展开更多

Pancreatic ductal adenocarcinoma(PDAC)is a devastating disease with a median overall survival time of5 mo and the five years survival less than 5%,a rate essentially unchanged over the course of the years.A well defined progression model of accumulation of genetic alterations ranging from single point mutations to gross chromosomal abnormalities has been introduced to describe the origin of this disease.However,due to the its subtle nature and concurring events PDAC cure remains elusive.Nuclear receptors(NR)are members of a large superfamily of evolutionarily conserved ligand-regulated DNA-binding transcription factors functionally involved in important cellular functions ranging from regulation of metabolism,to growth and development.Given the nature of their ligands,NR are very tempting drug targets and their pharmacological modulation has been widely exploited for the treatment of metabolic and inflammatory diseases.There are now clear evidences that both classical ligand-activated and orphan NR are involved in the pathogenesis of PDAC from its very early stages;nonetheless many aspects of their role are not fully understood.The purpose of this review is to highlight the striking connections that link peroxisome proliferator activated receptors,retinoic acid receptors,retinoid X receptor,androgen receptor,estrogen receptors and the orphan NR Nur,chicken ovalbumin upstream promoter transcription factorⅡand the liver receptor homologue-1 receptor to PDAC development,connections that could lead to the identification of novel therapies for this disease. 展开更多

Primary human T cells can be modified using viral and non-viral vectors to promote the specific targeting of cancer cells via the introduction of exogenous T-cell receptors(TCRs) or chimeric antigen receptors(CARs). This gene transfer displays the potential to increase the specificity and potency of the anticancer response while decreasing the systemic adverse effects that arise from conventional treatments that target both cancerous and healthy cells. This review highlights the generation of clinical-grade T cells expressing CARs for immunotherapy, the use of these cells to target B-cell malignancies and, particularly, the first clinical trials deploying the Sleeping Beauty gene transfer system, which engineers T cells to target CD19+ leukemia and non-Hodgkin's lymphoma. 展开更多

目的探讨小鼠实验性自身免疫性脑脊髓炎(EAE)模型中,外源性和内源性红细胞膜相关蛋白(ERMAP)通过白细胞介素6/信号转录及转录激活因子3(IL-6/STAT3)/维甲酸相关孤核受体γt/(ROR-γt)信号通路对辅助性T细胞17(Th17)细胞分化的影响。方... 目的探讨小鼠实验性自身免疫性脑脊髓炎(EAE)模型中,外源性和内源性红细胞膜相关蛋白(ERMAP)通过白细胞介素6/信号转录及转录激活因子3(IL-6/STAT3)/维甲酸相关孤核受体γt/(ROR-γt)信号通路对辅助性T细胞17(Th17)细胞分化的影响。方法流式细胞术验证不同浓度ERMAP-Ig融合蛋白功能;琼脂糖凝胶电泳鉴定ERMAP基因敲除小鼠。流式细胞术检测ERMAP-Ig融合蛋白在体外对Th17细胞分化影响。40只6周龄普通C57BL/6小鼠随机分为2组建立EAE模型:对照(control)-Ig和ERMAP-Ig组,每组20只;记录临床评分;流式细胞术检测EAE小鼠体内Th17细胞分化情况。40只6周龄已鉴定的野生型和ERMAP基因敲除小鼠分为2组建立EAE模型:ERMAP^(+/+)和ERMAP^(-/-)组,每组20只。记录临床评分;脊髓HE和固蓝(LFB)染色并进行组织学半定量评分。流式细胞术检测IL-17A+CD4+T细胞百分比;Western blotting检测IL-6、白细胞介素17(IL-17)、STAT3、磷酸化STAT3(p-STAT3)、ROR-γt蛋白水平表达;Real-time PCR检测IL-17、肿瘤坏死因子α(TNF-α)、IL-6、STAT3和ROR-γt的mRNA表达;ELISA检测细胞水平IL-17和TNF-α的表达。结果1.外源性ERMAP-Ig融合蛋白抑制体内外Th17细胞分化且减轻小鼠EAE症状。2.与对照组相比,ERMAP^(-/-)组EAE小鼠炎性浸润和脱髓鞘症状加重,Th17分泌IL-17A增加。3.内源性ERMAP基因敲除后,IL-17、TNF-α、IL-6、STAT3及ROR-γt表达均增加。差异均有统计学意义(P<0.05)。结论ERMAP可能通过IL-6/STAT3/ROR-γt信号通路调控Th17细胞分化,参与小鼠EAE发生发展。 展开更多

目的基于叉头框蛋白P3(FoxP3)、RAR相关孤儿受体γt(RORγt)及相关细胞因子探讨半夏泻心汤对慢性萎缩性胃炎(CAG)的干预机制。方法将48只大鼠随机分为空白组6只和模型制备组42只,模型制备组以脱氧胆酸钠加氨水联合法建立CAG模型。造模... 目的基于叉头框蛋白P3(FoxP3)、RAR相关孤儿受体γt(RORγt)及相关细胞因子探讨半夏泻心汤对慢性萎缩性胃炎(CAG)的干预机制。方法将48只大鼠随机分为空白组6只和模型制备组42只,模型制备组以脱氧胆酸钠加氨水联合法建立CAG模型。造模成功后,除去意外死亡及用于模型验证的12只大鼠,将剩余的30只大鼠随机分为模型组、半夏泻心汤低剂量组、半夏泻心汤中剂量组、半夏泻心汤高剂量组、叶酸组,各6只。给予空白组和模型组大鼠灌胃生理盐水,给予叶酸组大鼠灌胃叶酸,分别按照4.185 g/kg、8.370 g/kg、16.740 g/kg给予半夏泻心汤低剂量、半夏泻心汤中剂量、半夏泻心汤高剂量组大鼠灌胃半夏泻心汤,均连续干预12周。实验期间观察各组大鼠一般情况。给药结束后,取各组大鼠胃黏膜组织观察病理学变化及细胞凋亡情况,并检测FoxP3 mRNA、RORγt mRNA表达水平,取血清标本检测白细胞介素(IL)-10、IL-17、转化生长因子β(TGF-β)含量。结果与空白组相比,模型组大鼠毛色泛黄且光泽度下降,食欲降低,大便稀溏,肛周潮湿,体质量下降,胃黏膜固有层腺体数量减少,凋亡细胞增多,血清IL-10、IL-17、TGF-β含量及胃黏膜组织FoxP3 mRNA、RORγt mRNA表达水平均升高(均P<0.05)。与模型组比较,半夏泻心汤各剂量组和叶酸组大鼠毛色光泽度及大便性状有所改善,食欲增大,肛周毛发干燥,体质量增长,胃黏膜固有层腺体数量不同程度增加,凋亡细胞减少,血清IL-10、IL-17、TGF-β含量及胃黏膜组织FoxP3 mRNA、RORγt mRNA表达水平均下降(均P<0.05)。在半夏泻心汤各剂量组中,半夏泻心汤低剂量组上述指标改善最为明显。结论半夏泻心汤能够有效改善CAG大鼠的临床症状和病理改变,低剂量的半夏泻心汤可获得最佳的干预效果。半夏泻心汤干预CAG的机制可能与其下调大鼠胃黏膜Foxp3、RoRγt的mRNA表达及相关细胞因子的表达有关。 展开更多

目的探讨外周血单个核细胞(PBMC)中视黄酸相关孤儿核受体α(RORα)mRNA、白细胞介素17E(IL-17E)mRNA、叉头框蛋白P3(FOXP3)mRNA和髓源性抑制细胞(MDSC)百分比在直肠癌中的临床价值。方法选取直肠癌患者95例(直肠癌组)、健康体检者45名(... 目的探讨外周血单个核细胞(PBMC)中视黄酸相关孤儿核受体α(RORα)mRNA、白细胞介素17E(IL-17E)mRNA、叉头框蛋白P3(FOXP3)mRNA和髓源性抑制细胞(MDSC)百分比在直肠癌中的临床价值。方法选取直肠癌患者95例(直肠癌组)、健康体检者45名(正常对照组),收集所有研究对象的临床资料,检测所有研究对象PBMC中RORαmRNA、FOXP3 mRNA、IL-17E mRNA水平和MDSC百分比。结果直肠癌组FOXP3 mRNA、IL-17E mRNA相对表达量和MDSC百分比均高于正常对照组(P<0.05),RORαmRNA相对表达量低于正常对照组(P<0.05)。有淋巴转移、TNM分期Ⅲ期的直肠癌患者FOXP3mRNA、IL-17E mRNA相对表达量和MDSC百分比分别高于无淋巴转移、TNM分期Ⅰ~Ⅱ期的患者(P<0.05),RORαmRNA相对表达量分别低于无淋巴转移、TNM分期Ⅰ~Ⅱ期的患者(P<0.05)。不同性别、年龄、肿瘤直径的直肠癌患者之间RORαmRNA、FOXP3 mRNA、IL-17E mRNA相对表达量和MDSC百分比差异均无统计学意义(P>0.05)。结论直肠癌患者IL-17E mRNA、FOXP3 mRNA水平和MDSC显著升高,RORαmRNA水平显著降低,且与TNM分期和淋巴转移有关,或可作为直肠癌筛查和病情监测的指标。 展开更多

目的寻找结肠腺癌(COAD)免疫相关预后标志物,以期能够准确预测COAD患者预后。方法基于癌症基因组图谱(TCGA)数据库分析COAD患者免疫相关差异基因,通过加权基因共表达网络分析(WGCNA)与COX回归分析得到风险模型,根据风险值的中位值将患... 目的寻找结肠腺癌(COAD)免疫相关预后标志物,以期能够准确预测COAD患者预后。方法基于癌症基因组图谱(TCGA)数据库分析COAD患者免疫相关差异基因,通过加权基因共表达网络分析(WGCNA)与COX回归分析得到风险模型,根据风险值的中位值将患者分为高、低风险组,比较两者预后,在基因表达数据集(GEO)数据库中验证模型。结果通过分析共得到1015个免疫相关差异基因。COX回归分析得到由维甲酸受体相关孤核受体C(RORC)、富含亮氨酸重复序列Fli-I相互作用蛋白2(LRRFIP2)、半乳糖凝集素4(LGALS4)构成的风险模型,高风险组较低风险组预后较差并在GEO中得到验证,通过单因素与多因素COX分析发现模型可作为COAD患者独立预后风险模型。结论通过免疫相关预后差异基因建立模型,可以预测COAD患者预后。 展开更多

目的:探讨视黄酸相关孤儿受体α(RORα)对阿霉素(DOX)诱导的心肌损伤的改善作用及机制。方法:采用尾静脉注射RORα过表达慢病毒建立心肌RORα高表达模型。随机将小鼠分为对照组(NC组)、RORα过表达组、DOX组和RORα过表达+DOX组,每组20... 目的:探讨视黄酸相关孤儿受体α(RORα)对阿霉素(DOX)诱导的心肌损伤的改善作用及机制。方法:采用尾静脉注射RORα过表达慢病毒建立心肌RORα高表达模型。随机将小鼠分为对照组(NC组)、RORα过表达组、DOX组和RORα过表达+DOX组,每组20只。其中,DOX组和RORα过表达+DOX组一次性给予15 mg/(kg·d)DOX腹腔注射,NC组和RORα过表达组给予腹腔注射同等体积0.9%氯化钠溶液。2周后,采用超声检测左心室射血分数(LVEF)和左心室短轴缩短率(LVFS);采用相应试剂盒检测血浆血浆肌酸激酶(CK)和乳酸脱氢酶(LDH)活性;采用TUNEL法检测小鼠心肌凋亡情况;采用电镜观察心肌线粒体形态;采用比色法检测心肌活性氧簇(ROS)水平;采用Western blot检测RORα和锰超氧化物歧化酶(MnSOD)蛋白表达水平。结果:与NC组比较,DOX组LVEF和LVFS降低,CK、LDH活性增加,心肌细胞凋亡增加,心肌ROS水平增加,MnSOD蛋白表达降低(均P<0.05),并且心肌线粒体发生损伤。与DOX组比较,RORα过表达+DOX组LVEF和LVFS增加,CK、LDH活性下降,心肌细胞凋亡减少,心肌ROS水平降低,MnSOD蛋白表达增加(均P<0.05),并且心肌线粒体损伤有明显改善。结论:在DOX诱导的心肌损伤中,RORα可能通过调控MnSOD表达发挥改善心肌的作用。 展开更多
