Antioxidative and Neuroprotective Activities of the Pre-Germinated Brown Rice Extract

We evaluated the effects of pre-germinated brown rice extract (PGBR ex) with enhanced levels of GABA on proliferation and apoptosis of neuronal SK-N-SH cells line. Firstly, we used HPLC methods to study the level of γ-aminobutyric acid (GABA) in all rice extracts. We found that the concentration of GABA in the PGBR ex were 3 and 8 times higher than the GABA concentration in non-germinated brown rice (BR ex) and white rice (WR ex) compared with the stan- dard GABA respectively. Next we study the protective effects of brown rice extract by investigating various methods, we found that the effects of dose-dependent study by treated with PGBR ex, BR ex and WR ex at (0 - 4000 μg/ml). The data from MTT assay showed that the higher concentration of all rice extracts were not induced toxicity to SK-N-SH cells. To test the protective effect by study the viability of SK-N-SH cells. These results showed that PGBR ex and BR ex can protect cells by significantly increase cells survival up to 29.3% ± 0.01% and13.4% ± 0.07 % (p 56.9% ± 0.02 % (p < 0.05), compared with un- treated cells (control). Next study we test the effect of cells apoptotic by ROS assay and DNA fragmentation. The results showed that PGBR ex were definitely decrease the amount of ROS formation and had a little of DNA ladders comparable with condition that induced by 150 ?M H2O2. Our data indicating that PGBR ex with enhanced levels of GABA effectively inhibit SK-N-SH cells proliferation and apoptosis. These present results suggest that intake of PGBR and BR instead of WR is effective to protect cell proliferation and apoptosis which may be useful nutritional to prevent neuronal cells from neurodegenerative disease. 展开更多

Information on seed biology and germination cues of valuable species is lacking. We investigated seed germination of two Acacia species (Acacia tortilis (Forsk.) Hayne and Acacia oerfota (Forssk) schwein/), required for nitrogen fixation and rehabilitation of arid and semi-arid areas.(four pregermination seed treatments were applied in order to find the best treatment in germinating acacia species. The medium was L2 and three replicates were used. Seeds pretreated with sand paper and also with H2SO4 and then H2O2 had the highest germination percentage in both species. The lowest germination percentage resulted from soaking seeds in water for 48 h followed by soaking in H2SO4 for A. oerfota and from soaking in water for 24 h for A. tortilis. Because the use of sand paper is difficult and time consuming, we recommend pretreatment ofA. tortilis and A. oerfota seeds with H2SO4 and H2O2 before planting. Our study results are significant for conservation agencies with an interest in optimizing germination in arid zones for rehabilitation and reforestation. 展开更多

Seed Germination and Effects of Three Watering Regimes on the Growth of <i>Dialium guineense</i>(Wild) Seedlings

The effectiveness of three pre-germination treatments in breaking dormancy of the seeds of Dialum guineense and effects of three watering regimes on the growth performance of the seedlings were investigated. The pre-germination treatments were: soaking in hot water for 10 seconds and cooling down in cold water (T1), soaking in cold water for 24 hours (T2), soaking in running water for 48 hours (T3) and control (T4), while watering regimes include: watering once daily in the morning (W1), watering once every two days in the morning (W2) and watering once every three days in the morning (W3). The results showed that germination occurred first at 5 days after sowing (5 DAS) among the seeds soaked in cold water for 24 hours before sowing (T2), while the untreated seeds (control) took the longest period of 12 days before germination occurred. T1 had the highest germination value of 49.6% while T3 had the least of 31.2%. The effects of watering regimes were found to be significantly different on stem-collar diameter, leaf area and total dry weight (P < 0.05). The least significant difference (LSD) test showed that W2 and W3 supported the best growth performance. It is concluded from the results that T2 should be adopted for breaking the dormancy of Dialium guineense seeds and watering interval of three days would not dispose the seedlings to water-stress. 展开更多
