目的探讨桦木酸(betulinic acid,BA)在缺氧条件下对人前列腺癌骨转移PC⁃3细胞干性、侵袭、迁移、黏附及缺氧诱导因子⁃1α(hypoxia⁃inducible factor⁃1α,HIF⁃1α)表达的影响。方法采用CCK⁃8检测BA对前列腺癌骨转移细胞株PC⁃3细胞活力半... 目的探讨桦木酸(betulinic acid,BA)在缺氧条件下对人前列腺癌骨转移PC⁃3细胞干性、侵袭、迁移、黏附及缺氧诱导因子⁃1α(hypoxia⁃inducible factor⁃1α,HIF⁃1α)表达的影响。方法采用CCK⁃8检测BA对前列腺癌骨转移细胞株PC⁃3细胞活力半数抑制浓度(IC_(50)),再分为IC_(50)药物浓度BA组和对照组,检测BA对PC⁃3细胞增殖的影响;在常氧、缺氧及缺氧+IC_(50)(10μmol/L)BA条件下处理PC⁃3细胞,采用Transwell实验检测BA对细胞侵袭、迁移能力的影响;采用细胞黏附实验检测BA对细胞黏附能力的影响;采用肿瘤成球实验检测BA对细胞自我更新能力影响;采用PCR实验及Western blot实验检测BA对细胞表达HIF⁃1α、CD31、CD44的影响;采用酶联免疫吸附试验(ELISA)检测BA对细胞VEGF、IL⁃8、IL⁃1β、IL⁃6和TNF⁃α因子表达的影响。结果BA呈浓度和时间依赖性抑制PC⁃3细胞增殖,其IC_(50)为10μmol/L。PC⁃3细胞在缺氧环境中的干性、侵袭、迁移、黏附能力增加,而加入BA后PC⁃3细胞的干性、侵袭、迁移、黏附能力下降。Western blot实验结果显示,BA能抑制PC⁃3细胞中HIF⁃1α蛋白表达,同时抑制CD31、CD44蛋白表达(P<0.01);PCR实验结果显示,BA下调HIF⁃1α的mRNA表达,同时也下调CD31、CD44的mRNA表达(P<0.01)。ELISA试验结果显示BA可抑制VEGF、IL⁃8、IL⁃1β、IL⁃6和TNF⁃α因子的表达(P<0.01)。结论BA能抑制缺氧条件下PC⁃3细胞的干性、侵袭、迁移及黏附能力,其机制可能与BA下调HIF⁃1α、CD31、CD44表达,及抑制VEGF、IL⁃8、IL⁃1β、IL⁃6和TNF⁃α等因子表达有关。 展开更多

OBJECTIVE To study the changes in the matrix metalloproteinases-2 and 9 (MMP2, MMP9) induced by ^60Co γ-ray external irradiation of human prostate cancer PC-3 cells. METHODS Human prostate cancer PC-3 cells were irradiated with different doses of ^60Co γ-rays. Cell migration and invasiveness were evaluated and the expression of MMP2, and MMP9 was investigated by RT-PCR, Western blotting and flow cytometry(FCM). RESULTS Irradiation enchances invasive protential at the doses of 1,3 and 5 Gy,whereas it significantly inhibits cell migration. CONCLUSION The different doses of ^60Co γ-ray external irradiation for prostate cancer may have different effects through the changes of MMP2, and MMP9 expression. 展开更多

Objective:To study the mechanisms of pancreatic cancer treatment with Kanglaite combined Gemcitabine by investigating the relationship between the apoptosis and the expression of bcl-2, Bax and VEGF in pancreatic cancer cells.Methods:Nude mouse subcutaneous transplantation tumor model of Human PC-3 pancreatic cancer was established; the expressions of bcl-2, Bax and VEGF of transplantation tumor cell were determined; the earlier apoptosis rate of pancreatic cancer cell and the gross tumor volume were determined. Results:Kanglaite combined Gemcitabine remarkably decreased the protein expression of bcl-2,raised the expression of Bax,increased the apoptosis rate of the pancreatic cancer and contract the gross tumor volume. Kanglaite greatly decreased the protein expression of VEGF of the tumor cell. Conclusion:Therapeutic efficacy of Kanglaite combined Gemcitabine is far better than separate use of the two medicines in the pancreatic cancer transplantation tumor treatment. 展开更多

Effects of astaxanthin on oxidative stress induced by Cu^(2+) in prostate cells

Astaxanthin(AST), a carotenoid molecule extensively found in marine organisms and increasingly used as a dietary supplement, has been reported to have beneficial effects against oxidative stress. In the current paper, the effects of AST on viability of prostate cells were investigated by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide(MTT) assay; cell apoptosis and intracellular reactive oxygen species(ROS) levels were determined by flow cytometry; the mitochondrial membrane potential(MMP) was measured by fluorospectrophotometer; and activities of malondialdehyde(MDA), superoxide dismutase(SOD), catalase(CAT), and glutathione peroxidase(GSH-Px) were evaluated by a detection kit. The results show that copper ion(Cu^2+) induced apoptosis, along with the accumulation of intracellular ROS and MDA, in both prostate cell lines(RWPE-1 and PC-3). AST treatments could decrease the MDA levels, increase MMP, and keep ROS stable in RWPE-1 cell line. An addition of AST decreased the SOD, GSH-Px, and CAT activities in PC-3 cell line treated with Cu^2+, but had a contrary reaction in RWPE-1 cell lines. In conclusion, AST could contribute to protecting RWPE-1 cells against Cu^2+-induced injuries but could cause damage to the antioxidant enzyme system in PC-3 cells. 展开更多
