AISI 316L stainless steel sheets were used to develop double wall angle pyramid with aid of tungsten carbide tool. GRA coupled with PCA was used to plan the experiment conditions. Control factors such as Tool Diameter(TD), Step Depth(SD), Bottom Wall Angle(BWA), Feed Rate(FR) and Spindle Speed(SS) on Top Wall Angle(TWA) and Top Wall Angle Surface Roughness(TWASR) have been studied. Wall angle increases with increasing tool diameter due to large contact area between tool and workpiece. As the step depth, feed rate and spindle speed increase,TWASR decreases with increasing tool diameter. As the step depth increasing, the hydrostatic stress is raised causing severe cracks in the deformed surface. Hence it was concluded that the proposed hybrid method was suitable for optimizing the factors and response.展开更多

The present study demonstrated that the zucchini inbred line ‘19’ consistently flowered early, taking significantly fewer days to bloom the first female flower(DFF) than the inbred line ‘113’. Genetic analysis revealed that DFF, an inheritable quantitative trait, is controlled by multiple genes. Based on the strategy of quantitative trait locus(QTL) sequencing(QTL-seq) combined with linkage analysis, three QTLs for DFF were identified on chromosomes 4, 11, and 20. This study used additional F2 populations grown under different environmental conditions for QTL mapping analysis of DFF with insertion/deletion(InDel) markers to validate these results. Using the composite interval mapping(CIM) method of R/qtl software, we only identified one major locus under all environmental conditions, located in a 117-kb candidate region on chromosome 20. Based on gene annotation, gene sequence alignment, and qRT-PCR analysis, we found that the Cp4.1LG20g08050 gene encoding a RING finger protein may be a candidate gene for the opposite regulation of early flowering in zucchini. In summary, these results lay a foundation for a better understanding of early flowering and improving early flowering-based breeding strategies in zucchini.展开更多

The mutant had sparse branches and decreased number of florets from the base to the peak. Frequently, the florets at the top of the panicle did not develop completely. The underdeveloped florets often showed slender and white in their life cycle. Genetic analysis indicated that the mutant traits were controlled by a single recessive gene (temporarily designated as ah). ah gene controlled the development of inflorescence meristem and the flower organ. The florets of mutant showed degenerated lemma and palea. Stamens and lodicules were homeoticly transformed into pistils and palea/lemma-like structures, respectively. It seemed that ah mutant phenotypes of the homeotic conversions in lodicules and stamens were very similar to that of the B loss-of-function spwl gene reported previously in rice.展开更多

The unconditional and conditional analyses of genetic models and the corresponding statistic methods, including endospermic, cytoplasmic, and maternal plant genetic systems, were used to analyze the genetic relationships between protein content (PC) and the appearance quality traits of indica rice (Oryza sativa L.). The results from unconditional analysis indicated that PC was significantly correlated with the appearance quality traits of rice, except for the brown rice thickness (BRT). Only the genetic covariance between PC and the brown rice width (BRW) was positively correlative, whereas all the other pairwise traits were negatively correlative. The results from conditional analysis revealed that the weight of brown rice (WBR) or the amylose content (AC) could significantly affect the relationships between PC and the appearance quality traits of indica rice. The conditional analysis showed that WBR might negatively affect the relationships between PC and the brown rice length (BRL), BRW, or BRT through the geuotype x environmental (GE) interaction effects, but positively affected the relationships between PC and the ratio of brown rice length to width (RLW) or the ratio of brown rice length to thickness (RLT). The amylase content could positively affect the relationships between PC and BRL, RLW, RLT through the cytoplasmic effects and maternal additive effects, but negatively affected the relationships between PC and BRW.展开更多

[Method] Taking "Sanbei 98" and "Youyan 599" as ex- periment materials, correlation between yield and quality traits of Brassica napus L. cultivars was explored by orthogonal rotation method. [Result] The results showed that: 1 In both the two varieties-Sanbei98 and Youyan599, erucic acid content had the largest coefficient of variation, but its average content was lower than 2% in both the two varieties (0.90% in Sanbei98 and 1.24% in Youyan599), and the palmitic acid content had the smallest coefficient of variation. 2 In terms of the dif- ferences in quality traits at different yield levels, only the mean of eicosenoic acid content revealed significant difference in Sanbei98, and only the mean of seed-pro- tein content revealed significant difference in Youyan599, and there was no signifi- cant difference in any combined quality traits in both the two varieties. 3 The yield was negatively correlated with the erucic acid content or oil content, and posi- tively correlated with the content of seed-protein, linoleic acid, or linolenic acid, and the coefficient of correlation between the yield and either of the contents of glucosi- nolates, oleic acid, palmitic acid, stearic acid, and eicosenoic acid was smaller. ~, when the yield of a variety increased considerably, there was a certain decreasing trend in erucic acid content, oil content, the total amount of erucic chain fatty acids (El & Er), while there was an increasing trend in seed-protein content, linoleic acid content, linolenic acid content, saturated fatty acids (SFA), the total amount of fat and protein (P & F), unsaturation index (IUFA), unsaturated fatty acids (UFA), the total amount of 18 carbon fatty acids (18-C FA), unsaturated fatty acids [UFA/(Ei & Er) ], and the sum of oleic acid and linoleic acid (OI & Li). 5 When the seed-pro- tein content is increased, the quality of rape oil cake is also improved; when the unsaturated index is increased, the oil stability is decreased. The increase in the total amount of unsaturated fatty acids, oleic acid and linoleic acid helps to increase in the total amount of fatty acids absorbed by human bodies. [Conclusion] This study provides a theoretical foundation for the cultivation of high-yielding and top- quality hybrid rape cultivars.展开更多

文摘Shikimic acid/quinic acid hydroxy cinnamyl transferase(HCT)is one of the key enzymes in the phenylpropanoid pathway. However, the role of the HCT gene in chlorogenic acid(CGA)biosynthesis in peach fruit remains unclear. For this,we identified the accumulation pattern of CGA in four peach cultivars, cloned and characterized 11 PpHCT gene members, and further analyzed the expression patterns of these PpHCT genes during fruit development. The contents of CGAs in the four peach cultivars all exhibited a trend of increasing and then decreasing during the fruit growth and development. Moreover, the contents of CGAs in the peel and flesh were tissue-specific.Gene structure analysis indicated that the PpHCT genes were highly conserved, containing two exons and one intron. The protein structure analysis demonstrated that the PpHCT proteins contained two conserved motifs(HXXXD,DFGWG)and a transferase domain(PF02458),which belonged to the BAHD acyltransferase family. The cis-acting element analysis suggested that the promoters of PpHCT genes contained many light-related,hormone-related,stress-related,tissue-specific,and circadian-related elements, and they could participate in a variety of biological processes.Phylogenetic analysis showed that the HCT proteins of peach were closely related to the HCT proteins of plum and had a close evolutionary relationship. The qRT-PCR analysis indicated that the expression levels of PpHCT1 and PpHCT2 showed an opposite trend to the accumulation of CGA,whereas the expression levels of PpHCT4,PpHCT5,PpHCT7,PpHCT8,and PpHCT11 demonstrated the same trend as CGA accumulation. It was worth noting that only PpHCT4 and PpHCT5 were highly expressed in the two high-CGA cultivars but showed low levels of expression in the two low-CGA cultivars. Therefore,it was hypothesized that these two genes might be key genes to the synthesis of CGA in peach fruit. Those findings provide a theoretical basis for further study on the biological functions of the HCT gene and help to reveal the molecular mechanism of CGA.

文摘Early flowering promotes early maturity, production, and the capacity to counteract biotic and abiotic stresses, making it an important agronomic trait in zucchini. In summary, these results lay a foundation for a better understanding of early flowering and improving early flowering-based breeding strategies in zucchini.

文摘A factorial mating design in two environments was conducted using 7 cytoplasmic male sterile lines (A) and 5 restorer lines (R) along with their F1 (A × R) and F2 populations. The amylase content could positively affect the relationships between PC and BRL, RLW, RLT through the cytoplasmic effects and maternal additive effects, but negatively affected the relationships between PC and BRW.

文摘[Objective] This study aimed to investigate the correlation between yield and quality traits of rape. [Conclusion] This study provides a theoretical foundation for the cultivation of high-yielding and top- quality hybrid rape cultivars.

文摘Inter-simple sequence repeat(ISSR) molecular markers were applied to analyze the genetic diversity and clustering of 48 introduced and bred cultivars of Olea euyopaea L. Totally 106 DNA bands were amplified by 11 screened primers, including 99 polymorphic bands; the percentage of polymorphic loci was 93.40%, indicating a rich genetic diversity in Olea euyopaea L. germplasm resources. Based on Nei's genetic distances between various cultivars, a dendrogram of 48 cultivars of Olea euyopaea L. was constructed using unweighted pair-group(UPMGA)method,which showed that 48 cultivars were clustered into four main categories; 84.6% of native cultivars were clustered into two categories; most of introduced cultivars were clustered based on their sources and main usages but not on their geographic origins. This study will provide references for the utilization and further genetic improvement of Olea euyopaea L. germplasm resources.

文摘Both glial cells and glia scar greatly affect the development of spinal cord injury and have become hot spots in research on spinal cord injury treatment. The cellular deposition of dense extracellular matrix proteins such as chondroitin sulfate proteoglycans inside and around the glial scar is known to affect axonal growth and be a major obstacle to autogenous repair. These proteins are thus candidate targets for spinal cord injury therapy. Our previous studies demonstrated that 810 nm photo biomodulation inhibited the formation of chondroitin sulfate proteoglycans after spinal cord injury and greatly improved motor function in model animals. However, the specific mechanism and potential targets involved remain to be clarified. In this study,to investigate the therapeutic effect of photo biomodulation,we established a mouse model of spinal cord injury by T9 clamping and irradiated the injury site at a power density of 50 mW/cm~2 for 50 minutes once a day for 7 consecutive days.We found that photobiomodulation greatly restored motor function in mice and down regulated chondroitin sulfate proteoglycan expression in the injured spinal cord.Bioinformatics analysis revealed that photobiomodulation inhibited the expression of proteoglycan-related genes induced by spinal cord injury, and versican,a type of proteoglycan, was one of the most markedly changed molecules.Immunofluorescence staining showed that after spinal cord injury, versican was present in astrocytes in spinal cord tissue. The expression of versican in primary astrocytes cultured in vitro increased after inflammation induction, whereas photobiomodulation inhibited the expression of ve rsican.Furthermore,we found that the increased levels of p-Smad3,p-P38 and p-Erk in inflammatory astrocytes were reduced after photobiomodulation treatment and after delivery of inhibitors including FR 180204,(E)-SIS3,and SB 202190.This suggests that Sma d 3/Sox9 and MAP K/Sox9 pathways may be involved in the effects of photobiomodulation. In summary, our findings show that photobiomodulation modulates the expression of chondroitin sulfate proteoglycans, and versican is one of the key target molecules of photo biomodulation.MAPK/Sox9 and Smad3/Sox9 pathways may play a role in the effects of photo biomodulation on chondroitin sulfate proteoglycan accumulation after spinal cord injury.
