Monkeypox was declared a global health emergency by the World Health Organization,and as of March 2023,86,000 confirmed cases and 111 deaths across 110 countries have been reported.Its causal agent,monkeypox virus(MPV)belongs to a large family of double-stranded DNA viruses,Orthopoxviridae,that also includes vaccinia virus(VACV)and others.MPV produces two distinct forms of viral particles during its replication cycles:the enveloped viron(EV)that is released via exocytosis,and the mature viron(MV)that is discharged through lysis of host cells.This study was designed to develop multi-valent m RNA vaccines against monkeypox EV and MV surface proteins,and examine their efficacy and mechanism of action.Four m RNA vaccines were produced with different combinations of surface proteins from EV(A35R and B6R),MV(A29L,E8L,H3L and M1R),or EV and MV,and were administered in Balb/c mice to assess their immunogenicity potentials.A dynamic immune response was observed as soon as seven days after initial immunization,while a strong Ig G response to all immunogens was detected with ELISA after two vaccinations.The higher number of immunogens contributed to a more robust total Ig G response and correlating neutralizing activity against VACV,indicating the additive potential of each immunogen in generating immune response and nullifying VACV infection.Further,the m RNA vaccines elicited an antigen-specific CD4^(+)T cell response that is biased towards Th1.The m RNA vaccines with different combinations of EVand MV surface antigens protected a mouse model from a lethal dose VACV challenge,with the EV and MV antigens-combined vaccine offering the strongest protection.These findings provide insight into the protective mechanism of multi-valent m RNAvaccines against MPV,and also the foundation for further development of effective and safe m RNA vaccines for enhanced protection against monkeypox virus outbreak. 展开更多

Chemokines are cytokines that can promote the activation and migration of immune cells,and increase the recognition of antigen by antigen-presenting cells(APC).Previous studies showed that a DNA vaccine can induce humoral and cellular immune responses of flounder after immunization.To explore the improvement of chemokines on the efficiency of OmpK vaccine,two bicistronic DNA candidate vaccines were constructed and the immune responses they induced in the flounder were investigated by reverse transcription polymerase chain reaction(RT-PCR),indirect immunofl uorescent assay(IFA),H&E staining,fl ow cytometry(FCM),and quantifi cational real-time polymerase chain reaction(qRT-PCR).pBudCE4.1 plasmid as an expression vector,bicistronic DNA vaccines encoding OmpK gene and CC-motif ligand 4 gene(p-OmpK-CCL4),or Ompk gene and CC-motif ligand 19 gene(p-OmpK-CCL19)were successfully constructed.The results showed that two bicistronic DNA vaccines expressed Ompk protein of Vibrio anguillarum and CCL4/CCL19 proteins of fl ounder both in vitro and in vivo.After immunization,a large number of leucocytes in muscle were recruited at the injection site in treatment groups.The constructed vaccines induced signifi cant increases in CD4-1^(+) and CD4-2^(+) T lymphocytes,and sIgM^(+) B lymphocytes in peripheral blood,spleen,and head kidney.The percentage of T lymphocytes peaked on the 14^(th) post-vaccination day whereas that of B lymphocytes peaked in the 6^(th) post-vaccination week.Moreover,the expression profi les of 10 immune-related genes increased in muscles around the injection site,spleen,and head kidney.After the challenge,p-OmpK-CCL4 and p-OmpK-CCL19 conferred a relative percentage survival(RPS)of 74.1%and 63.3%,respectively,higher than p-OmpK alone(40.8%).In conclusion,both CCL4 and CCL19 can improve the protection of p-OmpK via evoking local immune response and then humoral and cellular immunity.CCL4 and CCL19 will be potential molecular adjuvants for use in DNA vaccines. 展开更多

Flavobacterium columnare causes columnaris disease in freshwater fi sh. In the present study, the antigenic regions of fi ve outer membrane proteins(OMPs), including zinc metalloprotease, prolyl oligopeptidase, thermolysin, collagenase and chondroitin AC lyase, were bioinformatically analyzed, fused together, and then expressed as a recombinant fusion protein in Escherichia coli. The expressed protein of 95.6 k Da, as estimated by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis, was consistent with the molecular weight deduced from the amino acid sequence. The purifi ed recombinant protein was used to vaccinate the grass carp, C tenopharyngodon idella. Following vaccination of the fi sh their Ig M antibody levels were examined, as was the expression of I g M, Ig D and Ig Z immunoglobulin genes and other genes such as MHC Iα and MHC I I β, which are also involved in adaptive immunity. Interleukin genes( IL), including I L- 1β, IL- 8 and I L- 10, and type I and type II interferon(I FN) genes were also examined. At 3 and 4 weeks post-vaccination(wpv), signifi cant increases in Ig M antibody levels were observed in the fi sh vaccinated with the recombinant fusion protein, and an increase in the expression levels of I g M, Ig D and Ig Z genes was also detected following the vaccinations, thus indicating that an adaptive immune response was induced by the vaccinations. Early increases in the expression levels of IL and IFN genes were also observed in the vaccinated fi sh. At four wpv, the fi sh were challenged with F. column a re, and the vaccinated fi sh showed a good level of protection against this pathogen, with 39% relative percent survival(RPS) compared with the control group. It can be concluded, therefore, that the fi ve OMPs, in the form of a recombinant fusion protein vaccine, induced an immune response in fi sh and protection against F. columnare. 展开更多

本文报告了含有赖型和七日热型抗原的钩端螺旋体(简称钩体)外膜菌苗免疫400余人,其中100μg组免疫200余人,200μg组免疫200余人,同时设菌体菌苗和安慰剂组各200余人作为对照。结果反应轻微,仅个别对象出现轻... 本文报告了含有赖型和七日热型抗原的钩端螺旋体(简称钩体)外膜菌苗免疫400余人,其中100μg组免疫200余人,200μg组免疫200余人,同时设菌体菌苗和安慰剂组各200余人作为对照。结果反应轻微,仅个别对象出现轻微的体温升高和局部红肿反应,无强反应发生。一针免疫后抗体显著升高,阳转率黄疸出血群赖型达98.13%以上,七日热群七日热型达96.26%以上,GMT分别为227.09和50.83以上,较菌体菌苗二针免疫抗体高2倍以上,且免疫后3个月下降亦较菌体菌苗为缓。100μg免疫后反应更为轻微,抗体水平满意,建议以每型100μg外膜苗扩大使用。 展开更多

Hepatitis C virus(HCV) infection is still a major public health problem worldwide since its first identification in 1989. Hepatitis C virus(HCV) infection is still a major public health problem worldwide since its first identification in 1989. At the start, HCV infection was post-transfusion viral infection, particularly in developing countries. Recently, due to iv drug abuse, HCV infection became number one health problem in well-developed countries as well. Following acute HCV infection, the innateimmune response is triggered in the form of activated coordinated interaction of NK cells, dendritic cells and interferon α. The acquired immune response is then developed in the form of the antibody-mediated immune response(ABIR) and the cell-mediated immune response(CMIR). Both are responsible for clearance of HCV infection in about 15% of infected patients. However, HCV has several mechanisms to evade these antivirus immune reactions. The current review gives an overview of HCV structure, immune response and viral evasion mechanisms. It also evaluates the available preventive and therapeutic vaccines that induce innate, ABIR, CMIR. Moreover, this review highlights the progress in recent HCV vaccination studies either in preclinical or clinical phases. The unsatisfactory identification of HCV infection by the current screening system and the limitations of currently available treatments, including the ineligibility of some chronic HCV patients to such antiviral agents, mandate the development of an effective HCV vaccine. 展开更多

Dengue virus(DENV)is a mosquito-borne virus with a rapid spread to humans,causing mild to potentially fatal illness in hundreds of millions of people each year.Due to the large number of serotypes of the virus,there remains an unmet need to develop protective vaccines for a broad spectrum of the virus.Here,we constructed a modified mRNA vaccine containing envelope domain III(E-DIII)and non-structural protein 1(NS1)coated with lipid nanoparticles.This multi-target vaccine induced a robust antiviral immune response and increased neutralizing antibody titers that blocked all four types of DENV infection in vitro without significant antibodydependent enhancement(ADE).In addition,there was more bias for Th1 than Th2 in the exact E-DIII and NS1-specific T cell responses after a single injection.Importantly,intramuscular immunization limited DENV transmission in vivo and eliminated vascular leakage.Our findings highlight that chimeric allogeneic structural and non-structural proteins can be effective targets for DENV vaccine and that they can prevent the further development of congenital DENV syndrome. 展开更多

目的确定致病性问号钩端螺旋体(钩体)属特异性外膜蛋白的抗原性和交叉免疫反应性,为研制通用性钩体基因工程疫苗及检测试剂盒提供依据。方法采用Ni-NTA亲和层析法,提取了4种钩体外膜蛋白主要基因型重组表达产物rLipL21、rLipL32/1和rLi... 目的确定致病性问号钩端螺旋体(钩体)属特异性外膜蛋白的抗原性和交叉免疫反应性,为研制通用性钩体基因工程疫苗及检测试剂盒提供依据。方法采用Ni-NTA亲和层析法,提取了4种钩体外膜蛋白主要基因型重组表达产物rLipL21、rLipL32/1和rLipL32/2、rLipL41/1和rLipL41/2、rOmpL1/1和rOmpL1/2,并用SDS-PAGE检测表达及提纯效果。上述重组蛋白常规皮下免疫家兔获得抗血清,显微镜凝集试验(MAT)检测各抗血清交叉凝集效价。采用盐变法提取了我国15群问号钩体参考标准株及非致病的双曲钩体PatocⅠ株的外膜蛋白。以兔抗血清为一抗,采用Western blot检测上述4种外膜蛋白自然表达情况及其免疫反应性。结果rLipL21、rLipL32/1和rLipL32/2、rLipL41/1和r LipL41/2、rOmpL1/1和rOmpL1/2表达量分别约占细菌总蛋白的10%、40%和35%、15%和10%、30%和15%,各重组蛋白SDS-PAGE提纯后均仅见单一的蛋白条带。重组蛋白兔抗血清在同一基因不同基因型表达产物之间有广泛的交叉免疫反应性,与不同钩体血清群的MAT效价为1:2-1:128。各血清群钩体外膜中均可检出上述4种外膜蛋白,但LipL21仅存在于问号钩体中。结论LipL21、LipL32、LipL41、OmpL,1均为钩体属特异性表面抗原,可作为通用性钩体疫苗及检测试剂盒的候选抗原。 展开更多

目的 观察一种结构新颖的HCV融合抗原DNA疫苗在BALB c小鼠的免疫效果 ,探讨其用于防治丙型肝炎的可行性。方法 用重叠延伸PCR拼接编码小鼠IgGkappa链信号肽和通用型辅助性T细胞表位PADRE的DNA片段 ,PCR分别扩增HCV核心抗原基因和包膜E... 目的 观察一种结构新颖的HCV融合抗原DNA疫苗在BALB c小鼠的免疫效果 ,探讨其用于防治丙型肝炎的可行性。方法 用重叠延伸PCR拼接编码小鼠IgGkappa链信号肽和通用型辅助性T细胞表位PADRE的DNA片段 ,PCR分别扩增HCV核心抗原基因和包膜E2抗原基因 ,将 3段基因插入真核表达载体pcDNA3.1,构成重组表达质粒pST CE2t,转染COS7细胞 ,免疫组化检测HCV抗原的表达。将pST CE2t和HCV核心抗原DNA疫苗pcDNA3.1core分别肌肉注射接种BALB c小鼠 ,检测小鼠的血清抗体、T细胞增殖和CTL反应。结果 pST CE2t可在COS7细胞内表达HCV核心抗原和E2抗原 ,接种于BALB c小鼠能有效诱导体液和细胞免疫应答 ,其中抗HCV核心抗原免疫应答的强度明显超过pcDNA3.1core ,且更趋向于TH1型免疫应答。结论 pST 展开更多
